Prevalence of illicit drugs detected in the urine of women of childbearing age in Alabama public health clinics.

Each year, it is estimated that from 350,000 to 739,000 U.S. infants are exposed in utero to one or more illicit drugs. To estimate the prevalence of and risk factors for illicit drug use by women of childbearing age in Alabama, during 2 months in 1991 the authors collected patient-reported histories, clinical histories, and urine specimens from 6,195 women statewide attending public health maternity clinics, family planning clinics, and a high-risk referral obstetrical clinic. Blind drug screening of urine specimens for marijuana, cocaine, opiates, barbiturates, and amphetamines was performed

The acidic regions of WASp and N-WASP can synergize with CDC42Hs and Rac1 to induce filopodia and lamellipodia.

The acidic (A) region of WASp family proteins is thought to represent a high-affinity binding site for Arp2/3 complex without activating properties. Here we show that GST-fused WASp-A and N-WASP-A, but not a WASP-A/W500S mutant, several truncated WASp-A constructs or WAVE1-A can pull down Arp2/3 complex from cell lysates. Significantly, WASp-A and N-WASP-A synergistically trigger formation of filopodia or lamellipodia when coinjected with sub-effective concentrations of V12CDC42Hs or V12Rac1, respectively, into macrophages. The ability of WASp family A region constructs to induce this effect i

Genome-wide characterization of the wheat cryptochrome/photolyase family identifies TaCRY1b as a lamina-joint-associated protein interacting with TaLG2 and TaLG2L.

BACKGROUND: Cryptochromes (CRYs) are blue light and ultraviolet A (UV-A) photoreceptors that play pivotal roles in regulating plant development and stress responses through mediating light signaling pathways. However, the evolutionary and functional characteristics of the cryptochrome/photolyase (CRY/PHL) family in wheat (Triticum aestivum L.) remain poorly understood. RESULTS: 57 CRY/PHL genes were identified across eight species, including 14 members in hexaploid wheat. Phylogenetic analysis classified CRY/PHLs into four subfamilies. All TaCRY/PHL proteins contain the conserved photolyase h

Incorporation of low-oxidation graphene oxide into electrospun poly(ε-caprolactone) /gelatin scaffolds for human embryonic stem cell-derived retinal pigment epithelium cell transplantation.

Scaffold-based retinal pigment epithelium (RPE) transplantation is a candidate approach to preserve or recover vision for macular degeneration. Electrospun poly(ε-caprolactone) (PCL)/Gelatin (Gel) scaffolds have demonstrated support for cellular proliferation and function, yet their inadequate mechanical performance compromises surgical handling and deliverability. In this study, we incorporated low-oxidation graphene oxide (GO) into electrospun PCL/Gel scaffolds and evaluated their performance for human embryonic stem cell (hESC)-derived RPE cell transplantation. We found that the incorporati

A signal peptide-guided approach towards in situ functionalization of bacterial nanocellulose in Komagataeibacter rhaeticus.

BACKGROUND: Bacterial nanocellulose (BC), produced by Komagataeibacter species, is an ideal scaffold for biological Engineered Living Materials (bioELMs) research. Current BC functionalization strategies often rely on secondary microbial hosts or post-production enzyme immobilization, limiting the scalability and modularity required for programmable bioELMs. Establishing a single-chassis system capable of simultaneous biopolymer synthesis and in situ functionalization remains a primary objective in bioELM research. This study addresses the need by benchmarking signal peptide-mediated protein t

TBC1D14 positively regulates autophagy induced by Brucella melitensis vaccine strain BA0711 in Sheep Leydig Cells.

Brucella is an intracellular Gram-negative bacterium that primarily infects the host reproductive and immune systems, inducing autophagy and facilitating pathogen replication. TBC (Tre2-Bub2-Cdc16) domain-containing proteins are important in membrane trafficking, cell polarity, and signal transduction as regulators of Rab small GTPases. Previously, we demonstrated that B. melitensis M5-90 modulates the expression of miR-146b-5p, which targets TBC1D14, in RAW264.7 cells. In this study, CRISPR-Cas9 was used to generate TBC1D14-knockout (KO) Sheep Leydig cells (SLCs), and B. melitensis BA0711 tre

Milk-derived extracellular vesicles enhance oocyte competence and embryo development during bovine in vitro maturation.

