Proteomic profiling of the monothiol glutaredoxin Grx3 reveals its global role in the regulation of iron dependent processes.

Iron is an essential nutrient required as a cofactor for many biological processes. As a fungal commensal-pathogen of humans, Candida albicans encounters a range of bioavailable iron levels in the human host and maintains homeostasis with a conserved regulatory circuit. How C. albicans senses and responds to iron availability is unknown. In model yeasts, regulation of the iron homeostasis circuit requires monothiol glutaredoxins (Grxs), but their functions beyond the regulatory circuit are unclear. Here, we show Grx3 is required for virulence and growth on low iron for C. albicans. To explore

A patient with allergic bronchopulmonary mycosis caused by Aspergillus fumigatus and Candida albicans.

Allergic Bronchopulmonary Mycosis (ABPM) is an exagregated immunologic response to fungal colonization in the lower airways. It may cause by many kinds of fungal, but Aspergillus fumigatus is the most common cause of ABPM, although other Aspergillus and other fungal organisms, like Candida albicans, have been implicated. Aspergllus fumigatus and Candida albicans may be found as outdoor and indoor fungi, and cause the sensitization, elicitation of the disease pathology, and its clinical manifestations. Several diagnostic procedurs may be impicated to support the diagnosis of ABPM caused by Aspe

Detection of Mucorales antigen in bronchoalveolar lavage samples using a newly developed lateral-flow device.

A murine IgG2b monoclonal antibody, named TG11, binding to an extracellular polysaccharide antigen secreted by all Mucorales fungi has been recently developed and integrated into a lateral-flow device (TG11-LFD). The aim of this study was to establish the clinical performance of TG11-LFD on bronchoalveolar lavage (BAL) fluids for the diagnosis of mucormycosis. Thirteen BAL samples from 13 patients with mucormycosis, all of which tested positive for Mucorales qPCR (Mucor/Rhizopus [n = 5], Lichtheimia [n = 2], Rhizomucor [n = 5], and Cunninghamella [n = 1]), were used to assess the TG11-LFD. Roles of Indole and Its Derivative in Modulating E. coli-Candida albicans Biofilm Formation.

Candida albicans is the causal agent of invasive candidiasis, which might be lethal in immunocompromised patients. Biofilm formation is considered a key virulence factor of C. albicans and is associated with its elevated resistance to antifungals. C. albicans and bacteria like E. coli are frequently found to form mixed biofilms on biotic or abiotic surfaces, rendering them more refractory to existing antifungals. To investigate how E. coli endogenous indole interplaying with exogenous IAA exerts modulatory effects on dual-species biofilm with C. albicans, an E. coli strain deficient in the ind

Candida albicans-stimulated hematopoietic stem and progenitor cells generate trained neutrophils with enhanced mitochondrial ROS production that defend against infection.

Central trained immunity, induced via reprogramming of hematopoietic stem and progenitor cells (HSPCs), mediates sustained heightened responsiveness of mature myeloid cells to secondary challenges. We have previously demonstrated that HSPCs use TLR2 and Dectin-1 to sense Candida albicans to induce the production of trained monocytes/macrophages to fight against secondary infection. Neutrophils play an important role in innate immunity and are critical for clearance of C. albicans. In this work, we used an in vitro model of mouse HSPC differentiation to investigate the functional phenotype of n

Evaluating the Role of Serum Beta-D-glucan Testing in Safely Reducing Antifungal Therapy in Critically Ill Patients: A Retrospective Study.

BACKGROUND AND AIMS: The role of (1→3)-β-D-glucan (BDG) testing in guiding antifungal therapy (AFT) in critically ill patients remains unclear. While BDG has a high negative predictive value (NPV), is it safe to withhold AFT in critically ill BDG-negative patients has not been well studied. PATIENTS AND METHODS: This retrospective cohort study analyzed BDG-negative intensive care unit (ICU) patients (<60 pg/mL) at a tertiary care hospital from March 2024 to January 2025. The ICU survival was compared between those who received AFT and those who did not. Propensity score matching (PSM) adjuste

Neonatal fungemia: a case series highlighting the threat posed by non-albicans Candida (NAC) species and a yeast-like filamentous fungus.

