Heart Failure Update: Diagnosis and Classification.

Heart failure (HF) is a complex clinical condition. The newer guidelines have phased out the designations of systolic and diastolic HF and replaced them with the more physiologically applicable classifications of HF with reduced or preserved ejection fraction (EF). Although the numbers of new patients within these two groups are similar, patient characteristics and risk factors often differ. There also are differences in the incidence of HF among racial and ethnic groups, with blacks having the highest incidence. Although survival has improved over time among patients with reduced EF, no signi

Emerging new biomarkers of preeclampsia.

Preeclampsia continues to plague some of the most vulnerable women and fetuses. It is surprisingly prevalent in developing and developed nations. According to the World Health Organization, hypertension during pregnancy is a leading cause of maternal mortality in industrialized countries at 16% and up to 25% in developing countries. As the pathogenesis of this disease is being unraveled, we are afforded new opportunities to develop novel biomarkers for early identification and prevention of disease. The angiogenic markers including soluble fms-like tyrosine kinase 1, placental growth factor, a

Proteomic profiling of regionalized proteins in rat epididymis indicates consistency between specialized distribution and protein functions.

The epididymis is a key structure of the male reproductive system; its function is to mature, transport, and store sperm. Most of the research examining the epididymis to date has been limited to the study of the secreted proteins involved in the maturation of spermatozoa. However, it is also very important to understand the protein components, regulation and function of the tissue itself since these are the basis for all of its physiological processes. We investigated the differential expression of proteins among the caput, corpus, and cauda regions of rat epididymis and considered the possib

The role of biomarkers across the care continuum in acute heart failure.

Acute heart failure (AHF) remains a global health challenge, characterized by high morbidity, mortality, and frequent rehospitalizations. The heterogeneity of clinical presentations in AHF and its complex underlying pathophysiology complicates diagnosis, risk stratification, and therapeutic decision-making. Biomarkers have reshaped the clinical landscape in AHF by providing objective, biologically-grounded insights that complement traditional clinical assessment. Natriuretic peptides are established as indispensable for diagnosis and prognosis, while troponins, soluble ST2, galectin-3, and eve

A2AR as a key target for immune microenvironment remodeling in prostate cancer.

This study elucidates the critical role of adenosine A2A receptor (A2AR) signaling in prostate cancer progression through comprehensive molecular characterization and clinical validation, demonstrating that A2AR overexpression in prostate cancer cells drives profound immunosuppression via coordinated upregulation of CD73-mediated adenosine production, subsequent activation of immunosuppressive pathways including ARG1, TGF-β, and IL-10 secretion, and induction of immune checkpoint molecules PD-L1 and Galectin-9, which collectively promote myeloid-derived suppressor cell expansion and CD8+ T cel

Molecular crosstalk between MASLD and IVDD revealed through integrated biomarker discovery analysis.

BACKGROUND: Non-alcoholic fatty liver disease (NAFLD), recently retermed as metabolic dysfunction-associated steatotic liver disease (MASLD), and intervertebral disc degeneration (IVDD) are major health burdens with rising prevalence. Despite affecting different organ systems, emerging evidence suggests potential molecular crosstalk between these conditions. However, the underlying mechanisms remain poorly understood. METHODS: We employed comprehensive bioinformatics analysis to investigate shared pathogenic mechanisms between MASLD and IVDD through integrated bulk and single-cell RNA sequenc

LGALS1-CD276 Paracrine axis between tumor and endothelial cells promotes tumor angiogenesis and progression in bladder cancer.

Bladder cancer, particularly muscle-invasive disease, has a high metastatic potential and limited treatment options, highlighting the need for new therapeutic targets. CD276 (B7-H3), a type I transmembrane protein of the B7 family, is traditionally considered an immunomodulatory ligand with an unidentified receptor. Here, we investigated the role of CD276 in bladder cancer and sought to identify its binding partner. Using a high-throughput human proteome microarray, we identified the secreted lectin galectin-1 (LGALS1) as a high-affinity binding partner of CD276, suggesting that CD276 may func

Extracellular vesicles as prognostic biomarkers: results of a neoadjuvant chemoimmunotherapy clinical trial in stage IIIA (N2) non-small-cell lung cancer (SAKK 16/14).

The introduction of immune checkpoint inhibitors has transformed cancer therapy. In non-small-cell lung cancer (NSCLC), immune-checkpoint blockade targeting programmed cell death 1 (PD-1) and its ligand PD-L1 has demonstrated substantial therapeutic efficacy and survival benefit. Recent evidence indicates that small extracellular vesicles (EVs), including exosomes, play a crucial role in modulating the tumor microenvironment and immune responses, potentially influencing immunotherapy efficacy. This study investigates longitudinal EV dynamics in patients with resectable stage IIIA (N2) NSCLC un

Vitamin D in the prevention of left ventricular remodelling after an acute myocardial infarction: a randomized clinical trial.

No clinical trial has yet assessed the effect of vitamin D on patients experiencing ST-segment elevation myocardial infarction (STEMI). In this multicentre, randomised (2:1), double-blind, placebo-controlled trial, 93 patients with anterior STEMI were randomised to calcifediol or placebo, for 12 months. The primary endpoint was left ventricular remodelling, defined as a 15% increase in left ventricular end-diastolic volume (EDV) on magnetic resonance imaging. Sixty-five patients were allocated to calcifediol and 28 to placebo. Calcidiol levels increased in the intervention group (20.0[8.4, 36.

