Genome mapping coupled with CRISPR gene editing reveals a P450 gene confers avermectin resistance in the beet armyworm.

The evolution of insecticide resistance represents a global constraint to agricultural production. Because of the extreme genetic diversity … avermectins recently evolved in the beet armyworm, threatening the sustainable use of this class of insecticides. Here, we report a high-quality chromosome-level assembly of the beet armyworm genome and use bulked segregant analysis (BSA) to identify the locus of avermectin resistance, which mapped on 15-16 Mbp of chromosome 17. Knockout of the CYP9A186 gene t …

Discovery of diamide derivatives bearing a flexible substituted benzylamine moiety as potential ryanodine receptor activator.

BACKGROUND: Lepidopteran pests have developed severe resistance to diamide insecticides, posing a huge challenge to effective pest control, necessitating the development of novel insecticides to achieve the effective comprehensive management of pests. Forty-two novel derivatives bearing substituted benzylamine moieties with chlorantraniliprole (CHL) as the lead compound were precisely design, synthesized, and evaluated for insecticidal activities against three Lepidopteran pests. RESULTS: The bioassay results showed that the LC50 values of compounds A26 and A40 against Mythimna separata were …

A whole transcriptomic approach provides novel insights into the molecular basis of organophosphate and pyrethroid resistance in Anopheles arabiensis from Ethiopia.

The development of insecticide resistance in malaria vectors is of increasing concern in Ethiopia because of its potential implications for … tance phenotype by either enhancing the cuticular barrier or promoting binding and sequestration of insecticides, respectively. These findings provide novel insights into the molecular basis of insecticide resistance in this lesser well-characterized major malaria vector species.

Mixture of neonicotinoid and fungicide affects foraging activity of honeybees.

The use of plant protection products (PPPs) is a major factor contributing to global insect decline. We here use the honeybee (Apis mellifer … he fungicide/insecticide mixture supports the notion that some fungicides can enhance the effect of insecticides, which did not lead to significant changes in behaviour when applied on their own. Our results emphasize the need for more studies on the interaction of different PPPs.

Flavin-Dependent Monooxgenase Confers Resistance to Chlorantraniliprole and Spinetoram in the Rice Stem Borer Chilo suppressalis Walker (Lepidoptera: Crambidae).

Understanding the role of flavin-containing monooxygenases (FMOs) in the genetic mechanisms of insecticide resistance is essential for developing effective management strategies against the rice stem borer, Chilo suppressalis. In this study, we identified five FMO genes in C. suppressalis, examined their expression patterns, and revealed overexpression of FMO3B and FMO3C in field populations resistant to multiple insecticides, including chlorantraniliprole and spinetoram. Functional characterization using transgenic Drosophila indicated that FMO3B and FMO3C do not confer resistance to abamecti…

Blackfly control: what choices after onchocerciasis?

Blackflies are reappearing in areas of West Africa where they used to be controlled with insecticides because they were vectors of the parasite Onchocerca volvulus. Even though they no longer transmit onchocerciasis in these areas they can hinder optimal land use through their biting behaviour. The authors discuss the problems associated with resuming the use of insecticides to control the blackfly and recommend that ground treatment be restricted to areas where it is likely to be effective on a continuing basis. In communities lacking technical and financial resources the only alternative con…

Targeting the GABA receptor for agricultural Pest control: from molecular mechanisms to novel NAM site inhibitors.

The ionotropic γ-aminobutyric acid receptor (GABAR) is a major inhibitory neurotransmitter receptor in the insect central nervous system and a proven target for multiple classes of insecticides. The recent discovery of the NAM binding site located at the transmembrane subunit interface has revitalized interest in this classic target, leading to the development of two novel classes of insecticides-isoxazolines and meta-diamides-that offer potent efficacy and lack cross-resistance with conventional agents. This review provides a comprehensive overview of the molecular basis of insect GABARs, inc…

Toxicity of copper (II) metal complexes with amino acids ligands: A study of activity in Aedes aegypti larvae.

The larval control of Aedes aegypti has been done mainly by insecticides, which cause resistance and environmental contamination and have a high-cost development and application. The need for alternative solutions to problems caused by synthetic insecticides has led to studies to find new, active, and less toxic compounds, such as metallo-insecticides. In the present study, the toxic activity of Cu(II) - complexes with the amino acids tryptophan, histidine, methionine, and taurine in Ae. aegypti larvae were evaluated. The Cu(II) - histidine complex showed no toxicity. For the Cu(II) - tryptoph…

miRNAs: A potentially valuable tool in pesticide toxicology assessment-current experimental and epidemiological data review.

miRNAs are responsible for the regulation of many cellular processes such as development, cell differentiation, proliferation, apoptosis, and tumor growth. Several studies showed that they can also serve as specific, stable, and sensitive markers of chemical exposure. In this review, current experimental and epidemiological data evidencing deregulation in miRNA expression in response to fungicides, insecticides or herbicides were analyzed. As shown by Venn's diagrams, miR-363 and miR-9 deregulation is associated with fungicide exposure in vitro and in vivo, while let-7, miR-155, miR-181 and mi…

Resistance to pirimiphos-methyl in West African Anopheles is spreading via duplication and introgression of the Ace1 locus.

