Rodents as vehicle for delivery of transgenic bacteria to make paratransgenic sand fly vectors of cutaneous leishmaniasis in field condition.

Vector-borne diseases, among them leishmaniasis, cause more than 700,000 deaths annually. The lack of an effective vaccination and the increasing resistance of sand flies to insecticides require the urgent development of innovative approaches to contain the disease. The use of engineered bacteria that express anti-parasite molecules (paratransgenesis) shows much promise. However, a challenge for implementation of this strategy is to devise means to introduce modified bacteria into sand flies in the field. In this study, we use rodent food bait as a delivery strategy to introduce two mCherry-fl

Promising antimalarial hits from phenotypic screens: a review of recently-described multi-stage actives and their modes of action.

Over the last two decades, global malaria cases caused by Plasmodium falciparum have declined due to the implementation of effective treatments and the use of insecticides. However, the COVID-19 pandemic caused major disruption in the timely delivery of medical goods and diverted public health resources, impairing malaria control. The emergence of resistance to all existing frontline antimalarials underpins an urgent need for new antimalarials with novel mechanisms of action. Furthermore, the need to reduce malaria transmission and/or prevent malaria infection has shifted the focus of antimala

A distinct sodium channel voltage-sensor locus determines insect selectivity of the spider toxin Dc1a.

β-Diguetoxin-Dc1a (Dc1a) is a toxin from the desert bush spider Diguetia canities that incapacitates insects at concentrations that are non-toxic to mammals. Dc1a promotes opening of German cockroach voltage-gated sodium (Nav) channels (BgNav1), whereas human Nav channels are insensitive. Here, by transplanting commonly targeted S3b-S4 paddle motifs within BgNav1 voltage sensors into Kv2.1, we find that Dc1a interacts with the domain II voltage sensor. In contrast, Dc1a has little effect on sodium currents mediated by PaNav1 channels from the American cockroach even though their domain II padd

Genetic structure of Aedes aegypti populations determined using pairwise comparisons.

The biological characteristics of Aedes aegypti (Diptera, Culicidae), which is a vector of dengue and yellow fever, make this organism a good model for studying population structure and the events that may influence it under the effect of human activity. We assessed the genetic variability of five A. aegypti populations using RAPD-PCR technique and six primers. Four populations were from Brazil and one was from the USA. A total of 165 polymorphic DNA loci were generated. Considering the six primers and the five populations, the mean value of inter-population genetic diversity (Gst) was 0.277,

Magnetic nanoparticles are highly toxic to chloroquine-resistant Plasmodium falciparum, dengue virus (DEN-2), and their mosquito vectors.

A main challenge in parasitology is the development of reliable tools to prevent or treat mosquito-borne diseases. We investigated the toxicity of magnetic nanoparticles (MNP) produced by Magnetospirillum gryphiswaldense (strain MSR-1) on chloroquine-resistant (CQ-r) and sensitive (CQ-s) Plasmodium falciparum, dengue virus (DEN-2), and two of their main vectors, Anopheles stephensi and Aedes aegypti, respectively. MNP were studied by Fourier-transform infrared spectroscopy and transmission electron microscopy. They were toxic to larvae and pupae of An. stephensi, LC50 ranged from 2.563 ppm (1s

Bedbugs: a primer for the health-system pharmacist.

PURPOSE: The history and life cycle of bedbugs, the identification and supportive treatment of bedbug bites, and the eradication and prevention of bedbug infestations are reviewed. SUMMARY: Cimex lectularius, known commonly as the bedbug, is a growing problem in the United States. While the bedbug population declined between the 1950s and the late 20th century, recent increases in international travel and immigration, the banning of dichlorodiphenyltrichloroethane, and insecticide resistance have caused a resurgence of bedbug infestations. Infestations can be identified based on clinical pres…

Advances in the diagnosis of human lymphatic filariases: a review.

The nematode parasites Wuchereria bancrofti, Brugia malayi and B. timori are the causative agents of human lymphatic filariasis. Of the estimated 90 million infections world-wide, W. bancrofti is responsible for over 80 million cases and is the only known aetiologic agent in the African Region. Numbers of infected persons are on the increase world-wide due to rural-urban migrations which result in mushrooming of shanty towns often encouraging formation of favourable mosquito breeding-sites. Development of insecticide resistance by the vector mosquitoes; the toxicity and high cost of available …

Genome-wide identification, phylogenetic analysis, and expression profiles of ATP-binding cassette transporter genes in the oriental fruit fly, Bactrocera dorsalis (Hendel) (Diptera: Tephritidae).

