Hepatitis C virus NS5B triggers an MDA5-mediated innate immune response by producing dsRNA without the replication of viral genomes.

During the replication of viral genomes, RNA viruses produce double-stranded RNA (dsRNA), through the activity of their RNA-dependent RNA polymerases (RdRps) as viral replication intermediates. Recognition of viral dsRNA by host pattern recognition receptors - such as retinoic acid-induced gene-I (RIG-I)-like receptors and Toll-like receptor 3 - triggers the production of interferon (IFN)-β via the activation of IFN regulatory factor (IRF)-3. It has been proposed that, during the replication of viral genomes, each of RIG-I and melanoma differentiation-associated gene 5 (MDA5) form homodimers f

Species-specific engagement of human nucleotide oligomerization domain 2 (NOD)2 and Toll-like receptor (TLR) signalling upon intracellular bacterial infection: role of Crohn's associated NOD2 gene variants.

Recognition of bacterial peptidoglycan-derived muramyl-dipeptide (MDP) by nucleotide oligomerization domain 2 (NOD2) induces crucial innate immune responses. Most bacteria carry the N-acetylated form of MDP (A-MDP) in their cell membranes, whereas N-glycolyl MDP (G-MDP) is typical for mycobacteria. Experimental murine studies have reported G-MDP to have a greater NOD2-stimulating capacity than A-MDP. As NOD2 polymorphisms are associated with Crohn's disease (CD), a link has been suggested between mycobacterial infections and CD. Thus, the aim was to investigate if NOD2 responses are dependent

Campylobacter jejuni drives MyD88-independent interleukin-6 secretion via Toll-like receptor 2.

Gastrointestinal disease caused by Campylobacter jejuni is characterized by localized inflammation and the destruction of the epithelial cell barrier that forms host innate protection against pathogens. This can lead to an imbalance in fluid transport across the gastrointestinal tract, resulting in severe diarrhea. The mechanisms of host cell receptor recognition of C. jejuni and downstream immune signaling pathways leading to this inflammatory disease, however, remain unclear. The aim of this study was to analyze the mechanisms involved in C. jejuni induction of the acute-phase inflammatory r

The antimicrobial peptide HBD-2 and the Toll-like receptors-2 and -4 are induced in synovial membranes in case of septic arthritis.

Septic arthritis is frequently observed especially in immune-compromised or chronically diseased patients and leads to functional impairment due to tissue destruction. Recently, production of antimicrobial peptides (AMP) was observed in articular cartilage after exposure to bacteria. This report examines the role of synoviocyte-derived AMPs in innate defense mechanisms of articular joints. Samples of healthy, low-grade synovialitis and septic synovial membranes were assessed for the expression of human beta-defensin-2 (HBD-2) and Toll-like receptor-2 and -4 (TLR) by immunohistochemistry and en

Binding of Macrophage Receptor MARCO, LDL, and LDLR to Disease-Associated Crystalline Structures.

Endogenous and exogenous crystalline structures are involved in various pathologies and diseases in humans by inducing sterile inflammation, mechanical stress, or obstruction of excretory organs. The best studied of these diseases is gout, in which crystallization of uric acid in the form of monosodium urate (MSU) mainly in synovial fluid of the joints leads to sterile inflammation. Though some of these diseases have been described for centuries, little is known about if and how the immune system recognizes the associated crystals. Thus, in this study we aimed at identifying possible recogniti

Neutrophil-derived ROS as a rapid functional biomarker: diagnostic and prognostic performance of the Leukocyte ImmunoTest in infection and sepsis.

BACKGROUND: Early diagnosis of sepsis remains a major clinical challenge due to the dynamic interplay between infection and host immune response. Conventional biomarkers often fail to capture the dynamic nature of immune activation. Neutrophil-derived reactive oxygen species (ROS), central to antimicrobial defense and tissue injury, may offer early insight into immune dysregulation. The Leukocyte ImmunoTest (LIT) is a rapid, bedside assay that quantifies neutrophil ROS production within minutes, providing a functional snapshot of innate immunity. METHODS: This prospective observational study

Innate immune responses in human hepatocyte-derived cell lines alter genotype 1 hepatitis E virus replication efficiencies.

