Comprehensive flavor profiling and neural perception of yeast extracts from five yeast sources via sensomics, machine learning, and EEG.

Yeast extract is widely used as a natural flavor enhancer, yet how yeast source shapes its taste, aroma, and neural perception remains insufficiently understood. Here, we compared yeast extracts from five industrially relevant sources (beer yeast, BY; bread yeast, BRY; Candida utilis, CUY; alcohol yeast, AY; wine yeast, WY) using an integrated sensory-chemical-neural framework. Sensory evaluation revealed clear source-dependent flavor signatures (p < 0.05), with CUY showing the strongest umami intensity. Solvent-assisted flavor evaporation (SAFE) coupled with GC-O-MS identified 120 volatile co

A Cross-Disease Microglial Transcriptional Program Characterizes Neurodegeneration and Highlights SPP1 as a Biomarker.

Microglial cells are key players in maintaining brain homeostasis and responding to pathological conditions. Their multifaceted roles in health and disease have garnered significant attention in the context of neurodegeneration. In recent years, single-cell transcriptomic techniques have provided unprecedented insights into microglial heterogeneity, revealing distinct subpopulations and gene expression patterns associated with neuroprotection or neurotoxicity. Here, we dissect the transcriptomic landscape of microglia by leveraging human single-nuclei RNA sequencing datasets from multiple neur

vClean: assessing virus sequence contamination in viral genomes.

Recent advancements in viral metagenomics and single-virus genomics have improved our ability to obtain the draft genomes of environmental viruses. However, these methods can introduce virus sequence contaminations into viral genomes when short, fragmented partial sequences are present in the assembled contigs. These contaminations can lead to incorrect analyses; however, practical detection tools are lacking. In this study, we introduce vClean, a novel automated tool that detects contaminations in viral genomes. By applying machine learning to the nucleotide sequence features and gene pattern

Fire risk to structures in California's Wildland-Urban Interface.

The destructive impacts of wildfires on people, property and the environment have dramatically increased, especially in the Wildland-Urban Interface (WUI) in California. In these areas structures are threatened by both approaching flames and lofted embers which spread fire into and within communities. While independent factors influencing structure fire protection are well known, their combined effects remain largely unquantified, limiting the accuracy of risk assessments and mitigation strategies. Here, we examine five major historical WUI fires-2017 Tubbs, 2017 Thomas, 2018 Camp, 2019 Kincad

Schizophrenia recognition via eye movement features in video paradigm.

Schizophrenia is a severe mental disorder characterized by abnormal eye movements. However, existing methods for detecting these abnormalities rely primarily on static stimuli that lack ecological validity. This study explores the potential of video paradigms for capturing eye movement patterns specific to schizophrenia. Forty patients with schizophrenia (SZ) and forty healthy controls (HCs) completed eye movement tests and cognitive assessments. The participants watched two types of videos: an animated video with low cognitive load and a documentary with high social cognitive load, from which

Harnessing scRNA-seq and bulk RNA-seq to identify CD39+ T cell genes for rheumatoid arthritis diagnosis and therapy.

Rheumatoid arthritis is a prevalent autoimmune disorder with an elusive pathogenesis, hindering early detection and therapeutic advancements. This study focuses on CD39+ T cells, which play a significant role in rheumatoid arthritis, to identify diagnostic and therapeutic biomarkers. We analyzed single-cell RNA sequencing data from rheumatoid arthritis patients to identify differentially expressed genes associated with CD39+ T cells. We then cross-referenced these differentially expressed genes with those from normal and rheumatoid arthritis samples to extract a CD39+ T cell gene signature. Fu

Single-cell transcriptomes identify patient-tailored therapies for selective co-inhibition of cancer clones.

Intratumoral cellular heterogeneity necessitates multi-targeting therapies for improved clinical benefits in advanced malignancies. However, systematic identification of patient-specific treatments that selectively co-inhibit cancerous cell populations poses a combinatorial challenge, since the number of possible drug-dose combinations vastly exceeds what could be tested in patient cells. Here, we describe a machine learning approach, scTherapy, which leverages single-cell transcriptomic profiles to prioritize multi-targeting treatment options for individual patients with hematological cancers

Interpersonal heart rate synchrony predicts effective information processing in a naturalistic group decision-making task.

Groups often outperform individuals in problem-solving. Nevertheless, failure to critically evaluate ideas risks suboptimal outcomes through so-called groupthink. Prior studies have shown that people who hold shared goals, perspectives, or understanding of the environment show similar patterns of brain activity, which itself can be enhanced by consensus-building discussions. Whether shared arousal alone can predict collective decision-making outcomes, however, remains unknown. To address this gap, we computed interpersonal heart rate synchrony, a peripheral index of shared arousal associated w

Delayed knowledge transfer: Cross-modal knowledge transfer from delayed stimulus to EEG for continuous attention detection based on spike-represented EEG signals.

Decoding visual and auditory stimuli from brain activities, such as electroencephalography (EEG), offers promising advancements for enhancing machine-to-human interaction. However, effectively representing EEG signals remains a significant challenge. In this paper, we introduce a novel Delayed Knowledge Transfer (DKT) framework that employs spiking neurons for attention detection, using our experimental EEG dataset. This framework extracts patterns from audiovisual stimuli to model brain responses in EEG signals, while accounting for inherent response delays. By aligning audiovisual features w

High Fidelity Machine-Learning-Assisted False Positive Discrimination in Loop-Mediated Isothermal Amplification Using Nanopore-Based Sizing and Counting.

