Regional difference in correlation of 5-HT4 receptor distribution with cholinergic transmission in the guinea pig stomach.

Localization and function of 5-HT4 receptors in the stomach were examined in mucosa-free preparations of antrum, corpus and fundus from guin … hyl-8-amino-7-chloro-1,4-benzodioxane-5-carboxylate. Thus, 5-HT4 receptors are located on myenteric cholinergic neurons in the antrum and corpus of guinea pig stomach and their activation evokes the release of acetylcholine.

Neuroprotective effect of rasagiline, a selective monoamine oxidase-B inhibitor, against closed head injury in the mouse.

UNLABELLED: The potential neuroprotective effects of rasagiline, N-propargyl-1R-aminoindan, a selective monoamine oxidase-B inhibitor and its inactive enantiomer TVP 1022, N-propargyl-1S-aminoindan were assessed against the sequelae of closed head injury in the mouse. Injury was induced in the left hemisphere under ether anaesthesia. Rasagiline (0.2 and 1 mg/kg) or TVP1022 (1 and 2 mg/kg) injected 5 min after injury accelerated the recovery of motor function and spatial memory and reduced the cerebral oedema by about 40-50%, (P < 0.01). The neuroprotective effects on motor function and spatial

Effects of substance P on gastric motility differ depending on the sites and vagal innervation in conscious dogs.

The effect of substance P on gastric motility was studied in conscious dogs by means of strain gauge force transducers chronically implanted on the gastric body, antrum, and a vagally-denervated fundic pouch. Intravenous infusion of substance P in the interdigestive state induced phasic contractions in the pouch and antrum. Atropine inhibited these contractions in the pouch and antrum. Hexamethonium enhanced substance P-induced contractions in the gastric antrum, but reduced those in the pouch. Pretreatment with phentolamine, propranolol, or naloxone did not affect substance P-induced contract

[Effect and its mechanism of substance P on contractile activity of isolated antral muscle strips of rat].

The present study was aimed at investigating the effect of substance P (SP) on the contractile activity of isolated antral muscle strips of rat and its underlying mechanism. Isolated strips were incubated in an organ bath into which SP was added with or without pretreatment of some antagonists or inhibitors. The results were as follows: (1) SP increased the contractile amplitude of the strips in a dose-dependent manner from 8 x 10(-11) to 8 x 10(-7) mol. At 4 x 10(-8) mol the amplitude was increased by 160.9 +/- 23.0%, while the automaticity of the strips was not affected. (2) This effect of S

High-affinity transport of choline and amino acid neurotransmitters in synaptosomes from brain regions after lesioning the nucleus basalis magnocellularis of young and aged rats.

Bilateral lesion of the nucleus basalis with ibotenic acid infusions in young and aged rats results in the degeneration of cholinergic neurons which innervate the cortex. As expected, high-affinity uptake of choline was decreased in the frontal cortex subsequent to the lesion. Twenty one days after surgery there was a significantly decrease of the transport rate of GABA, glutamate and glycine in the frontal cortex of young rats, but those activities showed a recovery six months after lesion. On the contrary, 12-month old rats lesioned with the same experimental protocol showed no recovery of t…

Genetic linkage analysis of nerve growth factor (beta) in familial Alzheimer's disease.

Recent studies have not shown linkage of late-onset (mean age, greater than 60 years) familial Alzheimer's disease (FAD) to the chromosome 21 locus reported linked to early-onset FAD. Beta nerve growth factor (beta-NGF) has been considered a candidate gene in the pathogenesis and therapy of FAD, based on its localization in the cortex and hippocampus and its ability to enhance the growth and survival of cholinergic neurons. A 1.5-kb fragment of the beta-NGF gene was used to detect a BglII restriction fragment length polymorphism, which was then used for linkage analysis. A total of 30 families…

Distribution of acetylcholinesterase in the limbic system of the brain of the rabbit.

The aim of the study is to present a detailed and thorough description of the acetylcholinesterase enzyme (AChE) content of the limbic structures of the rabbit, now commonly used in our laboratory for investigating the electrographic-behavioral correlations of the cholinergic limbic system. We use 'chemitrodes' for intracerebral mi croinjections of cholinomimetic drugs; a detailed histochemical study (using Koelle's method) of the distributions of AChE enzyme in the limbic system of the rabbit was deemed necessary. Such areas as the septum and the amygdala are particularly poorly described in

Exposing immature hippocampal neurons to excitotoxins reveals distinct transcriptome and protein regulation with induction of common survival signaling pathways.

Early life traumas lead to neuroprotection by preconditioning mechanisms. To determine which genes and pathways are most likely involved in specific adaptive effects, immature hippocampal cultures were exposed to a single high dose of glutamate (250 μM), NMDA (100 μM), or KA (300 μM) for 48 h (5-7 DIV) based on our prior "two hit" in vitro model of preconditioning. Transcriptome profiling and immunocytochemistry of gene candidates were performed 7 days later when cultured neurons mature (14 DIV). Many genes were up- and down- regulated involving distinct Ca2+-binding protein families, G-couple

GC-B Deficient Mice With Axon Bifurcation Loss Exhibit Compromised Auditory Processing.

