Gene expression profiling of key genes in hypothalamus-pituitary-gonad axis of rare minnow Gobiocypris rarus in response to EE2.

The 17α-ethinylestradiol (EE2), which could induce estrogenic effects, is found in different aquatic systems. The current study aimed to assess in vivo effects of short-term EE2 exposure on the transcriptional activity of genes in the brain and gonad tissues in order to characterize the mode of action of EE2 on the hypothalamus-pituitary-gonad axis in rare minnow (Gobiocypris rarus). The full length cDNAs of fshβ, lhβ, fshr and lhr were first characterized in G. rarus. The homology and phylogenetic analyses of the amino acid sequences revealed that these four genes share high identity in cypri

FSH mediated endocrine disruption potential of Dibutyl phthalate in cultured ovine ovarian granulosa cells: Cytotoxicity, oxidative stress, steroidogenesis, cell senescence, and expression of related key developmental genes.

Di-n-butyl phthalate (DBP) is a widely used phthalate ester recognized for its endocrine-disrupting properties and potential reproductive toxicity. The present study aimed to evaluate the concentration-dependent effects of DBP on morphology, cytotoxicity, oxidative stress, cell senescence, steroidogenesis and expression of related key developmental genes in cultured ovine granulosa cells in the presence of FSH. Granulosa cells were isolated from slaughterhouse-derived ovine ovaries and cultured in vitro with exposure to DBP at concentrations of 0, 1, 10, 25, 50, and 100 µM. Cytotoxic responses

Real-world study of medication-related osteonecrosis of the jaw from 2010 to 2023 based on Food and Drug Administration Adverse Event Reporting System.

Medication-related osteonecrosis of the jaw (MRONJ) is a rare and severe adverse drug reaction (ADR) commonly seen in people taking drugs prescribed for metastatic cancer and osteoporosis. Prior studies only analyzed this ADR utilizing the public database Food and Drug Administration Adverse Event Reporting System (FAERS) by different stages (2010-2014, 2015-2021); a more comprehensive study is needed to analyze the MRONJ cases and the associated drugs from a longer time span. We conducted a retrospective pharmacovigilance analysis for all reported MRONJ cases between 2010 and 2023 in FAERS, u

GPER in metabolic homeostasis and disease: molecular mechanisms, nutritional regulation, and therapeutic potential.

G protein-coupled estrogen receptor (GPER) significantly influences metabolic regulation by engaging multiple signaling pathways that affect overall metabolic health. Its structure and signaling mechanisms drive its diverse roles. Our team and others have successfully demonstrated that GPER modulates nutrient metabolism, spanning glucose, protein, and lipid metabolism, while also contributing to vitamin and mineral metabolism. Studies have linked GPER to metabolic disorders, including type 2 diabetes, nonalcoholic fatty liver disease, metabolic syndrome, and cardiovascular diseases, as well as

Tamoxifen- and Triptorelin-Induced Major Hypertriglyceridemia: A Case Report.

Tamoxifen, a selective estrogen receptor modulator (SERM), can have harmful side effects, such as hypertriglyceridemia, which can lead to acute pancreatitis. Meanwhile, triptorelin is an analog of natural GnRH (GnRHa), which may cause a small but significant increase in cholesterol and triglyceride (TG) levels. We describe below the case of a patient with breast cancer treated with Patey's operation, chemo-radiotherapy, and then with tamoxifen and triptorelin. After an exposure period of three months, she presented major hypertriglyceridemia at 56 g/L, total cholesterol at 13 g/L, LDL-choleste

Tamoxifen treatment ameliorates contractile dysfunction of Duchenne muscular dystrophy stem cell-derived cardiomyocytes on bioengineered substrates.

Duchenne muscular dystrophy (DMD) is a progressive genetic myopathy that leads to heart failure from dilated cardiomyopathy by early adulthood. Recent evidence suggests that tamoxifen, a selective estrogen receptor modulator widely used to treat breast cancer, ameliorates DMD cardiomyopathy. However, the mechanism of action of 4-hydroxytamoxifen, the active metabolite of tamoxifen, on cardiomyocyte function remains unclear. To examine the effects of chronic 4-hydroxytamoxifen treatment, we used state-of-the-art human-induced pluripotent stem cell-derived cardiomyocytes (iPSC-CMs) and a bioengi

Hormonal control of polyamine pools in experimental breast cancer in vivo: correlation with estrogen and progesterone receptor levels.

The present experiments were designed to test whether, in the hormone responsive N-nitrosomethyl-urea (NMU)-induced rat mammary tumor, polyamine levels are under hormonal control and whether they correlate with estrogen (ER) and progesterone (PgR) receptor content. We observed that tumor regression induced by ovariectomy was associated with a decline in putrescine (Pu), spermidine (Sd) and spermine (Sm). Administration of estradiol and perphenazine (to stimulate endogenous prolactin release) to castrated rats restored tumor growth and contents of Pu and Sd to control values in a time dependent

Effects of metal oxide inhalation on the transcription of some hormone receptors in the brain, examined in an in vivo mouse model.

Respirable metal oxide nanoparticles in welding fumes pose significant health risks upon inhalation, potentially leading to neurodegenerative diseases. While the exact mechanisms remain unclear, it is evident that metal oxide nanoparticles can disrupt cellular functions, including metabolism and inflammatory responses after crossing the blood-brain barrier (BBB). Our study investigates the impact of manual metal arc welding fumes on hormone receptor transcription in an in vivo mouse model. After collecting samples from six different brain regions at 24 and 96 h upon exposure, we focused on exp

The negative feedback actions of progesterone on gonadotropin-releasing hormone secretion are transduced by the classical progesterone receptor.

