Aging-driven metabolic abnormalities remodel intercellular communication through the gut-liver-heart axis and may promote coronary artery disease: the key role of bile acid metabolism.

Coronary artery disease (CAD) remains the leading cause of cardiovascular mortality worldwide and shows a strong age-dependence that classical risk-factor models do not fully explain. A growing body of work indicates that aging is closely associated with CAD and, in preclinical models, can promote it through immunometabolic remodeling of the gut-liver-heart axis, in which bile acid metabolism is proposed to act as a central molecular link. Here we integrate cellular, molecular, and clinical evidence to outline how aging perturbs this axis and sustains chronic vascular inflammation. At the cell

The pathologic findings of skin, lymph node, liver, and bone marrow in patients with adult-onset still disease: a comprehensive analysis of 40 cases.

Adult-onset Still disease (AOSD) is characterized by fever, skin rash, and lymphadenopathy with leukocytosis and anemia as common laboratory findings. We investigated the characteristic pathologic findings of skin, lymph node, liver, and bone marrow to assist in proper diagnosis of AOSD.Forty AOSD patients were included in the study. The skin (26 patients), lymph node (8 patients), liver (8 patients), or bone marrow biopsies (22 patients) between 1998 and 2013 were retrospectively analyzed. AOSD patients were diagnosed according to the Yamaguchi criteria after excluding common infections, hema

Advanced amphotericin-B gel formulation: An efficient approach to combat cutaneous leishmaniasis.

Several drug release studies are continuously carried out to minimize pharmacological limitations, such as insolubility in water and gastrointestinal irritability. This study focused on the development of gels for the administration of amphotericin-B (AmB) for the treatment of cutaneous leishmaniasis. Two gels were prepared from copolymers (polyglycerol and ε-caprolactone) incorporated with AmB. The gels were characterized by FTIR, NMR, TG, and DSC techniques. The incorporation of AmB into the copolymers showed that the medicine remained in the monomeric form, which is the least toxic. The AmB

Dietary advanced glycation end-products aggravate non-alcoholic fatty liver disease.

AIM: To determine if manipulation of dietary advanced glycation end product (AGE), intake affects non-alcoholic fatty liver disease (NAFLD) … protected from the additional harmful effects of the high AGE containing diet. Studies in isolated Kupffer cells showed that AGEs increase cell proliferation and oxidative stress, providing a likely mechanism through which these compounds contribute to liver injury. CONCLUSION: In the HFHC model of NAFLD, manipulation of dietary AGEs modulates liver injury, i …

Hypothermic oxygenated perfusion (HOPE) protects from biliary injury in a rodent model of DCD liver transplantation.

BACKGROUND & AIMS: The use of livers from donors after cardiac arrest (DCD) is increasing in many countries to overcome organ shortage. Due to additional warm ischemia before preservation, those grafts are at higher risk of failure and bile duct injury. Several competing rescue strategies by machine perfusion techniques have been developed with, however, unclear effects on biliary injury. We analyze the impact of an end-ischemic Hypothermic Oxygenated PErfusion (HOPE) approach applied only through the portal vein for 1h before graft implantation. METHODS: Rat livers were subjected to 30-min i

Genomic, cDNA and embryonic expression analysis of zebrafish IRF6, the gene mutated in the human oral clefting disorders Van der Woude and popliteal pterygium syndromes.

Van der Woude syndrome (VWS) and popliteal pterygium syndrome (PPS) are autosomal dominant clefting disorders recently discovered to be caused by mutations in the IRF6 (Interferon Regulatory Factor 6) gene. The IRF gene family consists of nine members encoding transcription factors that share a highly conserved helix-turn-helix DNA-binding domain and a less conserved protein-binding domain. Most IRFs regulate the expression of interferon-alpha and -beta after viral infection, but the function of IRF6 remains unknown. We have isolated a full-length zebrafish irf6 cDNA, which encodes a 492 amino

Increased susceptibility to liver injury after hemorrhagic shock in rats chronically fed ethanol: role of nuclear factor-kappa B, interleukin-6, and granulocyte colony-stimulating factor.

Chronic ethanol use preceding severe trauma and hemorrhagic shock (HS) is associated with an increased incidence of multiorgan failure (MOF) and death; however, the molecular basis for this increased susceptibility is unknown. We previously demonstrated that production of interleukin-6 (IL-6) and granulocyte colony-stimulating factor (G-CSF), mediated by nuclear factor-kappa B (NF-kappa B), each make essential contributions to organ injury and inflammation in a rodent model of controlled HS, and we proposed in this study to examine the hypothesis that the increased susceptibility to MOF after

Lanthanide "blockade" of antigen-presenting cells suppresses lymphocyte proliferation by inducing nitric oxide synthesis.

Introduction of antigen into the portal venous circulation can induce a state of antigen-specific tolerance. Reversal of this phenomenon by the administration of gadolinium chloride (GD) before portal venous inoculation with antigen has been thought to result from the inhibition of macrophage phagocytosis by GD, thereby permitting passage of antigen through the hepatic sinusoids into the systemic circulation where sensitization can occur. We recently demonstrated that i.v. GD also greatly enhances the ability of the hepatic macrophage, or Kupffer cell (KC), to suppress lymphocyte proliferation

Fucoidan attenuates alcohol-induced liver injury via PON1/HDL/NF-κB axis.

BACKGROUND: Paraoxonase 1 (PON1) exerts a protective effect against alcoholic liver disease (ALD) by regulating oxidative stress and inflammation. A fucoidan derived from Hizikia fusiformis, JHCF4, is associated with increased nuclear sterol regulatory element-binding protein 2 (SREBP 2) levels and higher PON1/high-density lipoprotein (HDL) expression. METHOD: PON1-silenced Kupffer cells were established using small-interfering RNA (siRNA), and protein blot analysis, TUNEL, JC-1, Hoechst 33342, and transmission electron microscopy (TEM) were used to determine the mechanism by which JHCF4 prot…

Simultaneous Occurrence of Wilson's Disease, Autoimmune Hepatitis, and Hereditary Hemochromatosis: A Diagnostic Challenge.

This is not surprising to detect iron overload in chronic liver diseases and end-stage liver diseases since Kupffer cells scavenge necrotic hepatocytes during the course of liver damage, leading to an increased serum iron level and transferrin saturation compatible with iron overload even in the absence of a genetic mutation suggestive of hereditary hemochromatosis. Therewith, a relative association has been found between some sorts of chronic liver diseases like non-alcoholic steatohepatitis and hepatitis C with human homeostatic iron regulator protein (HFE: High Fe2+) gene mutations. Schistosoma mansoni Infection-Induced Transcriptional Changes in Hepatic Macrophage Metabolism Correlate With an Athero-Protective Phenotype.

Hepatic macrophages play an essential role in the granulomatous response to infection with the parasitic helminth Schistosoma mansoni, but the transcriptional changes that underlie this effect are poorly understood. To explore this, we sorted the two previously recognized hepatic macrophage populations (perivascular and Kupffer cells) from naïve and S. mansoni-infected male mice and performed microarray analysis as part of the Immunological Genome Project. The two hepatic macrophage populations exhibited remarkably different genomic profiles. However, this diversity was substantially reduced f…
