Glial fibrillary acidic protein as an early marker of hepatic stellate cell activation in chronic and posttransplant recurrent hepatitis C.

Activated alpha-smooth muscle actin (alpha-SMA)-positive hepatic stellate cells (HSCs) are pericytes responsible for fibrosis in chronic liver injury. The glial fibrillary acidic protein (GFAP), commonly expressed by astrocytes in the central nervous system, is expressed in vivo in the liver in a subpopulation of quiescent stellate cells. In the rat, increased GFAP expression in the acute response to injury and down-regulation in the chronic response have been observed, whereas reports concerning GFAP expression in human liver are still conflicting. We investigated the utility of GFAP compared…

Prevention of diabetic vascular calcification by nifedipine, a dihydropyridine-based calcium channel blocker.

Vascular calcification is a common feature in advanced atherosclerosis and also a predictor of future cardiovascular events such as unstable … Furthermore, we have found that AGEs have the ability to induce the osteoblatic differentiation of pericytes, thus contributing to the development of vascular calcification as well. These observations suggest that the inhibition of AGE formation or blockade of the downstream signaling of RAGE may be a novel therapeutic target for the inhibition of vascular …

Calcium dobesilate attenuates vascular injury and the progression of diabetic retinopathy in streptozotocin-induced diabetic rats.

BACKGROUND: Diabetic retinopathy (DR) is a highly specific vascular complication of type 1 and type 2 diabetes mellitus. Calcium dobesilate … ortuosity, acellular capillaries, focal accumulations of capillaries and reduction of the number of pericytes in G1. The vascular changes observed in G2 seem to be intermediate between the control and the diabetic rats. CONCLUSIONS: We showed that long-term treatment with DOBE attenuated the progression of diabetic retinopathy and the alterations in vascul …

[Hemangiopericytoma (extrapleural solitary fibrous tumour). Exemplified and discussed on the basis of two cases].

BACKGROUND: Hemangiopericytoma was first mentioned in 1942. It is a very uncommon potentially malignant vascular tumour which can occur at every site of the body. According to the WHO, most of the tumours formerly diagnosed as hemangiopericytomas are considered to be extrapleural solitary fibrous tumours. The diagnosis of "hemangiopericytoma" is now only determined if a constant histological picture of hemangiopericytoma is present. The tumour can lead to lymphogenous or hematogenous metastasis. The major location of occurrence is the cutis and subcutis. It originates from the pericytes of the…

Expression of smooth muscle actin in osteoblasts in human bone.

It is well known that certain connective tissue cells (viz., dermal fibroblasts) can express the gene for a muscle actin--alpha-smooth muscl … s cell type, conferred by its progenitor cells: bone marrow stromal-derived stem cells, and perhaps pericytes and smooth muscle cells.

Villous capillary lesions of the placenta: distinctions between chorangioma, chorangiomatosis, and chorangiosis.

Chorangioma (CA), chorangiosis (CH), and chorangiomatosis (CM) are incompletely understood and overlapping villous capillary (VC) lesions be … litis. Finally, CH lacked the continuous perivascular layer of muscle-specific actin (MSA)-positive pericytes and the multifibrillar lattice-like reticulin pattern seen in both CA and CM. In conclusion, CA and localized CM are clinically and morphologically similar lesions distinct from CH. Diffuse multifocal CM is morphologically similar to CA and localize …

Peripheral nerve pathology in rats with streptozotocin-induced insulinoma.

Peripheral nerve structure was systematically examined in rats with insulinoma induced by streptozotocin (STZ). Normal Wistar rats, aged 3 m … els exhibited a narrowed vascular lumen with swollen endothelial cells and vacuolar degeneration of pericytes, suggesting an involvement of vascular changes in the neuropathic development. The present study demonstrated marked structural changes in both motor and sensory peripheral nerves of rats bearing experimentally induced insulinoma. We consider that a …

Insulin stimulates endothelin binding and action on cultured vascular smooth muscle cells.

Hyperinsulinemia has been implicated as a separate risk factor for the development of accelerated cardiovascular disease, but the mechanism … either treatment affected receptor affinity. Similarly, aortic smooth muscle cells, brain capillary pericytes, and kidney afferent arteriolar smooth muscle cells from rats made hyperinsulinemic in vivo each showed approximately a 2-fold increase in receptor number. This increase in receptor density probably resulted from the stimulation of receptor protein …

Primary malignant myopericytoma of the left atrium--a tumor of aggressive biological behavior: report of the first case and review of literature.

We report a case of malignant myopericytoma arising in the left atrium with brain, skeletal, and liver metastases, which, to our knowledge, is the first report of this rare entity in this anatomic location. A 52-year-old man presented with progressive blackening of his left field of vision. Magnetic resonance imaging and a computed tomography scan of the brain and thorax showed a heterogeneous mass in the right occipital lobe and a large left atrial floor mass. Excision of the atrial mass showed a circumscribed but unencapsulated malignant spindled neoplasm with a perivascular concentric cellu

Tumour angiogenesis regulation by the miR-200 family.

The miR-200 family is well known to inhibit the epithelial-mesenchymal transition, suggesting it may therapeutically inhibit metastatic biology. However, conflicting reports regarding the role of miR-200 in suppressing or promoting metastasis in different cancer types have left unanswered questions. Here we demonstrate a difference in clinical outcome based on miR-200's role in blocking tumour angiogenesis. We demonstrate that miR-200 inhibits angiogenesis through direct and indirect mechanisms by targeting interleukin-8 and CXCL1 secreted by the tumour endothelial and cancer cells. Using seve

Review of the effects of anti-angiogenic compounds on the epiphyseal growth plate.

