Tremor entrainment by patterned low-frequency stimulation.

High-frequency test stimulation for tremor suppression is a standard procedure for functional target localization during deep brain stimulation. This method does not work in cases where tremor vanishes intraoperatively, for example, due to general anaesthesia or due to an insertional effect. To overcome this difficulty, we developed a stimulation technique that effectively evokes tremor in a well-defined and quantifiable manner. For this, we used patterned low-frequency stimulation (PLFS), i.e. brief high-frequency pulse trains administered at pulse rates similar to neurons' preferred burst fr

Advances in risk prediction models for cancer-related cognitive impairment.

Cancer-related cognitive impairment (CRCI) has emerged as a significant long-term complication in cancer survivors, particularly those undergoing chemotherapy, radiotherapy, or targeted therapies. Despite advances in treatment, CRCI affects patients' quality of life, impacting their daily functioning, work capacity, and psychological well-being. In recent years, research has focused on identifying predictive factors for CRCI and developing risk prediction models to facilitate early intervention. This review summarizes the latest progress in CRCI risk prediction models, including traditional st

Fast, flexible analysis of differences in cellular composition with crumblr.

Changes in cell type composition play an important role in human health and disease. Recent advances in single-cell technology have enabled the measurement of cell type composition at increasing cell lineage resolution across large cohorts of individuals. Yet this raises new challenges for statistical analysis of these compositional data to identify changes in cell type frequency. We introduce crumblr (DiseaseNeurogenomics.github.io/crumblr), a scalable statistical method for analyzing count ratio data using precision-weighted linear mixed models incorporating random effects for complex study

Challenges in Analyzing Functional Epigenetic Data in Perspective of Adolescent Psychiatric Health.

The formative period of adolescence plays a crucial role in the development of skills and abilities for adulthood. Adolescents who are affected by mental health conditions are at risk of suicide and social and academic impairments. Gene-environment complementary contributions to the molecular mechanisms involved in psychiatric disorders have emphasized the need to analyze epigenetic marks such as DNA methylation (DNAm) and non-coding RNAs. However, the large and diverse bioinformatic and statistical methods, referring to the confounders of the statistical models, application of multiple-testin

Matching adolescents with a cannabis use disorder to multidimensional family therapy or cognitive behavioral therapy: treatment effect moderators in a randomized controlled trial.

BACKGROUND: In a recent randomized controlled trial (Hendriks et al., 2011), multidimensional family therapy (MDFT) and cognitive behavioral therapy (CBT) were equally effective in reducing cannabis use in adolescents (13-18 years old) with a cannabis use disorder (n=109). In a secondary analysis of the trial data, we investigated which pretreatment patient characteristics differentially predicted treatment effect in MDFT and CBT, in order to generate hypotheses for future patient-treatment matching. METHODS: The predictive value of twenty patient characteristics, in the area of demographic b

Artificial intelligence-powered models in predicting mortality in maternal, newborn, and children under five: a systematic review protocol.

BACKGROUND: Maternal, newborn, and under-five mortality remain significant global health challenges, with 287,000 maternal deaths reported in 2020 and 4.9 million under-five deaths in 2022. The use of artificial intelligence in prediction models has shown great potential in predicting these mortalities. Artificial intelligence (AI)-powered models are capable of processing complex datasets to identify non-linear relationships and patterns, potentially outperforming traditional statistical methods in predictive accuracy, thereby enabling early detections and interventions more specifically. Antibacterial activity of Limonium brasiliense (Baicuru) against multidrug-resistant bacteria using a statistical mixture design.

ETHNOPHARMACOLOGICAL RELEVANCE: Limonium brasiliense (Boiss.) Kuntze (Plumbaginaceae) is commonly known as "baicuru" or "guaicuru" and preparations of its dried rhizomes have been popularly used in the treatment of premenstrual syndrome and menstrual disorder, and as an antiseptic in genito-urinary infections. This study evaluated the potential antibacterial activity of rhizome extracts against multidrug-resistant bacterial strains using statistical mixture design. MATERIALS AND METHODS: The statistical design of four components (water, methanol, acetone, and ethanol) produced 15 different ex

Linear aspects of transformation from interictal epileptic discharges to BOLD fMRI signals in an animal model of occipital epilepsy.

Epileptic disorders manifest with seizures and interictal epileptic discharges (IEDs). The hemodynamic changes that accompany IEDs are poorly understood and may be critical for understanding epileptogenesis. Despite a known linear coupling of the neurovascular elements in normal brain tissues, previous simultaneous electroencephalography (EEG)-functional magnetic resonance imaging (fMRI) studies have shown variable correlations between epileptic discharges and blood oxygenation level-dependent (BOLD) response, partly because most previous studies assumed particular hemodynamic properties in no

Visual recognition limitations in multimodal large language models: A comparative analysis of histological image interpretation.

Multimodal large language models (LLMs) with image recognition capabilities have emerged as potential tools for medical image analysis, yet their performance in specialized domains like histology remains largely unexplored. The objective of this study was to systematically evaluate the performance of leading multimodal LLMs in histological image interpretation and assess their visual recognition capabilities. Four multimodal LLMs (GPT-4o, Claude Sonnet 4, Gemini 2.5 Flash, and Copilot) were evaluated using 144 histological images representing four tissue types (epithelial, connective, muscle,

Development and validation of machine learning predictive models for assessing dialysis adequacy in dialysis patients.

