Cortical networks for the production of spoken language in humans are organized by phonetic features1,2, such as articulatory parameters3,4 and vocal pitch5,6. Previous research has failed to find an equivalent forebrain representation in other species7-11. To investigate whether this functional organization is unique to humans, here we performed population recordings in the vocal production circuitry of the budgerigar (Melopsittacus undulatus), a small parrot that can generate flexible vocal output12-15, including mimicked speech sounds16. Using high-density silicon probes17, we measured the

The cerebellum is critical for motor timing control and error-driven motor learning. To reveal how the cerebellum transmits these process-relevant signals to the premotor cortex, we conducted two-photon calcium imaging of cerebellar-thalamocortical axons in the premotor cortex in male mice during a self-timing lever-pull task that required 1-1.7 s of waiting after cue onset. In non-expert sessions with many lever-pulls being made before the 1-s waiting, the axons of thalamic neurons that received cerebellar outputs exhibited larger transient activity immediately after the cue onset in post-err

The dorsal subiculum (dSub) and medial mammillary body (MM) are essential components of the extended hippocampal system, heavily implicated in spatial memory processes. However, the exact function of the prominent dSub-to-MM projection has remained ambiguous. Here, integrating c-fos mapping, fiber photometry, optogenetic manipulations and behavioral paradigms, we found that the monosynaptic connection between dSub and MM, rather than collaterals to other regions, is not only involved in spatial memory retrieval but also highly vulnerable to perturbations on activity level in both directions. I

While differences in patterns of functional connectivity and neural synchronization have been reported between individuals on the autism spectrum and neurotypical peers at various age stages, these differences appear to be subtle and may not be captured by typical quantitative measures of EEG. We used the dynamical systems approach to analyze resting-state EEG to investigate fine-grained spatiotemporal organization of brain networks in autistic and neurotypical young adults. While power spectra showed minimal group differences, autistic participants exhibited higher Lyapunov exponents (indicat

Understanding functional brain development during childhood and adolescence is essential for identifying typical neurodevelopmental trajectories. While resting-state fMRI (rs-fMRI) has become a key tool in developmental neuroscience, few studies have jointly examined multiple functional metrics to comprehensively characterize typical brain maturation across youth. We analyzed rs-fMRI data from 395 neurotypical participants aged 6-20 years from the ABIDE I and II datasets. Voxel-wise analyses were conducted using three complementary rs-fMRI metrics: fractional amplitude of low-frequency fluctua

A key question is how new semantic representations are formed in the human brain and how this may benefit from the hippocampal episodic memory system. Here, we describe the major effective connectivity between the hippocampal memory system and the anterior temporal lobe (ATL) semantic memory system in humans. Then, we present and model a theory of how semantic representations may be formed in the human ATL using slow associative learning in semantic attractor networks that receive inputs from the hippocampal episodic memory system. The hypothesis is that if one category of semantic representat

Changes in the frontostriatal system activity support individuals' perseverance in distressful thoughts and rigid, repetitive behaviours that define obsessive-compulsive disorder (OCD). Converging evidence from preclinical and clinical work suggests that OCD maps onto a functional imbalance in the ventral and dorsal frontostriatal circuits. However, the neural mechanisms supporting these dysregulations remain elusive, their association with symptom severity is unclear, and therapeutic interventions are limited. To address these gaps, we combined neuroimaging and behavioural data from individua

Much of the behavioral repertoire of humans and other vertebrates is learned and controlled through the function of brain circuits involving the cortex, thalamus and Basal Ganglia (for simplicity we will refer to this as the Cortico-Thalamo-Basal Ganglia, or CTBG, circuitry). As the name implies, these circuits include the different regions of cortex and thalamus, as well as BG subregions including the striatum, globus pallidus (GP), substantia nigra (SN)/ventral tegmental area (VTA), and the subthalamic nucleus (STN). This circuitry has developed evolutionarily to provide overarching control

BACKGROUND: Autism spectrum disorder (ASD) is associated with altered brain functional connectivity, but findings regarding the nature of these abnormalities remain inconsistent, partly due to methodological limitations and the disorder's intrinsic heterogeneity. This study aims to provide a comprehensive characterization of functional network alterations in preschool children with ASD by integrating low- and high-order functional connectivity (LOFC/HOFC), static and dynamic network analysis, and entropy-based state transition assessment.
METHODS: EEG data were collected from 32 children with

Sensory feedback is crucial for movement execution, especially in the highly specialized skilled hand use in humans and other primates. Extensive sensory loss of tactile and proprioceptive inputs from the hand initially results in severe motor deficits, but recovery occurs gradually over time. To determine how sensory loss from one hand affects evoked forelimb movements from sensorimotor cortical areas, lesions were selectively placed in the ascending somatosensory pathway in the dorsal column in the cervical spinal cord (C5, DCL) of three squirrel monkeys. After 1 year of postlesion recovery,

