Due to frequent viral antigenic change, current influenza vaccines need to be re-formulated annually to match the circulating strains for battling seasonal influenza epidemics. These vaccines are also ineffective in preventing occasional outbreaks of new influenza pandemic viruses. All these challenges call for the development of universal influenza vaccines capable of conferring broad cross-protection against multiple subtypes of influenza A viruses. Facilitated by the advancement in modern molecular biology, delicate antigen design becomes one of the most effective factors for fulfilling suc

Naturally acquired human infections with Plasmodium knowlesi are endemic to Southeast Asia. To determine the prevalence of P. knowlesi malaria in malaria-endemic areas of Thailand, we analyzed genetic characteristics of P. knowlesi circulating among naturally infected macaques and humans. This study in 2008-2009 and retrospective analysis of malaria species in human blood samples obtained in 1996 from 1 of these areas showed that P. knowlesi accounted for 0.67% and 0.48% of human malaria cases, respectively, indicating that this simian parasite is not a newly emergent human pathogen in Thailan

Aim of this study was to reconstruct the temporal and spatial phylodynamics of WNV-1a, the genotype to which the majority of European/Mediterranean viral strains belongs, by using sequences retrieved from public databases. WNV-1a isolates segregated into two major clades: the recent West Mediterranean sequences formed a single monophyletic group within clade A. Clade B included sequences from East Mediterranean and America. Phylogeographic analysis suggested that WNV-1a probably originated in sub-Saharan Africa in the early XXth century, and then spread northwards since the late 1970s, via two

OBJECTIVE: Fetal cells cross the feto-maternal barrier and circulate in maternal peripheral blood; thus, this study aimed to show the relationship between clinical evidence in pregnancy and qualitative feto-maternal barrier changes.
METHODS: The expression of fetal hemoglobin gamma chain messenger RNA (HbF-gamma mRNA) was measured by quantitative reverse transcriptase-polymerase chain reaction (RT-PCR) in maternal peripheral blood.
RESULTS: HbF-gamma mRNA was detected in all pregnant women after 5 weeks of gestation. In normal pregnancy, there were two peaks at 10 and 40 weeks, and a signifi

Macroprolactin is a complex of immunoglobulin G and monomeric prolactin with little biological activity in vivo. Macroprolactin cross-reacts in modern commercial prolactin assays, however, leading to pseudohyperprolactinaemia. This report is of three patients with macroprolactinaemia and the untoward consequences if this benign condition is misdiagnosed as genuine hyperprolactinaemia are discussed. One adult and one child without symptoms of hyperprolactinaemia were incidentally found to have elevated serum prolactin levels, one of whom had a pituitary incidentaloma. Repeat prolactin measureme

Doppler sonography is convenient and useful for evaluating intracranial lesions and hemodynamics, especially in the neonate. Recently we used power flow Doppler imaging (PF) to show vessels with a low flow velocity and a small caliber. The pulsed Doppler system (PD) in combination with PF can selectively visualize small vessels and slow blood flow, such as the steady flow conditions of the lenticulostriate artery (LSA). As PF can reveal the vessel crossing at a right angle to the ultrasonic beam, the bilateral middle cerebral arteries can be observed through the anterior fontanel under the sam

Bilateral arterial ducts represent an uncommon form of pulmonary or systemic blood supply associated with complex congenital cardiac disease. We sought to determine the role of high-resolution cross-sectional echocardiography and color flow Doppler ultrasonography in assessing this condition, as well as to describe the management and outcome in a group of patients. A retrospective review was conducted of 11 newborns identified over a 16-year period as having bilateral arterial ducts. Pulmonary atresia associated with non-confluent pulmonary arteries was the dominant lesion, with the heterotaxy

Mouse brain-derived, inactivated Japanese encephalitis (JE) vaccine has been internationally used for many years. It is believed that this vaccine made a great contribution to the reduction of JE patients in several countries. Mouse brain-derived, Beijing-1 and Nakayama JE vaccines induce high levels of neutralizing antibodies. High levels of induced antibodies are maintained at least for 3-4 yr. The induced antibodies are cross-reactive to heterologous strains; however, the neutralizing antibody titers against heterologous strains are usually lower than those against homologous strains. The effect of the hydroxyl radical on polyguanylic acid [poly(G)] was investigated with regard to progressive increase of autoantibodies against it in systemic lupus erythematosus (SLE) and progressive systemic sclerosis (PSS). Rabbits immunized with both native and ROS-poly(G) induced high titre antibodies. Immune IgG exhibited a high degree of specificity towards the immunogen, reiterated visually by a gel retardation assay. The induced antibodies showed a wide range of cross-reactivity with various synthetic polynucleotides exhibiting B-, A-, and allied conformations. The specificity of ind

One hundred seventy-eight children from three villages were studied in a cross-sectional fashion to evaluate the efficacy of conjunctival impression cytology (CIC) to characterize vitamin A status of individual children and populations of children and to examine the relationship of vitamin A status to nutritional status. Although children with abnormal CIC results had lower retinol concentrations than those with normal CIC results (P < 0.02), CIC exhibited poor sensitivity and specificity. Results of a CIC prevalence criterion were concordant with plasma retinol criteria in characterizing the

