The statistical power of genome-wide association (GWA) studies to detect risk alleles for human diseases is limited by the unfavorable ratio of SNPs to study subjects. This multiple testing problem can be surmounted with very large population sizes when common alleles of large effects give rise to disease status. However, GWA approaches fall short when many rare alleles may give rise to a common disease, or when the number of subjects that can be recruited is limited. Here, we demonstrate that this multiple testing problem can be overcome by a comparative genomics approach in which an initial

Cholera toxin (CT) travels as an intact AB(5) protein toxin from the cell surface to the endoplasmic reticulum (ER) of an intoxicated cell. In the ER, the catalytic A1 subunit dissociates from the rest of the toxin. Translocation of CTA1 from the ER to the cytosol is then facilitated by the quality control mechanism of ER-associated degradation (ERAD). Thermal instability in the isolated CTA1 subunit generates an unfolded toxin conformation that acts as the trigger for ERAD-mediated translocation to the cytosol. In this work, we show by circular dichroism and fluorescence spectroscopy that exp

We recently performed an autopsy on a premature female newborn with rhizomesoacromelic limb shortening of the upper and lower extremities, craniofacial dysmorphism, and chondrodysplasia punctata. A diagnosis of Conradi-Hunermann-Happle syndrome or X-linked dominant chondrodysplasia punctata was made based on elevated cholest-8(9)-ene-3beta-ol in serum and tissues. Molecular analysis of EBP, mutations of which are responsible for this malformation syndrome, revealed a monoallelic missense mutation, c.328 G>A (R110Q). We present this case as an illustration of an unusually severe manifestation o

In lymphocytes, the pineal hormone, melatonin, suppresses 5-lipoxygenase (5-LO) gene expression. Because circadian fluctuations in melatonin content are prominent in the pineal, we hypothesized that 5-LO mRNA level in this gland is greater when melatonin is low (day) than at night. Using the reverse transcription/polymerase chain reaction we assayed the levels of mRNAs coding for 5-LO, serotonin N-acetyltransferase (NAT), and for a constitutive gene, cyclophilin, in rat pineals obtained at 10:30-11:00 h (day) or 24:30-01:00 (night). Cyclophilin mRNA was not affected by circadian rhythm, wherea

It is believed that DOPA-negative melanocytes in the outer root sheath of the human hair follicle are activated, become identifiable by DOPA staining, and migrate into the epidermis during the repigmenting phase of vitiligo. These cells are difficult to identify, however, and otherwise have not been characterized. These cells are readily identified by immunofluorescence, immunohistochemistry, and immunoelectronmicroscopy using the antibodies NKI/beteb and A4F11, which recognize premelanosome-related antigens. The majority of the outer root sheath melanocytes were found in the mid to the upper

In this report we describe that the functional capacity of the mitochondrial oxidative phosphorylation system can be studied in cultured skin fibroblasts permeabilized with a limited amount of digitonin. By using a variety of different oxidizable substrates, information can be obtained on the functional activity of complex I, for instance, which is important since different diseases in man have recently been identified in which complex I is deficient. The method described may contribute to the biochemical characterization of patients suffering from one of a variety of encephalomyopathies.

Isozyme analysis was carried out on Onchocerca volvulus worms collected from Liberia, Ivory Coast, Burkina Faso and Sudan to see whether this technique could detect differences between forest and savannah populations of this parasite. A total of 243 forest and 189 savannah individual female worms were electrophoresed and stained for seven enzymes. Four showed some polymorphism, LDH, MDH, PGM and MPI and the other three, GAPDH, PEP and GPI were invariant. Statistical analysis of the results showed that the relative proportions of genotypes from within the different countries conformed to Hardy-

An Onchocerca sp. was isolated from the nuchal ligaments of 78 out of 79 naturally infected cattle in Southern Mexico. Infections were moderate to heavy, averaging 20-30 parasites per nuchal ligament. Microfilariae were concentrated in the skin of the head and neck, although small numbers were found in the skin of the withers, brisket and medial abdomen. High numbers of microfilariae were found in the skin around the eye, but not in the ocular tissues themselves a situation dissimilar to that found in human onchocerciasis. Morphologically, adult females, males and microfilariae were identified

