Tracking sero-molecular trends of swine brucellosis in Hawai'i and the central Pacific.

INTRODUCTION: Brucellosis is a zoonotic disease of mammals caused by bacterial species of the Brucella genus. The reservoir for disease is typically mammals, with species of Brucella found infecting amphibians, bats, and marine mammals. Brucella spp. can pass directly to humans through contact with infected animals or their products. Brucella spp. can cause chronic debilitating infections in mammals, including humans, and is associated with spontaneous abortions in infected animals, causing reduced fecundity. In Hawai'i, terrestrial species that could harbor Brucella spp. include swine, cattle…

Development of the integrated fish endocrine disruptor test-Part B: Implementation of thyroid-related endpoints.

Given the vital role of thyroid hormones (THs) in vertebrate development, it is essential to identify chemicals that interfere with the TH system. Whereas, among nonmammalian laboratory animals, fish are the most frequently utilized test species in endocrine disruptor research, for example, in guidelines for the detection of effects on the sex hormone system, there is no test guideline (TG) using fish as models for thyroid-related effects; rather, amphibians are used. Therefore, the objective of the present project was to integrate thyroid-related endpoints for fish into a test protocol combin…

Carry-Over Effects of Desiccation Stress on the Oxidative Status of Fasting Anuran Juveniles.

Amphibians are sensitive to deteriorating environmental conditions, especially during transition to a terrestrial environment which is full of uncertainties. Harsh conditions, such as desiccation during earlier stages, affect different larval traits with possible carry-over effects on juvenile and adult life histories. The first consequences of the effects can be seen in juveniles in the challenges to find food and the ability to survive without it in a terrestrial habitat. Body size and the internal energy reserves acquired during the larval phase play an important role in this period. Herein…

Insights into the evolutionary origin of the pineal color discrimination mechanism from the river lamprey.

BACKGROUND: Pineal-related organs in cyclostomes, teleosts, amphibians, and reptiles exhibit color opponency, generating antagonistic neural responses to different wavelengths of light and thereby sensory information about its "color". Our previous studies suggested that in zebrafish and iguana pineal-related organs, a single photoreceptor cell expressing both UV-sensitive parapinopsin and green-sensitive parietopsin generates color opponency in a "one-cell system." However, it remains unknown to what degree these opsins and the single cell-based mechanism in the pineal color opponency are con…

Strigea robusta causes polydactyly and severe forms of Rostand's anomaly P in water frogs.

BACKGROUND: Cases of polydactyly in natural populations of amphibians have attracted great interest from biologists. At the end of the 1940s, the French biologist Jean Rostand discovered a polymorphic syndrome in some water frog (Anura: Pelophylax) populations that included polydactyly and some severe morphological anomalies (he called it 'anomaly P'). The cause of this anomaly remains unknown for 70 years. In a previous study, we obtained anomaly P in the laboratory in tadpoles of water frogs that developed together with molluscs Planorbarius corneus (Mollusca: Gastropoda) collected in the fi…

Effect of exposure to contaminated pond sediments on survival, development, and enzyme and blood biomarkers in veined treefrog (Trachycephalus typhonius) tadpoles.

Sediments are important elements of aquatic ecosystems and in general sediments accumulate diverse toxic substances. Amphibians potentially have a greater risk of exposure to contaminants in sediments, and the test of sediments provides first lines of evidences. Sediment outdoor microcosm experiments were conducted to analyze biological endpoints (survival, development, growth, and morphological and organ malformation), enzyme activity (butyrylcholinesterase, BChE; glutathione-S-transferase, GST; and catalase, CAT) and blood biomarkers in veined treefrog Trachycephalus typhonius tadpoles, a wi…

Effects of feeding on arterial blood gases in the American alligator Alligator mississippiensis.

Reptiles habitually ingest large meals at infrequent intervals, leading to changes in acid-base status as the net secretion of acid to the stomach causes a metabolic alkalosis (the alkaline tide). In chronically cannulated and undisturbed amphibians and reptiles, the pH changes in arterial blood are, nevertheless, reduced by a concomitant respiratory acidosis (increased P(CO2) caused by a relative hypoventilation). Alligators (Alligator mississippiensis) have been reported to exhibit exceptionally large increases in plasma [HCO3(-)] following feeding, but these studies were based on blood samp…

Melatonin agonists induce phosphoinositide hydrolysis in Xenopus laevis melanophores.

Melatonin, the principal hormone of the vertebrate pineal gland, has been implicated in a variety of neurobiological processes such as circadian rhythmicity and reproductive function. One of the earliest described actions of melatonin was its ability to cause pigment translocation in the dermal melanophores of amphibians. Melatonin binding sites have been identified in the brain of many species and in pigmented tumour cell lines; however, the dermal melanophores of the frog Xenopus Laevis possess the highest known density of melatonin binding sites. These cells are the source from which a mela…

Photo-based individual identification is more reliable than visible implant elastomer tags or toe-tipping in young agile frogs.

In amphibian capture-recapture studies, commonly used individual-identification methods include toe clipping as well as less invasive alternatives of varying cost. Yet, choosing the best method for a study is challenging, because both the reliability for identification and the severity of adverse effects of a given mark type can greatly vary between species as well as life stages. We compared the reliability of three identification methods in young agile frogs (Rana dalmatina): clipping a single phalanx, injecting visible-implant-elastomer (VIE) tags (one of six colours) under the skin, and ph

It's not all about PFAS: Metal(loid)s are associated with physiological and metabolic changes in a native frog from conservation wetlands.

