Arginine methylation-enabled FUS phase separation with SMN contributes to neuronal granule formation.

Various ribonucleoprotein complexes (RNPs) often function in the form of membraneless organelles derived from multivalence-driven liquid-liquid phase separation (LLPS). Post-translational modifications, such as phosphorylation and arginine methylation, govern the assembly and disassembly of membraneless organelles. This study reveals that asymmetric dimethylation of arginine can create extra binding sites for multivalent Tudor domain-containing proteins like survival of motor neuron (SMN) protein, thereby lowering the threshold for LLPS of RNPs, such as fused in sarcoma (FUS). Accordingly, FUS

Automation of On-Resin Enrichment of S-Nitrosylated Proteins for Oxidized Cysteine-Selective cPILOT.

S-Nitrosylation (SNO) is a cysteine post-translational modification that increases with normal aging and is present in Alzheimer's disease and other aging-related illnesses. Detection of SNO-modified proteins can be challenging; however, we previously developed a robust quantitative proteomics approach termed "Oxidized Cysteine-Selective combined precursor isobaric labeling and isobaric tagging (OxcyscPILOT)" that allows for detection of endogenous SNO-modified proteins. OxcyscPILOT involves enrichment of SNO-modified proteins using a thiol-based resin. This enrichment is performed manually, a

Upregulated levels and pathological aggregation of abnormally phosphorylated Tau-protein in children with neurodevelopmental disorders.

The tubulin-associated unit (Tau) protein is an intrinsically disordered protein that plays a well-established role in promoting microtubule assembly and regulation of microtubule dynamics in neuronal axons at all stages of development. Identification of new interacting partners and different sub-cellular localizations of Tau in recent years led to the discovery of novel physiological functions in regulation of neuronal activity, neurogenesis, long-term depression, iron export and genomic integrity. In addition, Tau gene mutations, aberrant mRNA splicing and abnormal post-translational modific

RNA polymerase I promoter and splice acceptor site recognition affect gene expression in non-pathogenic Leishmania species.

Leishmania (Sauroleishmania) tarentolae has biotechnological potential for use as live vaccine against visceral leishmaniasis and as a system for the over expression of eukaryotic proteins that possess accurate post-translational modifications. For both purposes, new systems for protein expression in this non-pathogenic protozoan are necessary. The ribosomal RNA promoter proved to be a stronger transcription driver since its use yielded increased levels of recombinant protein in organisms of both genera Trypanosoma or Leishmania. We have evaluated heterologous expression systems using vectors

A computational in silico approach to predict high-risk coding and non-coding SNPs of human PLCG1 gene.

PLCG1 gene is responsible for many T-cell lymphoma subtypes, including peripheral T-cell lymphoma (PTCL), angioimmunoblastic T-cell lymphoma (AITL), cutaneous T-cell lymphoma (CTCL), adult T-cell leukemia/lymphoma along with other diseases. Missense mutations of this gene have already been found in patients of CTCL and AITL. The non-synonymous single nucleotide polymorphisms (nsSNPs) can alter the protein structure as well as its functions. In this study, probable deleterious and disease-related nsSNPs in PLCG1 were identified using SIFT, PROVEAN, PolyPhen-2, PhD-SNP, Pmut, and SNPS&GO tools.

Seminal plasma AnnexinA2 protein is a relevant biomarker for stallions which require removal of seminal plasma for sperm survival upon refrigeration†.

Some stallions yield ejaculates that do not tolerate conservation by refrigeration prior to artificial insemination (AI), showing improvement after removal of most of the seminal plasma (SP) by centrifugation. In this study, the SP-proteome of 10 different stallions was defined through high-performance liquid chromatography with tandem mass spectrometry and bioinformatic analysis in relation to the ability of the ejaculates to maintain semen quality when cooled and stored at 5°C. Stallions were classified into three groups, depending on this ability: those maintaining good quality after direct

Comparative interactomes of HSF1 in stress and disease reveal a role for CTCF in HSF1-mediated gene regulation.