Intratumoral 198Au Nanobrachytherapy Reduces Tumor Growth and Metastasis in a Murine Model of Triple-Negative Breast Cancer.

Nanobrachytherapy (NB) uses radioactive nanoparticles (NPs) to deliver localized ionizing radiation within tumors, whereas photobiomodulation therapy (PBM) employs low-intensity light to modulate cellular responses. Here, we evaluated NB alone and combined with PBM using 198AuNPs and red laser irradiation in a murine triple-negative breast cancer (TNBC) model. Female mice were inoculated with 4T1 cells in the lower left mammary fat pad and, once tumors reached ∼0.1 cm3, were assigned to three groups: NB (intratumoral injection of ∼200 μCi 198AuNPs), PBM+NB (single red laser session before NB),

Single-cell transcriptomics reveals phenotypic and functional alterations of erythroid progenitor cells in tuberculosis patients.

Anemia is a common complication of tuberculosis (TB), closely associated with impaired host immune function and poor prognosis; however, its impact on the dynamic alterations of erythroid progenitor cells (EPCs) remains largely unexplored. In this study, we integrated and analyzed single-cell transcriptomic data from peripheral blood mononuclear cells (PBMC) of healthy controls, latent tuberculosis (LTB), TB, and disseminated tuberculosis (DTB) cohorts, as well as 18F-FDG-labeled TB lung tissues and adjacent uninvolved regions. At single-cell resolution, we identified ten EPCs clusters within

Integrating bulk and single-cell RNA sequencing reveals intratumor heterogeneity phenotypes and immune infiltration in pancreatic cancer.

BACKGROUND: Although the significance of intratumoral heterogeneity (ITH) has been widely acknowledged in various cancers, its role in pancreatic cancer (PC) remains underexplored and warrants further investigation. METHODS: Pancreatic cancer transcriptomic data were acquired from the TCGA and GEO databases. The DEPTH2 algorithm, in combination with differential expression analysis, was used to identify genes associated with intratumoral heterogeneity (ITH). We applied univariate Cox regression analysis and multiple machine learning techniques to establish a reliable prognostic model. Patient

The effect of sodium-glucose cotransporter-2 inhibitors on cardiovascular biomarkers and left ventricular function in patients with type 2 diabetes and concomitant cardiovascular disease.

INTRODUCTION: Sodium-glucose cotransporter 2 inhibitors (SGLT2i) have demonstrated cardiovascular benefits in patients with type 2 diabetes mellitus (T2DM). The aim of this study was to assess the effects of SGLT2i therapy on heart failure (HF)-related biomarkers - galectin‑3 (GAL‑3) and N-terminal pro-B-type natriuretic peptide (NT‑proBNP) - as well as on echocardiographic parameters of cardiac function in patients with T2DM and established cardiovascular disease (CVD). MATERIAL AND METHODS: This prospective study included 61 patients with T2DM, all naïve to SGLT2i and with documented CVD. P

Dissecting the genetic association between abnormal sperm parameters and depression: a transcriptome-wide analysis of 157 participants.

PURPOSE: Depression often occurs in the males with semen abnormalities. Evidence suggested a genetic correlation between depression and the pathogenesis of abnormal sperm parameters, whereas the mechanisms remained unclear. METHODS: Genomic datasets of major depressive disorder (MDD) and abnormal sperm parameters were obtained from the Gene Expression Omnibus database. After screening the datasets, differentially expressed genes (DEGs) were identified. GO and pathway enrichment analyses, a protein-protein interaction network, and receiver operator characteristic curve analysis were conducted.

Targeting VSMCs to suppress macrophage-like phenotype switching that restrains atherogenesis by immunosuppressive oligodeoxynucleotide (A151) functionalized selenium nanoparticles.

Macrophage-like phenotype switching of vascular smooth muscle cells (VSMCs) is a crucial mechanism driving atherogenesis. Inhibition of a phenotype switch to macrophage-like cells is a promising strategy to prevent atherosclerosis (AS), and targeted nanotherapeutics represent one approach for implementing this strategy. To this end, we designed immunosuppressive oligodeoxynucleotide A151 functionalized selenium nanoparticles with a spearhead LacNAc (LN-A151-SeNPs) that target macrophage-like VSMCs. Nano characterization showed that the uniformity and stability of nanoparticles were optimized b

A structurally defined galactoglucan from Arisaema erubescens with a multi-target tumor-associated protein binding domain.

As targeted therapy assumes an increasingly pivotal role in comprehensive cancer treatment, there is a growing imperative to develop natural drugs that exhibit low toxicity and minimal side effects. Polysaccharides derived from the traditional Chinese herbal medicine Arisaema erubescens have been reported to show antitumor potential, however the key structural features and specific molecular targets responsible for their pharmacological effects are still vague. To address this question, a homogeneous polysaccharide TNX05 (Mw ≈ 9 kDa) was obtained and characterized from Arisaema erubescens. Str
