Both Dirofilaria immiti and Dipetalonema reconditum may be found in blood of infected dogs but it is not easy to distinguish D. immitis from D. reconditum in morphology. We cloned and sequenced the contiguous internal transcribed spacer (ITS) region, ITS1-5.8S-ITS2, of these two different parasites and published on GenBank as AF217800 for D. immiti and AF217801 for D. reconditum in this study. We designed two pairs of specific primers derived from ITS2 being used for polymerase chain reaction (PCR). The amplicons of ITS2 from D. immiti and D. reconditum are 302 and 348bp, respectively. The sensitive, rapid and species-specific diagnosis of Brugia infections in humans or animal models is important in determining the level of parasitemia and the efficacy of chemotherapy or vaccinations. The HhaI family of highly repeated DNA sequences from Brugia have been useful in polymerase chain reaction (PCR)-based diagnosis of brugian filarial infections in blood samples and in mosquitoes. A PCR assay was developed using a biotinylated primer, a non-biotinylated primer and a species-specific chemiluminescent probe [tris[2,2'bipyridine] ruthenium (II) chelate, TBR] to detect PCR amplified

Complete nucleotide sequences of the mitochondrial cytochrome b (Cytb) and cytochrome c oxidase subunit I (CO I) genes from various isolates of Taenia solium were examined. Eleven isolates were analyzed; two isolates from China, two isolates from Indonesia, one isolate each from India, Thailand, Mexico, Ecuador, Peru, Mozambique and Tanzania. In both genes, two isolates from Indonesia shared the same sequences. Similarly, the isolate from Mexico shared same sequences with that from Peru, and the isolate from Mozambique shared same sequences with that from Tanzania. Phylogenetic trees inferred

Chronic hyperactive dermatitis (sowda) in humans infected with the filarial parasite Onchocerca volvulus appears to reflect a hyperresponsiveness to parasite antigens. To identify antigens which play a role in this hyperresponsiveness an expression cDNA library of adult O. volvulus was screened with sera from patients with sowda. One further characterized cDNA clone, S1, consisting of 723 bp, surprisingly shows open reading frames (ORF) in both orientations. While a single ORF of 171 amino acids is present in sense orientation, a putative ORF of 95 AA is found in antisense orientation (aS1). W

Parasitic diseases are among the common health concerns in dogs and cats, with ascaridoid nematodes representing one of the major groups of zoonotic parasites. Three ascaridoids-Toxocara canis, Toxocara cati, and Toxascaris leonina-commonly infect these hosts. Effective diagnostic techniques are important for systematics, epidemiology, and clinical management. In this study, a multiplex PCR (mPCR) assay was developed for the simultaneous and specific identification of three ascaridoids. Specific primers targeting conserved mitochondrial regions were designed. Sensitivity was evaluated using se

Anthelmintic resistance is a growing threat to the treatment and control of parasitic nematode infections in both humans and animals. In the canine hookworm, Ancylostoma caninum, multi-anthelmintic drug-resistant (MADR) isolates have been identified, but the molecular mechanisms underlying resistance and the potential for these mechanisms to promote cross-resistance between drug classes remain unclear. In this study, we report evidence for a previously uncharacterized association between resistance to macrocyclic lactones (MLs) and benzimidazoles (BZs) in A. caninum. In a wild-type isolate wit

INTRODUCTION: Cystic echinococcosis (CE) is a zoonotic infection caused by the larvae of Echinococcus granulosus. Studies have indicated an indirect relationship between infection with parasites and many types of cancer, especially digestive tumors.
AIM: The following study aims to evaluate the anticancer activity of two antigens extracted from hydatid cyst antigens (extracted from cyst fluid and protoscoleces) of Echinococcus granulosus on the growth and proliferation of colorectal cancer cells compared to normal cells, and study their mechanism of action by DNA damage and protein expression

Cystic echinococcosis (CE), caused by Echinococcus granulosus sensu lato, is a significant zoonotic disease with profound public health and economic impacts. This study evaluated egg detection methods, molecular diagnostics, and post-treatment shedding dynamics in experimentally infected dogs. Three three-month-old male dogs were included in the study. Two experimental dogs (ED-1, ED-2) were orally infected with 20,000 E. granulosus protoscoleces, while one served as a control (CD). Stool samples were collected daily over 50 days post-infection and 30 days post-treatment. Detection of E. granu

BACKGROUND: Toxocara canis and Toxocara cati are parasitic nematodes that occur worldwide. As embryonated Toxocara spp. eggs in the environment pose a zoonotic risk, especially for children, optimal diagnostic approaches are necessary for effective disease response and management, including surveillance. However, little is known about the performance of different diagnostic protocols for detecting Toxocara spp. in the faeces of cats and dogs, hampering movement towards an optimal diagnostic process. This study aimed to compare detection methods, including a newly developed sequential sieving p

BACKGROUND: The genus Marshallagia (Family Haemonchidae, subfamily Ostertagiinae) contains multiple species of nematodes parasitising the abomasum (or duodenum) of ruminants, in particular of Caprinae. Male specimens have been described to be polymorphic with the frequent/major morphotype initially described in the genus Marshallagia while the minor/rare morphotype was initially often placed in the genus Grossospicularia. Due to common morphological features, certain pairs of morphotypes were suggested to belong to the same species such as Marshallagia marshalli/M. occidentalis. However, molec

