A new chemotype that opens the S2∗ pocket of the 3CL protease exhibits antiviral activity against SARS-CoV-2.

Coronaviruses have caused 3 pandemics in the past 2 decades. The SARS-CoV-2, the causative agent of the COVID-19 already led to more than 7 million deaths worldwide. The 3CL protease (3CLpro, Mpro) plays a key role in the replication of the SARS-CoV-2 and has been validated as promising therapeutic target as evidenced by the recent clinical use of anti-coronavirus drugs inhibitors of this protease. In addition, its high conservation among coronaviruses gives an opportunity to identify broad-spectrum anti-coronavirus compounds to combat not only SARS-CoV-2 but also emerging coronaviruses. Here

[Agonist-antagonist myoneural interface (AMI) : Innovative treatment option for lower limb amputees?].

The agonist-antagonist myoneural interface (AMI) is an innovative approach to restoring proprioception and achieving more intuitive motor control following limb loss. This cutting-edge technique replicates the natural biomechanical relationship between agonist and antagonist muscles, enabling bidirectional communication between a prosthesis and the user's peripheral nervous system. Through the transposition of neurovascularly pedicled agonist-antagonist muscle pairs, which are reconnected via an adapted tendon suture and positioned within a gliding mechanism, AMI generates proprioceptive feedb

The clinical use of epigenetics in psychiatry: a narrative review of epigenetic mechanisms, key candidate genes, and precision psychiatry.

The etiology of psychiatric disorders is complex, involving both genetic and environmental factors with emerging evidence suggesting that epigenetic modifications, including DNA methylation, histone modifications, and non-coding RNA regulation, significantly contribute to mental health. The epigenome influences the development of psychiatric disorders and human behavior and may be considered in clinical observations. Epigenetic changes have been well-established in BDNF, COMT, FKBP5, NR3C1, SLC6A4, and DRD2, genes associated with psychiatric disorders, including schizophrenia, major depressive

H6N6 Avian Influenza Virus Infection Induced Pyroptosis of M1 Macrophages by Activating Caspase-1.

The H6N6 avian influenza virus has expanded its host range from birds to mammals. Some strains can now bind to human-like receptors, raising concerns about human infection. Although H6N6 is a low-pathogenic avian influenza virus (LPAIV), it is unclear whether it triggers pyroptosis in human lungs, a process linked to cytokine storms in infections like H7N9. Here, we found that the chicken-origin H6N6 LPAIV can effectively replicate in and infect human alveolar macrophages and their M1 macrophages. Viral infection of M1 macrophages upregulated the mRNA levels of NLRP3, caspase-1, and Gasdermin

The experimentally evolved fluconazole-resistant clade II isolates of Candidozyma auris exhibit a distinct lipid compositional landscape, highlighting intraclade sphingolipid heterogeneity.

The intrinsic resistance of Candidozyma auris (C. auris) to antifungal drugs poses a major therapeutic challenge, with conventional resistance mechanisms providing only partial explanations. Sphingolipids (SLs), known for their interclade heterogeneity, play a crucial role in antifungal resistance. This study examined the SL landscape in two drug-susceptible clade II isolates, C-line and P-line, from distinct geographical origins, which were experimentally evolved to develop stable fluconazole (FLC) resistance. The progenitors displayed distinct SL profiles, P1 had higher PhytoCer and αOHPhyto

Description of a Virulent Systemic Feline Calicivirus Infection in a Kitten with Footpads Oedema and Fatal Pneumonia.

Feline calicivirus (FCV) is widespread in multi-cat environments and typically causes acute upper respiratory tract disease (URTD). FCV also causes outbreaks of virulent systemic disease (VSD), mainly in adults, with multiple organ involvement. In this study, an FCV-VSD infection was described in a less-one-month-old Maine Coon kitten originating from a cattery where an outbreak of FCV-URTD had previously been reported. After spontaneous death, post-mortem examination as well as histopathological, immunohistochemical, bacteriological and virological investigations were carried out. Pathologica

Regulatory Strategies for Accelerating the Translation of Gene Therapies to Clinical Practice: Focus on GMO Considerations.