Hepatitis E virus (HEV) is a significant health problem in developing countries causing sporadic and epidemic forms of acute viral hepatitis. Hepatitis E is a self-limiting disease; however, chronic HEV infections are being reported in immunocompromised individuals. The disease severity is more during pregnancy with high mortality (20-25%), especially in third trimester. Early cellular responses after HEV infection are not completely understood. We analyzed innate immune responses associated with genotype-I HEV replication in human hepatoma cell lines (Huh7, Huh7.5 and HepG2/C3A) using HEV rep

Microbial dysbiosis drives colorectal carcinogenesis via integrated inflammatory, metabolic, and biofilm pathways.

Colorectal cancer (CRC) arises from a multifaceted interplay among the intestinal microbiota, chronic inflammation, and host genomic instability, with microbial dysbiosis serving as an active driver rather than a by-product of malignant transformation. Genotoxic Escherichia coli (colibactin-positive), enterotoxigenic Bacteroides fragilis, and Fusobacterium nucleatum contribute to distinct stages of CRC progression by engaging the DNA-damage response and activating β-catenin-dependent Wnt signaling and NF-κB/STAT3 transcriptional programs controlling pro-inflammatory (IL-6, IL-8), pro-survival

Volatile Compounds in Feathers of the Scarlet Macaw (Ara macao) Reflect Sex and Pair-Bond Status.

Birds were traditionally considered to have a limited sense of smell; however, increasing evidence indicates that some taxa, including parrots, possess a functional olfactory system capable of detecting volatile compounds. These chemical cues may convey information related to identity, reproductive status, and mate quality. This study investigates the diversity of volatile compounds in the feathers of scarlet macaws (Ara macao) and examines whether their chemical composition varies according to sex, age, and pair-bond status, as well as whether plumage odor is associated with immune function.

Programmed inhibition of an innate immune receptor via de novo designed transmembrane proteins.

Transmembrane domains of immune complexes transmit precise signals across lipid bilayers. Probing their interactions has the potential to yield mechanistic insights relevant to therapeutic design. However, our capability to generate molecules that bind lipid-embedded sites is limited. Here, we created synthetic polypeptides targeting a prominent mediator of inflammatory signaling directly within membranes, Toll-like receptor 4 (TLR4), and clarify structural principles of its cross-membrane signaling mechanism. Doing so, we test emerging design principles for computationally encoding protein in

Parvovirus B19-associated arthropathy: a contemporary narrative review.

Parvovirus B19 is a common and frequently under-recognised cause of acute-onset inflammatory polyarthropathy in adults, with a clinical phenotype that can closely mimic early rheumatoid arthritis. This review synthesises the adult-focused literature on epidemiology, immunopathogenesis, clinical presentation, diagnostic strategy, and management of parvovirus B19-associated arthropathy, with attention to the renewed circulation observed in recent years and its implications for clinical practice. In adults, joint involvement typically manifests as abrupt, painful, symmetric polyarticular disease

A novel mannose-binding lectin homolog identified in the large yellow croaker (Larimichthys crocea) with evident binding and agglutinating activities.

Lectins are a large group of carbohydrate-binding proteins that widely participate in innate immune response, however, our understanding of its subfamily mannose-binding lectins (MBLs) is still in paucity for commercially important fish species such as the large yellow croaker (Larimichthys crocea). In this study, a novel MBL homolog (designated as galactose-binding lectin, LcGalBL-1, due to its galactose preference) was identified in L. crocea through transcriptomic screening. In addition to bioinformatic characterization, expression profiling of LcGalBL-1 in different tissues and upon pathog

Optimal development of apoptotic cells-mimicking liposomes targeting macrophages.

Macrophages are multifunctional innate immune cells that play indispensable roles in homeostasis, tissue repair, and immune regulation. However, dysregulated activation of macrophages is implicated in the pathogenesis of various human disorders, making them a potential target for treatment. Through the expression of pattern recognition and scavenger receptors, macrophages exhibit selective uptake of pathogens and apoptotic cells. Consequently, the utilization of drug carriers that mimic pathogenic or apoptotic signals shows potential for targeted delivery to macrophages. In this study, a serie

Analysis of critical protein-protein interactions of SARS-CoV-2 capping and proofreading molecular machineries towards designing dual target inhibitory peptides.

