The Janus kinase, JAK3 plays an important role in interleukin-2 (IL-2)-dependent signal transduction and proliferation of T lymphocytes. Our findings show that prostaglandin E2 (PGE2) can inhibit upregulation of JAK3 protein in naive T cells and can downregulate its expression in primed cells. Reduction in JAK3 was selective because expression of other tyrosine kinases (JAK1, p56(lck), and p59(fyn)) and signal transducer and activator of transcription (STAT)5, which are linked to IL-2 receptor (IL-2R) signaling pathway, were not affected. Inhibition of JAK3 may be controlled by intracellular c

BACKGROUND: The therapeutic outcomes for rheumatoid arthritis (RA) remain unsatisfactory. Neutrophil migration and activation in the synovium are pivotal drivers of RA pathology. Sidaxue (SX), a traditional Miao herbal formula, has shown anti-RA potential, yet its mechanism concerning neutrophil migration is unclear.
PURPOSE: This study aims to evaluate the effects of SX on neutrophil migration in RA and to uncover the novel molecular mechanisms.
METHODS: We integrated bulk RNA sequencing (RNA-seq) and single-cell RNA-seq (scRNA-seq) with Mendelian randomization (MR) to pinpoint critical tar

BACKGROUND: Post-traumatic stress disorder (PTSD) is associated with dysregulation of the hypothalamic-pituitary-adrenal (HPA) axis and the sympatho-adrenomedullary system, leading to immune imbalance and alterations in microbiota composition. Stress-induced disruption of intestinal barrier integrity may promote bacterial translocation to peripheral organs, including the spleen. Aripiprazole (ARI), an atypical antipsychotic proposed for PTSD treatment, also modulates the immune system and microbiota. This study investigated the effects of ARI on splenic microbiota composition and splenic neuro

The stress-induced keratin 16 intermediate filament gene, KRT16, and K16 protein are spatially restricted to the suprabasal layers of the epidermis and widely used as biomarkers in psoriasis, hidradenitis suppurativa, atopic dermatitis, and other inflammatory disorders. Pathogenic variants in KRT16 cause pachyonychia congenita (PC), a rare genetic condition in which tissue homeostasis is disrupted in palmoplantar epidermis, nail, oral mucosa, and other KRT16-expressing epithelial appendages. How K16 may regulate the molecular mechanisms underlying these conditions is poorly understood. Here, w

Interleukin-19 like (IL-19L) is a member of the interleukin-10 family and plays a crucial role in inflammation, immune regulation, and other biological processes. Although the characteristics and functions of IL-10 family members in mammals have been elucidated, those of IL-19L in fish remain poorly understood. In this study, we cloned the IL-19L gene from the large yellow croaker (Larimichthys crocea), designated as LcIL-19L. The open reading frame (ORF) of LcIL-19L comprises 684 base pairs, encoding 227 amino acids, and lacks a signal peptide. It possesses a typical IL-10 domain and six cons

BACKGROUND: Endometrial cancer lacks robust immune-linked biomarkers that can be translated into tractable therapeutic strategies. WNK4, a member of the with-no-lysine (WNK) kinase family, is involved in cancer-relevant signaling pathways; however, its immunological significance in endometrial cancer has yet to be defined.
METHODS: Immune deconvolution and gene-immune correlation analyses were performed to relate WNK4 to immune-cell-associated signatures. WNK4 expression was assessed in clinical endometrial cancer tissues and cell lines by immunoblotting. Macrophage polarization was modeled u

Protein kinase C (PKC) signalling has been shown to be dysregulated in various cancers including acute lymphoblastic leukemia (ALL). We have previously determined that changes in the expression levels of SLC43A3-encoded equilibrative nucleobase transporter 1 (ENBT1) can significantly alter 6-mercaptopurine (6-MP) toxicity in ALL cells. 6-MP is a common drug used in ALL chemotherapy. Furthermore, it has been reported that activation of PKC by phorbol 12-myristate 13-acetate (PMA) impacts nucleobase uptake via an ENBT1-like transporter in Lilly Laboratories Culture-Porcine Kidney 1 (LLC-PK1) cel

