Per- and polyfluoroalkyl substances and type 2 diabetes risk: Evidence from metabolomics and triglyceride-glucose index-related pathways.

Per- and polyfluoroalkyl substances (PFAS), including emerging alternatives, have been implicated in type 2 diabetes (T2D), but epidemiological evidence remains inconsistent and the underlying mechanisms are unclear. We aimed to investigate the associations of individual and mixed PFAS exposure with T2D risk and to explore the roles of triglyceride-glucose (TyG)-related indices and metabolomic alterations. A total of 1647 participants (749 participants with T2D and 898 controls) were included. Multivariable logistic regression and weighted quantile sum (WQS) regression were used to evaluate th

BMSC-derived extracellular vesicles affect gluconeogenesis and lipogenesis by releasing 5'-tRF-GlyCCC to improve MAFLD insulin sensitivity.

TsRNAs (tRNA-derived small RNAs) also known as tRNA-derived small RNAs, are a relatively new type of non-coding RNAs that have demonstrated promising effect in treating various liver diseases. However, the function of small extracellular vesicles (sEVs) secreted by human bone marrow mesenchymal stem cells (BMSCs) in safeguarding against metabolic-associated fatty liver disease (MAFLD) is still uncertain. In this research, we explored the effects of BMSCs sEVs on lipid metabolism using Palmitic Acid (PA)-induced HepG2 cells, both in the presence and absence of the sEVs inhibitor GW4869. Pandora

Rutaecarpine Attenuates Hepatic Lipid Accumulation and Oxidative Stress by Activating Autophagy Through the AMPK/ULK1 Pathway.

Non-alcoholic fatty liver disease (NAFLD) is a prevalent metabolic disorder disease in modern society, primarily caused by abnormal fat accumulation in the liver. This study aims to uncover the effects and potential mechanisms by which Rutaecarpine (Rut) alleviates lipid accumulation and oxidative damage in hepatocytes. Oleic acid and palmitic acid were used to induce lipid accumulation in AML12 cells, and Tyloxapol was used to induce hepatic lipid accumulation in mice. Then, various concentrations of Rut were administered. The mechanisms were further explored by using the Nrf2 inhibitor ML385

Dendrobium officinale polysaccharide ameliorates high-fat diet-induced hepatic lipid metabolic disorder via the SIRT6/PGC-1α signaling axis.

AIMS: This study aims to explore the potential therapeutic effect of Dendrobium officinale polysaccharide (DOP) on non-alcoholic fatty liver disease (NAFLD) induced by high-fat diet (HFD), and to elucidate the underlying mechanism involving the SIRT6/PGC-1α signaling axis and the regulation of the gut microbiota. METHODS: We extracted and characterized DOP. We established a rat model of NAFLD induced by HFD and evaluated the efficacy of DOP by integrating multi-omics techniques (transcriptomics, metabolomics) and 16S rRNA sequencing. To verify the specific role of SIRT6, we introduced the SIR

Single-cell analysis identifies CPT1a-associated metabolic remodeling in human NK cells during COVID-19.

Natural killer (NK) cells are critical for early antiviral immunity, yet their metabolic regulation during acute human viral infection remains incompletely understood. We analyzed NK cell activation and metabolic reprogramming in 47 vaccinated individuals with mild breakthrough SARS-CoV-2 infection and 20 matched healthy control subjects. COVID-19 patients exhibited elevated plasma interferon α and NK cell activation markers (CD69, CD38), alongside increased basal STAT5 phosphorylation, consistent with IL-15-mediated signaling. Functionally, NK cells from infected subjects displayed heightened

Plastic poisoning the next generation: omics-anchored partial-adverse outcome pathway affords a glimpse into how gestational polystyrene nano-plastics exposure compromises adult male offspring reproduction.

