Cysteine metabolic circuitries: druggable targets in cancer.

To enable survival in adverse conditions, cancer cells undergo global metabolic adaptations. The amino acid cysteine actively contributes to cancer metabolic remodelling on three different levels: first, in its free form, in redox control, as a component of the antioxidant glutathione or its involvement in protein s-cysteinylation, a reversible post-translational modification; second, as a substrate for the production of hydrogen sulphide (H2S), which feeds the mitochondrial electron transfer chain and mediates per-sulphidation of ATPase and glycolytic enzymes, thereby stimulating cellular bio

[Bone metabolism and cardiovascular function update. The estimation of bone material quality in the concept of bone and vascular linkage].

It has become clear that a reduction in sex hormones from middle age onwards, increasing age, and an increase in oxidative stress related to lifestyle-related diseases can also reduce bone material properties in terms of collagen post-translational modification, crosslink formation. These changes lead to both qualitative and quantitative abnormalities in collagen, which is the major bone matrix protein. The intermolecular cross-link formation of collagen, which regulates bone-material attributes, is a mechanism independent of bone remodeling. In other words, cross-link formation is controlled

SUMOylation occurs in acute kidney injury and plays a cytoprotective role.

SUMOylation is a form of post-translational modification where small ubiquitin-like modifiers (SUMO) are covalently attached to target proteins to regulate their properties. SUMOylation has been demonstrated during cell stress and implicated in cellular stress response. However, it is largely unclear if SUMOylation contributes to the pathogenesis of kidney diseases, such as acute kidney injury (AKI). Here we have demonstrated a dynamic change of protein SUMOylation in ischemic and cisplatin nephrotoxic AKI in mice. In rat kidney proximal tubular cells (RPTC), cisplatin-induced SUMOylation was

Fumarate hydratase inactivation in renal tumors: HIF1α, NRF2, and "cryptic targets" of transcription factors.

Biallelic inactivation of fumarate hydratase(FH) causes type 2 papillary renal cell carcinoma (PRCC2), uterine fibroids, and cutaneous leimyomas, a condition known as hereditary leiomyomatosis and renal cell cancer(HLRCC). The most direct effect of FH inactivation is intracellular fumarate accumulation. A majority of studies on FH inactivation over the past decade have focused on the theory that intracellular fumarate stabilizes hypoxia-inducible factor 1α(HIF1A) through competitive inhibition of HIF prolyl hydroxylases. Recently, a competing theory that intracellular fumarate activates nuclea

Precise regulation of porcupine activity is required for physiological Wnt signaling.

Gradients of diverse Wnt proteins regulate development, renewal, and differentiation. Porcupine (PORCN) is a membrane-bound O-acyltransferase that is required for post-translational modification of all Wnts to enable their transport, secretion, and activity. Mutations in PORCN are associated with focal dermal hypoplasia (FDH), whereas gene deletion causes embryonic lethality in mice. To study the protein in more detail, zinc finger nucleases were used to edit the PORCN genomic locus, establishing two HT1080 fibrosarcoma clones null for PORCN activity that facilitate the study of PORCN structur

Simvastatin reduces plasma membrane caveolae and caveolin-1 in uterine leiomyomas.

AIMS: Uterine leiomyomas, or fibroids, are estrogen dependent benign tumor in women, however, they have limited treatment options. Simvastatin, a drug commonly used to treat high cholesterol. Recently we demonstrated that simvastatin alters estrogen signaling by reducing the expression and trafficking of the estrogen receptor-α (ER-α) in human uterine leiomyoma cells. Caveolae are invaginations of the plasma membrane where ER-α is known to localize and directly interacts with the caveolar protein caveolin-1 (CAV1). This study examines the effects of simvastatin on plasma membrane caveolae and

Cloning, expression and location of RNase9 in human epididymis.

