Role of the Antigen Capture Pathway in the Induction of a Neutralizing Antibody Response to Anthrax Protective Antigen.

Toxin neutralizing antibodies represent the major mode of protective immunity against a number of toxin-mediated bacterial diseases, including anthrax; however, the cellular mechanisms that lead to optimal neutralizing antibody responses remain ill defined. Here we show that the cellular binding pathway of anthrax protective antigen (PA), the binding component of anthrax toxin, determines the toxin neutralizing antibody response to this antigen. PA, which binds cellular receptors and efficiently enters antigen-presenting cells by receptor-mediated endocytosis, was found to elicit robust anti-P

Validating community concerns of menstrual changes associated with COVID-19 vaccination using a self-controlled case series analysis of real-world data.

OBJECTIVES: Stories of menstrual changes occurring post COVID-19 vaccination have abounded, with many affected persons expressing frustration their concerns were not being heard. In an era where misinformation is rampant and can fuel vaccine hesitancy it is imperative to address and validate community concerns. We aimed to investigate evidence of increased menstrual disturbances associated with COVID-19 vaccination. METHODS: We adopted a two-pronged approach; firstly, scrutinising social-media for discussions on menstrual changes associated with COVID-19 vaccination using our deep learning fr

Sweet complexity: O-linked protein glycosylation in pathogenic Neisseria.

The genus Neisseria, which colonizes mucosal surfaces, includes both commensal and pathogenic species that are exclusive to humans. The two pathogenic Neisseria species are closely related but cause quite different diseases, meningococcal sepsis and meningitis (Neisseria meningitidis) and sexually transmitted gonorrhea (Neisseria gonorrhoeae). Although obvious differences in bacterial niches and mechanisms for transmission exists, pathogenic Neisseria have high levels of conservation at the levels of nucleotide sequences, gene content and synteny. Species of Neisseria express broad-spectrum O-

Genetics, Structure, and Function of Group A Streptococcal Pili.

Streptococcus pyogenes (Group A Streptococcus; GAS) is an exclusively human pathogen. This bacterial species is responsible for a large variety of infections, ranging from purulent but mostly self-limiting oropharynx/skin diseases to streptococcal sequelae, including glomerulonephritis and rheumatic fever, as well as life-threatening streptococcal toxic-shock syndrome. GAS displays a wide array of surface proteins, with antigenicity of the M protein and pili utilized for M- and T-serotyping, respectively. Since the discovery of GAS pili in 2005, their genetic features, including regulation of

Discovery of protease inhibitors from bacteria as novel adjuvants for oral vaccine formulations.

INTRODUCTION: There is a need for mucosal vaccines that can fight pathogens at the site of infection. At present, there are no approved adjuvants for mucosal vaccines. Among different immunization routes, oral delivery is the natural choice because of its ease of administration. However, oral administration has two main drawbacks: proteolytic digestion and immune tolerance. METHODS: In this study, a systematic in silico screening of putative protease inhibitors (PIs) from bacteria to identify novel oral vaccine adjuvants was conducted. Selected candidates were then evaluated for their ability

Ether lipid vesicle-based antigens impart protection against experimental listeriosis.

BACKGROUND: Incidence of food-borne infections from Listeria monocytogenes, a parasite that has adapted intracellular residence to avoid antibody onslaught, has increased dramatically in the past few years. The apparent lack of an effective vaccine that is capable of evoking the desired cytotoxic T cell response to obliterate this intracellular pathogen has encouraged the investigation of alternate prophylactic strategies. It should also be noted that Archaebacteria (Archae) lipid-based adjuvants enhance the efficacy of subunit vaccines. In the present study, the adjuvant properties of archaeo

Synergistic Immunoenhancement of Recombinant PBP2a Vaccine Candidate by Biogenic Selenium Nanoparticles and MF59 Against Methicillin-Resistant Staphylococcus Aureus.