The ATP-binding cassette (ABC) is the largest transporter gene family and the genes play key roles in xenobiotic resistance, metabolism, and development of all phyla. However, the specific functions of ABC gene families in insects is unclear. We report a genome-wide identification, phylogenetic, and transcriptional analysis of the ABC genes in the oriental fruit fly, Bactrocera dorsalis (Hendel). We identified a total of 47 ABC genes (BdABCs) from the transcriptomic and genomic databases of B. dorsalis and classified these genes into eight subfamilies (A-H), including 7 ABCAs, 7 ABCBs, 9 ABCCs

Evaluation of the effectiveness of Aquatain, Bacillus thuringiensis var. israelensis, and Temephos on Anopheles arabiensis and Anopheles stephensi larvae in the laboratory and field settings.

BACKGROUND: The main tools to control malaria vectors in sub-Saharan Africa are long-lasting insecticidal nets and indoor residual spraying. However, their sustainability is threatened by the emergence of insecticide resistances, behavioral avoidance and presence of outdoor biting mosquito populations. Thus, complementary interventions such as larval source management, which includes larviciding, are required to achieve better results in malaria vector control. This study aimed to evaluate the effectiveness of three larvicides (Aquatain AMF®, Temephos and Bacillus thuringiensis var. israelensi…

The impact of Bacillus thuringiensis var. israelensis (Vectobac® WDG) larvicide sprayed with drones on the bio-control of malaria vectors in rice fields of sub-urban Kigali, Rwanda.

BACKGROUND: The core vector control tools used to reduce malaria prevalence are currently long-lasting insecticidal nets (LLINs), and indoor residual spraying (IRS). These interventions are hindered by insecticide resistance and behavioural adaptation by malaria vectors. Thus, for effective interruption of malaria transmission, there is a need to develop novel vector control interventions and technologies to address the above challenges. Larviciding using drones was experimented as an innovative tool that could complement existing indoor interventions to control malaria. METHODS: A non-random…

Mosquito larvicidal potential of Solanum xanthocarpum leaf extract derived silver nanoparticles and its bio-toxicity on non-target aquatic organism.

BACKGROUND & OBJECTIVES: Mosquitoes are insects of public health importance that act as a vector to transmit various vector-borne diseases in humans including dengue, malaria, filariasis and yellow fever. The continually employed synthetic insecticides have developed resistance in mosquitoes. Nano-based botanical insecticides can be considered as the best alternative due to several advantages like being simple, non-pathogenic, biodegradable and safe to the environment. The present work reported the maximum larvicidal potential of green synthesized silver nanoparticles (AgNPs) derived from the

Larvicidal activity of Ricinus communis extract against mosquitoes.

BACKGROUND & OBJECTIVES: Vector control strategies play significant role in reducing the transmission of malaria, dengue and other vector-borne diseases. The control of vector population using synthetic insecticides has resulted in development of insecticide resistance and negative effects on humans and environment. The present investigation evaluated the larvicidal potential of methanol, dichloromethane and hexane extracts of leaves and seeds of Ricinus communis (castor) plant against the early IV instar larvae of the dengue vector, Aedes aegypti, and malaria vector, Anopheles culicifacies. …

Structural and Biological Evidence for Acetylcholinesterase-Targeted Insecticidal Activity in Musca domestica: A 4.5  Å Distance Correlation From Novel Benzenesulfonamide Derivatives.

House fly (Musca domestica) control faces mounting resistance to conventional insecticides, driving interest in novel chemistries targeting acetylcholinesterase (AChE). We examined eight benzenesulfonamide derivatives originally developed as carbonic anhydrase inhibitors to explore their potential as AChE-targeting insecticides and to identify structural determinants of biological activity. Using a high-quality homology model of M. domestica AChE, we performed blind docking, refined docking to the peripheral anionic site, 100 ns molecular dynamics simulations, and molecular mechanics-poisson-b

Knockdown of Clavesin family genes NlClvs1l, NlClvs1t, and NlClvs2l in Nilaparvata lugens reveals their potential as novel targets for pest control strategies.

The brown planthopper (BPH) Nilaparvata lugens is one of the most destructive pests of rice, and its management has primarily relied on chemical insecticides. Currently, the chemical management of BPH is facing challenges due to the development of pesticide resistance. RNA interference (RNAi) provides attractive alternative to chemical insecticides, provided that suitable target genes are identified. Clavesin (clathrin vesicle-associated Sec14 protein) proteins are involved in multiple biological processes, including endocytosis, oxidative stress response, and the regulation of lysosome morpho
