Guidelines for the Investigation of Mediating Variables in Business Research.

Business theories often specify the mediating mechanisms by which a predictor variable affects an outcome variable. In the last 30 years, investigations of mediating processes have become more widespread with corresponding developments in statistical methods to conduct these tests. The purpose of this article is to provide guidelines for mediation studies by focusing on decisions made prior to the research study that affect the clarity of conclusions from a mediation study, the statistical models for mediation analysis, and methods to improve interpretation of mediation results after the resea

Separability of histopathology foundation model embeddings of stimulated Raman histology images for lung carcinoma subtyping.

Histopathology foundation models trained on hematoxylin and eosin (H&E) whole-slide images have recently emerged as versatile encoders for downstream diagnostic tasks, but their transferability to out-of-domain imaging modalities such as stimulated Raman histology (SRH) is unknown. Here, we retrospectively evaluated whether embeddings from histopathology foundation models enable intraoperative SRH-based classification of lung cancer patients into (i) non-small cell lung cancer (NSCLC) versus pulmonary metastasis (MET) and (ii) adenocarcinoma (AC) versus squamous cell carcinoma (SCC). For NSCLC

Computer models versus reality: how well do in silico models currently predict the sensitization potential of a substance.

National legislations for the assessment of the skin sensitization potential of chemicals are increasingly based on the globally harmonized system (GHS). In this study, experimental data on 55 non-sensitizing and 45 sensitizing chemicals were evaluated according to GHS criteria and used to test the performance of computer (in silico) models for the prediction of skin sensitization. Statistic models (Vega, Case Ultra, TOPKAT), mechanistic models (Toxtree, OECD (Q)SAR toolbox, DEREK) or a hybrid model (TIMES-SS) were evaluated. Between three and nine of the substances evaluated were found in the

Sufficient Sample Size and Power in Multilevel Ordinal Logistic Regression Models.

For most of the time, biomedical researchers have been dealing with ordinal outcome variable in multilevel models where patients are nested in doctors. We can justifiably apply multilevel cumulative logit model, where the outcome variable represents the mild, severe, and extremely severe intensity of diseases like malaria and typhoid in the form of ordered categories. Based on our simulation conditions, Maximum Likelihood (ML) method is better than Penalized Quasilikelihood (PQL) method in three-category ordinal outcome variable. PQL method, however, performs equally well as ML method where fi

Mapping common psychiatric disorders: structure and predictive validity in the national epidemiologic survey on alcohol and related conditions.

CONTEXT Clinical experience and factor analytic studies suggest that some psychiatric disorders may be more closely related to one another, as indicated by the frequency of their co-occurrence, which may have etiologic and treatment implications. OBJECTIVE To construct a virtual space of common psychiatric disorders, spanned by factors reflecting major psychopathologic dimensions, and locate psychiatric disorders in that space, as well as to examine whether the location of disorders at baseline predicts the prevalence and incidence of disorders at 3-year follow-up. DESIGN, SETTING, AND PATIENT

Hypothesis testing in semiparametric additive mixed models.

We consider testing whether the nonparametric function in a semiparametric additive mixed model is a simple fixed degree polynomial, for example, a simple linear function. This test provides a goodness-of-fit test for checking parametric models against nonparametric models. It is based on the mixed-model representation of the smoothing spline estimator of the nonparametric function and the variance component score test by treating the inverse of the smoothing parameter as an extra variance component. We also consider testing the equivalence of two nonparametric functions in semiparametric addi

Population pharmacokinetic and pharmacodynamic analysis in allergic diseases.

In this chapter, we introduce the concepts and methodologies of population analysis as applied to analyzing pharmacokinetic and pharmacodynamic data. One of the key determining characteristics of the population approach is that through it, one seeks not only to characterize deterministic trends in the data, but also to identify and estimate the magnitudes of the important sources of variability within the data. The first section of this chapter provides an introduction to the primary concepts of, and motivation for, population modeling by way of a hypothetical case study. Then, the various met

Effect of urethral compliance on the steady state p-Q relationships assessed with a mechanical analog of the male lower urinary tract.

