Unveiling the Bioactive Potential of Shilajit: Integrated Chemical Profiling and Antimicrobial Evaluation via Chromatographic, Spectroscopic, and In Silico Approaches.

Shilajit is a herbomineral exudate widely used in traditional medicine and composed mainly of humic substances and low-molecular-weight orga … heterogeneous morphology and elemental composition. In silico ADMET predictions suggested favorable gastrointestinal absorption and low predicted toxicity for most compounds. Antimicrobial assays demonstrated stronger antibacterial activity of the RF compared to whole Shilajit, while antifungal activity remained limited. Overall, the findings highlight the biological signifi …

Microbial ecology, functional implications, and associated factors influencing poultry intestinal health.

Gut health is a dynamic phenomenon regulated by the balanced interaction among gastrointestinal tract morphological structure, resident microbiota, and available nutrients, producing an integrated barrier that ensures efficient physiological functions, enhanced immune competence and optimal productivity. Gut health is recognised as a crucial factor for optimum management, farming economy, and sustainable commercial poultry production. Poultry gut health has become a central focus, particularly following restrictions applied on the use of antimicrobial growth promoters in poultry production due

Preparation and evaluation of plumbagin-loaded lipid cubic liquid crystal nano-drug delivery system.

To address the poor water solubility and low oral absorption of plumbagin (PL), an anti-hepatocellular carcinoma (HCC) agent, we developed lipidic cubic liquid crystalline nanoparticles (LCNPs) as an oral delivery system to enhance its druggability. PL-loaded LCNPs(PL-LCNPs)were prepared by the ethanol injection method, and optimized via single-factor tests combined with Box-Behnken response surface methodology (BBD-RSM). In vitro release was evaluated by dynamic dialysis, while in vivo fate was assessed by small animal imaging and rat pharmacokinetics. The optimized PL-LCNPs exhibited a parti

Marine-Derived Proteins: Individual Variability in Health Effects Across Protein Structure, Host Genetics, and Gut Microbiota.

Marine-derived proteins are increasingly used in nutritional interventions, but their health effects show substantial interindividual variability, challenging one-size-fits-all approaches. This review proposes a framework based on a triadic interaction model to explain personalized physiological responses to marine proteins. It considers three main determinants. First, the structural characteristics of marine proteins, such as higher order structure, surface charge, solubility, and processing-induced modifications (e.g., enzymatic hydrolysis, heating), shape their digestion behavior and the pr

Re-engineering insulin for oral delivery: structural modifications, advanced formulation strategies, and future directions.

Oral insulin delivery represents a transformative approach to diabetes management, offering improved patient compliance and physiological insulin delivery patterns compared to subcutaneous injection. However, multiple gastrointestinal barriers, including enzymatic degradation, mucus entrapment, epithelial impermeability, and first-pass metabolism, have limited oral bioavailability to below 1% for unmodified insulin. This review comprehensively examines contemporary strategies to overcome these barriers. We analyze structural modifications of insulin, including PEGylation, lipidation, cyclizati

Mycophenolate mofetil induces angiogenesis disorders in zebrafish embryos by inhibiting the mitogen-activated protein kinase signaling pathway.

As a commonly used immunosuppressant, mycophenolate mofetil (MMF) is widely prescribed after organ transplantation because it is not toxic to the liver and kidney. However, its use is associated with certain gastrointestinal toxicity and increased risk of opportunistic infections. Meanwhile, whether it affects vascular development is unclear. Herein, a zebrafish model was utilized to evaluate whether MMF affects vascular development, and the results showed that MMF exhibited teratogenicity, shortened the body length, caused pericardial edema, and delayed yolk sac absorption in zebrafish. In ad

Chronic Fasciola hepatica infection in a patient with cutaneous visceral loxoscelism: paediatric case report.

