Immunity against microbes depends on recognition of pathogen-associated molecular patterns by innate receptors. Signaling pathways triggered by Brucella abortus DNA involves TLR9, AIM2, and stimulator of IFN genes (STING). In this study, we observed by microarray analysis that several type I IFN-associated genes, such as IFN-β and guanylate-binding proteins (GBPs), are downregulated in STING knockout (KO) macrophages infected with Brucella or transfected with DNA. Additionally, we determined that STING and cyclic GMP-AMP synthase (cGAS) are important to engage the type I IFN pathway, but only

Scrub typhus is a potentially lethal infection that is caused by the obligate intracellular bacterium Orientia tsutsugamushi The roles of Toll-like receptor 2 (TLR2) and TLR4 in innate recognition of O. tsutsugamushi have not been elucidated. By overexpression of TLR2 or TLR4 in HEK293 cells, we demonstrated that TLR2, but not TLR4, recognizes heat-stable compounds of O. tsutsugamushi that were sensitive to treatment with sodium hydroxide, hydrogen peroxide, and proteinase K. TLR2 was required for the secretion of tumor necrosis factor alpha (TNF-α) and interleukin-6 (IL-6) by dendritic cells.

The lectin pathway of complement is an important component of innate immunity. Its activation has been thought to occur via recognition of pathogens by mannan-binding lectin (MBL) or ficolins in complex with MBL-associated serine protease (MASP)-2, followed by MASP-2 autoactivation and cleavage of C4 and C2 generating the C3 convertase. MASP-1 and MASP-3 are related proteases found in similar complexes. MASP-1 has been shown to aid MASP-2 convertase generation by auxiliary C2 cleavage. In mice, MASP-1 and MASP-3 have been reported to be central also to alternative pathway function through acti

Receptors for the Fc portion of IgG (Fc gamma Rs) integrate the innate and acquired components of immunity by coupling the specific recognition of IgG Abs to the activation of phagocytic leukocytes. Knowledge of the molecular mechanisms that regulate phagocyte stimulation by Fc gamma Rs may permit therapeutic modulation to augment immunoprotective aspects and minimize damage to host tissues in diverse inflammatory diseases. Since phospholipase D (PLD) has been linked to the stimulation of cytotoxic leukocyte responses, we characterized Fc gamma R-dependent activation of PLD in human macrophage

UNLABELLED: STING is a protein in the cytosolic DNA and cyclic dinucleotide sensor pathway that is critical for the initiation of innate responses to infection by various pathogens. Consistent with this, herpes simplex virus 1 (HSV-1) causes invariable and rapid lethality in STING-deficient (STING(-/-)) mice following intravenous (i.v.) infection. In this study, using real-time bioluminescence imaging and virological assays, as expected, we demonstrated that STING(-/-) mice support greater replication and spread in ocular tissues and the nervous system. In contrast, they did not succumb to cha

Air pollution is a major environmental risk factor contributing to the development, exacerbation, and persistence of asthma. A wide range of airborne pollutants, including particulate matter (PM), ozone (O₃), nitrogen dioxide, sulfur dioxide, diesel exhaust particles (DEPs), volatile organic compounds, and biological contaminants, can interact with the airway epithelium and immune system, leading to dysregulated inflammatory responses. Increasing evidence indicates that pollutant exposure induces epithelial injury, disrupts epithelial barrier integrity, promotes oxidative stress, and activates

Tissue repair and regeneration represent multifaceted processes involving a coordinated interplay of diverse cell types, growth factors, and cytokines. Following tissue injury, innate immune cells serve as the primary cellular responders that infiltrate the damaged microenvironment, clear cellular debris, and undergo phenotype transitions essential for establishing a proregenerative environment. To facilitate functional recovery, three-dimensional scaffolds are conventionally used to give physical signals and structural support. However, these constructs frequently encounter barriers in host-s

Immune checkpoint blockade (ICB) produces durable tumor control in a subset of patients, yet resistance is usually interpreted through tumor-intrinsic lesions or suppression within the tumor microenvironment (TME). We propose that ICB resistance can also be organized as failure of a multiscale antitumor immune circuit comprising local immune execution, regional immune education in tumor-draining lymph nodes (TDLNs), and systemic immune calibration by the host. In this model, "hierarchical" denotes nested functional dependence rather than one-way anatomical control: local killing depends probab

Brucellosis is a worldwide zoonotic disease caused by Brucella spp. Human infection often results from direct contact with tissues from infected animals or by consumption of undercooked meat and unpasteurised dairy products, causing serious economic losses and public health problems. The male genitourinary system is a common involved system in patients with brucellosis. Among them, unilateral orchitis and epididymitis are the most common. Although the clinical and imaging aspect of orchi-epididymitis caused by brucellosis have been widely described, the cellular and molecular mechanisms involv