Vector population control using insecticides is a key element of current strategies to prevent malaria transmission in Africa. The introduction of effective insecticides, such as the organophosphate pirimiphos-methyl, is essential to overcome the recurrent emergence of resistance driven by the highly diverse Anopheles genomes. Here, we use a population genomic approach to investigate the basis of pirimiphos-methyl resistance in the major malaria vectors Anopheles gambiae and A. coluzzii. A combination of copy number variation and a single non-synonymous substitution in the acetylcholinesterase…

Potential sources of Triatoma infestans reinfesting peridomiciles identified by morphological characterization in Los Llanos, La Rioja, Argentina.

The presence of Triatoma infestans in habitats treated with insecticides constitutes a frequent problem in endemic areas. Basing our study on the hypothesis that descendants of a residual population should be more similar to the pre-treatment population than to any other, we compared the indications of two quantitative morphological approaches. This study seeks to find the origin of 247 T. infestans from three populations found in two chicken coops and a goat corral after treatment with insecticides. The results obtained by quantitative morphology suggest that the T. infestans found between th…

Susceptibility of head lice (Pediculus humanus capitis) to pediculicides in Australia.

Infestation with head lice, Pediculus humanus capitis, is a worldwide problem, especially among primary (elementary) school children. Although studies in many different countries indicate lower levels of susceptibility to certain insecticides than expected ("resistance"), there is no empirical data from Australia. Data on the susceptibility of head lice to malathion, pyrethrums and permethrin were collected from four schools in Brisbane and one school in northern Queensland. Since no completely susceptible strain of head lice was available and head lice are difficult to keep in culture, a comp…

Comparative analyses of the effects of sublethal doses of emamectin benzoate and tetrachlorantraniliprole on the gut microbiota of Spodoptera frugiperda (Lepidoptera: Noctuidae).

Spodoptera frugiperda (J. E. Smith) is an important invasive pest that poses a serious threat to global crop production. Both emamectin benzoate (EB) and diamide insecticides are effective insecticides used to protect against S. frugiperda. Here, 16S rRNA sequencing was used to characterize the gut microbiota in S. frugiperda larvae exposed to EB or tetrachlorantraniliprole (TE). Firmicutes and Proteobacteria were found to be the dominant bacterial phyla present in the intestines of S. frugiperda. Following insecticide treatment, larvae were enriched for species involved in the process of inse…

Comparison of techniques for detection of domestic infestations with Triatoma infestans in Brazil.

The detection of infestations of Triatominae in houses in regions endemic for Chagas's disease is a criterion frequently used in the selecti … for a long time, this also limits the value of the information obtained regarding the evaluation of insecticides. On the basis of the results presented, the flushing-out method using a systematic search of the house was found to be the most suitable sampling technique, but it is recognized that this method still has limitations.

Pesticides related to land use in watersheds of the Great Lakes basin.

In this study, we evaluated the distribution and concentrations of a range of neonicotinoid insecticides (NNIs) and other insecticides, fungicides, biocides and selected herbicides in watersheds that drain into the lower Great Lakes in Ontario, Canada. Polar Organic Chemical Integrative Samplers (POCIS) were deployed in 18 watersheds during late May to late June of 2016. Grab samples were also collected in 7 of these watersheds. There was generally good agreement between the time-weighted average concentrations of pesticides estimated from the POCIS and the concentrations detected in grab samp…

Concentrations and loads of suspended sediment-associated pesticides in the San Joaquin River, California and tributaries during storm events.

Current-use pesticides associated with suspended sediments were measured in the San Joaquin River, California and its tributaries during two storm events in 2008. Nineteen pesticides were detected: eight herbicides, nine insecticides, one fungicide and one insecticide synergist. Concentrations for the herbicides (0.1 to 3,000 ng/g; median of 6.1 ng/g) were generally greater than those for the insecticides (0.2 to 51 ng/g; median of 1.5 ng/g). Concentrations in the tributaries were usually greater than in the mainstem San Joaquin River and the west side tributaries were higher than the east sid…

The logic of visceral leishmaniasis control.

Mathematical models are used to compare the effectiveness of various untested, unused, and undeveloped methods for controlling canine and human zoonotic visceral leishmaniasis (ZVL), including insecticides, vaccines, killing serologically positive and sick dogs, and drugs. For given percentage changes in control parameters, insecticides are the most effective control method. Where transmission occurs peridomestically and vectors are accessible to treatment, as in parts of tropical America, insecticides are expected to reduce the incidence of human ZVL even more effectively than they reduce the…