Gene therapy has revolutionized modern medicine by offering innovative treatments for genetic disorders, cancers, and immune-related conditions through technologies such as viral vector delivery, genome editing, and genetically modified cell therapies. Despite significant advancements, the classification of gene therapy medicinal products (GTMPs) as genetically modified organisms (GMOs) under EU legislation imposes significant regulatory burdens, hindering early and timely patient access to such therapies. Current GMO regulations, originally designed for agricultural biotechnology, require env

BioIron: Origin, Chemical Properties, and Biological Functions.

Iron is a highly abundant element, essential for life on earth and integral to various biological processes. It originated from stellar nucleosynthesis, particularly in supernovae type Ia, which produced iron used in planetary formation. Life's emergence on earth, about 3.5 billion years ago, occurred in an oxygen-free atmosphere, with iron playing a pivotal role in early biochemical reactions. Hydrothermal vents, rich in iron-sulfur (Fe-S) minerals, provided the conditions for prebiotic chemistry and CO2 fixation, potentially driving the synthesis of key organic compounds. Iron-sulfur cluster

Rebamipide Attenuates Lupus Nephritis by Enhancing Antioxidative Defense in Podocytes: Evidence from a Lupus-Prone Mouse Model.

Systemic lupus erythematosus (SLE) is a prototypical autoimmune disease that affects various organs, including the kidneys. Despite recent advancements, effective treatment options for renal involvement in SLE remain limited. Rebamipide, originally developed as a gastroprotective agent, has been reported to exert immunomodulatory effects in rheumatic diseases. Here, we aimed to evaluate the therapeutic potential of rebamipide in SLE using an animal model and to elucidate its mechanisms of action. We administered rebamipide or vehicle control to lupus-prone MRL/lpr mice and evaluated its effica

From stem cells to organoids in thyroid: Useful tools or a step for cell therapy?

Organoids are three-dimensional tissue cultures derived from stem cells. They replicate the complexity of different cell types in an organ and can survive in specific media. They also have the capacity to proliferate and self-renew. Thyroid organoids have now been established using a variety of cell sources, including murine or human embryonic stem cells, pluripotent stem cells, adult thyroid-derived stem cells, and even fetal thyroids. Experimental designs to obtain thyroid organoids depend on the originating cell types and whether a forced (transient or permanent) overexpression of two impor

A deep insight into the sialome of the house fly, Musca domestica, infected with the salivary gland hypertrophy virus (MdSGHV).

The house fly, Musca domestica, serves as a mechanical vector for numerous pathogens, posing a significant risk to human and animal health. More than two decades ago, the Musca domestica salivary gland hypertrophy virus (MdSGHV) was discovered, infecting both males and females flies and disrupting mating and the reproductive process. While MdSGHV can infect various tissues, its primary replication site is the house fly salivary gland. It is well established that arthropod salivary glands play an important role not only in acquiring food but also in transmitting pathogens. Therefore, understand

Extracellular vesicles in viral disease management.

INTRODUCTION/BACKGROUND: Extracellular vesicles (EVs) are non-replicating lipid-bilayered bodies that are naturally released by a cell that aid in various biological functions including cell-to-cell communication. They resemble the cells that they originate from, mimicking their composition and contents. The shared Endosomal Sorting Complex Required for Transport (ESCRT) mechanism between virions and EVs allows EVs to aid in the dispersion and infection of viruses. SCOPE/OBJECTIVES: The aim of this review is to encapsulate important studies that highlight the potential use of EVs in diagnosis

Modeling sporadic Alzheimer's disease in mice by combining Apolipoprotein E4 risk gene with environmental risk factors.

INTRODUCTION: Developing effective treatment for Alzheimer's disease (AD) remains a challenge. This can be partially attributed to the fact that the mouse models used in preclinical research largely replicate familial form of AD, while majority of human cases are sporadic; both forms differ widely in the onset and origin of pathology, therefore requiring specific/targeted treatments. METHODS: In this study, we aimed to model sporadic AD in mice by combining two of the many risk factors that are strongly implicated in AD: ApoE4, a major genetic risk factor, together with an inflammatory stimul

Current View on Major Natural Compounds Endowed with Antibacterial and Antiviral Effects.