Methicillin-resistant Staphylococcus aureus remains a major global health concern due to its resistance to β-lactam and multiple other classes of antibiotics. Herein, we evaluated a recombinant penicillin-binding protein 2a (r-PBP2a) vaccine candidate formulated with biogenic selenium nanoparticles (SeNPs) and the MF59 adjuvant. While r-PBP2a was purified under denaturing conditions and subsequently refolded for use as an immunogen, biogenic selenium nanoparticles were prepared and characterized in terms of morphology, charge, and safety. Female BALB/c mice were assigned to experimental groups

Design, development, and assessment of a novel multi-peptide vaccine targeting PspC, PsaA, and PhtD proteins of Streptococcus pneumoniae.

Pneumococcus is the top cause of diseases such as pneumonia/meningitis, and of secondary infections after viral respiratory diseases like COVID-19/flu. Pneumococcal protein-based vaccines consisting of proteins with various functions in virulence might provide a qualified alternative for present vaccines. In this project, PspC, PsaA, and PhtD proteins were considered to anticipate B/T-cell epitopes using immunoinformatics to develop 4 multi-peptide constructs (C, A, and D individual constructs, and a fusion construct CAD). We tested whether vaccination with CAD is able to elicit more efficient

A novel mAb broadly neutralizes SARS-CoV-2 VOCs in vitro and in vivo, including the Omicron variants.

Novel immune escape variants have emerged as severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) continues to spread worldwide. Many of the variants cause breakthrough infections in vaccinated populations, posing great challenges to current antiviral strategies targeting the immunodominance of the receptor-binding domain within the spike protein. Here, we found that a novel broadly neutralizing monoclonal antibody (mAb), G5, provided efficient protection against SARS-CoV-2 variants of concern (VOCs) in vitro and in vivo. A single dose of mAb G5 could significantly inhibit the viral bu

[Novel vaccines against M. tuberculosis].

CDC and ACET in U.S.A. reported that novel vaccines instead of BCG are required for the protection against infection of Mycobacterium tuberculosis worldwide. However, no novel vaccine for clinical use has not yet been developed in the world including U.S.A. and Europe. We have developed two novel tuberculosis (TB) vaccines; a DNA vaccine combination expressing mycobacterial heat shock protein 65 (HSP 65) and interleukin-12 (IL-12) by using the hemagglutinating virus of Japan (HVJ)-liposome (HSP 65 + IL-12/HVJ). Identification of Salmonella enterica serovar Enteritidis mutants overproducing OMVs as vaccine candidates.

Salmonella enterica serovar Enteritidis (S. Enteritidis) remains one of the predominant serotypes causing infections in both humans and animals. In poultry farms, it is frequently reported as the leading cause of infection. Therefore, the development of vaccines against S. Enteritidis is considered a key strategy to control bacterial infections in poultry. Outer membrane vesicles (OMVs) have emerged as promising vaccine candidates for preventing bacterial infections in both animals and humans. However, the natural yield of OMVs from wild-type Salmonella is low, limiting large-scale application

Leveraging genomic conservation in emerging SARS-CoV-2 variants for precision diagnostics, vaccines, and RNAi therapeutics.

BACKGROUND: The ongoing evolution of SARS-CoV-2 has led to the emergence of numerous variants, challenging the efficacy of existing diagnostics, vaccines, and therapeutics. Despite this genomic variability, several regions of the viral genome remain highly conserved, offering stable targets for pan-variant intervention strategies. METHODS: This study employed a multi-pronged approach integrating immunoinformatics, RNA interference (RNAi), and molecular diagnostics to design mutation-resilient tools against SARS-CoV-2. Conserved genomic regions-including NSP7-NSP16 (except NSP15), ORF6, E, M,

Genome-Guided Discovery of Vaccine Targets for a Multi-Epitope Construct Against Multidrug-Resistant Enterobacter cloacae.