Urban wetlands provide essential habitats for wildlife but face increasing contamination from anthropogenic sources, including but not limited to per- and polyfluoroalkyl substances (PFAS) and metal(loid)s. Accumulation of these contaminants are known to impair organism biological functions. This study quantified PFAS and metal(loid) concentrations in liver tissue of motorbike frogs (Ranoidea moorei) along a contamination gradient of conservation significant urban wetlands in Perth, Western Australia, and assessed relationships of liver concentrations with whole-organism health metrics and met

Cave-dwelling phlebotomine sand flies (Diptera: Psychodidae: Phlebotominae) in Thailand: population composition and pathogen detection of Bartonella and Trypanosoma.

BACKGROUND: Leishmaniasis is an emerging vector-borne disease that occurs in Thailand. Although Leishmania (Mundinia) parasites, the causative agents of the disease have been identified, the vectors of the disease remain unidentified. In the present study, we collected sand flies from three caves located in endemic areas of leishmaniasis, including Lampang and Chiang Rai in northern Thailand, and Songkhla in southern Thailand. METHODS: Female sand flies were identified on the basis of morphological characteristics and confirmed by cytochrome c oxidase subunit I (COI) sequencing. Sand fly DNA

Classification of pleurodire polystomes (Platyhelminthes, Monogenea, Polystomatidae) revisited with the description of two new genera from the Australian and Neotropical Realms.

Polystomatids are platyhelminth parasites that infect mostly amphibian and chelonian hosts. Polystomatid of testudines were, for more than seven decades, classified in the three genera - Neopolystoma Price, 1939, Polystomoides Ward, 1917 and Polystomoidella Price, 1939. The genus delimitation was primarily based on the absence of hamuli in Neopolystoma, the presence of one pair of hamuli in Polystomoidella, and two pairs in Polystomoides. From 2016 to 2020, five new genera were erected - namely Uropolystomoides Tinsley and Tinsley, 2016, Uteropolystomoides Tinsley, 2017, Apaloneotrema Du Preez

Two New Species of Telorchis (Digenea: Telorchiidae) from a Green Turtle, Chelonia Mydas (Cheloniidae), from the Upper Texas Coast with a Key to North American Species of Telorchis.

Sea turtles are difficult to sample because of their protected status; however, museum collections and sea turtle stranding networks provide unique opportunities for parasitological research. Four gastrointestinal tracts from stranded, endangered green turtles, Chelonia mydas, were collected between 1993 and 1995 from the upper Texas coast and opportunistically sampled for parasite fauna. Two new species of Telorchis, a common freshwater amphibian and reptilian intestinal parasite genus, were found and described. Telorchis marinus n. sp. differs from Telorchis mydas n. sp. by its short body le

Impact of acute stress on antimicrobial polypeptides mRNA copy number in several tissues of marine sea bass (Dicentrarchus labrax).

BACKGROUND: In comparison to higher vertebrates, fish are thought to rely heavily on innate immune system for initial protection against pathogen invasion because their acquired immune system displays a considerably poor immunological memory, and short-lived secondary response. The endogenous antimicrobial polypeptides (AMPPs) directly and rapidly killing pathogens such as bacteria, fungi, parasites, and viruses are included within the realm of innate defenses. In addition to piscidins, AMPPs that in recent years have been shown to be commonly linked to innate defense, are histones and their p

Regulation of oocyte maturation in the rainbow trout,Salmo gairdneri: role of cyclic AMP in the mechanism of action of the maturation inducing steroid (MIS), 17α-hydroxy, 20β-dihydroprogesterone.

In fish, oocyte maturation (resumption of meiosis after completion of vitellogenesis and before ovulation) is triggered by maturation inducing steroids (MIS) which generally appear to be secreted in the ovary in response to stimulation by a pituitary maturational gonadotropin. Converging data from different laboratories show that 17α-hydroxy, 20β-dihydroprogesterone (17α, 20β-OH-P) is the principal MIS in salmonoids; but clear identification remains to be done in other taxonomic groups. The experiments reported here in the rainbow troutSalmo gairdneri examine the possible involvement of oocyte

Na+ transport by rabbit urinary bladder, a tight epithelium.

By in vitro experiments on rabbit bladder, we reassessed the traditional view that mammalian urinary bladder lacks ion transport mechanisms. Since the ratio of actual-to-nominal membrane area in folded epithelia is variable and hard to estimate, we normalized membrane properties to apical membrane capacitance rather than to nominal area (probably 1 muF approximately 1 cm2 actual area). A new mounting technique that virtually eliminates edge damage yielded resistances up to 78,000 omega muF for rabbit bladder, and resistances for amphibian skin and bladder much higher than those usually reporte

Characterization and tissue distribution of Lhx9 and Lhx9α in Chinese giant salamander Andrias davidianus.

Lhx9 is an LIM (named for the first three proteins in which the domain was found, Lin-11, Isl1 and Mec-3) homeodomain protein involved in development and differentiation of the gonad. In this study, we isolated the full-length Lhx9 and Lhx9α from Andrias davidianus, detected the tissue distribution and analysed the methylation of the promoters. We identified Lhx9 of 1411 bp and Lhx9α of 1153-bp length, differing in the 3'-flanking region, encoding 399 and 330 amino acids, respectively. The Lhx9 gene was detected primarily in liver, ovary and heart with moderate expression in brain, pituitary,