Adult worms of Erschoviorchis anuiensis sp. n., parasites of the pancreas and liver of birds, were found by feeding the Muscovy ducks Cairina moschata dom. with freshwater fish (Phoxinus percnurus) from the Amur River basin (Russia). The trematodes obtained differ from the only previously known representative of the genus, E. lintoni by the large size of the ventral sucker, testes and ovary, the shape of the ovary (three-lobed vs irregular oval for E. lintoni), and the degree of vitellarium development (well-developed vitellarium with numerous follicles vs weakly developed vitelline fields for

A survey of the parasite fauna of freshwater fishes from the Wet Tropics Bioregion in Queensland, Australia, revealed the presence of a new species of Stemmatostoma Cribb, 1986 (Digenea: Cryptogonimidae). Stemmatostoma cribbi n. sp. is described from the intestine and pyloric caeca of 2 species of grunter (Terapontidae), Hephaestus fuliginosus (Macleay) and Hephaestus tulliensis (De Vis), and the Jungle perch (Kuhliidae), Kuhlia rupestris (Lacepède), collected from the Barron and Mulgrave-Russell River drainage divisions in tropical north Queensland, Australia. Stemmatostoma cribbi is primaril

BACKGROUND: Next generation sequencing (NGS) technologies are extensively used to dissect the molecular mechanisms of host-parasite interactions in human pathogens. However, ecological studies have yet to fully exploit the power of NGS as a rich source for formulating and testing new hypotheses.
METHODS: We studied Eurasian perch (Perca fluviatilis) and its eye parasite (Trematoda, Diplostomidae) communities in 14 lakes that differed in humic content in order to explore host-parasite-environment interactions. We hypothesised that high humic content along with low pH would decrease the abundan

BACKGROUND: Echinococcosis is a chronic zoonosis caused by tapeworms of the genus Echinococcus. Treatment of the disease is often expensive and complicated, sometimes requiring extensive surgery. Ultrasonographic imaging is currently the main technique for diagnosis, while immunological analysis provides additional information. Confirmation still needs pathological analysis. However, these diagnostic techniques generally detect infection in late stages of the disease. An accurate, early and non-invasive molecular diagnostic method is still unavailable.

DNA-binding proteins play critical roles in many major processes such as development and sexual biology of Schistosoma mansoni and are important for the pathogenesis of schistosomiasis. Chromatin immunoprecipitation (ChIP) experiments followed by sequencing (ChIP-seq) are useful to characterize the association of genomic regions with posttranslational chemical modifications of histone proteins. Challenges in the standard ChIP protocol have motivated recent enhancements in this approach, such as reducing the number of cells required and increasing the resolution. In this chapter, we describe th

Avitellina tapeworms are common intestinal parasites of ruminants with a worldwide distribution. In Senegal, only Avitellina centripunctata tapeworm has been reported to date, and genetic diversity was previously confirmed by enzymatic analysis. This study aims to clarify the diversity of Avitellina tapeworms isolated from sheep and cattle in Senegal. In total, 613 adult Avitellina tapeworms were collected from sheep and cattle. Morphological analysis by the light microscopy and scanning electron microscopy identified three Avitellina "morphospecies": A. centripunctata and Avitellina sp.2 were

Adult schistosomes, parasitic flatworms that cause the tropical disease schistosomiasis, have always been considered to be homolactic fermenters and, in their energy metabolism, strictly dependent on carbohydrates. However, more recent studies suggested that fatty acid β-oxidation is essential for egg production by adult female Schistosoma mansoni. To address this conundrum, we performed a comprehensive study on the lipid metabolism of S. mansoni. Incubations with [14C]-labelled fatty acids demonstrated that adults, eggs and miracidia of S. mansoni did not oxidise fatty acids, as no 14CO2 prod

There is a paucity of information on hookworm species in humans, domestic animals and wildlife in southern Africa. Our study aimed to identify hookworm species from stray dogs, humans, and selected wildlife from South Africa. A total of 356 faecal samples were screened for the presence of hookworm-like eggs and subsequently coproculture from the positive samples was carried out to obtain larvae. Hookworm-like eggs were detected in 23.03% (82/356) of samples. Of these samples, 78/296 were from dogs, 3/50 from humans and 1/10 from wildlife. DNA was then isolated from the larvae of 55 positive sa

For field-identification of taeniid cestodes in canine animals in Tibetan area, loop-mediated isothermal amplification (LAMP) assays for Echinococcus multilocularis, E. shiquicus, Taenia hydatigena, T. multiceps, T. pisiformis and T. crassiceps were developed and evaluated along with the reported assay for E. granulosus. The LAMP assays showed specific reaction with their corresponding target species DNA with the detection limit of 1 to 10 pg. Moreover, the assays for E. granulosus, E. multilocularis, T. hydatigena and T. multiceps could detect DNA extracted from 3 or more eggs of their corres

Complete mitochondrial genomes and nuclear rRNA operons of eight geographically distinct isolates of the Asian fish tapeworm Schyzocotyle acheilognathi (syn. Bothriocephalus acheilognathi), representing the parasite's global diversity spanning four continents, were fully characterised using an Illumina sequencing platform. This cestode species represents an extreme example of a highly invasive, globally distributed pathogen of veterinary importance with exceptionally low host specificity unseen elsewhere within the parasitic flatworms. In addition to eight specimens of S. acheilognathi, we ful
