"Shaker" calf syndrome: a newly recognized inherited neurodegenerative disorder of horned Hereford calves.

The clinical and pathological features of a newly recognized, inherited neurodegenerative disorder in horned Hereford calves are described. The disorder is expressed in newborns by tremulous shaking of the head, body and tail, difficulty in rising, a wobbly spastic gait, and aphonia. Transient improvement is followed by deterioration and progressive spastic paraplegia. Generalized tremors can be induced easily by a variety of stimuli, and spinal reflexes may be exaggerated or depressed. The major pathological finding is an excessive accumulation of neurofilaments within neurons of the central,

In vitro evidence that covalent crosslinking of neurofilaments occurs in gamma-diketone neuropathy.

We have postulated that the toxic neuropathies associated with neurofilament-filled axonal swellings have a common pathogenesis, the covalent crosslinking of neurofilaments during anterograde transport. The newly described gamma-diketone, 3,4-dimethyl-2,5-hexanedione (DMHD), is a more potent analogue of the toxic metabolite of n-hexane, 2,5-hexanedione. The axonal swellings observed in DMHD toxicity are in the proximal axon, as seen in intoxication with beta, beta'-iminodipropionitrile, rather than in the distal axon, where neurofilamentous swellings are observed in n-hexane, carbon disulfide,

Characterization of antineurofilament autoantibodies in Creutzfeldt-Jakob disease.

The antineurofilament antibodies found in the serum of a chimpanzee with experimental Creutzfeldt-Jakob disease reacted specifically with the 200,000-dalton polypeptide of the purified neurofilament triplet. They also reacted strongly with thoroughly characterized neurofilament swellings of proximal axons of spinal cord motoneurons from beta, beta' iminodipropionitrile (IDPN)-intoxicated rats.

[Cholesteatoma, oto- and tympanosclerosis: their general mechanism of tissue destruction (author's transl)].

The connective tissue contains extracellular enzymes for the turnover of the ground substance. If this enzyme system gets out of control there is development of a complex metabolic disturbance of all tissue components. The lysosomal dysfunction is the basis of the general mechanism of tissue destruction in cholesteatoma, oto- and tympanosclerosis. This process seems most enhanced in cholesteatoma. Emphasis is placed on the role of extracellular lysosomes (= matrix vesicles).

Ultrastructural aspects of mucinous (colloid) breast carcinoma.

Seven cases of mucinous (colloid) breast carcinoma were studied by electron microscopy. In addition to the predictably abundant mucosubstance production, the following observations merit elaboration: 1) absence of myoepithelial differentiation and basal lamina deposition, 2) notably developed cytoplasmic filamentous systems and relatively scarce lysosomes, 3) frequent and apparently well-developed intercellular junctions, and 4) a distinct paucity of vessels in the tumors' stroma. The rather favorable clinical prognosis of mucinous breast carcinoma despite the absence of myoepithelial differen

[Ultrastructural investigation on muscles of patients with talipes equinovarus (author's transl)].

Authors in course of soft tissue release of children with talipes equinovarus between the age of 9 months and 4 years performed a muscle biopsy in 23 cases from the so called club foot muscles and from the peronei. The results of electronmicroscopic investigation are described. The muscle is not as a hole affected, but the pathologic parts are scattered among the intact ones. Fatty degeneration was noted after long termed redression in plaster, together with the fat connective tissue also appeared. The most severe alterations were observed on the contractile elements, consistent seemingly with

Adhesion gateway: decoding mechanobiological cues for cytoskeletal reconfiguration.

Cellular adhesion is a central determinant of functional three-dimensional tissue structures. The challenges lie in restoring stable cell-to-cell and cell-to-matrix adhesion from dissociated cells without compromising cell integrity and impairing the ability of cells to form functional 3D structures. Recent breakthroughs have elucidated diverse, force-sensitive mechanisms for inducing cellular adhesion, providing a foundation for advanced therapeutic and engineering applications. This review provides a comprehensive analysis of recent work on individual mechanosensing and mechanotransduction p

A 2D fluid-based model for individual cell crawling.

This paper presents the analysis and numerical simulations of a two-dimensional model of single-cell crawling migration, focusing on intracellular dynamics. The fluid-based model relies on the interaction between the actin cytoskeleton and a generic molecular regulator of its activity. The model is formulated as a Poisson problem with Darcy's law, coupled with a nonlocal advection-diffusion equation for the molecular concentration, where the non-local term involves boundary values. Existence and uniqueness of resting and travelling stationary states are established in certain parameter domains…

Aberrant erythrocyte rigidity in JAK2V617F myeloproliferative neoplasms underlies concurrent thrombosis and hemorrhage.

Thrombotic events are the leading cause of death in myeloproliferative neoplasms (MPNs) driven by the JAK2 Val617→Phe (JAK2V617F) mutation. … n. Proteomic and lipidomic analysis of Jak2V617F erythrocytes revealed the loss of key membrane and cytoskeleton proteins, a sparse spectrin cytoskeletal network, increased membrane lipid content, and deep plasma membrane invaginations that were consistent with biaxial cytoskeletal stretching and altered lipid packing. These JAK2V617F-driven changes in erythro …

Beyond cytoskeletal regulation: Rho-associated, coiled-coil-containing protein kinase 2 in kidney inflammation and fibrosis.

