The mode of action of dimeticone 4% lotion against head lice, Pediculus capitis.

BACKGROUND: Treatment of head lice using physically acting preparations based on silicones is currently replacing insecticide use due to widespread resistance to neurotoxic agents. It has been postulated that some products act by asphyxiation, although the limited experimental evidence and the anatomy of the louse respiratory system suggest this is unlikely. RESULTS: Observation over several hours of lice treated using 4% high molecular weight dimeticone in a volatile silicone base showed that, although rapidly immobilised initially, the insects still exhibited small movements of extremities

Drivers for the emergence and re-emergence of vector-borne protozoal and bacterial diseases.

In recent years, vector-borne parasitic and bacterial diseases have emerged or re-emerged in many geographical regions causing global health and economic problems that involve humans, livestock, companion animals and wild life. The ecology and epidemiology of vector-borne diseases are affected by the interrelations between three major factors comprising the pathogen, the host (human, animal or vector) and the environment. Important drivers for the emergence and spread of vector-borne parasites include habitat changes, alterations in water storage and irrigation habits, atmospheric and climate

Abamectin resistance in Drosophila is related to increased expression of P-glycoprotein via the dEGFR and dAkt pathways.

Many insects have evolved resistance to abamectin but the mechanisms involved in this resistance have not been well characterized. P-glycoprotein (P-gp), an ATP-dependent drug-efflux pump transmembrane protein, may be involved in abamectin resistance. We investigated the role of P-gp in abamectin (ABM) resistance in Drosophila using an ABM-resistant strain developed in the laboratory. A toxicity assay, Western blotting analysis and a vanadate-sensitive ATPase activity assay all demonstrated the existence of a direct relationship between P-gp expression and ABM resistance in these flies. Our ob

A research agenda for malaria eradication: vector control.

Different challenges are presented by the variety of malaria transmission environments present in the world today. In each setting, improved control for reduction of morbidity is a necessary first step towards the long-range goal of malaria eradication and a priority for regions where the disease burden is high. For many geographic areas where transmission rates are low to moderate, sustained and well-managed application of currently available tools may be sufficient to achieve local elimination. The research needs for these areas will be to sustain and perhaps improve the effectiveness of cur

Assessment of molecular markers for anti-malarial drug resistance after the introduction and scale-up of malaria control interventions in western Kenya.

BACKGROUND: Although it is well known that drug pressure selects for drug-resistant parasites, the role of transmission reduction by insecticide-treated bed nets (ITNs) on drug resistance remains unclear. In this study, the drug resistance profile of current and previous first-line anti-malarials in Kenya was assessed within the context of drug policy change and scale-up of ITNs. National first-line treatment changed from chloroquine (CQ) to sulphadoxine-pyrimethamine (SP) in 1998 and to artemether-lumefantrine (AL) in 2004. ITN use was scaled-up in the Asembo, Gem and Karemo areas of western

Organophosphate and carbamate susceptibility profiling of Anopheles gambiae sl. across different ecosystems in southern Benin.

BACKGROUND: To overcome the spread of high pyrethroid resistance in the main malaria vectors and malaria disease persistence, it is crucial to look for effective and better resistance management strategies. Understanding the phenotypic profile of Anopheles gambiae sl. against alternatives insecticides like organophosphates and carbamates is crucial. METHODS: Anopheles larvae and pupae were collected from the breeding sites in rice fields, pineapple crop areas, and peri-urban areas. WHO susceptibility tests were conducted on unfed female mosquitoes aged 3-5 days old. Mosquitoes were exposed to

Fine-scale spatio-temporal dynamics of kdr mutation V1016G in Aedes albopictus along a transect encompassing the Rome metropolitan area.

Pyrethroid insecticides are largely exploited in Italy to reduce Aedes albopictus nuisance and to discontinue autochthonous transmission of exotic arboviruses. V1016G mutation in the voltage-gated sodium channel gene is strongly associated with resistance to pyrethroids in this species. Here, we provide an unprecedented high-resolution spatiotemporal analysis of the distribution/frequency of the V1016G mutation over an 11-year period in Ae. albopictus populations sampled across a transect encompassing the eco-geographical variability of Mediterranean Europe, including Rome, the only European m

Resistance evolution to basil essential oil: how selection pressure shapes tolerance and fitness costs in Aphis craccivora.

Essential oils (EOs) are increasingly proposed as sustainable alternatives to synthetic insecticides, yet their long-term evolutionary effects remain poorly understood. This study examined how two contrasting selection regimes with basil essential oil (Ocimum basilicum; estragole-rich chemotype) selection regimes, a stepwise escalation regime (+ 15% per generation) and a repeated constant-exposure regime with stabilisation phases at F7-F8 and F14-F15, influenced tolerance evolution and associated fitness costs over 15 generations. Stepwise selection led to limited tolerance (nymphs RRF10/F0 =

Ectoparasite management in extensively grazed sheep and cattle in New Zealand: Current strategies and future innovations.