INTRODUCTION: Patients with chronic kidney disease (CKD) and diabetes mellitus face a heightened risk of cardiovascular complications and infections, potentially exacerbated by dysregulated NETosis. Given the superior survival rates observed with hemodiafiltration (HDF) over high-flux hemodialysis (HD; HFHD) and the documented NETosis dysregulation in HD and in patients with diabetes mellitus, this study aimed to investigate the impact of dialysis modality on NETosis activity in patients on HD, stratified by diabetes status.
METHODS: A total of 20 patients on HD (10 with diabetes, 10 without

Heat shock protein beta-1 may be involved in regulating ferroptosis in cells. The expression of heat shock protein beta-1 is upregulated after stroke; however, the underlying mechanism of action of heat shock protein beta-1 in cerebral ischemia/reperfusion injury remains unclear. Here, using both in vivo and in vitro models of ischemic injury—middle cerebral artery occlusion/reperfusion in C57BL/6J mice and oxygen-glucose deprivation/reoxygenation in BV-2 microglial cells—we observed that heat shock protein beta-1 overexpression significantly reduced infarct volume, mitigated neuronal loss, an

BACKGROUND AND AIMS: Alcohol-associated liver disease (ALD) is one of the leading causes of severe liver disease with limited pharmacological treatments for alcohol-associated steatohepatitis. CD44, a glycoprotein mainly expressed in immune cells, has been implicated in multiple inflammatory diseases but has never been studied in the ALD context. We therefore studied its contribution to alcohol-associated steatohepatitis development in mice and its expression in patients with ALD.
APPROACH AND RESULTS: Here, we report that liver CD44 expression is associated with liver injury and inflammation

BACKGROUND: Lifelong exercise is associated with beneficial immune adaptations, but the extent to which these adaptations manifest during an acute inflammatory challenge remains unclear. Therefore, we aimed to compare the inflammatory responses to ex vivo whole blood and peripheral blood mononuclear cells [PBMCs] cultures from young and master athletes, before and after a single bout of moderate-intensity exercise.
METHODS: Young (n=7; 22 ± 4 years) and master (n=12; 52 ± 9 years) female and male athletes with similar performance levels performed a 30-minute bout of moderate-intensity exercis

BACKGROUND: Sepsis is a common critical condition that can lead to multiple organ injury. Sepsis-induced acute respiratory distress syndrome (ARDS) is frequently an important cause of poor prognosis and is associated with high mortality rates, despite existing therapeutic interventions. Neutrophil infiltration and extracellular traps (NET) are implicated in acute lung injury (ALI) and ARDS following sepsis. As circulating neutrophils infiltrate infected tissues, they come into direct contact with vascular endothelial cells (ECs). Although the ability of NETs to induce endothelial damage is wel

Magnesium hydride (MgH2) is a hydrogen storage material that is known for its high capacity and safety and is capable of releasing hydrogen in a controlled manner when administered orally. This release of hydrogen has been associated with a range of biological effects, including anti-inflammatory properties, antioxidant activity, and protection of the intestinal barrier. Previous research has shown that neutrophil extracellular traps (NETs) play a role in the dysfunction of the intestinal barrier in conditions such as sepsis and critical illnesses. However, it remains unclear as to whether MgH

AIM: The high glucose (HG) environment in diabetic periodontitis aggravates the damage of periodontal tissue. Pyroptosis has been shown to be positively correlated with the severity of periodontitis, including macrophage pyroptosis. O-GlcNAcylation is a posttranslational modification that is involved in the pathogenesis of periodontitis. However, whether HG regulates macrophage pyroptosis through O-GlcNAcylation remains uncertain. This study aimed to investigate the effect of HG on the O-GlcNAcylation level of a pyroptosis regulator GSDME in macrophages to further probe the mechanisms of diabe