BACKGROUND: Mammalian spermatozoa become fully motile and fertile during transit through the luminal fluid of the epididymis. At least 200 proteins are present in the epididymal lumen, but the potential roles of these luminal proteins in male fertility are unknown. Investigation of the function of these proteins will elucidate the mechanism of sperm maturation, and also provide new drug targets for male contraception. We cloned RNase9 from a human epididymis cDNA library for characterization and analysis of its functions. FINDINGS: It was predicted that human RNase9 gene was located on chromo

Amino Acid-Driven Gold Nanochaperone Platforms for Alzheimer's Therapy: A Mechanistic and Design Framework for Protein Misfolding Modulation.

Alzheimer's disease (AD) is a progressive neurodegenerative disorder characterized by the misfolding and aggregation of amyloid-β (Aβ) peptides and tau proteins, leading to synaptic dysfunction and neuronal degeneration. Despite decades of therapeutic development, currently available treatments provide limited disease modification, largely due to the structural heterogeneity of Aβ aggregates, poor blood-brain barrier penetration and insufficient targeting of toxic oligomeric intermediates. Recently, nanotechnology-based strategies have emerged as promising approaches to modulate pathological p

Therapeutic targeting of tauopathies: From druggable biology to precision intervention.

Tauopathies encompass a heterogeneous group of neurodegenerative disorders characterized by the transition of tau from a microtubule-stabilizing protein into pathogenic, seed-competent species. Increasing evidence has redefined tau from a descriptive pathological hallmark to a pharmacologically actionable target across multiple disease stages. Therapeutic opportunities are now emerging across several interconnected modules, including microtubule-associated protein tau (MAPT) gene dysregulation, 3-repeat:4-repeat (3 R:4 R) tau imbalance and pathogenic post-translational modification (PTM). Protein C-Mannosylation and C-Mannosyl Tryptophan in Chemical Biology and Medicine.

C-Mannosylation is a post-translational modification of proteins in the endoplasmic reticulum. Monomeric α-mannose is attached to specific Trp residues at the first Trp in the Trp-x-x-Trp/Cys (W-x-x-W/C) motif of substrate proteins, by the action of C-mannosyltransferases, DPY19-related gene products. The acceptor substrate proteins are included in the thrombospondin type I repeat (TSR) superfamily, cytokine receptor type I family, and others. Previous studies demonstrated that C-mannosylation plays critical roles in the folding, sorting, and/or secretion of substrate proteins. A C-mannosylati

Destruction or Reconstruction: A Subtle Liaison between the Proteolytic and Signaling Role of Protein Ubiquitination in Spermatogenesis.

Ubiquitination is one of the most diverse forms of protein post-translational modification that changes the function of the landscape of substrate proteins in response to stimuli, without the need for "de novo" protein synthesis. Ubiquitination is involved in almost all aspects of eukaryotic cell biology, from the best-studied role in promoting the removal of faulty or unnecessary proteins by the way of the ubiquitin proteasome system and autophagy-lysosome pathway to the recruitment of proteins in specific non-proteolytic signaling pathways, as emerged by the more recent discoveries about the

Multimodal MALDI Imaging Mass Spectrometry Reveals Spatially Correlated Lipid and Protein Changes in Mouse Heart with Acute Myocardial Infarction.

Acute myocardial infarction (MI) is a cardiovascular disease that remains a major cause of morbidity and mortality worldwide despite advances in its prevention and treatment. During acute myocardial ischemia, the lack of oxygen switches the cell metabolism to anaerobic respiration, with lactate accumulation, ATP depletion, Na+ and Ca2+ overload, and inhibition of myocardial contractile function, which drastically modifies the lipid, protein, and small metabolite profile in the myocardium. Imaging mass spectrometry (IMS) is a powerful technique to comprehensively elucidate the spatial distribut

The hydroxyl radical in lens nuclear cataractogenesis.

Cataract is the major cause of blindness; the most common form is age-related, or senile, cataract. The reasons for the development of cataract are unknown. Here we demonstrate that nuclear cataract is associated with the extensive hydroxylation of protein-bound amino acid residues, which increases with the development of cataract by up to 15-fold in the case of DOPA. The relative abundance of the oxidized amino acids in lens protein (assessed per parent amino acid) is DOPA > o- and m-tyrosine > 3-hydroxyvaline, 5-hydroxyleucine > dityrosine. Nigrescent cataracts, in which the normally transpa

Cellular and molecular mechanisms of neuronal degeneration: Amyotrophic lateral sclerosis, parkinsonism-demantia, and Alzheimer disease.