Nowadays, infectious diseases of bacterial and viral origins represent a serious medical problem worldwide. In fact, the development of antibiotic resistance is responsible for the emergence of bacterial strains that are refractory even to new classes of antibiotics. Furthermore, the recent COVID-19 pandemic suggests that new viruses can emerge and spread all over the world. The increase in infectious diseases depends on multiple factors, including malnutrition, massive migration of population from developing to industrialized areas, and alteration of the human microbiota. Alternative treatmen

Study of different heterocycles showing significant anti-severe acute respiratory syndrome 2 activity in vitro and in vivo.

BACKGROUND AND AIM: With the emergence of severe acute respiratory syndrome-related coronavirus (SARS-CoV-2), antiviral drug development has gained increased significance due to the high incidence and potentially severe complications of the resulting coronavirus infection. Heterocycle compounds, acting as antimetabolites of DNA and RNA monomers, rank among the most effective antiviral drugs. These compounds' antiviral effects on various SARS-CoV-2 isolates, as found in existing data collections, form the basis for further research. The aim of this study was to examine the possible antiviral ef

A Review: Biomechanical Aspects of the Fallopian Tube Relevant to its Function in Fertility.

The fallopian tube (FT) plays a crucial role in the reproductive process by providing an ideal biomechanical and biochemical environment for fertilization and early embryo development. Despite its importance, the biomechanical functions of the FT that originate from its morphological aspects, and ultrastructural aspects, as well as the mechanical properties of FT, have not been studied nor used sufficiently, which limits the understanding of fertilization, mechanotrasduction, and mechanobiology during embryo development, as well as the replication of the FT in laboratory settings for infertili

Amino Acids at Positions 156 and 332 in the E Protein of the West Nile Virus Subtype Kunjin Virus Classical Strain OR393 Are Involved in Plaque Size, Growth, and Pathogenicity in Mice.

The West Nile virus (WNV) subtype Kunjin virus (WNVKUN) is endemic to Australia. Here, we characterized the classical WNVKUN strain, OR393. The original OR393 strain contained two types of viruses: small plaque-forming virus (SP) and large plaque-forming virus (LP). The amino acid residues at positions 156 and 332 in the E protein (E156 and E332) of SP were Ser and Lys (E156S/332K), respectively, whereas those in LP were Phe and Thr (E156F/332T). SP grew slightly faster than LP in vitro. The E protein of SP was N-glycosylated, whereas that of LP was not. Analysis using two recombinant single-m

When practice does not make a perfect - paradoxical learning curve in schizophrenia and bipolar disorder revealed by different serial reaction time task variants.

INTRODUCTION: Our previous studies identified a paradoxical implicit motor learning curve in schizophrenia (SZ) and bipolar disorder (BD) patients. This study aimed to verify whether those previously observed deficits may be captured by a new version of the ambidextrous serial reaction time task (SRTT), prepared for use in the MRI. METHODS: This study involved 186 participants. A total of 97 participants (33 BD, 33 SZ, and 31 healthy controls, HCs) completed the original, unlimited time response variant of SRTT. A total of 90 individuals (30 BD, 30 SZ, and 30 HCs) underwent a newer, limited r

Further validation of the Multidimensional Version of the Fear-of-Self Questionnaire in a Persian non-clinical and clinical obsessive-compulsive disorder sample.

Previous research has shown that vulnerable self-themes and feared self-perceptions may play an important role in the development and maintenance of obsessive-compulsive disorder (OCD). In particular, the recently validated Multidimensional Version of the Fear-of-Self Questionnaire (FSQ-MV) has shown strong relationships with OCD symptoms independent of cognitive constructs and negative mood in non-clinical samples. The current study aimed to further evaluate the validity and reliability of a Persian version of the FSQ-MV in OCD patients (N = 300), as well as non-clinical individuals (N = 300)