INTRODUCTION: Neonatal fungemia represents a substantial contributor to morbidity and mortality in 25-30% of neonatal intensive care units (NICUs), particularly affecting preterm or low-birth-weight infants. The immune systems of neonates, particularly those who are preterm, are underdeveloped, making them highly susceptible to infections like fungemia. CASES: In the neonatal intensive care unit (NICU), bloodstream infections (BSIs) were identified in 18 out of 72 cases, accounting for 25% of the total. Of these, five cases were attributed to fungal infections, constituting 27.7% of all infec

Intracranial infections of Candida albicans and Mycobacterium intracellulare associated with CARD9 deficiency: case report and review of literature.

BACKGROUND: Invasive candidiasis, most commonly caused by Candida albicans, poses a significant mortality risk and is challenging to treat. Non-tuberculous mycobacterial infections are opportunistic and linked to immune impairment. Caspase recruitment domain-containing protein 9 (CARD9) represents a class of proteins that incorporates the caspase recruitment domain, and its deficiency follows a strict autosomal recessive inheritance pattern, resulting in an impaired immune response. CASE PRESENTATION: A 51-year-old male who was admitted to the hospital 3 years ago because of recurrent fever a

The Combination of 3-Hydrazinoquinoxaline-2-Thiol with Thymoquinone Demonstrates Synergistic Activity Against Different Candida Strains.

INTRODUCTION: Candida is the primary cause of invasive fungal disease, candidiasis, especially in developed nations. The increasing resistance observed in multiple antibiotics, coupled with the prolonged process of creating new antibiotics from the ground up, emphasizes the urgent requirement for innovative methods and new compounds to combat Candida infections. Employing a treatment strategy that combines antibiotics can improve efficacy, broaden the spectrum of targeted fungal, and reduce the chances of resistance emergence. This approach shows potential in tackling the escalating problem of

Impact of Obesity on Echinocandin Effectiveness in Treating Candida Infections: A Retrospective Observational Cohort Study.

INTRODUCTION: Echinocandins are used to treat invasive candidiasis (IC), with FDA-approved doses indicated for both obese and non-obese patients. Pharmacokinetic (PK) studies have identified subtherapeutic exposure in obese patients receiving standard doses (SDs) of echinocandins. However, research on clinical outcome differences of echinocandins' SDs between obese and non-obese patients is lacking. Therefore, this study aimed to evaluate the effectiveness of echinocandins' SDs in obese compared to normal-weight patients with IC. PATIENTS AND METHODS: This retrospective cohort study was condu

Cdc14 phosphatase contributes to cell wall integrity and pathogenesis in Candida albicans.

The Cdc14 phosphatase family is highly conserved in fungi. In Saccharomyces cerevisiae, Cdc14 is essential for down-regulation of cyclin-dependent kinase activity at mitotic exit. However, this essential function is not broadly conserved and requires only a small fraction of normal Cdc14 activity. Here, we identified an invariant motif in the disordered C-terminal tail of fungal Cdc14 enzymes that is required for full enzyme activity. Mutation of this motif reduced Cdc14 catalytic rate and provided a tool for studying the biological significance of high Cdc14 activity. A S. cerevisiae strain e

Encephalartos villosus Lem. Displays a Strong In Vivo and In Vitro Antifungal Potential against Candida glabrata Clinical Isolates.

Recently, Candida glabrata has been recognized as one of the most common fungal species that is highly associated with invasive candidiasis. Its spread could be attributed to its increasing resistance to antifungal drugs. Thus, there is a high need for safer and more efficient therapeutic alternatives such as plant extracts. Here, we investigated the antifungal potential of Encephalartos villosus leaves methanol extract (EVME) against C. glabrata clinical isolates. Tentative phytochemical identification of 51 metabolites was conducted in EVME using LC-MS/MS. EVME demonstrated antifungal activi

Graphene oxide nanosheets increase Candida albicans killing by pro-inflammatory and reparative peritoneal macrophages.

Graphene oxide (GO) is a new nanomaterial with different potential biomedical applications due to its excellent physicochemical properties and ease of surface functionalization. Macrophages play key roles in the control of fungal infections preventing invasive candidiasis by both limiting the growth of the opportunistic fungal pathogen Candida albicans and activating other immune effector cells. In order to know if macrophages maintain their immunocompetence against this microorganism after GO uptake, we have evaluated the interactions at the interface of GO nanosheets, macrophages and Candida