Sensory axon T-like branching (bifurcation) in neurons from dorsal root ganglia and cranial sensory ganglia depends on the molecular signaling cascade involving the secreted factor C-type natriuretic peptide, the natriuretic peptide receptor guanylyl cyclase B (GC-B; also known as Npr2) and cGMP-dependent protein kinase I (cGKI, also known as PKGI). The bifurcation of cranial nerves is suggested to be important for information processing by second-order neurons in the hindbrain or spinal cord. Indeed, mice with a spontaneous GC-B loss of function mutation (Npr2cn/cn ) display an impaired bifur

The Enteric Nervous System: An Integrative Hub From Development to Systemic Physiology.

The enteric nervous system (ENS) constitutes the largest and most functionally autonomous subdivision of the peripheral nervous system in vertebrates. It orchestrates stereotyped motor programs, secretomotor reflexes, and neuroimmune responses without central input. This review synthesizes evidence from developmental biology, mucosal immunology, and comparative physiology to advance a framework of enteric allostatic plasticity: the proposition that ENS functional resilience arises not from hard-wired reflex arcs but from a conserved capacity for transcriptional and morphological remodeling ins

Neuronal regulation of myenteric interstitial cells of Cajal (ICC-MY) in the proximal colon.

Interstitial cells of Cajal (ICC) generate contractile patterns of colonic motility. We investigated innervation of ICC within the plane of the myenteric plexus (ICC-MY) in proximal colon using mice expressing GCaMP6f in ICC. ICC-MY generated localized Ca2+ transients that couple to activation of ANO1 channels, a Ca2+-activated Cl- conductance. ICC are electrically coupled to SMCs, so activation or suppression of currents in ICC affects excitability of SMCs. ICC-MY displayed tonic inhibition, as the neurotoxin, TTX, increased the frequency of Ca2+ transients. Tonic inhibition was mimicked by a

Metalloproteinase-9 in the ocular surface of patients with implanted Boston type 1 keratoprosthesis.

In Huntington's disease (HD), a hereditary neurodegenerative disorder, striatal medium-sized spiny neurons undergo degenerative changes. In contrast, large cholinergic interneurons (LCIs) are relatively spared. However, their ability to release acetylcholine (ACh) is impaired. The present experiments examined morphological and electrophysiological properties of LCIs in the R6/2 mouse model of HD. R6/2 mice show a severe, rapidly progressing phenotype. Immunocytochemical analysis of choline acetyltransferase-positive striatal neurons showed that, although the total number of cells was not chang

Current Drug Targets in Alzheimer's Associated Memory Impairment: A Comprehensive Review.

Alzheimer's disease (AD) is the most prevalent form of dementia among geriatrics. It is a progressive, degenerative neurologic disorder that causes memory and cognition loss. The accumulation of amyloid fibrils and neurofibrillary tangles in the brain of AD patients is a distinguishing feature of the disease. Therefore, most of the current therapeutic goals are targeting inhibition of beta-amyloid synthesis and aggregation as well as tau phosphorylation and aggregation. There is also a loss of the cholinergic neurons in the basal forebrain, and first-generation therapeutic agents were primaril…

Single-cell transcriptomic analysis of neuroepithelial cells and other cell types of the gills of zebrafish (Danio rerio) exposed to hypoxia.

The fish gill is a multifunctional organ involved in numerous physiological processes, such as gas exchange and sensing of hypoxia by respiratory chemoreceptors, called neuroepithelial cells (NECs). Many studies have focused on zebrafish (Danio rerio) to investigate the structure, function and development of the gills, yet the transcriptomic profile of most gill cells remains obscure. We present the results of a comprehensive transcriptomic analysis of the gills of zebrafish using single-cell RNA sequencing (scRNA-seq). Gill cells from ETvmat2:EGFP zebrafish were individually labelled before s

Nicotine Self-Administration Induces Plastic Changes to Nicotinic Receptors in Medial Habenula.

Chronic nicotine upregulates nicotinic acetylcholine receptors (nAChRs) throughout the brain, and reducing their activity may promote somatic and affective states that lead to nicotine seeking. nAChRs are functionally upregulated in animal models using passive nicotine administration, but whether/how it occurs in response to volitional nicotine intake is unknown. The distinction is critical, as drug self-administration (SA) can induce neurotransmission and cellular excitability changes that passive drug administration does not. In this study, we probed the question of whether medial habenula (

Antibiotic-induced dysbiosis of the microbiota impairs gut neuromuscular function in juvenile mice.

BACKGROUND AND PURPOSE: Gut microbiota is essential for the development of the gastrointestinal system, including the enteric nervous system (ENS). Perturbations of gut microbiota in early life have the potential to alter neurodevelopment leading to functional bowel disorders later in life. We examined the hypothesis that gut dysbiosis impairs the structural and functional integrity of the ENS, leading to gut dysmotility in juvenile mice. EXPERIMENTAL APPROACH: To induce gut dysbiosis, broad-spectrum antibiotics were administered by gavage to juvenile (3weeks old) male C57Bl/6 mice for 14 day

Engineering novel spinal circuits to promote recovery after spinal injury.

We have developed an innovative way to establish a functional bridge around a spinal lesion. We disconnected the T13 nerve from its muscle targets, leaving the proximal end intact. The cut end was inserted either into an intact spinal cord, to assess regeneration of T13 axons into the cord and synapse formation with spinal neurons, or caudal to a hemisection at L2/3, to assess restoration of function below the injury. Four to 28 weeks later, anterograde tracers indicated that axons from the inserted T13 nerve regenerated into the ventral horn, the intermediate zone, and dorsal horn base, both