The formation of new blood vessels from a pre-existing vascular bed, termed "angiogenesis," is of critical importance for the growth and development of the animal since it is required for the growth of the skeleton during endochondral ossification, development and cycling of the corpus luteum and uterus, and for the repair of tissues during wound healing. "Vasculogenesis," the de novo formation of blood vessels is also important for the proper function and development of the vascular system in the embryo. New blood vessel formation is a prominent feature and permissive factor in the relentless

Vascular ossification-calcification in metabolic syndrome, type 2 diabetes mellitus, chronic kidney disease, and calciphylaxis-calcific uremic arteriolopathy: the emerging role of sodium thiosulfate.

BACKGROUND: Vascular calcification is associated with metabolic syndrome, diabetes, hypertension, atherosclerosis, chronic kidney disease, and end stage renal disease. Each of the above contributes to an accelerated and premature demise primarily due to cardiovascular disease. The above conditions are associated with multiple metabolic toxicities resulting in an increase in reactive oxygen species to the arterial vessel wall, which results in a response to injury wound healing (remodeling). The endothelium seems to be at the very center of these disease processes, acting as the first line of d

Sequence of events in the glomerular capillary wall at the onset of proteinuria in passive Heymann nephritis.

Proteinuria in passive Heymann nephritis (PHN) results from complement-mediated glomerular injury, since complement depletion with cobra venom factor (CVF) prevents proteinuria. However, there are no comprehensive morphological studies identifying the sites of injury leading to onset of proteinuria. To address this issue, we attempted to locate sites of injury involved in the onset of proteinuria in PHN. PHN was induced in intact Munich-Wistar rats (PHN-rats, examined at days 3, 5, and 7) and in complement-depleted rats (CVF treated, PHN-CVF-rats, examined at days 3 and 5). The distribution of

Subretinal fibrosis in neovascular age-related macular degeneration: From clinical features to fibrotic mechanisms and therapeutic opportunities.

Neovascular age-related macular degeneration (nAMD) is a leading cause of irreversible central vision loss worldwide. Although intravitreal anti-vascular endothelial growth factor therapy has substantially improved short-term outcomes, long-term vision is frequently limited by the development of subretinal fibrosis. This progressive scarring process occurs in a substantial proportion of patients and represents a major determinant of permanent visual impairment during long-term follow-up. Subretinal fibrosis arises from a complex interaction among structural injury, inflammatory signaling, and

Optical Coherence Tomography Abnormalities in NOTCH3 Mutation Carriers: A Matched Case-Control Study From UK Biobank.

OBJECTIVES: This study aims to identify retinal biomarkers of NOTCH3 mutation carriers using optical coherence tomography (OCT) data from the UK Biobank. METHODS: We conducted a cross-sectional, matched case-control study of individuals with or without NOTCH3 mutation from the UK Biobank between 2006 and 2010. All participants had macular OCT scan and cognitive assessment. Cases were identified based on pathogenic/likely pathogenic NOTCH3 mutations and 1:1 matched with controls based on age, sex, hypertension, diabetes mellitus, and cigarette smoking status. Baseline characteristics and cogni

Procedure-related vasospasm and reperfusion vulnerability during mechanical thrombectomy.

Mechanical thrombectomy (MT) has reshaped the treatment of acute ischemic stroke caused by large-vessel occlusion (LVO), yet successful angiographic reperfusion does not reliably translate into meaningful neurological recovery. Landmark randomized trials and the Highly Effective Reperfusion Evaluated in Multiple Endovascular Stroke Trials (HERMES) patient-level meta-analysis established MT as standard of care, but they also brought into focus the persistent problem of incomplete tissue-level rescue despite technically successful recanalization. Procedure-related vasospasm is usually handled as

Radiation retinopathy: a review.

PURPOSE: Radiation retinopathy (RR) and radiation maculopathy (RM) are common, vision-threatening complications of ocular and periocular radiotherapy, particularly after treatment of uveal melanoma. They result from progressive occlusive microangiopathy leading to retinal ischemia, macular edema, and visual decline. With the increasing use of plaque brachytherapy and proton beam therapy, understanding their epidemiology, risk factors, and management is essential to improve outcomes. RESULTS: RR and RM arise from radiation-induced endothelial and pericyte injury causing blood-retinal barrier b

Bridging the gap: Integrating oxidative stress profiling with retinal imaging for precision management of diabetic retinopathy.

Diabetic retinopathy (DR) remains a primary cause of vision impairment in working-age adults, driven by chronic hyperglycemia-induced oxidative stress that exacerbates retinal microvascular alterations, including pericyte loss, endothelial dysfunction, and capillary occlusion. This narrative review summarizes evidence suggesting that specific oxidative signatures may precede detectable microvascular alterations, suggesting a "metabolic-memory" imprint on retinal cellular homeostasis. This paradigm shift implicates oxidative stress biomarkers not merely as correlates of disease but as predictiv

Fibroblast State Remodelling and Stromal Depletion Shape the Ageing Microenvironment of Human Penile Skin.

Disentangling intrinsic (chronological) skin ageing from ultraviolet-driven photoaging remains challenging because most human skin single-cell studies are biassed towards sun-exposed sites and non-genital anatomy. Here, we profiled histologically normal human penile skin, a typically UV-shielded mucocutaneous niche, to characterise ageing-associated remodelling in a specialised cutaneous microenvironment. We analysed 137 880 cells from 13 healthy male donors (13-63 years) and identified 12 major cell types. Donor-aware compositional profiling revealed pronounced age-associated stromal depletio