Multidrug-resistant (MDR) Enterobacter cloacae has emerged as a serious public health threat, particularly in hospital settings, due to its ability to cause severe infections and rapidly acquire antibiotic resistance. The limited effectiveness of current antibiotics highlights the urgent need for alternative preventive strategies such as vaccination. In this study an immunoinformatics-based approach was employed to design a multi epitope vaccine (MEV) targeting the conserved and surface exposed outer membrane protein A (OmpA) of MDR E. cloacae. The entire proteome was screened to identify suit

Immunoinformatics-based design of artificial chimeric proteins as universal vaccine candidates against foot-and-mouth disease virus serotypes A, O, and SAT2.

Foot-and-mouth disease virus (FMDV) remains a major constraint to livestock health due to its high mutation rate and serotype diversity. Currently, FMDV vaccines, primarily inactivated whole-virus formulations, have significant limitations, including limited cross-protection, high production costs, and potential biosafety risks. To address the need for broad-spectrum protection, this study aimed to design a universal vaccine candidate by rationally constructing artificial chimeric proteins (ACPs) integrating conserved structural (VP1-VP3) and non-structural (3 A, 3 C) proteins from the predomi

Diltiazem inhibits SARS-CoV-2 cell attachment and internalization and decreases the viral infection in mouse lung.

The continuous emergence of severe acute respiratory coronavirus 2 (SARS-CoV-2) variants and the increasing number of breakthrough infection cases among vaccinated people support the urgent need for research and development of antiviral drugs. Viral entry is an intriguing target for antiviral drug development. We found that diltiazem, a blocker of the L-type calcium channel Cav1.2 pore-forming subunit (Cav1.2 α1c) and an FDA-approved drug, inhibits the binding and internalization of SARS-CoV-2, and decreases SARS-CoV-2 infection in cells and mouse lung. Cav1.2 α1c interacts with SARS-CoV-2 spi

Dihydrolipoamide acetyltransferase is a key factor mediating adhesion and invasion of host cells by Mycoplasma synoviae.

Mycoplasma synoviae is a significant avian pathogen responsible for chronic respiratory diseases, arthritis, and infectious synovitis in chickens and turkeys. These infections result in substantial economic losses to the global poultry industry. Dihydrolipoamide acetyltransferase (E2) is a multifunctional protein that plays an indispensable role in energy metabolism and redox balance and is also a key virulence factor of various pathogens. In this study, we used the avian pathogen M. synoviae as a model to identify the role of the E2 protein in the colonization and invasion of host cells. Firs

Susceptibility of naturally human papillomavirus type 16 major capsid protein L1 variants to vaccines and predictions of future evolutionary trends.

Human papillomavirus type 16 (HPV16) causes over 50% of HPV-related cervical cancers. With the widespread use of virus-like particle (VLP)-based HPV prophylactic vaccines, investigating the differences in viral assembly, infectivity and antigenic properties among different variants is highly important for further eliminating diseases caused by HPV. We analyzed HPV16 L1 protein sequences from the NCBI database and identified 14 representative clusters. Pseudovirus (PsV) formation, infectivity, and susceptibility to vaccines were assessed for these variants, and molecular dynamics (MD) simulatio

Evaluation of the contribution of the minor envelope complex glycoprotein 3 ectodomain to the porcine reproductive and respiratory syndrome virus 1 neutralizing antibody response.

Porcine reproductive and respiratory syndrome viruses (PRRSVs) cause significant economic losses to the global swine industry. Current live attenuated vaccines are unable to effectively control PRRSV infection, and strain variability, driven by rapid mutation, complicates the development of improved vaccines. Identification of the targets of broadly neutralizing antibodies is crucial for guiding next-generation vaccine design, yet PRRSV glycoproteins bearing conserved neutralizing epitopes remain poorly defined. To address this, recombinant soluble forms of PRRSV-1 envelope glycoproteins, GP2,