Progesterone (P) powerfully inhibits gonadotropin-releasing hormone (GnRH) secretion in ewes, as in other species, but the neural mechanisms underlying this effect remain poorly understood. Using an estrogen (E)-free ovine model, we investigated the immediate GnRH and luteinizing hormone (LH) response to acute manipulations of circulating P concentrations and whether this response was mediated by the nuclear P receptor. Simultaneous hypophyseal portal and jugular blood samples were collected over 36 hr: 0-12 hr, in the presence of exogenous P (P treatment begun 8 days earlier); 12-24 hr, P imp

Targeting Peroxisome Proliferator-Activated Receptor γ to Increase Estrogen-Induced Apoptosis in Estrogen-Deprived Breast Cancer Cells.

Peroxisome proliferator-activated receptor γ (PPARγ) is an important transcription factor that modulates lipid metabolism and inflammation. However, it remains unclear whether PPARγ is involved in modulation of estrogen (E2)-induced inflammation, thus affecting apoptosis of E2-deprived breast cancer cells, MCF-7:5C and MCF-7:2A. Here, we demonstrated that E2 treatment suppressed the function of PPARγ in both cell lines, although the suppressive effect in MCF-7:2A cells was delayed owing to high PPARγ expression. Activation of PPARγ by a specific agonist, pioglitazone, selectively blocked the i

Distribution and molecular drivers of the 21-gene recurrence score in early breast cancer with low, intermediate, and high estrogen receptor expression.

BACKGROUND: Estrogen receptor (ER)-low human epidermal growth factor receptor 2 (HER2)-negative breast cancer shares similar pathological and molecular features with triple-negative breast cancer, yet the utility of the 21-gene assay in this population remains uncertain. This study aimed to evaluate the distribution and molecular drivers of the 21-gene recurrence score (RS) in early breast cancer with low, intermediate, and high ER expression. METHODS: ER-positive, HER2-negative early breast cancer patients with 21-gene testing results from Shanghai Jiao Tong University Breast Cancer Database

GDF15 modulates mitochondrial content and differentiation-associated metabolic remodeling in skeletal muscle cells.

Growth differentiation factor 15 (GDF15) is a distant member of the transforming growth factor-β (TGF-β) superfamily increasingly implicated in metabolic regulation. Evidence points to GDF15 as a myokine that regulates systemic energy homeostasis, yet its role in skeletal muscle remains unclear. Here, we investigated whether GDF15 modulates mitochondrial phenotype during myogenesis using mouse C2C12 myoblasts and complementary models. Bioinformatic analyses of GDF15 interaction networks and gene ontology terms revealed enrichment for pathways related to cell differentiation and metabolic regul

Amlodipine with letrozole as maintenance post-chemotherapy: targeting calcium calcineurin calmodulin pathway and aromatase inhibitor in disseminated endometrioid adenocarcinoma of the endometrium-a case report.

World is now clinging to immune checkpoint inhibitor in almost every type of cancer. PD-1 and PD-L1 inhibitor are taking over the treatment algorithm. But cost is an issue; we have a perspective to target energy mechanism of cytotoxic T cells to modulate the representation of PD-1 receptor so as to increase immunogenicity of cytotoxic T cells. Cellular energy mechanism is largely dependent on calcium channels and pharmacological modulation of it may help in modulating response of cytotoxic T cell. We hereby present a case of endometroid adenocarcinoma of endometrium successfully treated with C

A pilot study on the use of a 7-day course of letrozole followed by misoprostol for the termination of early pregnancy up to 63 days.

BACKGROUND: Our previous randomized controlled study showed that the complete abortion rate in termination of pregnancy up to 63 days with the combined use of letrozole for 3 days followed by vaginal misoprostol was significantly higher than that of misoprostol alone. A positive correlation was observed between the basal estradiol level and the failure rate. We performed this pilot study to assess if a longer course of letrozole followed by misoprostol would improve the estradiol suppression and the complete abortion rate of pregnancy up to 63 days. STUDY DESIGN: Twenty subjects requesting le

[Genetic predisposition for breast cancer: BRCA1 and BRCA2 genes].

The discovery of genes BRCA1 and BRCA2 has led to the introduction of genetic tests more complex every time for the evaluation of the hereditary cancer risk, among others. In the present paper we explore the criteria to decide when to run the testing for the genes, as well as the implications for the treatment of patients who are identified with them.

Improved antitumoral properties of pure antiestrogen RU 58668-loaded liposomes in multiple myeloma.

In most of multiple myeloma (MM) cells, the "pure" antiestrogen (AE) RU 58668 (RU) induced either a G1-arrest (LP-1, OPM-2, NCI-H929, U266 cells) or apoptosis (RPMI 8226 cells). In RPMI 8226 cells, RU activates a caspase-dependent cell death pathway leading to the release of cytochrome c, the decrease of the essential MM survival factor Mcl-1, the cleavage of Bid and the activation of caspases-3 and -8. Incorporation of RU in pegylated cholesterol-containing liposomes allowed a controlled RU release, improving its anti-proliferative and apoptotic effects in cells. In RPMI 8226 xenografts, i.v.

A nuclear receptor corepressor modulates transcriptional activity of antagonist-occupied steroid hormone receptor.

Synthetic steroid hormone antagonists are clinically important compounds that regulate physiological responses to steroid hormones. The antagonists bind to the hormone receptors, which are ligand-inducible transcription factors, and modulate their gene-regulatory activities. In most instances, a steroid receptor, such as progesterone receptor (PR) or estrogen receptor (ER), is transcriptionally inactive when complexed with an antagonist and competitively inhibits transactivation of a target steroid-responsive gene by the cognate hormone-occupied receptor. In certain cellular and promoter conte
