Meta-Analysis of Face-Validity Indicators in Automatically Detected Dog Sleep Spindles.

Visual inspection as the golden standard for sleep spindle detection has received mixed support and may not be equally viable across species and experimental conditions. Here we propose a novel intermediate strategy for approximating the quality of a detector and subject our own algorithm for spindle detection in dogs to the procedure. Automatic detections across the full range of our data (excluding only data sets with high subject overlap) were analyzed for their compliance with three face validity criteria, originally discovered by expert scorers in human EEG. These criteria are the shortne

The effects of plasma protein levels on the risk of endometriosis: A Mendelian randomization study.

Endometriosis is a prevalent gynecologic disorder that significantly impacts women's health. However, its underlying pathogenesis remains unknown. This study aimed to ascertain causal associations between plasma protein levels and endometriosis using a Mendelian randomization (MR) design. Plasma protein genome-wide association studies (GWAS) data originating from the UK Biobank Pharma Proteomics Project (UKB-PPP) were utilized as exposure variables. Endometriosis GWAS data from the FinnGen study and UKB study served as outcome variables at the discovery and replication stages, respectively. Su

Minicircle DNA Vaccines: Overcoming Delivery and Expression Barriers in Next-Generation Immunization.

DNA vaccines have emerged as a promising immunization platform, offering key advantages over conventional vaccine approaches, including superior stability, a favorable safety profile, rapid and flexible antigen design, and scalable manufacturing. However, their clinical efficacy has remained limited, primarily due to inefficient cellular uptake, poor endosomal escape, and degradation of the plasmid DNA within host cells. Recent advances have highlighted minicircle DNA (mcDNA) as a next-generation alternative to conventional plasmid vectors. mcDNA constructs are compact, backbone-free episomal

Short-Term Feeding on Ecologically Distinct Dietary Plants Is Associated with Gut-Sample Bacterial and Archaeal Profiles in Adult Anoplophora glabripennis.

Ecological control studies of the Asian longhorned beetle (Anoplophora glabripennis) distinguish susceptible, resistant, and dead-end trap trees as functionally different plant categories. We used shotgun metagenomics to examine bacterial and archaeal profiles detected in adult gut samples after 72 h exposure to three dietary plants or prolonged water-only starvation. The study included 24 metagenomes, with three biological replicates per DietGroup × SexGroup combination. No time-zero gut samples were available, so the observed patterns remain superimposed on the beetles' field history. The re

Sliding-before-breaking governs deformation in chitinous extracellular matrices reinforced by strong, fatigue-resistant chitin.

Many fibrous biological materials combine high strength with the ability to undergo permanent shape change, a balance that remains difficult to replicate synthetically. In twisted, plywood-like architectures, the origin of irreversible deformation remains unresolved: does strain arise from covalent bond scission or from coordinated interfacial processes across hierarchical scales? Here, we use the chitin-based protective tubes of vestimentiferan deep-sea tubeworms (Siboglinidae: Vestimentifera) as a model system to address this question. By integrating multiscale simulations with experimental

Temporal Declines in Accuracy of the Donor Risk Index for Liver Transplantation.

INTRODUCTION: The Liver Donor Risk Index (LDRI) has long served as the principal tool for quantifying donor risk in liver transplantation, integrating donor and graft characteristics to estimate graft failure. However, shifts in donor demographics and the widespread adoption of machine perfusion technologies may have reduced its relevance to contemporary practice. We therefore evaluated the contemporary predictive performance of the LDRI for 90-d graft survival. METHODS: Using the UNOS registry, we examined 49,220 liver transplant recipients across three time periods, replicating the methodol

Somatic Mosaicism of the Brain: The Functional Implications of Post-Zygotic Mutations to Neural Circuits.

The human brain is a genomic mosaic shaped by post-zygotic mutations that accumulate from the earliest embryonic divisions through aging. This review synthesizes current evidence on the origins, detection, functional consequences and disease relevance of neural somatic mosaicism. We find that somatic variants-including single-nucleotide variants, copy-number variations, mobile element insertions, structural rearrangements and aneuploidies-arise through distinct molecular mechanisms such as replication errors, oxidative stress, defective DNA repair and transposable element activity, each leavin

Immature squamous metaplastic epithelium of Weber's glandular duct: a significant origin of HPV-independent oropharyngeal squamous cell carcinoma in the base of the tongue.

PURPOSE: This study investigated the anatomical and pathological mechanisms of immature squamous metaplastic epithelium in Weber's glandular ducts at the tongue base in human papillomavirus (HPV)-independent oropharyngeal squamous cell carcinoma (OPSCC). METHODS: An analysis of 80 tongue base carcinoma cases clarified the anatomical connection to Weber's glands. Histological and immunohistochemical studies of these glandular ducts identified areas susceptible to cancer. A mouse model using 4-nitroquinoline-N-oxide was employed to simulate carcinogenic development. RESULTS: 42.5% of cases wer

Multi-target tracking for star sensor based on CenterTrack deep learning model.