AIMS: Using a lumped parameter theoretical model of bladder outlet function, we previously explored the relationship between the tube law of the urethra and the pressure-flow characteristics during micturition [Mijailovich et al., 2004]. To validate this theoretical model, we constructed a mechanical analog of the male lower urinary tract that incorporated elements simulating all relevant parameters of the theoretical model. In addition, we determined the effect of alterations in compliance of the flow-controlling zone on these relationships. METHODS: In the mechanical analog, the bladder nec

Trifluoperazine causes a disturbance in glycerophospholipid monolayers containing phosphatidylserine (PS): effects of pH, acyl unsaturation, and proportion of PS.

We have studied the interaction of trifluoperazine (TFP) with monolayers of various glycerophospholipids at 37 degrees C. TFP (1-10 microM) had little effect on surface pressure/molecular area isotherms in monolayers (on pure water) of dipalmitoylphosphatidylcholine (DPPC) and dipalmitoylphosphatidylethanolamine but greatly increased the mean molecular area (mma) of dipalmitoylphosphatidylserine; the increment was greatest between 0 and 1 microM, and a further increase to 10 microM TFP gave only a slight increase in mma. With phosphatidylserine (PS)-containing stearoyl and varying acyls in the

Efficient multi-fidelity computation of blood coagulation under flow.

Clot formation is a crucial process that prevents bleeding, but can lead to severe disorders when imbalanced. This process is regulated by the coagulation cascade, a biochemical network that controls the enzyme thrombin, which converts soluble fibrinogen into the fibrin fibers that constitute clots. Coagulation cascade models are typically complex and involve dozens of partial differential equations (PDEs) representing various chemical species' transport, reaction kinetics, and diffusion. Solving these PDE systems computationally is challenging, due to their large size and multi-scale nature.

A modified Ising model of Barabási-Albert network with gene-type spins.

The central question of systems biology is to understand how individual components of a biological system such as genes or proteins cooperate in emerging phenotypes resulting in the evolution of diseases. As living cells are open systems in quasi-steady state type equilibrium in continuous exchange with their environment, computational techniques that have been successfully applied in statistical thermodynamics to describe phase transitions may provide new insights to the emerging behavior of biological systems. Here we systematically evaluate the translation of computational techniques from s

Interrogating Emergent Transport Properties for Molecular Motor Ensembles: A Semi-analytical Approach.

Intracellular transport is an essential function in eucaryotic cells, facilitated by motor proteins-proteins converting chemical energy into kinetic energy. It is understood that motor proteins work in teams enabling unidirectional and bidirectional transport of intracellular cargo over long distances. Disruptions of the underlying transport mechanisms, often caused by mutations that alter single motor characteristics, are known to cause neurodegenerative diseases. For example, phosphorylation of kinesin motor domain at the serine residue is implicated in Huntington's disease, with a recent st

Deterministic models of inhalational anthrax in New Zealand white rabbits.

Computational models describing bacterial kinetics were developed for inhalational anthrax in New Zealand white (NZW) rabbits following inhalation of Ames strain B. anthracis. The data used to parameterize the models included bacterial numbers in the airways, lung tissue, draining lymph nodes, and blood. Initial bacterial numbers were deposited spore dose. The first model was a single exponential ordinary differential equation (ODE) with 3 rate parameters that described mucociliated (physical) clearance, immune clearance (bacterial killing), and bacterial growth. At 36 hours postexposure, the

Characterization of the fungal microbiota (mycobiome) in healthy and dandruff-afflicted human scalps.

