Underpinning regions associated with nitrogen use efficiency in Brassica juncea L. Czern: using associative transcriptomics approach.

BACKGROUND: Breeding crops for enhanced understanding of apparent nitrogen use efficiency (NUE) requires understanding of phenotypic variations and underlying genetics. Improving NUE in Indian mustard is considered important to reduce nitrogen (N) fertilizer use without sacrificing crop yield. In the present study, 260 genotypes of Brassica juncea were field evaluated for seed yield and NUE-related traits at three N levels: N0 (no added nitrogen), N50 (50 kg/ha nitrogen), and N100 (100 kg/ha nitrogen) for two years. RESULTS: N response in plants varied depending on the dose of nitrogen applie

Public health implications of multidrugresistant and methicillinresistant Staphylococcus aureus in retail oysters.

Methicillin-resistant Staphylococcus aureus (MRSA) is a major challenge for food safety and public health. This study aimed to investigate the potential role of oysters sold in Egypt as a source for Staphylococcus aureus (S. aureus), MRSA, and multidrug-resistant methicillin-resistant Staphylococcus aureus (MDR-MRSA). It also examined the occurrence of tsst-1 virulence gene and assessed the associated zoonotic risks. Thirty-three pooled fresh oyster samples were acquired from different retail fish markets in Egypt. S. aureus was identified by conventional culture-based and molecular methods. A

Association analysis of mitochondrial DNA heteroplasmic variants: Methods and application.

We rigorously assessed a comprehensive association testing framework for heteroplasmy, employing both simulated and real-world data. This framework employed a variant allele fraction (VAF) threshold and harnessed multiple gene-based tests for robust identification and association testing of heteroplasmy. Our simulation studies demonstrated that gene-based tests maintained an appropriate type I error rate at α = 0.001. Notably, when 5 % or more heteroplasmic variants within a target region were linked to an outcome, burden-extension tests (including the adaptive burden test, variable threshold

High carriage of plasmid-mediated quinolone resistance (PMQR) genes by cefotaxime-resistant Escherichia coli recovered from surface-leaking sanitary sewers.

There is a rapid rise in the incidence of quinolone resistant bacteria in Nigeria. Most studies in Nigeria have focused on isolates from the clinical settings, with few focusing on isolates of environmental origin. This study aimed to investigate the antibiogram and carriage of plasmid-mediated quinolone resistance (PMQR) genes by quinolone-resistant isolates obtained from a pool of cefotaxime-resistant Escherichia coli (E. coli) recovered from sewage leaking out of some surface-leaking sanitary sewers in a University community in Nigeria. Isolation of E. coli from the sewage samples was done

Loss of a Candidate Biliary Atresia Susceptibility Gene, add3a, Causes Biliary Developmental Defects in Zebrafish.

OBJECTIVES: Biliary atresia (BA) is a progressive fibroinflammatory cholangiopathy affecting the bile ducts of neonates. Although BA is the leading indication for pediatric liver transplantation, the etiology remains elusive. Adducin 3 (ADD3) and X-prolyl aminopeptidase 1 (XPNPEP1) are 2 genes previously identified in genome-wide association studies as potential BA susceptibility genes. Using zebrafish, we investigated the importance of ADD3 and XPNPEP1 in functional studies. METHODS: To determine whether loss of either gene leads to biliary defects, we performed morpholino antisense oligonuc

Polymorphisms of excision repair gene XPD Lys751Gln and hOGG1 Ser326Cys might not be associated with hepatocellular carcinoma risk: a meta-analysis.

Источник пометил публикацию как отозванную

The xeroderma pigmentosum group D (XPD) and human 8-oxoguanine glycosylase 1 (hOGG1) genes have been suggested to play an important role in the pathogenesis of hepatocellular carcinoma (HCC). However, the results have been inconsistent. In this study, we performed a meta-analysis to clarify the associations of polymorphisms of XPD and hOGG1 genes with HCC risk. Published literature from PubMed, EMBASE, and Chinese National Knowledge Infrastructure were retrieved. Pooled odds ratio (OR) with 95 % confidence interval (CI) was calculated using a fixed- or random-effects model. Seven studies (1,95

Generalist genes analysis of DNA markers associated with mathematical ability and disability reveals shared influence across ages and abilities.

BACKGROUND: The Generalist Genes Hypothesis is based upon quantitative genetic findings which indicate that many of the same genes influence diverse cognitive abilities and disabilities across age. In a recent genome-wide association study of mathematical ability in 10-year-old children, 43 SNP associations were nominated from scans of pooled DNA, 10 of which were validated in an individually genotyped sample. The 4927 children in this genotyped sample have also been studied at 7, 9 and 12 years of age on measures of mathematical ability, as well as on other cognitive and learning abilities.

