Effects of AFQ056 on language learning in fragile X syndrome.

BACKGROUNDFXLEARN, the first-ever large multisite trial of effects of disease-targeted pharmacotherapy on learning, was designed to explore a paradigm for measuring effects of mechanism-targeted treatment in fragile X syndrome (FXS). In FXLEARN, the effects of metabotropic glutamate receptor type 5 (mGluR5) negative allosteric modulator (NAM) AFQ056 on language learning were evaluated in 3- to 6-year-old children with FXS, expected to have more learning plasticity than adults, for whom prior trials of mGluR5 NAMs have failed.METHODSAfter a 4-month single-blind placebo lead-in, participants wer

Language-informed treatment recommendations in psychiatry: bridging the gap in individual predictability.

Treatment recommendation systems in psychiatry perform suboptimally, largely due to low individual-level predictability from conventional clinical predictors. We argue that language captured through clinical interviews and electronic health records provides a rich, workflow-native substrate for estimating individualized treatment effects. We highlight opportunities for language-informed systems using modern language models and outline key challenges that must be addressed for safe and scalable deployment.

How much do language and context affect the accuracy of AI health advice?

OBJECTIVES: Large language models (LLMs) are increasingly used for health advice, but evidence on how performance varies across languages, topics and sources remains limited. We assess linguistic and contextual variation in Artificial Intelligence (AI) health-claim verification. METHODS: We evaluated seven widely accessible LLMs using UK and European Union (EU) health-claim registers and 9088 journalist-vetted PUBHEALTH claims. The register-language analysis used authorised entries translated into 21 languages; status-labelled UK/EU extensions added non-authorised entries for diagnostic check…

Engineering of Generative Artificial Intelligence and Natural Language Processing Models to Accurately Identify Arrhythmia Recurrence.

BACKGROUND: Large language models (LLMs) such as Chat Generative Pre-trained Transformer (ChatGPT) excel at interpreting unstructured data from public sources, yet are limited when responding to queries on private repositories, such as electronic health records (EHRs). We hypothesized that prompt engineering could enhance the accuracy of LLMs for interpreting EHR data without requiring domain knowledge, thus expanding their utility for patients and personalized diagnostics. METHODS: We designed and systematically tested prompt engineering techniques to improve the ability of LLMs to interpret…

Modern resources for intrinsic disorder predictions: protein language models, deep learning, meta-servers, and databases.

Computational prediction of intrinsic disorder in protein sequences is an impactful and growing research area, recently infused with deep learning and protein language models, prompting the need to assess the impact of these advancements. We systematically surveyed 128 disorder predictors, many of which are accurate, and some that have been cited thousands of times. We demonstrated that recent methods utilizing protein language models outperform those that do not, particularly when combined with deep learning, yielding substantial gains in predictive quality. We place these observations within

Toward Sensor-to-Text Generation: Leveraging LLM-Based Video Annotations for Stroke Therapy Monitoring.

Stroke-related impairment remains a leading cause of long-term disability, limiting individuals' ability to perform daily activities. While wearable sensors offer scalable monitoring solutions during rehabilitation, they struggle to distinguish functional from non-functional movements, and manual annotation of sensor data is labor-intensive and prone to inconsistency. In this paper, we propose a novel framework that uses large language models (LLMs) to generate activity descriptions from video frames of therapy sessions. These descriptions are aligned with concurrently recorded accelerometer s…

Advancing Adverse Drug Reaction Prediction with Deep Chemical Language Model for Drug Safety Evaluation.

The accurate prediction of adverse drug reactions (ADRs) is essential for comprehensive drug safety evaluation. Pre-trained deep chemical language models have emerged as powerful tools capable of automatically learning molecular structural features from large-scale datasets, showing promising capabilities for the downstream prediction of molecular properties. However, the performance of pre-trained chemical language models in predicting ADRs, especially idiosyncratic ADRs induced by marketed drugs, remains largely unexplored. In this study, we propose MoLFormer-XL, a pre-trained model for enco

Dorsal language stream anomalies in an inherited speech disorder.

Speech disorders are highly prevalent in the preschool years, but frequently resolve. The neurobiological basis of the most persistent and severe form, apraxia of speech, remains elusive. Current neuroanatomical models of speech processing in adults propose two parallel streams. The dorsal stream is involved in sound to motor speech transformations, while the ventral stream supports sound/letter to meaning. Data-driven theories on the role of these streams during atypical speech and language development are lacking. Here we provide comprehensive behavioural and neuroimaging data on a large nov

An Evidence-Constrained LLM Framework for Automated Abdominal CTA Aneurysm Analysis and Reporting.

Abdominal computed tomography angiography (CTA) aneurysm analysis requires simultaneous localization, measurement, and anatomical attribution of multiple candidate lesions across a continuous three-dimensional vascular network, followed by translation of these findings into clinically reviewable CTA report drafts. Existing automated approaches largely focus on segmentation, measurement, or candidate detection, whereas direct large language model (LLM) interpretation of CTA images remains vulnerable to factual errors, hallucinations, and safety concerns. We propose an evidence-constrained large