Hyperendemic Pacific foci of amyotrophic lateral sclerosis (ALS) and parkinsonism-dementia (PD) represent naturally occurring models of late-onset chronic degenerative diseases that occur in different cultures, in different ecological zones, and among genetically divergent populations. These diseases occur among the Chamorros of the Mariana Islands, among the Auyu and Jakai of southern West New Guinea, and among the Japanese from the Kill Peninsula of Honshu Island. Accumulating evidence supports the hypothesis that a defect in mineral metabolism may be etiologically involved. Toxic and essent

Hydrogen Sulfide as a Mediator of Protein Persulfidation and Motility Regulation in Mammalian Sperm.

AIMS: Hydrogen sulfide (H2S) is a signaling molecule implicated in diverse physiological processes, yet its role in sperm function remains poorly understood. This study characterizes protein persulfidation, a post-translational modification mediated by H2S in boar spermatozoa, tracks its distribution during sperm maturation, and assesses whether H2S influences sperm motility during capacitation. METHODS: Persulfidation was localized using fluorescent labeling and confirmed by Western blotting across epididymal and post-ejaculatory stages. LC-MS/MS identified persulfidated proteins, which were

Palmitoylation Induced Activation of SMAD3 Exacerbates Colitis by Promoting Tissue-resident Memory T Cells Differentiation.

Tissue-resident memory T cells (TRM cells) have been shown to play an instrumental role in driving the onset and relapse of inflammatory bowel diseases (IBD). However, the underlying mechanism of TRM cells differentiation and its regulation in intestines remain to be unveiled. Mothers against decapentaplegic homolog 3 (SMAD3) is translocated from nucleus to membrane and activated in response to transforming growth factor beta (TGF-β), which is a key cytokine in the process of TRM cells polarization. Protein O-Mannosyltransferases in Development and Disease: Evolutionary and Functional Insights from Model Organisms.

Protein O-mannosylation is an evolutionarily conserved post-translational modification essential for neuromuscular development and physiology. The obligate heteromeric complex of protein O-mannosyltransferase 1 and 2 (POMT1/2) initiates O-mannosylation of α-dystroglycan and a limited number of known additional substrates in the endoplasmic reticulum. In humans, defects in these enzymes disrupt dystroglycan function, leading to impaired extracellular matrix interactions and causing a spectrum of severe congenital muscular dystrophies termed dystroglycanopathies. However, mounting evidence indic

PermaPhosPeptide Technology: Recombinant Production of Permanently Phosphorylated Oligopeptides for 14-3-3 Protein Interactomic Studies.

Phosphorylation is a prevalent post-translational modification that controls many signaling pathways by regulating protein-protein interactions. Traditionally, these interactions are studied with chemically synthesized phosphopeptides, which are often expensive and prone to dephosphorylation, or in vitro kinase reactions, which often give incomplete or off-target phosphorylation. Here, using genetic code expansion with amber-codon-directed incorporation of a nonhydrolyzable phosphoserine analog (nhpS) that is autonomously produced in E. coli, we developed a technology to produce PermaPhosPepti

Comparative analysis of differentially acetylated proteins between Antarctic white-blooded Chionodraco hamatus and red-blooded Trematomus bernacchii.

Protein acetylation is a critical post-translational modification that regulates a wide range of cellular functions. The Antarctic icefish, which lack haemoglobin and functional red blood cells, serves as a unique model to explore the impact of acetylation on haematopoiesis and cold-adaptive epigenetic mechanisms. Despite its relevance, the specific acetylation mechanisms in Antarctic fish remain poorly understood. In this study, we utilized a comprehensive methodology combining tandem mass tag labelling, high-performance liquid chromatography fractionation, Kac antibody affinity enrichment an