Down syndrome (DS) is the most common genetic cause of intellectual disability, but our understanding of white matter microstructure in children with DS remains limited. Previous studies have reported reductions in white matter integrity, but nearly all studies to date have been conducted in adults or relied solely on diffusion tensor imaging (DTI), which lacks the ability to disentangle underlying properties of white matter organization. This study examined white matter microstructural differences in 7- to 12-year-old children with DS (n = 23), autism (n = 27), and typical development (n = 50

The cerebellum acts as a forward internal model to predict motor outcomes, compare them with sensory feedback, and generate prediction errors that refine prediction accuracy. Our physiological understanding of cerebellar function during motor control derives predominantly from animal experiments and clinical observations in patients with disorders of the cerebellum or its connections with the cerebrum and spinal cord. Here, we report a human electrophysiology-based investigation of cerebello-thalamo-cortical pathway activity during motor error detection and correction. Participants performed a

OBJECTIVES: Functional neurological disorder (FND) is associated with altered self-processing - the neural processes that underpin the individual's self-agency and sense of self. This study used resting-state functional magnetic resonance imaging to examine three nested meta-analytically defined self-processing brain networks in 28 children with mixed FND symptoms and 31 healthy controls.
METHODS: Regions of interest (ROIs) for each layer of brain network analysis were defined using the nested hierarchical model of self-processing developed by Northoff and colleagues specifying the insula (in

The dorsal and ventral hippocampus have distinct processing properties, but it remains unclear if interneuron subtypes differ in connectivity along the dorsoventral axis. Oriens lacunosum-moleculare (OLM) interneurons, identified by the Chrna2 gene, are known to regulate memory processes differently along this axis. OLMɑ2 cells bidirectionally modulate risk-taking behavior, while ventral hippocampal medial prefrontal cortex (mPFC)-projecting neurons regulate approach and avoidance behaviors. Using rabies virus-mediated monosynaptic retrograde tracing, we show that OLMɑ2 cells receive different

The anterior (DA) and posterior parts of the deltoid (DP) show alternating contraction during shoulder flexion and extension movements. It is expected that an inhibitory spinal reflex between the DA and DP exists. In this study, spinal reflexes between the DA and DP were examined in healthy human subjects using post-stimulus time histogram (PSTH) and electromyogram averaging (EMG-A). Electrical conditioning stimulation was delivered to the axillary nerve branch that innervates the DA (DA nerve) and DP (DP nerve) with the intensity below the motor threshold. In the PSTH study, the stimulation t

The functional organization of the frontal lobe is a source of debate, focusing on broad functional subdivisions, large-scale networks, or local refined specificities. Multiple neurocognitive models have tried to explain how functional interactions between cingulate and lateral frontal regions contribute to decision making and cognitive control, but their neuroanatomical bases remain unclear. We provide a detailed description of the functional connectivity between cingulate and lateral frontal regions using resting-state functional MRI in rhesus macaques. The analysis focuses on the functional

The executive control process of monitoring information in working memory depends on the mid-dorsolateral prefrontal cortical region (cytoarchitectonic areas 46 and 9/46) in interaction with the hippocampal memory system. Anatomical studies demonstrated strong connectivity between the mid-dorsolateral prefrontal cortex and the medial parietal area PGm that lies on the precuneus. Area PGm is also strongly connected with the attentional system on the lateral inferior parietal lobule (area PG) and the limbic retrosplenial/posterior cingulate region that interacts with the hippocampal memory syste

The supraspinal descending pain modulatory system (DPMS) shapes pain perception via monoaminergic modulation of sensory information in the spinal cord. However, the role and synaptic mechanisms of descending noradrenergic signaling remain unclear. Here, we establish that noradrenergic neurons of the locus coeruleus (LC) are essential for supraspinal opioid antinociception. While much previous work has emphasized the role of descending serotonergic pathways, we find that opioid antinociception is primarily driven by excitatory output from the ventrolateral periaqueductal gray (vlPAG) to the LC.

BACKGROUND: Parenting is a common and potent environmental factor influencing adolescent anxiety. Yet, the underlying neurobiological susceptibility signatures remain elusive. Here, we used a longitudinal twin neuroimaging study to investigate the brain network integration and its heritable relation to underpin the neural differential susceptibility of adolescent anxiety to parenting environments.
METHODS: 216 twins from the Beijing Twin Study completed the parenting and anxiety assessments and fMRI scanning. We first identified the brain network integration involved in the influences of pare