Mycobacterium tuberculosis (Mtb) has been known to evade host innate immunity by manipulating macrophage function. Interleukin-1β (IL-1β) is a cytokine secreted by macrophages as a consequence of inflammasome activation. Mtb can inhibit activation of the NLRP3 and AIM2 inflammasomes and subsequent pyroptosis. The capacity of Mtb to manipulate other types of inflammasomes is unknown. In this study, we investigated whether Mtb or the nontuberculous mycobacteria Mycobacterium kansasii (Mkan) and Mycobacterium smegmatis (Msmeg) can inhibit the NLRP1 or pyrin inflammasomes. We show that none of the

Hemiconvulsion-hemiplegia-epilepsy (HHE) syndrome is a rare pediatric epilepsy syndrome characterized by prolonged focal febrile seizures, postictal hemiparesis, and progressive unilateral brain injury, often followed by chronic epilepsy. We report a previously healthy 10-month-old girl who presented with a prolonged left-sided focal fever-associated seizure. She tested positive for SARS-CoV-2 but did not meet criteria for multisystem inflammatory syndrome in children. On admission, she had left-sided flaccid hemiparesis. Brain MRI showed mild diffusion restriction and marked hyperperfusion of

BACKGROUND: Mutations in the LMNA gene, which encodes the nuclear envelope proteins lamins A and C, account for 6% to 8% of all cases of congenital dilated cardiomyopathy. LMNA-related dilated cardiomyopathy is among the most severe forms of dilated cardiomyopathy, with limited treatment options due to an incomplete understanding of its molecular mechanisms.
METHODS: We generated an inducible, cardiomyocyte-specific Lmna deletion mouse model (cKO) and conducted comprehensive bulk, single-nucleus, and spatial transcriptomic analyses on left ventricular free wall tissue from cKO and controls la

BACKGROUND: In solid organ transplantation, the intestine remains the most challenging. Previous studies have linked NOD2 genetic variation to intestinal transplantation (ITx) outcomes. Since then, a larger set of inflammatory bowel disease-associated genetic variants (IBDGVs) has been identified. This study aims to explore the prevalence and association of these IBDGVs with ITx outcomes.
METHOD: Clinical data and DNA from 150 donor/recipient pairs were collected from 5 ITx centers (Leuven, Cambridge, Paris, Madrid, Buenos Aires). Genotyping-by-sequencing of 540 IBDGVs was performed, and gene

Lectins are carbohydrate-binding proteins with diverse biological functions, particularly their roles in innate immunity through the recognition of pathogen-associated molecular patterns and mediation of non-specific immune responses. This study investigates the bioactivities of mucus lectins derived from Lissachatina fulica (LFLs). The LFLs were purified using ion exchange chromatography and analyzed for toxicity, antimicrobial, antioxidant, antitumor, and insecticidal properties. SDS-PAGE experiment revealed protein bands of approximately 70.0 ± 1.0 and 49.0 ± 1.0 kDa. Hemagglutination activ

With rising life expectancy, more elderly individuals are undergoing surgery, highlighting the need to understand postoperative neurological complications. Studies have linked general anesthesia (GA) to olfactory dysfunction (OD) and cognitive decline following operation (OP), though mechanisms remain unclear. Using aged C57BL/6 mice subjected to laparotomy and 2-hour isoflurane exposure, we assessed behavioral and molecular outcomes through functional tests and bulk RNA sequencing (RNAseq). We observed persistent OD in the weeks following GA/OP, along with reduced limb strength and motor coor

In recent years, chimeric antigen receptor (CAR) T-cell therapy has emerged as a groundbreaking approach in cancer immunotherapy. Particularly in hematologic malignancies, such as B-cell acute lymphoblastic leukemia (B-ALL), B cell lymphomas and multiple myeloma. CAR-T therapy has demonstrated remarkable clinical efficacy, leading to the approval of several CAR-T cell products and offering significant benefits to numerous leukemia patients. Despite these successes, the application of CAR-T cells in solid tumors remains limited due to significant challenges, including immunosuppressive tumor mi

Antigenic cell fragments, pathogen-associated molecular patterns, and other immunostimulants in bacterial lysates or extracts may induce local and systemic immune responses in specific and nonspecific paradigms. Based on current knowledge, this review aimed to determine whether bacterial lysate has comparable functions in infectious diseases and cancer treatment. In infectious diseases, including respiratory and urinary tract infections, immune system activation by bacterial lysate can identify and combat pathogens. Commercially available bacterial lysates, including OM-85, Ismigen, Lantigen B

Toll-like receptors (TLRs) are pattern recognition receptors of the innate immunity. TLRs are known to mediate both antitumor effects and tumorigenesis. TLRs are abundant in many cancers, but their expression in small bowel neuroendocrine tumors (SB-NETs) is unknown. We aimed to characterize the expression of TLRs 1-9 in SB-NETs and lymph node metastases and evaluate their prognostic relevance. The present study included 125 patients with SB-NETs, of whom 95 had lymph node metastases, from two Finnish hospitals. Tissue samples were stained immunohistochemically for TLR expression, assessed bas