Loop-mediated isothermal amplification (LAMP) is a rapid, sensitive, and cost-effective method for developing point-of-care nucleic acid testing due to its isothermal nature. Yet, LAMP can suffer from the issue of false positives, which can compromise the specificity of the results. LAMP false positives typically arise due to contamination, nonspecific amplification, and nonspecific signal reporting (intercalating dyes, colorimetric, turbidity, etc.). While dye-labeled primers or probes have been introduced for multiplexed detection and enhanced specificity in LAMP assays, they carry the risk

Tai Chi Expertise Classification in Older Adults Using Wrist Wearables and Machine Learning.

Tai Chi is a Chinese martial art that provides an adaptive and accessible exercise for older adults with varying functional capacity. While Tai Chi is widely recommended for its physical benefits, wider adoption in at-home practice presents challenges for practitioners, as limited feedback may hamper learning. This study examined the feasibility of using a wearable sensor, combined with machine learning (ML) approaches, to automatically and objectively classify Tai Chi expertise. We hypothesized that the combination of wrist acceleration profiles with ML approaches would be able to accurately

Impact of the COVID-19 pandemic on dengue in Brazil: Interrupted time series analysis of changes in surveillance and transmission.

Measures to curb the spread of SARS-CoV-2 impacted not only COVID-19 dynamics, but also other infectious diseases, such as dengue in Brazil. The COVID-19 pandemic disrupted not only transmission dynamics due to changes in mobility patterns, but also several aspects of surveillance, such as care seeking behavior and clinical capacity. However, we lack a clear understanding of the overall impact on dengue in different parts of Brazil and the contribution of individual causal drivers. In this study, we estimated the gap between expected and observed dengue cases in each Brazilian state from March

Seeking Solace in Gambling: The Cycle of Gambling and Intimate Partner Violence Against Women Who Gamble.

This study explored women's gambling in response to male intimate partner violence (IPV). Twenty-four women were recruited through service providers and online advertising. All women had been victimised by IPV and all experienced problems relating to the gambling on electronic gaming machines (EGMs). Thematic analysis of their in-depth interviews identified three major themes. The main pattern of gambling and IPV (Theme 1) was where ongoing coercive control preceded the woman's gambling. Situational violence in response to gambling was also observed. Regardless of temporal sequence, a self-per

A computer-aided approach for automatic detection of breast masses in digital mammogram via spectral clustering and support vector machine.

Breast cancer continues to be a widespread health concern all over the world. Mammography is an important method in the early detection of breast abnormalities. In recent years, using an automatic Computer-Aided Detection (CAD) system based on image processing techniques has been a more reliable interpretation in the illustration of breast distortion. In this study, a fully process-integrated approach with developing a CAD system is presented for the detection of breast masses based on texture description, spectral clustering, and Support Vector Machine (SVM). To this end, breast Regions of In

Readmission prediction using deep learning on electronic health records.

Unscheduled 30-day readmissions are a hallmark of Congestive Heart Failure (CHF) patients that pose significant health risks and escalate care cost. In order to reduce readmissions and curb the cost of care, it is important to initiate targeted intervention programs for patients at risk of readmission. This requires identifying high-risk patients at the time of discharge from hospital. Here, using real data from over 7500 CHF patients hospitalized between 2012 and 2016 in Sweden, we built and tested a deep learning framework to predict 30-day unscheduled readmission. We present a cost-sensitiv

Neural markers of negative symptom outcomes in distributed working memory brain activity of antipsychotic-naive schizophrenia patients.

Since working memory deficits in schizophrenia have been linked to negative symptoms, we tested whether features of the one could predict the treatment outcome in the other. Specifically, we hypothesized that working memory-related functional connectivity at pre-treatment can predict improvement of negative symptoms in antipsychotic-treated patients. Fourteen antipsychotic-naive patients with first-episode schizophrenia were clinically assessed before and after 7 months of quetiapine monotherapy. At baseline, patients underwent functional magnetic resonance imaging while performing a verbal n-

Exploring the diagnostic potential of EEG theta power and interhemispheric correlation of temporal lobe activities in Alzheimer's Disease through random forest analysis.

BACKGROUND: Considering the prevalence of Alzheimer's Disease (AD) among the aging population and the limited means of treatment, early detection emerges as a crucial focus area whereas electroencephalography (EEG) provides a promising diagnostic tool. To date, several studies indicated EEG dataset-based models sporting high diagnostic power in distinguishing patients with AD from healthy controls (HC). However, exploration into which features play a crucial role in the diagnosis remains limited. METHODS: This study investigates the diagnostic capabilities of EEG for distinguishing patients w

Household Clustering of High-Risk Contacts in Smear-Positive TB Patient Families: Evidence for Hotspot Households and Risk Stratification in Rural Eastern Cape.

BACKGROUND: Household contacts of smear-positive tuberculosis (TB) patients face an elevated risk of infection and disease progression, particularly young children and individuals living in overcrowded households. Despite WHO recommendations for systematic contact screening and provision of TB preventive therapy (TPT), implementation remains suboptimal in high-burden rural areas. This study aimed to develop a practical framework for identifying and prioritizing high-risk families by examining demographic predictors, household clustering, and machine learning-based risk models. METHODS: A tota