Heat shock transcription factor 1 (HSF1) orchestrates cellular stress protection by activating or repressing gene transcription in response to protein misfolding, oncogenic cell proliferation, and other environmental stresses. HSF1 is tightly regulated via intramolecular repressive interactions, post-translational modifications, and protein-protein interactions. How these HSF1 regulatory protein interactions are altered in response to acute and chronic stress is largely unknown. To elucidate the profile of HSF1 protein interactions under normal growth and chronic and acutely stressful conditio

Impact of Ser81 phosphorylation on alanine: glyoxylate aminotransferase associated with Primary hyperoxaluria type I.

Phosphorylation is a fundamental post-translational modification that contributes to the dynamic control of protein function and stability. More than 300,000 site-specific phosphorylation sites have been detected across > 20,000 human proteins, yet only a small fraction (~ 5%) have been experimentally characterized, and their roles in health and disease remain largely unexplored. In particular, the functional consequences of most phosphorylation events in metabolic enzymes remain unknown. Here, we investigated the functional impact of modifying Ser81 in alanine:glyoxylate aminotransferase (AGT

Edaravone attenuates carbonyl-stress-driven ApoA-I carboxymethylation and macrophage cholesterol-handling dysfunction in diabetic atherosclerosis.

Diabetes increases atherosclerotic risk, but how carbonyl stress impairs high-density lipoprotein (HDL) function remains unclear. We investigated a glyoxal-apolipoprotein A-I (ApoA-I) pathway and evaluated edaravone as a carbonyl-stress modulator. In a clinically stratified cohort, plasma protein-bound Nε-(carboxymethyl)lysine (CML) and glyoxal increased progressively across controls, diabetes without coronary artery disease, and diabetes with coronary artery disease. Under short-term exposure, glyoxal generated more ApoA-I-bound CML than other carbonyl species. Site-resolved liquid chromatogr

Succinylation modification: a potential therapeutic target in stroke.

Stroke is a leading cause of mortality and disability worldwide. Ischemic cell death triggered by the compromised supply of blood oxygen and glucose is one of the major pathophysiology of stroke-induced brain injury. Impaired mitochondrial energy metabolism is observed minutes after stroke and is closely associated with the progression of neuropathology. Recently, a new type of post-translational modification, known as lysine succinylation, has been recognized to play a significant role in mitochondrial energy metabolism after ischemia. However, the role of succinylation modification in cell m

S-Nitrosylation of Ras Mediates Nitric Oxide-Dependent Post-Injury Neurogenesis in a Seizure Model.

AIMS: Nitric oxide (NO) is involved in the upregulation of endogenous neurogenesis in the subventricular zone and in the hippocampus after injury. One of the main neurogenic pathways activated by NO is the extracellular signal-regulated kinase (ERK)/mitogen-activated protein kinase (MAPK) pathway, downstream of the epidermal growth factor receptor. However, the mechanism by which NO stimulates cell proliferation through activation of the ERK/MAPK pathway remains unknown, although p21Ras seems to be one of the earliest targets of NO. Here, we aimed at studying the possible neurogenic action of

Melon Seed Proteins: From Waste to Wellness-Functionality and Opportunities in Future Foods.

The global population growth and the transformation of dietary structure are intensifying the contradiction between protein supply and demand, so there is an urgent need to develop sustainable alternative protein sources. Melon seeds, as a major by-product of the melon processing industry, are estimated to have an annual recyclable volume of approximately 738,000 tons. They are often discarded, causing resource waste and environmental pressure. Due to its well-balanced amino acid profile, melon seed proteins are regarded as a valuable source of nontraditional proteins. Building upon and extend

Stathmin reduction and cytoskeleton rearrangement in rat nucleus accumbens in response to clozapine and risperidone treatment - Comparative proteomic study.

The complex network of anatomical connections of the nucleus accumbens (NAc) makes it an interface responsible for the selection and integration of cognitive and affective information to modulate appetitive or aversively motivated behaviour. There is evidence for NAc dysfunction in schizophrenia. NAc also seems to be important for antipsychotic drug action, but the biochemical characteristics of drug-induced alterations within NAc remain incompletely characterized. In this study, a comprehensive proteomic analysis was performed to describe the differences in the mechanisms of action of clozapi

BpSRK1-mediated phosphorylation of BpERF3 relieves HDA6-dependent chromatin repression to enhance drought tolerance in birch.