Extracellular vesicles (EVs) have been explored as bioactive components across various fields, including tissue repair, cell culture systems, and therapeutic delivery. In reproductive biology, EVs are investigated as regulators of oocyte maturation and embryo development; however, the effects of milk-derived EVs on bovine oocyte maturation remain poorly understood. This study evaluated milk-derived EV supplementation during bovine in vitro maturation (IVM) on oocyte competence and subsequent embryonic development. Cumulus-oocyte complexes were matured in IVM medium supplemented with milk-deriv

Effects of combined exposure to polyethylene and oxidized polycyclic aromatic hydrocarbons on growth, development, and neurobehavior in Zebrafish.

Oxygenated polycyclic aromatic hydrocarbons (OPAHs) are a class of anthropogenic, persistent, and highly toxic PAH contaminants associated with developmental toxicity, 9-fluorenone (9-FLO) is a typical member of the OPAH family. Due to its ketone group, it has higher polarity, which results in increased solubility in water and greater potential for transport via atmospheric particles or water bodies. Polyethylene (PE), an amorphous polymer, is characterized by high diffusivity, high permeability, and a large internal molecular free volume, which confers a strong absorption capacity for organic

Rapid autofluorescence flow cytometric analysis of agonist-induced neutrophil and eosinophil polarization reveals novel insights into 5-oxo-ETE-mediated granulocyte activation.

Minimizing unintended granulocyte activation while measuring functional responsiveness is essential, as the use of external probes, antibodies, or fluorescent dyes can potentially alter cellular responsiveness. To address this, we employed an antibody-free flow cytometry approach that measures forward scatter (FSC) to detect variations in cell-size, morphology, and shape; some key indicators of neutrophil and eosinophil activation. Human peripheral blood neutrophils, containing contaminating eosinophils, were isolated using discontinuous Percoll gradients and pre-treated with receptor antagoni

Macrocyclic dihydropyridine analogs as pan-BET BD2-preferred inhibitors.

Starting from dihydropyridopyrimidine benzyl ester 1, we pursued a macrocyclization strategy by linking its two aryl rings, hypothesizing that decreasing the conformational flexibility of the ester side chain would increase bromodomain and extra-terminal (BET) protein affinity and selectivity. We prepared 14 analogs and tested them in our fluorescent polarization (FP) assay for BRDT-1 and BRD4-1 affinity. Based on their Ki values, we selected compounds 6b (BRDT-1 Ki = 1.05 μM and BRD4-1 Ki = 0.68 μM) and 6d (BRDT-1 Ki = 0.86 μM and BRD4-1 Ki = 0.70 μM) for further testing. Differential Scannin

Pursuing the holy grail of thermally activated delayed fluorescence emitters: a molecular strategy for reducing the energy gap and enhancing spin-orbit coupling.

The beauty and complexity of organic materials stem from the intricate interplay of their structural effects, where the same substituent can prove highly beneficial in one position yet entirely redundant in another. In this study, we demonstrate how a single halogen atom, strategically positioned, can significantly enhance the emissive properties of thermally activated delayed fluorescence (TADF) emitters. To address the inherent "low singlet-triplet energy gap (ΔE ST) equals low spin-orbit coupling (SOC)" tradeoff in donor-acceptor (DA) emitters, we propose leveraging heavy-atom substitution

IL-1β-Mediated Immunometabolic Adaptation in Corneal Epithelial Cells.

PURPOSE: As an external mucosal surface, the corneal epithelium is subject to a barrage of stressors that are known to trigger inflammation. IL-1β, a master regulator of inflammation, is secreted into the preocular tear film by ocular surface epithelial cells and infiltrating immune cells. While increased levels of IL-1β have been associated with corneal disease, the effects of IL-1β on mitochondrial function in corneal epithelial cells (CECs) is unknown. METHODS: To investigate the effects of IL-1β on mitochondrial function, telomerase immortalized human CECs were cultured in either 50 ng/mL

Cannabidiol Alleviates Neurological Deficits After Traumatic Brain Injury by Improving Intracranial Lymphatic Drainage.

