Absence of fenfluramine-induced anorexia and reduced c-Fos induction in the hypothalamus and central amygdaloid complex of serotonin 1B receptor knock-out mice.

Fenfluramine, a serotonin releaser and uptake inhibitor, has been widely prescribed as an appetite suppressant. Despite its popular clinical use, however, the precise neural pathways and specific 5-HT receptors that account for its anorectic effect have yet to be elucidated. To test the hypothesis that stimulation of 5-HT1B receptors is required for the anorectic effect of fenfluramine, we assessed food intake in wild-type and 5-HT1B knock-out mice. Next, to determine possible brain structures and pathways that may contribute to the 5-HT1B-mediated effects of fenfluramine, we studied by immuno

Long-term consequences of subtle stimuli during the first twenty-four hours of seizure-induced brain injury.

Chronically epileptic (induced by a single systemic injection of lithium and pilocarpine about 30 days before the experiment began) male rats were trained within a radial maze while they were administered either GABA-pentin (Neurontin), or prednisolone or given no treatment. There was no significant improvement in learning or memory between the groups. Numbers of trials per day were positively correlated with the time required to display the overt stereotyped forelimb clonus after the single pilocarpine injection. The numbers of correct trials completed during the first few days of acquisition

GABA antagonist and benzodiazepine partial inverse agonist reduce motivated responding for ethanol.

Brain gamma-aminobutyric acid (GABA) systems have long been associated with the behavioral actions of ethanol. This study investigated the effects of GABAergic agents on ethanol reinforcement. Rats were trained to orally self-administer ethanol in a 30-min, free-choice operant task. Responses at one of two levers produced contingent access to ethanol (10% w/v) or water. Pretreatment with RO 15-4513, a benzodiazepine inverse agonist (0.375 to 3.0 mg/kg ip), selectively reduced responses for ethanol, and a higher dose of RO 15-4513 (6.0 mg/kg) reduced both ethanol and water responses. Self-admin

Self-induced vomiting and laxative and diuretic use among teenagers. Precursors of the binge-purge syndrome?

Bulimia (binge-purge syndrome) is a recently described but apparently common eating disorder. Purging behaviors associated with bulimia can cause serious medical complications. Prevalence data on purging behaviors are lacking for younger adolescents. A survey was conducted with 1,728 tenth-grade students to assess their attitudes about eating, dieting, weight control, and frequency of purging. Height, weight, and skin-fold thicknesses were also measured. Thirteen percent reported purging behavior. Female purgers outnumbered male purgers 2 to 1. Male purgers were significantly heavier than male

Influence of eating behaviors on short-term weight loss by orlistat and anorectic agent.

Little data exists concerning whether eating behaviors determine the response to orlistat treatment, especially with added anorectic agents. This study was a sub-investigation of a 12-week randomized controlled trial for the additive effect of orlistat on sibutramine treatment. The analysis presented here was restricted to 98 women who had fulfilled the protocol. The Dutch eating behavior questionnaire and three-factor eating questionnaire were used to assess eating behaviors. Scores of emotional eating, external eating, disinhibition and hunger are significantly interrelated. Using multiple l

[Food-procuring stereotype movements is accompanied by changes of c-Fos gene expression in the amygdala and modulation of heart rate in rats].

The distribution of Fos-immunoreactive (Fos-ir) and NADPH Diaphorase reactive (NADPH-dr-) neurons in the different subnuclei of amygdala and insular cortex (on the level -2,12 to -3,14 mm from bregma), and the associated changes of heart rate (HR) in intact, food-deprivated and executed food-procuring movements of rats were studied. In comparison with other groups of animals, the mean number of the Fos-ir neurons in the central nucleus of amygdala (Ce) and the insular cortex (GI/DI) at all studied levels was significantly greater in the executed food-procuring movements in rats. The main focus

Behavioral satiety sequence in a genetic mouse model of obesity: effects of ghrelin receptor ligands.