Chronic kidney disease (CKD), histologically defined by glomerulosclerosis, tubular atrophy, interstitial fibrosis, and capillary rarefaction, represents a profound challenge in cellular pathology. This review synthesizes recent advances to position Rho-associated, coiled-coil-containing protein kinase 2 (ROCK2) as a master regulator of maladaptive cellular remodeling in CKD. We delineate the compartment-specific histopathological roles of ROCK2, demonstrating its function as a central signaling integrator that transduces mechanical, metabolic, and inflammatory stimuli into a convergent profib

Myosin II-independent contraction of actin filaments in membrane nanotubes.

The ability to generate active stresses within filamentous actin matrices is a fundamental and evolutionarily conserved process driving locomotion and morphogenetic changes in cells. The generation of pushing forces by actin polymerization is reasonably well understood, and is known to drive lamellipodia based motility and filopodial extension. Actin filaments decorated with myosin motors can also generate contractile stresses as in the cell cortex or in cytokinetic rings. In this article we use membrane nanotubes pulled out of axonal shaft to investigate actin dynamics and force generation. W

SASH1 as a Context-Dependent Multi-Docking Scaffold Linking Receptor Signaling to Cytoskeletal Dynamics.

SASH1 (SAM [sterile alpha motif] and SH3 [SRC-homology-3] domain-containing protein 1) is a multidomain scaffold implicated in pigmentation, innate immunity, receptor signaling, cytoskeletal dynamics, vascular biology, and tumor suppression. Although genetic and expression studies link SASH1 dysfunction to diverse diseases, a unifying mechanistic framework has remained elusive. Here, we synthesize current knowledge of SASH1 structure, interaction networks, and biological functions across cell types and disease contexts. SASH1 contains an intrinsically disordered SPIDER (SLy Proteins Associated

We also discuss the translational implications of targeting the proposed neural receptor-DOCK-cytoskeleton axis, while emphasizing that such applications remain speculative and require substantial experimental validation before any clinical translation can be justified.

Tailored Xenogeneic-Free Polymer Surface Promotes Dynamic Migration of Intestinal Stem Cells.

Regenerative therapy employing intestinal stem cells (ISCs) holds promise for epithelial restoration. However, it requires scalable and robust, but fully defined culture platforms free of xenogeneic components. Most existing systems, however, typically rely on Matrigel, which complicates clinical translation and limits understanding of stem cell-material interactions. Here, a poly(ethyleneglycoldimethacrylate) (pEGDMA)-based synthetic culture surface is exposed to N2 plasma treatment for tailored modulation of surface properties. This modification enhances surface wettability and introduces N-

Identification of hub genes and transcription factors in patients with primary gout complicated with atherosclerosis.

Evidence shows that primary gout is prone to develop to atherosclerosis, but the mechanism of its occurrence is still not fully clarified. T … alyses indicated that DEGs were mostly enriched in chemokine signaling pathway, regulation of actin cytoskeleton, and TNF signaling pathway. CytoScape demonstrated 11 hub genes and two gene cluster modules. The immune infiltration analysis showed that the expression of DEGs in gout was significantly upregulated in activated CD4 T cells, gamma delta T cells, T …

Dock2 generates characteristic spatiotemporal patterns of Rac activity to regulate neutrophil polarisation, migration and phagocytosis.

INTRODUCTION: Rac-GTPases and their Rac-GEF activators play important roles in neutrophil-mediated host defence. These proteins control the adhesion molecules and cytoskeletal dynamics required for neutrophil recruitment to inflamed and infected organs, and the neutrophil effector responses that kill pathogens. METHODS: Here, we used live cell TIRF-FRET imaging in neutrophils from Rac-FRET reporter mice with deficiencies in the Rac-GEFs Dock2, Tiam1 or Prex1/Vav1 to evaluate if these proteins activate spatiotemporally distinct pools of Rac, and to correlate patterns of Rac activity with the n

The N-Terminal Part of the 1A Domain of Desmin Is a Hot Spot Region for Putative Pathogenic DES Mutations Affecting Filament Assembly.

Desmin is the major intermediate filament protein of all three muscle cell types, and connects different cell organelles and multi-protein complexes such as the cardiac desmosomes. Several pathogenic mutations in the DES gene cause different skeletal and cardiac myopathies. However, the significance of the majority of DES missense variants is currently unknown, since functional data are lacking. To determine whether desmin missense mutations within the highly conserved 1A coil domain cause a filament assembly defect, we generated a set of variants with unknown significance and systematically a

A lamin A/C variant causing striated muscle disease provides insights into filament organization.

The LMNA gene encodes the A-type lamins, which polymerize into ∼3.5-nm-thick filaments and, together with B-type lamins and associated proteins, form the nuclear lamina. Mutations in LMNA cause a wide variety of pathologies. In this study, we analyzed the nuclear lamina of embryonic fibroblasts from LmnaH222P/H222P mice, which develop cardiomyopathy and muscular dystrophy. Although the organization of the lamina appeared unaltered, there were changes in chromatin and B-type lamin expression. An increase in nuclear size and consequently a relative reduction in heterochromatin near the lamina al