PURPOSE: The assessment of dialysis adequacy is of great clinical importance. However, it depends on the nonlinear effects of numerous confounding factors and is therefore difficult to predict using traditional statistical methods. In this study, we used Random Forest (RF), Extreme Gradient Boosting (XGBoost), and Least Absolute Shrinkage and Selection Operator Regression (LASSO) to assess dialysis adequacy. METHODS: A training set (70%) and a test set (30%) were randomly selected from the 264 dialysis patient case records collected for this study. We compared the machine learning models with

Mixed effects models but not t-tests or linear regression detect progression of apathy in Parkinson's disease over seven years in a cohort: a comparative analysis.

INTRODUCTION: While there is an interest in defining longitudinal change in people with chronic illness like Parkinson's disease (PD), statistical analysis of longitudinal data is not straightforward for clinical researchers. Here, we aim to demonstrate how the choice of statistical method may influence research outcomes, (e.g., progression in apathy), specifically the size of longitudinal effect estimates, in a cohort. METHODS: In this retrospective longitudinal analysis of 802 people with typical Parkinson's disease in the Luxembourg Parkinson's study, we compared the mean apathy scores at

Differential Performance of Machine Learning Models in Prediction of Procedure-Specific Outcomes.

BACKGROUND: Procedure-specific complications can have devastating consequences. Machine learning-based tools have the potential to outperform traditional statistical modeling in predicting their risk and guiding decision-making. We sought to develop and compare deep neural network (NN) models, a type of machine learning, to logistic regression (LR) for predicting anastomotic leak after colectomy, bile leak after hepatectomy, and pancreatic fistula after pancreaticoduodenectomy (PD). METHODS: The colectomy, hepatectomy, and PD National Surgical Quality Improvement Program (NSQIP) databases wer

A benchmark study of vision and pathology foundation models for computational pathology.

To advance precision medicine in pathology, artificial intelligence (AI)-driven foundation models must generalize across diverse datasets, tissues, and clinical tasks. However, their comparative performance and generalizability in computational pathology remain incompletely characterized. Here, we benchmark 32 AI foundation models across four categories, including general vision models (VM), general vision-language models (VLM), pathology-specific vision models (Path-VM), and pathology-specific vision-language models (Path-VLM), using slide- and patch-level tasks from The Cancer Genome Atlas (

Evaluating methods for testing publication bias in meta-analyses with confounding covariates: A simulation study.

Selective publication of favorable results can bias meta-analytic estimates and threaten the validity of meta-analyses. Many methods that detect and adjust for publication bias perform poorly under substantial between-study heterogeneity. Heterogeneity complicates publication bias assessment because study characteristics may be associated with both effect size estimates and standard errors. These characteristics can induce an effect-precision association that regression-based diagnostics may misattribute to selective publication when such characteristics are omitted. However, it remains unclea

Frequency Band Analysis of Multiple Stationary Time Series.

The frequency-domain properties of biomedical signals offer valuable insights into health and functioning of underlying physiological systems. The power spectrum, which characterizes these properties, is often summarized by partitioning frequencies into standard bands and averaging power within bands. These summary measures are regularly used for analysis in practice, but are not guaranteed to optimally retain differences in power spectra across signals from different participants. We propose a data-adaptive method for identifying frequency band summary measures that preserve spectral variabil

Nothing About Us Without Us: Centering Lived Experience as the AMPD Moves Forward.

The alternative model for personality disorders (AMPD) is approaching readiness for inclusion in the main section of the DSM, yet research informing its development has largely overlooked the perspectives of those who receive personality disorder diagnoses. This commentary argues that centering lived experience is essential for evaluating the AMPD's diagnostic language, given evidence that certain trait terminology (notably "manipulativeness") is perceived as highly stigmatizing. The AMPD's Criterion A, which emphasizes personality functioning rather than dispositional traits, may offer a less

A Neutral Comparison of Multivariate Hypothesis Tests for Parallel Group Designs in Rare Disease Settings.

Multivariate endpoints are increasingly used in clinical studies on rare diseases, where sample sizes tend to be small and distributional assumptions are often violated. In such settings, commonly applied parametric multivariate methods, such as classical MANOVA approaches, may suffer from inflated type-I error or reduced statistical power. Therefore, a neutral simulation-based comparison of four methodologically different approaches for the analysis of multivariate outcomes in parallel group designs was conducted. The four approaches included the nonparametric R package npmv (using permutatio

Effects of the Minimum Number of Usable Time Points in Bootstrapped Data Sets on f 2 Confidence Interval Using Bootstrap Methodology for the Comparison of Dissolution Profiles.

To use the conventional f 2 method to compare dissolution profiles, at least 3 time points should be included; however, when the bootstrap f 2 method is applied, there are no clear criteria in regulatory guidelines in this regard. The effects of applying the minimum number of time points on the accuracy, precision, type I error (TIE), and statistical power have been investigated in the current study. Four scenarios were simulated where the reference product (R) dissolves at different rates. For each scenario, 17 population dissolution profiles of the test product (T) were simulated with pred

A Bayesian Adaptive Design for Assessing Treatment Effect Consistency in Bridging Studies.

Bridging studies serve as an efficient approach to evaluate the applicability of findings from an original study to a new population. We propose a Bayesian adaptive design for bridging studies with time-to-event endpoints, aiming to assess the consistency of treatment effects with those observed in the original study. The design adopts a group sequential framework, using posterior probabilities at interim and final analyses to evaluate the evidence for consistency between the bridging and original studies. If strong evidence of consistency or inconsistency is observed, the trial may be stopped
