The shufflon of Salmonella enterica serovar Typhi regulates type IVB pilus-mediated bacterial self-association.

Previously, it was shown that type IVB pili encoded by the Salmonella enterica serovar Typhi pil operon are used to facilitate bacterial entry into human intestinal epithelial cells in vitro and that such entry is inhibited by purified prepilin (pre-PilS) protein (X.-L. Zhang, I. S. M. Tsui, C. M. C. Yip, A. W. Y. Fung, D. K.-H. Wong, X. Dai, Y. Yang, J. Hackett, and C. Morris, Infect. Immun. 68:3067-3073, 2000). The pil operon concludes with a simple shufflon, and a recombinase gene product (Rci) inverts DNA in the C-terminal region of the pilV gene to allow synthesis of two distinct PilV pro

Conventional alpha beta T cells are sufficient for innate and adaptive immunity against enteric Listeria monocytogenes.

We have begun to dissect the cellular requirements for generation of immunity against enteric infection by Listeria monocytogenes using a novel T(-) B(-) NK(-) mouse strain (mice double deficient for the common cytokine receptor gamma-chain (gamma(c)) and the recombinase-activating gene-2 (RAG2/gamma(c) mice). Initial experiments showed that C57BL/6 mice and alymphoid RAG2/gamma(c) mice had similar kinetics of bacterial accumulation in the spleen, liver, and brain early after intragastric L. monocytogenes infection (up to day 3), calling into question the physiologic role of gut-associated lym

RAD51 in Breast Cancer: From Vulnerability to Resistance.

Homologous recombination deficiency (HRD) has transformed the therapeutic landscape of breast cancer through the clinical success of poly(ADP-ribose) polymerase (PARP) inhibitors and platinum-based chemotherapy. Central to this vulnerability is radiation sensitivity 51 (RAD51), the recombinase that executes homologous DNA repair and stabilizes stalled replication forks. In breast cancer susceptibility type 1 (BRCA1)- and breast cancer susceptibility type 2 (BRCA2)-mutant tumors, impaired RAD51 loading produces profound sensitivity to DNA-damaging agents. However, accumulating evidence indicate

Epicardial Contributions to Fibro-Inflammatory Signaling in a Pkp2-Deficient Arrhythmogenic Cardiomyopathy Model.

BACKGROUND: Arrhythmogenic cardiomyopathy (ACM) is an inherited disease that is characterized by lethal ventricular arrhythmias stemming from myocyte dysfunction. ACM is associated with considerable subepicardial fibrosis and inflammation with right ventricle predominance. Most cases of gene-positive ACM are caused by a desmosome protein mutation, with plakophilin-2 (Pkp2) mutations being the most common. We hypothesized that Pkp2-deficiency in epicardium-derived cells (EPDCs) contributes to fibro-inflammatory signaling and ACM pathogenesis. METHODS: We developed transgenic mice that lack Pkp

Challenges in Developing a Microglial NF-Kappa B Conditional Mouse Model for Glioblastoma Studies.

Tumor microenvironments (TME) play a crucial role in the formation, progression, and drug resistance of many cancers, including glioblastoma (GBM). Microglia are the resident innate immune cells of the central nervous system and are associated with brain cancers like GBM. However, there are limited studies about the specific effects that microglia have on GBM tumors. One possible way to reduce GBM tumor progression is by targeting key signaling pathways in microglia, such as the nuclear factor-kappa B (NF-κB) signaling pathway, which has been implicated in both microglia/macrophage phenotype p

Efficient Construction of Recombinant CHO Cells for Stable Expression of Therapeutic Proteins Based on New Hotspot 'LOC'.

Efficient generation of stably express therapeutic recombinant proteins in Chinese hamster ovary (CHO) cell lines remains as a major challenge in biologics development. Among available strategies, CRISPR/Cas9-mediated homology-directed repair (HDR) has emerged as a highly efficient method for inserting genes of interest (GOIs) into predefined genomic 'hotspot.' In this study, we report such a 'hotspot' (LOC site) which has discovered previously by our research team, which is a non-coding RNA region within the CHO-K1 genome at position 1689 of NW_003626341.1, located in the LOC103162981 gene. Trafficking Glycogen Nanoparticles through Lymph Node Tissues for the Delivery of Small and Large Bioactive Molecules.

Targeted delivery of therapeutics to lymph nodes (LNs) via minimally invasive subcutaneous injection offers promise to treat B and T cell malignancies, latent HIV-1 reservoirs, and cancer metastasis. However, achieving therapeutic drug levels in subcutaneous tissues and LNs is challenging because of the poor retention and accumulation of soluble drugs. Engineered nanoparticles (NPs) provide a platform for prolonging drug stability and retention in the subcutaneous space and LNs, acting as reservoirs for the sustained release of drugs through the lymphatic system. Yet, their clinical translatio

Characterization of the recombination activity in the pituitary and ovary of Prop1-iCre knockin mice.

