BACKGROUND: Mood disorders, including anxiety and depression, are frequently diagnosed in multiple sclerosis (MS) patients, even independently of the disabling symptoms associated with the disease. Anatomical, biochemical, and pharmacological evidence indicates that type-1 cannabinoid receptor (CB1R) is implicated in the control of emotional behavior and is modulated during inflammatory neurodegenerative diseases such as MS and experimental autoimmune encephalomyelitis (EAE).
METHODS: We investigated whether CB1R could exert a role in anxiety-like behavior in mice with EAE. We performed behav

Dementia with Lewy bodies (DLB) patients frequently experience well formed recurrent complex visual hallucinations (RCVH). This is associated with reduced blood flow or hypometabolism on imaging of the primary visual cortex. To understand these associations in DLB we used pathological and biochemical analysis of the primary visual cortex to identify changes that could underpin RCVH. Alpha-synuclein or neurofibrillary tangle pathology in primary visual cortex was essentially absent. Neurone density or volume within the primary visual cortex in DLB was also unchanged using unbiased stereology. M

Tobacco usage typically precedes illicit drug use in adolescent and young adult populations. Several animal studies suggest nicotine increases the risk for subsequent cocaine abuse, and may be a negative prognostic factor for treatment of cocaine addiction; i.e., a "gateway drug". Neurotensin (NT) is a 13-amino acid neuropeptide that modulates dopamine, acetylcholine, glutamate, and GABA neurotransmission in brain reward pathways. NT69L, a NT(8-13) analog, blocks behavioral sensitization (an animal model for psychostimulant addiction) to nicotine, and nicotine self-administration in rats. The

Salicylate affects central respiratory control. The inspiratory neurons are the most important component of the medullary respiratory control center because they modulate the final motor output via the phrenic nerve. We investigated changes in burst rate, intraburst firing frequency, and membrane properties of inspiratory neurons in the isolated brainstem after the administration of salicylate. Newborn rat brainstem-spinal cord preparations were superfused with salicylate. Whole-cell recordings were performed from inspiratory neurons. Application of 1 mM salicylate caused an increase in the in

The involvement of GABA mediated neurotransmission in the central control of respiration was investigated by administration of the specific GABAA receptor agonist muscimol and the specific GABAA receptor antagonist biculline into the fourth cerebral ventricle of the rabbit. Cycle-triggered averaging of the phrenic nerve activity (PNA) was used to quantify drug-induced changes of the central respiratory pattern. Muscimol reduced the peak amplitude of PNA and increased the duration of the respiratory phases. High amounts of muscimol led to a long-lasting but reversible central apnea. Bicuculline

OBJECTIVE: Pathogenic variants in the calcium/calmodulin-dependent protein kinase II B gene (CAMK2B) have been associated with neurodevelopmental disorders, including epilepsy, yet the mechanisms underlying cortical dysfunction remain largely unclear. Building on our previous clinical report of a patient carrying the CaMKIIβ P213L variant and our prior characterization of the corresponding mouse models, we investigated how P213L-associated CaMKIIβ insufficiency alters cortical network dynamics and susceptibility to pentylenetetrazol (PTZ)-induced seizures in vivo.

Neurodegenerative diseases, such as Alzheimer's disease (AD), are characterized by oxidative imbalance, leading to memory deficits and cognitive impairments. Aluminium chloride (AlCl3), a neurotoxin found in certain foods and medications, disrupts neurotransmitter systems, thereby exacerbating cognitive decline. Current drug development strategies aim to counter these effects through cholinesterase inhibition, activation of GABAergic transmission, the use of antioxidants, and the promotion of neuroprotection. This work was conducted to assess the neuroprotective properties of the aqueous extra

Depression is characterized by a dysregulated brain-gut axis. Xiaoyao San (XYS), a classic Traditional Chinese Medicine formula for soothing Liver and strengthening Spleen, is clinically effective in alleviating depression. However, the systems-level mechanisms by which XYS coordinates gut-brain communication to exert its antidepressant effects remain insufficiently understood. This study aimed to systematically elucidate the antidepressant mechanisms of XYS, with a focus on identifying a key gut-derived metabolic pathway that modulates prefrontal cortex (PFC) function. A mouse model of depres

PURPOSE: To investigate how glutamate and glutamine levels are established in the aqueous humor by identifying the transporters and metabolism pathways that contribute to the differential accumulation of glutamate and glutamine between the distinct epithelial cell layers that constitute the ciliary body.
METHODS: Postembedding immunohistochemistry and silver intensification were used to quantify the relative distributions of glutamate, glutamine, and related amino acids (aspartate, alanine, GABA, and glycine) in the pigmented (PE) and nonpigmented (NPE) epithelial cells of the ciliary body. F

Serotonergic (5-HT) cells in the rat dorsal raphe nucleus (DRN) appear in topographically organized groups. Based on cellular morphology, expression of other neurotransmitters, afferent and efferent connections and functional properties, 5-HT neurons of the DRN have been grouped into six cell clusters. The subdivisions comprise the rostral, ventral, dorsal, lateral, caudal and interfascicular parts of the DRN. In addition to 5-HT cells, neurons containing γ-aminobutyric acid (GABA), glutamate, dopamine, nitric oxide and the neuropeptides corticotropin-releasing factor, substance P, galanin, ch