Ubiquitous environmental nano-plastics (NPs) can enter organisms via multiple pathways, posing potential health risks; however, the adverse effects of gestational polystyrene NPs (PS-NPs) exposure on adult offspring reproductive function and underlying mechanisms remain unclear. The adverse outcome pathway (AOP) framework models toxic events from molecular to organismal levels. This study employed multi-omics analysis to construct a putative partial-AOP network, revealing insights into the mechanism of intergenerational reproductive impairment of PS-NPs in adult offspring. Gestational PS-NPs e

Functional Role of Fatty Acid Synthase for Signal Transduction in Core-Binding Factor Acute Myeloid Leukemia with an Activating c-Kit Mutation.

Background/Objectives: Acute myeloid leukemia (AML) is a rare hematological malignancy with a poor prognosis. Activating c-Kit (CD117) mutations occur in 5% of de novo AML and 30% of core-binding factor (CBF) AML, leading to worse clinical outcomes. Posttranslational modifications, particularly with myristic and palmitic acid, are crucial for various cellular processes, including membrane organization, signal transduction, and apoptosis regulation. However, most research has focused on solid tumors, with limited understanding of these mechanisms in AML. Fatty acid synthase (FASN), a key palmit

Curcumol ameliorates high-fat diet-induced hepatic fibrosis via dual regulation of FXR-CREB and Rab18-mediated hepatic stellate cell lipophagy.

Curcumol, a bioactive constituent derived from Rhizoma Curcumae roots, possesses anti-inflammatory and anti-viral properties. This study investigated its therapeutic effects on lipophagy in hepatic stellate cells (HSCs) and lipid accumulation in hepatocytes within a model of high-fat diet-induced hepatic fibrosis, along with the underlying molecular mechanisms. Initially, curcumol treatment significantly attenuated lipid droplets (LDs) degradation and suppressed HSC activation, effects potentially associated with the inhibition of lipophagy. These outcomes were partially reversed by Rab18 over

Lycopene Alleviates Endoplasmic Reticulum Stress in Steatohepatitis through Inhibition of the ASK1-JNK Signaling Pathway.

Lycopene has been proven to alleviate nonalcoholic steatohepatitis (NASH), but the precise mechanisms are inadequately elucidated. In this study, we found a previously unknown regulatory effect of lycopene on the apoptosis signal-regulating kinase 1 (ASK1) signaling pathway in both in vivo and in vitro models. Lycopene supplementation (3 and 6 mg/kg/day) exhibited a significant reduction in lipid accumulation, inflammation, and fibrosis of the liver in mice fed with a high-fat/high-cholesterol diet or a methionine-choline-deficient diet. RNA sequencing uncovered that the mitogen-activated prot

Imeglimin Halts Liver Damage by Improving Mitochondrial Dysfunction in a Nondiabetic Male Mouse Model of Metabolic Dysfunction-Associated Steatohepatitis.

Imeglimin promotes glucose-stimulated insulin secretion in the pancreas in a glucose-dependent manner and inhibits gluconeogenesis in the liver. Meanwhile, imeglimin can improve mitochondrial function in hepatocytes. We used a nondiabetic metabolic dysfunction-associated steatohepatitis (MASH) model to examine the effects of imeglimin on MASH independent of its glucose-lowering action. Mice fed a choline-deficient high-fat diet (CDA-HFD) were orally administered imeglimin (100 and 200 mg/kg twice daily), and MASH pathophysiology was evaluated after 8 weeks. Moreover, an in vitro study investig

Phytocannabinoids promote viability and functional adipogenesis of bone marrow-derived mesenchymal stem cells through different molecular targets.

The cellular microenvironment plays a critical role in the maintenance of bone marrow-derived mesenchymal stem cells (BM-MSCs) and their subsequent cell lineage differentiation. Recent studies suggested that individuals with adipocyte-related metabolic disorders have altered function and adipogenic potential of adipose stem cell subpopulations, primarily BM-MSCs, increasing the risk of heart attack, stroke or diabetes. In this study, we explored the potential therapeutic effect of some of the most abundant non-euphoric compounds derived from the Cannabis sativa plant (or phytocannabinoids) inc