Detecting and tracking non-cooperative targets in sequential star sensor images poses several challenges in rapid expanding space involving mega constellations, commercial space activities: poor real-time performance in target acquisition, inadequate generalization to different target speeds, and dependence on the star sensor's attitude priors. To address these challenges, an improved multi-target tracking deep learning model based on the CenterTrack model is proposed in this paper. A sophisticated training and testing dataset that closely replicates real on-orbit star sensor images is constru

Targeting the roots of myeloid malignancies with T cell receptors.

Myeloid malignancies are clonal diseases of haematopoietic stem cell or haematopoietic progenitor cell origin, for which allogeneic haematopoietic stem cell transplantation remains the only curative treatment for most patients. However, the severe side effects and high relapse rates underscore the need for novel therapies. The success of adoptive transfer of chimeric antigen receptor (CAR) T cells targeting B cell-specific cell surface molecules in B cell cancers has not been replicated in myeloid malignancies. T cells engineered to express cancer-directed T cell receptors (TCRs) could provide

Organoid development and applications in gynecological cancers: the new stage of tumor treatment.

Gynecologic cancers (GCs), including cervical cancer (CC), ovarian cancer (OC), endometrial cancer (EC), as well as vulvar and vaginal cancers, represent major health threats to women, with increasing incidence rates observed globally. Conventional treatments, such as surgery, radiation therapy, and chemotherapy, are often hindered by challenges such as drug resistance and recurrence, contributing to high mortality rates. Organoid technology has emerged as a transformative tool in cancer research, offering in vitro models that closely replicate the tumor cell architecture and heterogeneity of

Anhedonia as a Transdiagnostic Symptom Across Psychological Disorders: A Network Approach.

BACKGROUND: Anhedonia is apparent in different mental disorders and is suggested to be related to dysfunctions in the reward system and/or affect regulation. It may hence be a common underlying feature associated with symptom severity of mental disorders. METHODS: We constructed a cross-sectional graphical Least Absolute Shrinkage and Selection Operator (LASSO) network and a relative importance network to estimate the relationships between anhedonia severity and the severity of symptom clusters of major depressive disorder (MDD), anxiety sensitivity (AS), attention-deficit hyperactivity disor

VEGF in Tears as a Biomarker for Exudative Age-Related Macular Degeneration: Molecular Dynamics in a Mouse Model and Human Samples.

Vascular endothelial growth factor (VEGF) is a key mediator of exudative age-related macular degeneration (eAMD), yet non-invasive biomarkers for disease monitoring remain limited. This study evaluates VEGF levels in human tear fluid as a potential biomarker for eAMD and investigates the molecular dynamics of VEGF in a laser-induced choroidal neovascularization (lCNV) mouse model. Tear VEGF levels were quantified using proximity qPCR immunoassays in eAMD patients (n = 29) and healthy controls (n = 21) and correlated with optical coherence tomography (OCT) findings. Molecular analyses, includin

Premature ovarian insufficiency: addressing the most controversial issues.

It was a true honor to be invited as President to deliver the Pieter Van Keep oration at the 2024 International Menopause Society (IMS) Congress in Melbourne, Australia. There was never any doubt that the topic of my lecture would be premature ovarian insufficiency (POI). POI is a condition which has been neglected for far too long given its greater global prevalence than originally estimated and its implications to the sufferers, both in terms of quality of life and long-term well-being. The hope is that we are now entering a new era of awareness given the recent update of the European Societ

Modeling environmental inhalant exposure in rheumatoid arthritis.

The mucosal origins hypothesis posits that environmental inhalant exposures, including cigarette smoke (CS) and crystalline silica (c-silica), trigger immune responses in the lung mucosa, an extra-articular site, which precede initiating events of rheumatoid arthritis (RA) pathogenesis in distant joints. Epidemiological data strongly associates these exposures with RA risk, especially in genetically susceptible individuals carrying HLA-DRB1 alleles, and with the production of autoantibodies such as anti-citrullinated peptide antibodies (ACPA) and rheumatoid factor (RF). However, establishing c

Creating a Genetic Toolbox for the Carbon-Fixing, Nitrogen-Fixing and Dehalogenating Bacterium Xanthobacter autotrophicus.

Xanthobacter autotrophicus is a metabolically flexible microorganism with two key features: (1) The organism has adapted to grow on a wide variety of carbon sources including CO2, methanol, formate, propylene, haloalkanes and haloacids; and (2) X. autotrophicus was the first chemoautotroph identified that could also simultaneously fix N2, meaning the organism can utilize CO2, N2, and H2 for growth. This metabolic flexibility has enabled use of X. autotrophicus for gas fixation, the creation of fertilizers and foods from gases, and the dehalogenation of environmental contaminants. Despite the w

Triplex H-DNA structure: the long and winding road from the discovery to its role in human disease.

H-DNA is an intramolecular DNA triplex formed by homopurine/homopyrimidine mirror repeats. Since its discovery, the field has advanced from characterizing the structure in vitro to discovering its existence and role in vivo. H-DNA interacts with cellular machinery in unique ways, stalling DNA and RNA polymerases and causing genome instability. The foundational S1 nuclease and chemical probing technologies originally used to show H-DNA formation have been updated and combined with genome-wide sequencing methods for large-scale mapping of secondary structures. There is evidence for triplex H-DNA