We present the case of a 9-year-old girl from the Peruvian Andean Region (Ancash), diagnosed with fascioliasis, who was also suffering from severe malnutrition. Initially, she was referred for treatment of skin necrosis caused by cutaneous visceral loxoscelism, but during hospitalisation she developed persistent vomiting and pancreatitis. Magnetic resonance cholangiography showed dilation of the intrahepatic and common bile ducts. Endoscopic retrograde cholangiopancreatography identified an adult Fasciola spp. specimen. She also expelled adult worms of Ascaris lumbricoides during vomiting. Del

Effects of fluoroquinolone antibiotics on extracellular matrix-related phenotypes in tendon cells.

Fluoroquinolones (FQ) are amongst the most widely prescribed antibiotics used to treat a wide range of infections because of their excellent gastrointestinal absorption, superior tissue penetration and broad-spectrum activity. However, FQs can cause rare, though serious, collagen-associated adverse reactions, including tendinopathy, and there is still no clear framework for understanding the phenotypes relating to how tenocyte-produced extracellular matrix (ECM) and related molecules change in response to FQs. Here, we show carrier-mediated intracellular uptake of the FQ ciprofloxacin by mouse…

Oral co-delivery of quercetin and anthocyanins via dual-ligand nanoparticles alleviate obesity and associated metabolic disorders.

Obesity is a global epidemic driven in part by poor oral bioavailability of bioactive food compounds. Here, we develop dual-ligand-functionalized nanoparticles (NPs) for the oral co-delivery of hydrophilic anthocyanins (ACN) and hydrophobic quercetin (Que) to combat obesity. The system is assembled from lactobionic acid-modified chitosan (LACS) and glycyrrhetinic acid-modified casein (GACA), forming a core-shell structure that protects both compounds against gastrointestinal degradation and affords pH-responsive colloidal stability. In Caco-2/HT29-MTX-E12 co-culture models, the LACS-ACN-GACA-Q

Oral colon-targeted responsive chitosan/pectin-based nanoparticles propels the application of tofacitinib in colitis therapy.

Tofacitinib (Tof), a commercially available pan-Janus kinases inhibitor, is approved for the treatment of moderate to severe ulcerative colitis. However, its clinical application is limited due to dose-dependent systemic side effects. The present study aims to develop an efficient oral colon-targeted drug delivery systems using prebiotic pectin (Pcn) and chitosan (Csn) polysaccharides as a shell, with Tof loaded into a Bovine Serum Albumin (BSA) core, and improving it with chondroitin sulfate (Chs), thus constructing Tof@BSA-Chs-CP nanoparticles (NPs). Our results suggest that the pH-sensitive

Contribution of Vasoactive Intestinal Peptide to the Depressant Effects of Glucagon-like Peptide-2 on Neurally Induced Contractile Responses in Mouse Ileal Preparations.

Glucagon-like peptide-2 (GLP-2) has been reported to cause gastrointestinal relaxation by interfering with enteric inhibitory neurotransmitters, including vasoactive intestinal peptide (VIP). However, the involvement of VIP in the GLP-2's actions on isolated ileal preparations has never been explored. In this study, we investigated whether VIP contributes to the inhibitory effects of GLP-2 on spontaneous and neurally evoked contractions in mouse ileal segments. Functional experiments showed that VIP, as well as GLP-2, depresses both spontaneous and electrically induced contractile responses. T

Investigation of the preparation of iron sulfate-loaded niosomes by an experimental novel method.

Iron is a crucial nutrient for our bodies, and its absence can lead to serious health issues like anemia. Unfortunately, oral iron supplements are frequently poorly absorbed and can lead to gastrointestinal discomfort due to iron's oxidative properties. However, using nanocarrier systems to encapsulate iron can mitigate these issues, enhancing its absorption and protecting it from oxidation, ultimately improving its effectiveness. In this research, niosomes loaded with iron sulfate were prepared using surfactant span 60 and Tween 80 via thin film hydration method. Cholesterol and 1-Dodecanol w
