Integrative genomics analysis identifies promising SNPs and genes implicated in tuberculosis risk based on multiple omics datasets.

More than 10 GWASs have reported numerous genetic loci associated with tuberculosis (TB). However, the functional effects of genetic variants on TB remains largely unknown. In the present study, by combining a reported GWAS summary dataset (N = 452,264) with 3 independent eQTL datasets (N = 2,242) and other omics datasets downloaded from public databases, we conducted an integrative genomics analysis to highlight SNPs and genes implicated in TB risk. Based on independent biological and technical validations, we prioritized 26 candidate genes with eSNPs significantly associated with gene expres

Mendelian randomization analysis of celiac GWAS reveals a blood expression signature with diagnostic potential in absence of gluten consumption.

Celiac disease (CeD) is an immune-mediated enteropathy with a strong genetic component where the main environmental trigger is dietary gluten, and currently a correct diagnosis of the disease is impossible if gluten-free diet (GFD) has already been started. We hypothesized that merging different levels of genomic information through Mendelian randomization (MR) could help discover genetic biomarkers useful for CeD diagnosis. MR was performed using public databases of expression quantitative trait loci (QTL) and methylation QTL as exposures and the largest CeD genome-wide association study cond

Application of genomic bioinformatic tools in clinical psychiatry and counseling.

Genomic medicine has introduced transformative tools for psychiatric practice, facilitating the diagnosis of complex cases through the identification of genetic variants. The gene encodes a key component of the histone methyltransferase complex. Mutations in this gene have been linked to neurodevelopmental disorders and schizophrenia, particularly in cases with early onset and cognitive impairment. The aim of this article is to describe the application of genomic tools and international criteria for interpreting genetic pathogenicity in psychiatric clinical practice, using as a model. Based on

Advancing precision hepatology for rare paediatric liver diseases.

UNLABELLED: Rare genetic liver disorders pose a significant burden due to their chronic nature, early morbidity, and frequent need for liver transplantation. Despite advances in diagnostic tools, these conditions are often misdiagnosed or diagnosed late due to non-specific symptoms, which overlap with more common aetiologies. The advent of next-generation sequencing has revolutionised the diagnosis of these disorders, enabling precise genetic identification, reducing diagnostic delays, enabling application of targeted therapies, and informing prognosis and family counselling. This opinion arti

[Current state of research on the role of gene polymorphisms in the development of urolithiasis in different popu- lations].

Urolithiasis is a common recurrent urological disease that continues to show increasing global incidence. Its pathogenesis is multifactorial, and the evident role of genetic factors highlights the importance of identifying gene polymorphisms associated with susceptibility to stone formation. We performed an analysis of published data evaluating the contribution of individual gene polymorphisms to urolithiasis development in populations across different countries. This review included publications from Russian and international databases (2013-2024). Both review articles and original research s

A polygenic risk score model for psoriasis based on the protein interactions of psoriasis susceptibility loci.

INTRODUCTION: Polygenic Risk Scores (PRS) are an emerging tool for predicting an individual's genetic risk to a complex trait. Several methods have been proposed to construct and calculate these scores. Here, we develop a biologically driven PRS using the UK BioBank cohort through validated protein interactions (PPI) and network construction for psoriasis, incorporating variants mapped to the interacting genes of 14 psoriasis susceptibility (PSORS) loci, as identified from previous genetic linkage studies. METHODS: We constructed the PPI network via the implementation of two major meta-databa

Diagnosis and Risk Prediction of Dilated Cardiomyopathy in the Era of Big Data and Genomics.

Dilated cardiomyopathy (DCM) is a leading cause of heart failure and life-threatening ventricular arrhythmias (LTVA). Work-up and risk stratification of DCM is clinically challenging, as there is great heterogeneity in phenotype and genotype. Throughout the last decade, improved genetic testing of patients has identified genotype-phenotype associations and enhanced evaluation of at-risk relatives leading to better patient prognosis. The field is now ripe to explore opportunities to improve personalised risk assessments. Multivariable risk models presented as "risk calculators" can incorporate
