Mutations in MRR1 that confer fluconazole resistance in Candida parapsilosis and Candida albicans confer cross-resistance to manogepix.

OBJECTIVES: Fluconazole resistance has been documented in Candida albicans and Candida parapsilosis, both leading causes of invasive candidiasis, driven in part by mutations in MRR1 that encodes a transcription factor that regulates drug efflux pump expression. A correlation has been observed between susceptibilities to fluconazole and manogepix, the active moiety of fosmanogepix, a promising new antifungal with activity against most Candida species. We aimed to determine whether MRR1-activating mutations driving fluconazole resistance in these species conferred cross-resistance to manogepix.

Clinical characteristics and treatment response of chronic disseminated candidiasis in patients with hematological disorders.

Chronic disseminated candidiasis (CDC) is an invasive fungal infection typically affecting patients with hematological diseases and severe neutropenia, associated with increased mortality. However, there is a global shortage of clinical evidence on CDC. We retrospectively analyzed clinical data from 49 CDC patients over the past decade. Clinical characteristics of primary hematological diseases, CDC diagnosis, treatment and response evaluations were included. Clinical factors associated with CDC remission and patients' survival were analyzed. The majority of patients had hematological malignan

Discovery of petroselinic acid with in vitro and in vivo antifungal activity by targeting fructose-1,6-bisphosphate aldolase.

BACKGROUND: The incidence of invasive fungal diseases (IFDs), represented by Candida albicans infection, is increasing year by year. However, clinically available antifungal drugs are very limited and encounter challenges such as limited efficacy, drug resistance, high toxicity, and exorbitant cost. Therefore, there is an urgent need for new antifungal drugs. PURPOSE: This study aims to find new antifungal compounds from plants, preferably those with good activity and low toxicity, and reveal their antifungal targets. METHODS: In vitro antifungal activities of compounds were investigated usi

Fluconazole-resistant Candida parapsilosis: A new emerging threat in the fungi arena.

Candida parapsilosis is a leading cause of invasive candidiasis in southern Europe, Latin America and Asia. C. parapsilosis has been mostly considered susceptible to triazoles, but fluconazole resistance is on the rise in some countries. The main mechanism related to fluconazole resistance is the presence of ERG11p substitutions, dominated by the Y132F amino acid substitution. Isolates harbouring this substitution mimic C. auris given that they may cause hospital outbreaks, become endemic, and emerge simultaneously in distant areas around the world. At the moment, Spain is experiencing a brusq

Preparation and In Vitro Evaluation of Alginate Microparticles Containing Amphotericin B for the Treatment of Candida Infections.

Invasive candidiasis (IC) remains as a major cause of morbidity and mortality in critically ill patients. Amphotericin B (AmB) is one of the most effective antifungal agents commonly used to treat this infection. However, it induces severe side effects such as nephrotoxicity, cardiac alterations, nausea, fever, and liver damage. The utilization of drug delivery systems has been explored to overcome these limitations. Several AmB lipid formulations have been developed and are currently available in the market. Although they have the ability to reduce the main side effects of free AmB, their hig

Antifungal effect of titanium oxide nanoparticles on Candida glabrata internalized in human macrophages.

BACKGROUND: Candida glabrata (Nakaseomyces glabrata) is an emergent human fungal opportunist pathogen. Recently, it was considered by the WHO as a fungal priority pathogen due to the high percentage of mortality (20-50%) from invasive candidiasis. C. glabrata shows high resistance to azoles, and in recent years, echinocandin resistance has been rising. Additionally, C. glabrata can evade immunological responses and manipulate macrophage activity to survive within these immune cells successfully. Titanium oxide (TiO2) is a photocatalytic material with recognized antimicrobial properties with an

The role of manganese in morphogenesis and pathogenesis of the opportunistic fungal pathogen Candida albicans.

Metals such as Fe, Cu, Zn, and Mn are essential trace nutrients for all kingdoms of life, including microbial pathogens and their hosts. During infection, the mammalian host attempts to starve invading microbes of these micronutrients through responses collectively known as nutritional immunity. Nutritional immunity for Zn, Fe and Cu has been well documented for fungal infections; however Mn handling at the host-fungal pathogen interface remains largely unexplored. This work establishes the foundation of fungal resistance against Mn associated nutritional immunity through the characterization

A Preliminary in vitro and in vivo Evaluation of the Effect and Action Mechanism of 17-AAG Combined With Azoles Against Azole-Resistant Candida spp.

Invasive candidiasis is the primary reason for the increased cases of mortality in a medical environment. The resistance spectra of Candida species to antifungal drugs have gradually expanded. Particularly, the resistance spectra of Candida auris are the most prominent. Hsp90 plays a protective role in the stress response of fungi and facilitates their virulence. In contrast, Hsp90 inhibitors can improve the resistance of fungi to antifungal drugs by regulating the heat resistance of Hsp90, which destroys the integrity of the fungal cell walls. Hsp90 inhibitors thus offer a great potential to

Antimicrobial peptides: Defending the mucosal epithelial barrier.

The recent epidemic caused by aerosolized SARS-CoV-2 virus illustrates the importance and vulnerability of the mucosal epithelial barrier against infection. Antimicrobial proteins and peptides (AMPs) are key to the epithelial barrier, providing immunity against microbes. In primitive life forms, AMPs protect the integument and the gut against pathogenic microbes. AMPs have also evolved in humans and other mammals to enhance newer, complex innate and adaptive immunity to favor the persistence of commensals over pathogenic microbes. The canonical AMPs are helictical peptides that form lethal por

Antifungal and immunomodulatory activities of dihydroartemisinin-loaded chitosan nanoparticles against fluconazole‑resistant Candida tropicalis.

OBJECTIVES: Invasive fungal infections, such as those caused by fluconazole-resistant Candida tropicalis, pose an important threat to immunocompromised patients, with mortality rates ranging from 55 to 60%. The rise of clinically resistant C. tropicalis has compounded the difficulties in treatment, highlighting the urgent need for the development of novel antifungal agents. This study builds on previous research to evaluate the antifungal efficacy of dihydroartemisinin-loaded chitosan nanoparticles (DHA-CS NPs) against fluconazole-resistant C. tropicalis and elucidates their mechanisms of acti

New insights on Drug's design against candidiasis on the fructose biphosphate aldolase (Fba1) and the pyruvate kinase (Pk) of Candida glabrata.

Candida glabrata is well known to be the second most common cause of invasive candidiasis (IC) within immunocompromised and hospitalized patients, after Candida albicans. Candida species adhere to host cells and implanted medical devices by means of cell wall proteins (CWP), of which the moonlight proteins have recently been described and are of particular importance because they have been identified in response to various virulence and/or pathogenic factors. Among the identified CWP moonlights, fructose-bisphosphate aldolase (Fba1) and pyruvate kinase (Pk) have been observed to confer immune