Traumatic brain injury (TBI) persists as a substantial clinical dilemma, largely because of the absence of effective treatments. This challenge is exacerbated by the hindered clearance of intracranial metabolic byproducts and the continual accrual of deleterious proteins. The glymphatic system (GS) and meningeal lymphatic vessels (MLVs), key elements of the intracranial lymphatic network, play critical roles in the clearance of harmful substances. Cannabidiol (CBD) has shown promise in reducing metabolite overload and bolstering cognitive performance in various neurodegenerative diseases. The

Exploring oxidative stress and endothelial dysfunction as a mechanism linking bisphenol S exposure to vascular disease in human umbilical vein endothelial cells and a mouse model of postnatal exposure.

BACKGROUND: Structural analogues used to replace bisphenol A (BPA) since the introduction of new regulatory restrictions are considered emerging environmental toxicants and remain understudied with respect to their biological actions and health effects. Studies reveal a link between BPA exposure and vascular disease in human populations, whereas the vascular effects of BPA substitutes remain largely unknown. OBJECTIVES: To determine the effect of BPS, a commonly used BPA substitute, on redox balance, nitric oxide (NO) availability and microvascular NO-dependent dilation. METHODS: In human um

High-Fidelity Drug-Induced Liver Injury Screen Using Human Pluripotent Stem Cell-Derived Organoids.

BACKGROUND & AIMS: Preclinical identification of compounds at risk of causing drug induced liver injury (DILI) remains a significant challenge in drug development, highlighting a need for a predictive human system to study complicated DILI mechanism and susceptibility to individual drug. Here, we established a human liver organoid (HLO)-based screening model for analyzing DILI pathology at organoid resolution. METHODS: We first developed a reproducible method to generate HLO from storable foregut progenitors from pluripotent stem cell (PSC) lines with reproducible bile transport function. The

Self-assembly polymorphism of 2,7-bis-nonyloxy-9-fluorenone: solvent induced the diversity of intermolecular dipole-dipole interactions.

In this present work, a scanning tunneling microscope (STM) operated under ambient conditions was utilized to probe the self-assembly behavior of 2,7-bis-nonyloxy-9-fluorenone (F-OC9) at the liquid-solid (l/s) interface. On the highly oriented pyrolytic graphite (HOPG) surface, two-dimensional (2D) polymorphism with diversity of intermolecular dipole interactions induced by solvent was found. Solvents ranged from hydrophilic solvating properties with high polarity, such as viscous alkylated acids, to nonpolar alkylated aromatics and alkanes. 1-Octanol and dichloromethane were used to detect th

Pronounced interferon-alpha production from plasmacytoid dendritic cells in patients with Behçet's disease following CpG D ODN stimulation.

OBJECTIVE: Several lines of evidence point to a polarized T-helper-1 (Th1) immune response in Behçet's disease (BD). Interferon (IFN)-alpha which has an ability to promote strong Th1 type immune response has been shown to increase in patients with BD. In order to clarify if plasmacytoid dendritic cells (pDCs) abnormally respond to a stimulus in patients with BD, we investigated the levels of intracellular IFN-alpha and beta in pDCs with or without CpG D ODN stimulation. METHODS: The study population consisted of 8 patients with clinically active BD, 8 ankylosing spondilitis (AS) patients havi

Protoporphyrin-induced cholestasis in the isolated in situ perfused rat liver.

The pathogenesis of liver disease in protoporphyria has been presumed to result from the hepatic deposition of protoporphyrin. To examine the effects of protoporphyrin on hepatic bile flow and histopathology, studies were performed employing an isolated, in situ, rat liver perfusion system. Rat livers in the control group were perfused with 0-80 mumol sodium taurocholate/h. Rat livers in the experimental group were perfused with sodium taurocholate and (a) sufficient quantities of protoporphyrin to produce maximal canalicular secretion and (b) perfusate protoporphyrin concentrations of 0.01, 0

Distinct modes of migration position oligodendrocyte precursors for localized cell division in the developing spinal cord.

Establishment of the cytoarchitecture of the central nervous system reflects the stereotyped cell migration and proliferation of precursor cells during development. In vitro analyses have provided extensive information on the control of proliferation and differentiation of oligodendrocyte precursors (OPCs), but less is known about the migratory behavior of these cells in vivo. Here we utilize a transgenic mouse line expressing enhanced green fluorescent protein (EGFP) under the proteolipid protein promoter (PLP-EGFP mice) to visualize directly the behaviors of OPCs in developing spinal cord sl