The pituitary gland plays a central role in the regulation of development, metabolism, stress, reproduction, and homeostasis in the mammalians. All the endocrine cells secreting distinct pituitary hormones are derived from Prop1-expressing progenitors. Prop1 promoter-driven Cre transgenic mice are useful in the field of pituitary biology based on the Cre/LoxP system; however, their ectopic activity complexes data interpretation. Here, we demonstrate the robust and specific recombination activity of a Prop1-iCre knockin line in pituitary endocrine cells as well as in ovarian granulosa cells. Pr

Factors associated with and kinetics of anti-IFN-α autoantibodies in RAG1/2 deficiency.

BACKGROUND: Autoantibodies against IFN-α (anti-IFN-α) have been reported in recombinase activating gene (RAG) deficiency, attributed to impaired central and peripheral T-cell/B-cell tolerance. However, the clinical features, especially viral infections, associated with these autoantibodies at baseline, their kinetics over time, and their response to hematopoietic cell transplantation are not well defined. OBJECTIVE: We described the clinical and immunologic findings linked to anti-IFN-α IgG in RAG deficiency and tracked its kinetics longitudinally, including in those who underwent hematopoiet

Paratuberculosis in small ruminants in the Sudan: prevalence and risk factors.

BACKGROUND: Paratuberculosis (PTB), caused by Mycobacterium avium subsp. paratuberculosis (MAP), is a contagious and chronic enteric disease of ruminants and many non-ruminants leading to emaciation and death of the animal. PTB is poorly investigated in sheep and goats in Sudan, where these animals contribute significantly to food security and poverty alleviation as sources of income. They also play an important role in the national economy through animal exports. The objectives of this study were to investigate the prevalence of PTB and associated risk factors in small ruminants. Blood and fa

Detection of P. malariae using a new rapid isothermal amplification lateral flow assay.

BACKGROUND: While Plasmodium falciparum and Plasmodium vivax cause the majority of malaria cases and deaths, infection by Plasmodium malariae and other Plasmodium species also causes morbidity and mortality. Current understanding of these infections is limited in part by existing point-of-care diagnostics that fail to differentiate them and have poor sensitivity for low-density infections. Accurate diagnosis currently requires molecular assays performed in well-resourced laboratories. This report describes the development of a P. malariae diagnostic assay that uses rapid, isothermal recombinas

Phenotypic quantification of Nphs1-deficient mice.

BACKGROUND: Steroid-resistant nephrotic syndrome (SRNS) is the second most frequent cause of chronic kidney disease in children and young adults. The most severe form of steroid-resistant nephrotic syndrome is congenital nephrotic syndrome Finnish type (CNSF), caused by biallelic loss-of-function variants in NPHS1, encoding nephrin. Since each of the 68 monogenic causes of steroid-resistant nephrotic syndrome represents a rare cause of the disease, tailoring therapeutic interventions to multiple molecular targets remains challenging, suggesting gene replacement therapy (GRT) as a viable altern

Cre-loxP System-Based Mouse Model for Investigating Graves' Disease and Associated Orbitopathy.

Background: Graves' disease (GD), one of the most common forms of autoimmune thyroid disorders, is characterized by hyperthyroidism caused by antibodies (Abs) against the extracellular A-subunit of the thyrotropin receptor (TSHR). Various approaches have been used to create mouse models of GD, including transfected fibroblasts and immunization with plasmids or adenoviruses expressing human TSHR A-subunit (hTSHR A-subunit). These models, however, require repeated immunization and produce inconsistent results. In this study, we established a novel Cre-loxP system-based mouse model that is able t

A toolkit for assembly of targeting clones for C. elegans transgenesis.

Transgenic worms are a key resource for C. elegans researchers dissecting molecular pathways using this simple metazoan model system. Transgenes provide an avenue to visualize developmental events, cellular processes as well as real-time signal events in live animals using genetically encoded sensors. Generation of these tools has become increasingly efficient with the advent of numerous integration methods including transposon, CRISPR and recombinase-mediated integration. A growing limitation in transgene production is the assembly of the targeting constructs used to direct insertion of seque

Siponimod exerts neuroprotective effects on the retina and higher visual pathway through neuronal S1PR1 in experimental glaucoma.

Sphingosine-1-phosphate receptor (S1PR) signaling regulates diverse pathophysiological processes in the central nervous system. The role of S1PR signaling in neurodegenerative conditions is still largely unidentified. Siponimod is a specific modulator of S1P1 and S1P5 receptors, an immunosuppressant drug for managing secondary progressive multiple sclerosis. We investigated its neuroprotective properties in vivo on the retina and the brain in an optic nerve injury model induced by a chronic increase in intraocular pressure or acute N-methyl-D-aspartate excitotoxicity.