In recent years, the emergence of severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2), as the cause of the coronavirus disease (COVID-19) global pandemic, and its variants, especially those with higher transmissibility and substantial immune evasion, have highlighted the imperative for developing novel therapeutics as sustainable solutions other than vaccination to combat coronaviruses (CoVs). Beside receptor recognition and virus entry, members of the SARS-CoV-2 replication/transcription complex are promising targets for designing antivirals. Here, the interacting residues that media

Molecular Docking and Dynamics Simulation Studies of a Dataset of NLRP3 Inflammasome Inhibitors.

BACKGROUND: The organism's defense against aggressive agents is performed by the innate immune system via activation of pattern-recognition receptors (PRRs). Initially, these agents are recognized by the immune system, resulting in the inflammatory response that activates the pathogen elimination and tissue repair. Inflammasomes are macromolecules related to the host's response to endo or exogenous aggressive agents. Thus, inflammation mediated by inflammasomes plays an important role in the pathogenesis of diseases, such as neurodegenerative disorders, autoimmune diseases, and type 2 diabetes

RNA-Seq Analysis Reveals the Role of Omp16 in Brucella-Infected RAW264.7 Cells.

Brucellosis is an endemic zoonotic infectious disease in the majority of developing countries, which causes huge economic losses. As immunogenic and protective antigens at the surface of Brucella spp., outer membrane proteins (Omps) are particularly attractive for developing vaccine and could have more relevant role in host-pathogen interactions. Omp16, a homolog to peptidoglycan-associated lipoproteins (Pals), is essential for Brucella survival in vitro. At present, the functions of Omp16 have been poorly studied. Here, the gene expression profile of RAW264.7 cells infected with Brucella suis

Staphylococcal infections and infertility: mechanisms and management.

Infertility is a subject of worldwide concern as it affects approximately 15% of couples. Among the prime contributors of infertility, urogenital bacterial infections have lately gained much clinical importance. Staphylococcal species are commensal bacteria and major human pathogens mediating an array of reproductive tract infections. Emerging evidences are 'bit by bit' revealing the mechanisms by which Staphylococci strategically disrupt normal reproductive functions. Staphylococcal species can directly or through hematogenous routes can invade the reproductive tissues. In the testicular cell

Effect of Natural Compounds on NK Cell Activation.

Natural killer (NK) cells are lymphocytes of the innate immune system that survey the body for stressed and abnormal cells. The integration of signals that they receive through various inhibitory and activating cell surface receptors controls their activation and ability to kill target cells and produce cytokines. In this manner, phenotypically and functionally distinct subsets of NK cells help protect against microbial infections and cancer and shape the adaptive immune response. NK cells can use two different mechanisms to kill their targets, either by cytotoxic granule exocytosis or by indu

Cathepsin B-Deficient Mice Resolve Leishmania major Inflammation Faster in a T Cell-Dependent Manner.

A critical role for intracellular TLR9 has been described in recognition and host resistance to Leishmania parasites. As TLR9 requires endolysosomal proteolytic cleavage to achieve signaling functionality, we investigated the contribution of different proteases like asparagine endopeptidase (AEP) or cysteine protease cathepsins B (CatB), L (CatL) and S (CatS) to host resistance during Leishmania major (L. major) infection in C57BL/6 (WT) mice and whether they would impact on TLR9 signaling. HSV-2 genome recognition by nuclear cGAS instigates IFN-β production and influences inflammasome activation during de novo infection in HFF cells.

INTRODUCTION: Type I interferon response, specifically, the cGAS-cGAMP-STING axis that results in IFN-β response, is well known for its complex roles early during viral infection. Previous reports suggest that HSV-1 DNA in Thp-1 cells and HIV-2 dsDNA in DCs and macrophages could be sensed by cGAS. The nuclear DNA sensor IFI16's viral DNA sensing leads to its acetylation, cytoplasmic translocation and STING activation and inflammasome activation. Although cGAS is known to be associated with IFI16 in the nucleus, however, during HSV-2 infection, the role of nuclear cGAS in viral DNA sensing, inf