Drought tolerance in plants requires rapid conversion of water-deficit signals into protective transcriptional and epigenetic programs. Although ERF transcription factors (TFs) are central regulators of stress-responsive gene expression, how drought-induced kinase signaling is coupled to chromatin regulation and antioxidant gene activation remains unclear. Here, we identify the ERF TF BpERF3 as a phosphorylation-dependent regulator of drought tolerance in birch (Betula platyphylla). Using transgenic and CRISPR-Cas9 mutant lines, protein interaction assays, immunoaffinity fluorescent electropho

Epithelial SLC25A20 Deficiency Drives Immunosuppressive Neutrophil Remodeling via ARG1 Palmitoylation to Promote Colorectal Cancer.

While lipid metabolic reprogramming is increasingly recognized as a substantial driver of tumor microenvironment (TME) remodeling, how dysregulated fatty acid oxidation (FAO) contributes to immune evasion in colorectal cancer (CRC) progression remains obscure. Here, we identify the mitochondrial carnitine-acylcarnitine translocase solute carrier 25 member 20 (SLC25A20), an essential conduit for FAO, as a key regulator of CRC progression through neutrophil phenotypic remodel. Mechanistically, SLC25A20 deficiency in CRC cells blunts mitochondrial long-chain fatty acid (LCFA) import and FAO, trig

Arginine methylation as a regulatory ratchet in cancer: From substrate selection to malignant-state stabilization.

Arginine methylation, catalyzed by the protein arginine methyltransferase (PRMT) family, is a widespread post-translational modification yet its regulatory logic in cancer remains incompletely framed. Unlike rapidly reversible modifications, arginine methylation operates through a persistence-prone, forward-biased logic shaped by the lack of a broadly acting demethylase system, multisite substrates, and ongoing methylation flux. Here, we propose a regulatory ratchet framework in which PRMT-dependent methylation can reinforce cellular states through cumulative and network-level effects rather t

SMAD4 Palmitoylation Drives a Metabolic-Transcriptional Circuit to Promote Tumorigenesis and Confers Radiosensitivity in Pancreatic Cancer.

Pancreatic ductal adenocarcinoma (PDAC) is characterized by frequent SMAD4 inactivation and profound lipid metabolic rewiring, yet how these processes intersect especially in SMAD4+ PDAC remains elusive. Here, we identify palmitoylation as a previously unrecognized post-translational modification of SMAD4. Combining biochemical labeling, mutagenesis, and functional assays, we demonstrate that SMAD4 is palmitoylated at cysteine 363 by the acyltransferase ZDHHC22 and depalmitoylated by APT2. Mechanistically, palmitoylation enhances SMAD4 protein stability, facilitates the interaction of SMAD4 wi

NRF2 at the crossroads of Parkinson's disease and aging: Mechanistic insights and translational perspectives.

Parkinson's disease (PD) is a multifactorial neurodegenerative disorder characterized by dopaminergic neuronal loss, α-SYNUCLEIN aggregation, mitochondrial dysfunction, oxidative stress, and neuroinflammation. The transcription factor NRF2 (nuclear factor erythroid 2-related factor 2) orchestrates cellular defense mechanisms by controlling genes involved in antioxidant responses, detoxification, and proteostasis. Impaired NRF2 signaling in PD amplifies oxidative damage, protein misfolding, and inflammatory cascades, whereas NRF2 activation confers broad neuroprotection. This review summarizes

The Impact of Glycosylation on the Conformational Ensembles of β-, δ-, and γ Sarcoglycans.

Glycosylation is a pivotal post-translational modification that influences protein folding, stability, and interactions, with direct implications for muscular dystrophy pathogenesis and emerging gene therapies. Sarcoglycans (SGs), β-, δ-, γ-, and α-subunits of the dystrophin-glycoprotein complex (DGC), contain essential N-linked glycosylation sites, and mutations that disrupt glycan attachment, destabilize the complex and cause limb-girdle muscular dystrophy. Yet, the structural consequences of SG glycosylation remain poorly defined due to the absence of experimental sarcoglycan complex struct