A simple and quantitative enzyme-linked immunosorbent assay (ELISA) has been developed for the detection of antigens of larval Taenia solium in the cerebrospinal fluid (CSF) of patients with cerebral cysticercosis. Another ELISA was developed for detecting antibodies in CSF against larval antigens. The examination of sixteen patients with clinical diagnosis of cerebral cysticercosis revealed that eleven patients had both circulating larval antigens and anti-larval IgG (but not IgM) antibodies in their cerebrospinal fluids. Of these patients, those with surgically and histologically confirmed i

Onchocerciasis is one of the major filarial diseases affecting humans and a leading cause of blindness. Control of the disease by chemotherapy and by elimination of the vector is not feasible in most areas of endemicity. The host immune response is thought to play a major role in the pathogenesis of complications. However, there is no clear evidence of protective immunity to reinfection in individuals who continue to be exposed to infective larvae. Antigens of Onchocerca volvulus are complex and show extensive cross-reactivity with other filarial parasites of humans and animals. Infection in h

A 3-year-old boy with pancytopenia and a paucity of circulating blast cells was found to have acute megakaryoblastic leukaemia. Histopathologic investigation of the bone marrow biopsy disclosed replacement by megakaryoblasts and mild-to-moderate reticulin fibrosis, the megakaryocytic origin of these cells was confirmed by their staining properties and by cross-reactivity with rabbit anti-rat platelet serum. Treatment with adriamycin and cytosine arabinoside induced a complete remission of this otherwise rapidly fatal disease. Before chemotherapy, the patient's platelets showed decreased aggreg

Cardiac fibrosis is a pathological process that arises as a compensatory response to injury or mechanical stress and is a common feature across diverse cardiovascular diseases. Here, we review recent advances in the translational biology of human cardiac fibrosis, highlighting insights from single-cell studies of human cardiac fibroblasts and genetic analyses of fibrotic phenotypes using cardiac imaging. We discuss the heterogeneity and origins of cardiac fibroblasts, the role of immune-fibroblast cross-talk in fibrogenesis, and the utility of imaging and circulating biomarkers for assessing f

Antigen release is a critical step in initiating antitumor immune responses, yet its regulation during metastasis is not well understood. Here we show that circulating tumor cells undergoing vascular migration produce migrasomes that serve as a metastasis-specific mechanism of antigen release. These migrasomes are enriched in tumor-associated antigens, including cancer-testis and mutated antigens, and are captured efficiently by antigen-presenting cells in secondary lymphoid organs, where they undergo cross-presentation to elicit CD8+ T cell-mediated immune responses that constrain metastatic

Crimean-Congo hemorrhagic fever virus (CCHFV) was first isolated in the Democratic Republic of the Congo (DRC) in 1956. To date, only 3 sporadic human cases have been reported in the DRC, and data on CCHFV infection in livestock, which are key players in transmission, are scant. We conducted a cross-sectional seroepidemiological study on archived human and animal serum samples collected from 25 provinces across the DRC. Samples were tested using an ELISA detecting CCHFV nucleoprotein-specific antibodies. The seroprevalence of CCHFV infection in humans was 4.4% (55/1,239) and in domestic rumina

Dengue fever is a mosquito-borne disease prevalent in tropical and subtropical regions, with its prevalence expanding due to increased global travel. The dengue virus, the causative agent of dengue fever, often co-circulates in the form of four distinct serotypes. Cross-reactive antibodies generated during a primary infection pose a significant risk during secondary infections with different serotypes, and fully protective vaccines and antiviral drugs are yet to be developed. Over the past decade, advances in antibody technology have led to the isolation of numerous monoclonal antibodies again

The extent to which dengue virus has been circulating globally and especially in Africa is largely unknown. Testing available blood samples from previous cross-sectional serological surveys offers a convenient strategy to investigate past dengue infections, as such serosurveys provide the ideal data to reconstruct the age-dependent immunity profile of the population and to estimate the average per-capita annual risk of infection: the force of infection (FOI), which is a fundamental measure of transmission intensity. In this study, we present a novel methodological approach to inform the size a

Differentiation and maturation of megakaryocytes occur in close association with the cellular and extracellular components of the bone marrow. These processes are characterized by the gradual appearance of essential structures in the megakaryocyte cytoplasm such as a polyploid and polylobulated nucleus, an internal membrane network called demarcation membrane system (DMS) and the dense and alpha granules that will be found in circulating platelets. In this article, we describe a standardized protocol for the in situ ultrastructural study of murine megakaryocytes using transmission electron mic

BACKGROUND: The pathophysiology of Ebola virus disease (EVD) is still poorly understood. This study aimed at identifying soluble biomarkers that inform on disease mechanisms.
METHODS: Fifty-four soluble mediators of the immune, coagulation, and endothelial system were measured in baseline and follow-up samples from hospitalized patients with EVD, using Luminex technology. Cross-sectional expression levels and changes over time were correlated with outcome.
RESULTS: Levels of circulating proinflammatory cytokines and chemokines, as well as markers of endothelial dysfunction and coagulopathy,