BACKGROUND: Airway inflammation plays a major role in the development and progression of chronic obstructive pulmonary disease (COPD). Quercetin, which has potent antioxidant and anti-inflammatory properties reduces lung inflammation in mice displaying COPD-like lung disease. Previously we showed that quercetin was safely tolerated up to 2000 mg/day. We conducted a pilot Phase II clinical trial to examine the effects of quercetin on inflammation in COPD patients.
METHODS: Fourteen COPD patients with ≥ 10 pack-year smoking history and CRP > 3.0 mg/L were randomized in 1:2 ratio to either place

Leber congenital amaurosis type 2 (LCA2) is an inherited retinal disorder, with severe vision impairment in children, progressing to complete blindness in the later stages of life. The current approved treatment involves Adeno-associated virus (AAV) vector serotype 2-based subretinal delivery of human RPE65 gene. However, long-term follow-up have reported gradual loss of phenotypic response. To overcome this, we have pursued strategies aimed at improving the transduction efficacy of the vector, optimizing the transgene for enhanced protein expression and co-delivering the therapeutic vector al

BACKGROUND: Giardia duodenalis and Cryptosporidium spp. are globally significant enteric protozoa responsible for waterborne diarrheal disease with important public health and zoonotic implications. Conventional genotyping approaches based on Sanger sequencing frequently underestimate mixed infections and minority variants, limiting the resolution of environmental molecular surveillance. This study aimed to characterize the presence and genetic diversity of both pathogens in surface water and wastewater samples from the Río Pasto basin (Nariño, southwestern Colombia) using a multilocus amplico

UNLABELLED: During infection, bacterial pathogens rely on secreted virulence factors to manipulate the host cell. However, in gram-positive bacteria, the molecular mechanisms underlying the folding and activity of these virulence factors after membrane translocation are not clear. Here, we solved the protein structures of two secreted parvulin and two secreted cyclophilin-like peptidyl-prolyl isomerase (PPIase) ATP-independent chaperones found in gram-positive streptococcal species. The extracellular parvulin-type PPIase, PrsA in Streptococcus pneumoniae and Streptococcus mutans maintain dimer

Disulfide bond formation has a central role in protein folding of both eukaryotes and prokaryotes. In bacteria, disulfide bonds are catalyzed by DsbA and DsbB/VKOR enzymes. First, DsbA, a periplasmic disulfide oxidoreductase, introduces disulfide bonds into substrate proteins. Then, the membrane enzyme, either DsbB or VKOR, regenerate DsbA's activity by the formation of de novo disulfide bonds which reduce quinone. We have previously performed a high-throughput chemical screen and identified a family of warfarin analogs that target either bacterial DsbB or VKOR. In this work, we expressed func

Transmissible gastroenteritis virus (TGEV), a member of the coronavirus family, is the pathogen responsible for transmissible gastroenteritis, which results in mitochondrial dysfunction in host cells. Previously, we identified 123 differentially expressed circular RNAs (cRNA)from the TGEV-infected porcine intestinal epithelial cell line jejunum 2 (IPEC-J2). Previous bioinformatics analysis suggested that, of these, circBIRC6 had the potential to regulate mitochondrial function. Furthermore, mitochondrial permeability transition, a key step in the process of mitochondrial dysfunction, is known

In the cell, the conformations of nascent polypeptide chains during translation are modulated by both the ribosome and its associated molecular chaperone, trigger factor. The specific interactions that underlie these modulations, however, are still not known in detail. Here, we combine protein engineering, in-cell and in vitro NMR spectroscopy, and molecular dynamics simulations to explore how proteins interact with the ribosome during their biosynthesis before folding occurs. Our observations of α-synuclein nascent chains in living Escherichia coli cells reveal that ribosome surface interacti

Aspirin is one of the most commonly used nonsteroidal anti-inflammatory drugs. Various potential pharmacological effects of aspirin, such as anticancer, antibacterial activity, and prolonging life expectancy have been discovered. However, the mechanism of aspirin is not fully elucidated. Herein, the effects of aspirin on fatty acid metabolism in yeast cell model Saccharomyces cerevisiae were studied. The results showed that aspirin can induce lipid accumulation and reduce the unsaturated fat index in cells. The assessment of cell membrane integrity demonstrated that aspirin caused damage to th