Comparative gene expression analysis of ovarian carcinoma and normal ovarian epithelium by serial analysis of gene expression.

Despite the poor prognosis of ovarian cancer and the importance of early diagnosis, there are no reliable noninvasive biomarkers for detection in the early stages of disease. Therefore, to identify novel ovarian cancer markers with potential utility in early-stage screening protocols, we have undertaken an unbiased and comprehensive analysis of gene expression in primary ovarian tumors and normal human ovarian surface epithelium (HOSE) using Serial Analysis of Gene Expression (SAGE). Specifically, we have generated SAGE libraries from three serous adenocarcinomas of the ovary and, using novel

Large-scale identification of genes required for full virulence of Staphylococcus aureus.

Gene products required for in vivo growth and survival of microbial pathogens comprise a unique functional class and may represent new targets for antimicrobial chemotherapy, vaccine construction, or diagnostics. Although some factors governing Staphylococcus aureus pathogenicity have been identified and studied, a comprehensive genomic analysis of virulence functions will be a prerequisite for developing a global understanding of interactions between this pathogen and its human host. In this study, we describe a genetic screening strategy and demonstrate its use in screening a collection of 6

Two Drosophila beta tubulin isoforms are not functionally equivalent.

We have tested the functional capacity of different beta tubulin isoforms in vivo by expressing beta 3-tubulin either in place of or in addition to beta 2-tubulin in the male germ line of Drosophila melanogaster. The testes-specific isoform, beta 2, is conserved relative to major metazoan beta tubulins, while the developmentally regulated isoform, beta 3, is considerably divergent in sequence. beta 3-tubulin is normally expressed in discrete subsets of cells at specific times during development, but is not expressed in the male germ line. beta 2-Tubulin is normally expressed only in the postmi

Macrophages generated in 3D mechanical microenvironment contribute to recruiting Peritonitis-associated neutrophils in vivo.

BACKGROUD: The influence of scaffolds with bone marrow (BM) niche-like mechanical properties on the stemness and lineage differentiation of hematopoietic stem cells (HSCs) in vitro has been studied. Previous research has demonstrated that 3D collagen hydrogels can regulate the differentiation trajectory of late myeloid progenitors, leading to the specialization of '3D-macrophages' that express various chemokine genes, including Cxcl2 and Cd14. Comprehensive transcriptional profiles in single-cell level have characterized the interaction between 3D-macrophages and neutrophil clusters through th

Presence of a cryptic Onchocerca species in black flies of northern California, USA.

BACKGROUND: Black flies (Diptera: Simuliidae) serve as arthropod vectors for various species of Onchocerca (Nematoda: Onchocercidae) that may be associated with disease in humans, domestic animals, and wildlife. The emergence of zoonotic Onchocerca lupi in North America and reports of cervid-associated zoonotic onchocerciasis by Onchocerca jakutensis highlight the need for increased entomological surveillance. In addition, there is mounting evidence that Onchocerca diversity in North America is far greater than previously thought, currently regarded as Onchocerca cervipedis species complex. Th

Efficacy and safety of vosoritide in children with achondroplasia: a systematic review and meta-analysis.

UNLABELLED: Achondroplasia is caused by a gain-of-function mutation in the FGFR3 gene. Vosoritide activates the NPR-B receptor to inhibit the overactive FGFR3 signaling pathway. We aim to pool the efficacy and safety outcomes of vosoritide in children with genetically confirmed achondroplasia who are receiving the approved dose of 15 μg/kg/day. We searched five databases up to February 10, 2026. A systematic review and single-arm meta-analysis were conducted in accordance with the PRISMA guidelines. The primary outcomes were annualized growth velocity (AGV), height gain, and change in height Z

The CYP4G15-SORD-like-associated pathway promotes ovarian ZIKV infection and egg-associated viral RNA detection in Aedes albopictus.

Zika virus (ZIKV) can persist in mosquito populations through both horizontal and vertical transmission, and successful ovarian infection is a prerequisite for transgenerational spread. However, the mosquito host factors that regulate ovarian permissiveness and vertical transmission remain poorly characterized. Here, we investigated the role of cytochrome P450 4G15 (CYP4G15) in ZIKV infection and vertical transmission in Aedes albopictus and identified an associated downstream pathway involving SORD-like, a predicted sorbitol dehydrogenase-like gene. Expression profiling revealed that CYP4G15