The human scalp harbors a vast community of microbial mutualists, the composition of which is difficult to elucidate as many of the microorganisms are not culturable using current culture techniques. Dandruff, a common scalp disorder, is known as a causative factor of a mild seborrheic dermatitis as well as pityriasis versicolor, seborrheic dermatitis, and atopic dermatitis. Lipophilic yeast Malassezia is widely accepted to play a role in dandruff, but relatively few comprehensive studies have been reported. In order to investigate fungal biota and genetic resources of dandruff, we amplified t

On the global stability of a generalized cholera epidemiological model.

In this paper, we conduct a careful global stability analysis for a generalized cholera epidemiological model originally proposed in [J. Wang and S. Liao, A generalized cholera model and epidemic/endemic analysis, J. Biol. Dyn. 6 (2012), pp. 568-589]. Cholera is a water- and food-borne infectious disease whose dynamics are complicated by the multiple interactions between the human host, the pathogen, and the environment. Using the geometric approach, we rigorously prove the endemic global stability for the cholera model in three-dimensional (when the pathogen component is a scalar) and four-di

Thermodynamic and infrared analyses of the interaction of chlorpromazine with phospholipid monolayers.

An investigation has been made of the interaction between chlorpromazine (CPZ) and monolayers of 1,2-dipalmitoyl-sn-3-glycerophosphatidylcholine (DPPC) and 1,2-dipalmitoyl-sn-3-glycero[phospho-rac-(1-glycerol)] (DPPG), both at the air/water interface and in transferred Langmuir-Blodgett films. The Gibbs free energy, DeltaG, and the compressibility modulus (C(S)(-1)), obtained from the surface pressure isotherms, indicated changes in the in-plane interactions of CPZ/DPPG mixed monolayers, with positive values of DeltaG. The arrangement of CPZ in the zwitterionic DPPC monolayers causes a weaker

Statistical procedures for calculating the residual virulence of Brucella abortus strain 19 (S19) and Brucella melitensis strain Rev 1 vaccines in mice: theoretical basis and practical applications.

Regular control of the biological quality of live Brucella abortus strain 19 (S19) and B. melitensis strain Rev 1 vaccines is essential for the successful management of ruminant brucellosis in affected countries. The reference procedures recommended by the OIE (World organisation for animal health) and the European Pharmacopoeia include the determination of residual virulence, expressed as the recovery time 50 (RT50), of the tested (problem) vaccine in a reference mouse model compared with the RT50 of the corresponding reference strains in the same assay. The underlying statistical procedure a

Evaluating allograft risk models in organ transplantation: Understanding and balancing model discrimination and calibration.

In the field of organ transplantation, the accurate assessment of donor organ quality is necessary for efficient organ allocation and informed consent for recipients. A common approach to organ quality assessment is the development of statistical models that accurately predict posttransplant survival by integrating multiple characteristics of the donor and allograft. Despite the proliferation of predictive models across many domains of medicine, many physicians may have limited familiarity with how these models are built, the assessment of how well models function in their population, and the

Applicability of simplified computational models in prediction of peak wall stress in abdominal aortic aneurysms.

In the paper impact of different material models on the calculated peak wall stress (PWS) and peak wall rupture risk (PWRR) in abdominal aortic aneurysms (AAAs) is assessed. Computational finite element models of 70 patient-specific AAAs were created using two different material models - a realistic one based on mean population results of uniaxial tests of AAA wall considered as reference, and a 100 times stiffer artificial model. The calculated results of PWS and PWRR were tested to evaluate statistical significance of differences caused by the non-realistic material model. It was shown that

Using Natural Language Processing Methods to Predict Topics Included in 2019 Ohio Syphilis Disease Intervention Specialist Records.

BACKGROUND: Free-text notes in disease intervention specialist (DIS) records may contain relevant information for sexual transmitted infection control. In their current form, the notes are not analyzable without manual reading, which is labor-intensive and prone to error. METHODS: We used natural language processing methods to analyze 2019 Ohio DIS syphilis records with nonmissing notes (n = 1987). We identified 21 topics relevant for transmission and case investigations. We manually coded these records to create "gold standard" labels for each topic (0 = topic not present, 1 = topic present)