Ectoparasites can negatively affect animal health and welfare and incur financial cost to farmers due to losses in animal productivity and performance, and the cost of chemicals and labour used to prevent and/or treat infestations. Therefore, effective management and control are needed to ensure good animal health, welfare and performance. A range of tools are currently available to manage ectoparasites including insecticides, genetics and integrated management strategies. However, emerging challenges such as climate change and chemical resistance are likely to influence how we manage ectopara

Synergistic enhancement of emamectin benzoate and Metarhizium anisopliae against late-instar Spodoptera frugiperda by promoting fungal pathogenicity.

Spodoptera frugiperda, a globally invasive pest, has developed severe resistance to chemical insecticides, necessitating sustainable control strategies. This study investigated the synergistic interaction between emamectin benzoate (EB) and the entomopathogenic fungus Metarhizium anisopliae CQMa421 against fourth-instar larvae. Bioassays revealed a synergistic ratio (SR) of 4.19 when EB was combined with CQMa421 spores (2 × 107 spores/mL), reducing EB usage by 76.1 %. CQMa421 did not cause significant effects on S. frugiperda, whereas emamectin benzoate (EB) significantly disrupted feeding, de

Evaluation of pepper cultivars with different heat units on the feeding preference of pepper weevil (Coleoptera: Curculionidae).

Pepper weevil, Anthonomus eugenii Cano, is a devastating pest of pepper that is typically controlled, to varying degrees, with chemical insecticides. Additional tools are needed to manage this pest, including host plant resistance. Field and laboratory studies were conducted over 4 yr to determine pepper weevil preferences for pepper hosts with different Scoville heat units (SHU): bell (0 SHU), jalapeño (2,500 to 8,000 SHU), habanero (100,000 to 350,000 SHU), ghost (855,000 to 1,041,427 SHU), and scorpion (1,200,000 to 2,000,000 SHU) peppers. In field trials, jalapeño had the highest number of

Evaluation of Yellow Fever Virus Infection in Aedes aegypti Mosquitoes from Pakistan with Distinct Knockdown Resistance Genotypes.

BACKGROUND: Yellow fever (YF) is an acute hemorrhagic disease endemic to Africa and Latin America; however, no cases have been reported in Asian regions with high Aedes aegypti infestation. Factors such as environmental conditions and genetic variations in the yellow fever virus (YFV) strains and mosquito populations may explain this absence. Mosquito populations have undergone strong selective pressure owing to the excessive use of insecticides. This pressure has led to the spread of alterations, such as knockdown-resistant mutations (kdr), which, while conferring resistance to pyrethroids, a

Larvicidal Activity of Carbon Black against the Yellow Fever Mosquito Aedes aegypti.

The yellow fever mosquito Aedes aegypti is one of the deadliest animals on the planet because it transmits several medically important arboviruses, including Zika, chikungunya, dengue, and yellow fever. Carbon-based nanoparticles (CNPs) derived from natural sources have previously been shown to have toxic effects on mosquito larvae and offer a potential alternative to chemical insecticides such as pyrethroids, for which mosquitoes have evolved resistance. However, CNPs derived from industrial sources, such as carbon black, have not previously been evaluated as larvicides. Here, we evaluate the

Trypanotolerance in cattle and prospects for the control of trypanosomiasis by selective breeding.

Tsetse-transmitted trypanosomiasis is one of the major constraints on the expansion of the livestock and agricultural industries in Africa. The disease affects animals and man, with direct and indirect losses estimated in billions of dollars annually. Because of the phenomenon of antigenic variation, no vaccine is available. Current prophylactic efforts must rely on tsetse control by the use of insecticides and on trypanocidal drugs. However, recent advances in our knowledge of tsetse and trypanosome biology are offering hope for alternative methods of trapping tsetse, new drugs and even vacci

A costing analysis of an intermittent biolarviciding intervention with limited epidemiological effect in Tanga Region, Tanzania.

BACKGROUND: Malaria vector control strategies in sub-Saharan Africa have largely relied on the use of Insecticide Treated-mosquito Nets and Indoor Residual Spraying. However, after substantial reductions in malaria burden associated with the scale-up of these interventions, little progress has been achieved over the last decade. With the increase in insecticide resistance and outdoor biting of mosquitoes, larviciding offers the possibility to be an additional malaria control tool. This study is part of a comprehensive evaluation and aimed to assess prospectively the financial cost of a large-s…