Social memory, the ability to recall and comprehend social interactions, depends on hippocampal circuits, particularly the dorsal CA2. Disruptions in inhibitory transmission within CA2 have been implicated in neuropsychiatric and neurodevelopmental disorders, including schizophrenia and autism spectrum disorder. However, the extrahippocampal mechanisms regulating CA2-dependent social memory remain poorly understood. The medial septum (MS), a major source of hippocampal afferents involved in network synchronization and information processing, has emerged as a potential modulator of social memor

Emotional intelligence (EI) has emerged as a critical neurocognitive construct linking affective processing, social behavior, and adaptive functioning. This integrative narrative review synthesizes multidisciplinary findings to map the biological underpinnings of EI across genetic, epigenetic, neuroanatomical, and neurophysiological domains. Drawing on evidence from neuroimaging, molecular genetics, neurochemistry, and brain connectivity studies, the review suggests EI as a product of dynamic interactions between prefrontal-limbic circuits, neurotransmitter systems, and environmentally sensiti

Plant-derived exosome-like vesicles (PDEVs) have emerged as promising oral delivery carriers, yet their application is severely limited by gastrointestinal instability and poor intestinal absorption. To overcome these barriers, we developed a biohybrid nanoplatform by sequentially coating wheat-seedling PDEVs with trimethyl chitosan (TMC) and carboxymethyl chitosan (CMC) via layer-by-layer self-assembly. This engineering strategy confers enhanced physicochemical stability, effectively protecting the payload against gastric degradation while enabling targeted intestinal release. Mechanistically

The leaves of Shiliangcha (an alternative tea, Chimonanthus salicifolius), a perennial bush cultivated in Lishui, Zhejiang, have been used for thousands of years by the She ethnic group as an herb and tea. Chimonanthus salicifolius flavonoids (CsFE) have exhibited remarkable antiaging properties. Therefore, we established a d-galactose (d_Gal)-induced aging mouse model to investigate the effect of CsFE on the central nervous system (CNS) of aging hosts. Supplementation with CsFE effectively alleviated symptoms of aging in mice, including weight loss, declining learning and memory capacity, blo

INTRODUCTION: Primary dysmenorrhea, defined by painful menstruation in the absence of pelvic disorders, often requires treatment. Traditional options such as NSAIDs and oral contraceptives have limitations. This systematic review and meta-analysis evaluate the efficacy of Valeriana officinalis (Valerian) in managing primary dysmenorrhea.
METHODS: We conducted a comprehensive literature search (2000-November 2023) for RCTs comparing valerian to placebo or NSAIDs in primary dysmenorrhea. The primary outcome was a reduction in dysmenorrhea pain. Risk of bias was assessed using Cochrane's tool.

Sleep deprivation (SD) induces significant neurobiological changes, including oxidative stress, neuroinflammation, and behavioural impairments. This study was designed as a proof of concept to assess the potential for modulating the effects of SD through a short-term seven-day administration of Cornus mas (C. mas) in a rapid eye movement (REM) SD rodent paradigm. Adult male Wistar rats were randomised in four groups (n = 7): control, C. mas (CM), sleep deprivation (SD), and sleep deprivation with C. mas (SD + CM). Behaviourally, SD induced hyperactivity and hyperlocomotion. SD determined histo

Panic disorder is more frequent in women than in men. In women, vulnerability to panic is enhanced during the late luteal phase of the menstrual cycle. At this time secretion of progesterone and its neuroactive metabolite allopregnanolone (ALLO), which acts as a positive allosteric modulator of the actions of GABA at GABAA receptors, decline sharply. In female rats, responsiveness to a hypoxic panicogenic challenge increases during the late diestrus (LD) phase as ALLO concentration in the brain falls. During LD, short-term treatment with fluoxetine at a low dose (1.75 mg/kg i.p.) blocked panic

BACKGROUND: Mechanoreceptor activation modulates GABA neuron firing and dopamine (DA) release in the mesolimbic DA system, an area implicated in reward and substance abuse. The lateral habenula (LHb), the lateral hypothalamus (LH), and the mesolimbic DA system are not only reciprocally connected, but also involved in drug reward. We explored the effects of mechanical stimulation (MS) on cocaine addiction-like behaviors and the role of the LH-LHb circuit in the MS effects. MS was performed over ulnar nerve and the effects were evaluated by using drug seeking behaviors, optogenetics, chemogeneti

PURPOSE: Inhibitory synaptic currents from fast-spiking neurons (FSNs), a typical gamma-aminobutyric acid (GABA)ergic interneuron in the cerebral cortex, to pyramidal neurons are facilitated by insulin. FSNs frequently show electrical synapses to FSNs, however, the effect of insulin on these electrical synapses is unknown. The aim of this study was to evaluate effects of insulin on electrical synaptic potentials between FSNs.
METHODS: Electrical synaptic potentials via gap junctions between FSNs were recorded to examine how insulin modulates these potentials in the rat insular cortex (IC).