Behavioral satiety sequence (BSS) is a useful paradigm to assess the effects of orexigenic and anorexigenic profiles of novel pharmacological and genetic manipulations in rodents. To date, no studies have described the satiety profile of leptin-deficient ob/ob mice, an important animal model of obesity in this task. Furthermore, no studies have described changes in the BSS after treatment with ghrelin receptor ligands, which have become an attractive therapeutic target in obesity drug discovery efforts. BSS testing was carried out in ob/ob mice and their lean controls. After baseline analysis,

Effect of neuropeptide B on feeding behavior is influenced by endogenous corticotropin-releasing factor activities.

Decrease in food intake is commonly seen in the mammalian response to stress. Corticotropin-releasing factor (CRF) is a factor that modulates feeding behavior under these stressful conditions. We previously reported that neuropeptide B (NPB) exerts strong synergistic anorectic effects in mice when co-administered with CRF. Tissue distributions of neuropeptide B/W receptor-1 (NPBWR1) and NPB suggest that this neuropeptide system also plays a role in regulating emotion and stress responses. In this study, we examined whether the effects of NPB are altered by endogenous CRFergic tone. During the

A comparison of the effects of psychotomimetics and anxiolytics on punished and unpunished responding maintained by fixed interval schedules of food reinforcement in the rat.

Characterization of anxiolytic drugs often employs conflict paradigms in which the drug effects on punished and unpunished responding can be compared. In this study, a fixed interval schedule generating a range of baseline response rates allowed comparison of the effects of anxiolytic drugs with those of psychotomimetic drugs on equivalent and differing rates of punished and unpunished responding. The first response made by the rat after a 40-s fixed interval elapsed resulted in food pellet delivery. In punished intervals, signalled by the illumination of stimulus lamps above each lever, a 0.6

Use of amphetamine-like appetite suppressants: a cross-sectional survey in Southern Brazil.

In 1994 a cross-sectional survey was undertaken in Pelotas, Southern Brazil, to assess the prevalence of amphetamine-like appetite suppressant use. 1,277 adults were interviewed, and the prevalence of anorectic drug use was 1.3% (95% CI = 0.7-1.9): 15 women and one man, mainly from higher socioeconomic groups. Most of the users (81%) had a medical prescription. Forty-one different drugs had been prescribed. Mean length of use was 8.7 months. These findings are discussed in terms of the overreliance on anorectics as aids to dieting, the dangers of polypharmacy, and the risks of long-term use.

Electrical activity of the reticular formation during aversive and appetitive conditioning in rats.

Evoked potentials (EP) to a conditioned stimulus (CS) were recorded from several parts of the reticular formation (RF), the midbrain central gray (CG), the occipital cortex (OC), the basolateral amygdala and the ventrobasal thalamus during appetitive and aversive classical conditionings in rats. In addition, single unit activity to rewarding CS (+) and punishing CS (-) in a single discriminative classical conditioning was recorded from RF, OC, CG and the tectum. In the EP study, peak-to-peak (p-p) amplitudes, especially of the pontine RF, increased in the initial phase of pre-conditioning, in

Maternal capital predicts investment in infant growth and development through lactation.

INTRODUCTION: Maternal capital (MC) is a broad term from evolutionary biology, referring to any aspects of maternal phenotype that represent resources available for investment in offspring. We investigated MC in breastfeeding mothers of late preterm and early term infants, examining its relationship with infant and breastfeeding outcomes. We also determined whether MC modified the effect of the relaxation intervention on these outcomes. METHODS: The data was collected as part of a randomized controlled trial investigating breastfeeding relaxation in 72 mothers of late preterm and early term i

Hypothalamic-pituitary-adrenal axis, neuroendocrine factors and stress.

The stress system coordinates the adaptive responses of the organism to stressors of any kind.(1). The main components of the stress system are the corticotropin-releasing hormone (CRH) and locus ceruleus-norepinephrine (LC/NE)-autonomic systems and their peripheral effectors, the pituitary-adrenal axis, and the limbs of the autonomic system. Activation of the stress system leads to behavioral and peripheral changes that improve the ability of the organism to adjust homeostasis and increase its chances for survival. The CRH and LC/NE systems stimulate arousal and attention, as well as the meso
