Ethyl maltol disrupt iron homeostasis in SH-SY5Y neuroblastoma cell line.

Ethyl Maltol (EM) is a commonly used flavoring compound and has been reported to bind iron and facilitate iron transport. Since EM is membrane permeable, the potential that it disrupts intracellular iron homeostasis was investigated. EM increased the labile iron pool in SH-SY5Y cells and increased iron-responsive protein activity using a reporter assay in the HEK293 cells. EM induced the expression of transferrin receptor 1 messenger RNA (mRNA) and decreased the expression of ferritin light chain protein in SH-SY5Y cells. Expression of the iron-responsive amyloid precursor protein attenuated t

Identification of genetic polymorphisms modulating nausea and vomiting in two series of opioid-treated cancer patients.

Nausea and vomiting are often associated with opioid analgesia in cancer patients; however, only a subset of patients develop such side effects. Here, we tested the hypothesis that the occurrence of nausea and vomiting is modulated by the genetic background of the patients. Whole exome sequencing of DNA pools from patients with either low (n = 937) or high (n = 557) nausea and vomiting intensity, recruited in the European Pharmacogenetic Opioid Study, revealed a preliminary association of 53 polymorphisms. PCR-based genotyping of 45 of these polymorphisms in the individual patients of the same

TRRAP is a central regulator of human multiciliated cell formation.

The multiciliated cell (MCC) is an evolutionarily conserved cell type, which in vertebrates functions to promote directional fluid flow across epithelial tissues. In the conducting airway, MCCs are generated by basal stem/progenitor cells and act in concert with secretory cells to perform mucociliary clearance to expel pathogens from the lung. Studies in multiple systems, including Xenopus laevis epidermis, murine trachea, and zebrafish kidney, have uncovered a transcriptional network that regulates multiple steps of multiciliogenesis, ultimately leading to an MCC with hundreds of motile cilia

Molecular, serological and epidemiological observations after a suspected outbreak of plague in Nyimba, eastern Zambia.

Plague is a re-emerging zoonotic disease caused by the bacterium Yersinia pestis. The disease has caused periodic global devastation since the first outbreak in the 6th century. Two months after a suspected plague outbreak in Nyimba district, samples were collected from 94 livestock (goats and pigs), 25 rodents, 6 shrews and 33 fleas. Enzyme-linked immunosorbent assay (ELISA) and polymerase chain reaction (PCR) techniques were used to investigate the presence of Y. pestis, which showed that 16.0% (4/25) of rodents, 16.7% (1/6) of shrews (Crocidura spp) and 6.0% (5/83) of goats were positive fo

Genes and Pathways Involved in Adult Onset Disorders Featuring Muscle Mitochondrial DNA Instability.

Replication and maintenance of mtDNA entirely relies on a set of proteins encoded by the nuclear genome, which include members of the core replicative machinery, proteins involved in the homeostasis of mitochondrial dNTPs pools or deputed to the control of mitochondrial dynamics and morphology. Mutations in their coding genes have been observed in familial and sporadic forms of pediatric and adult-onset clinical phenotypes featuring mtDNA instability. The list of defects involved in these disorders has recently expanded, including mutations in the exo-/endo-nuclease flap-processing proteins MG

Sox9 coordinates a transcriptional network in pancreatic progenitor cells.

During pancreas development, both the exocrine and endocrine lineages differentiate from a common pool of progenitor cells with similarities to mature pancreatic duct cells. A small set of transcription factors, including Tcf2, Onecut1, and Foxa2, has been identified in these pancreatic progenitor cells. The Sry/HMG box transcription factor Sox9 is also expressed in the early pancreatic epithelium and is required for normal pancreatic exocrine and endocrine development in humans. In this study, we found Sox9 in mice specifically expressed with the other progenitor transcription factors in both

Genetic basis of tuberculosis susceptibility in India.

Tuberculosis is a complex disease resulting from the responses of immunological, genetic and environmental factors to the chronic infectious agent, Mycobacterium tuberculosis. Several genetic factors have been implicated in host disease susceptibility and the prevalence of a disease in a population may be equal to the product of the frequencies of the susceptible alleles present in the population living in an endemic area. The endogamous, sympatrically isolated gene pools, exposed to the highly infectious environmental load of India, is an ideal model to study tuberculosis susceptibility. Our …

Efficient Myoinositol biosynthesis from glucose through gluconate bypass of glycolysis.

Microbial myo-inositol (MI) production typically requires deletion of conditionally essential genes such as phosphoglucose isomerase (pgi in E. coli) to increase pools of the precursor glucose-6-phosphate (G6P). This disrupts growth and has necessitated complex glycerol co-feeding strategies. Here, we address this limitation by implementing a synthetic gluconate-bypass (GBP) central metabolism that establishes an alternative route by which glucose-derived carbon enters central metabolism, allowing robust growth despite pgi deletion. However, pgi deletion alone was insufficient to improve MI pr

Operationalizing accreditation standards for laboratory-developed next-generation sequencing validation of CFTR amplicon-based assays.

INTRODUCTION: Application of College of American Pathologists (CAP) standards for molecular next-generation sequencing assays, typically high-complexity laboratory-developed tests, requires assay-specific, principled translation. We sought to systematically apply CAP laboratory-developed test validation requirements for next-generating sequencing to targeted CFTR gene sequencing on an Illumina iSeq100 Sequencing System and to address assay-specific translational guidance gaps. METHODS: We evaluated the analytic performance (accuracy, precision, analytic sensitivity, and specificity) and other

The potential of MRI radiomics based on extrapulmonary metastases in predicting EGFR mutations: a systematic review and meta-analysis.

BACKGROUND: Epidermal growth factor receptor (EGFR) gene mutations can lead to distant metastasis in non-small cell lung cancer (NSCLC). When the primary NSCLC lesions are removed or cannot be sampled, the EGFR status of the metastatic lesions are the potential alternative method to reflect EGFR mutations in the primary NSCLC lesions. This review aimed to evaluate the potential of magnetic resonance imaging (MRI) radiomics based on extrapulmonary metastases in predicting EGFR mutations through a systematic reviews and meta-analysis. Dermanyssus gallinae as a pathogen vector: Phylogenetic analysis and associated health risks in pigeons.

Dermanyssus gallinae (D. gallinae) is a nocturnal, blood-feeding ectoparasite that primarily infests poultry, causing significant economic losses. This study aimed to identify poultry red mites and detect associated pathogens using morphological and molecular techniques. Mites and blood samples were collected from naturally infected pigeons. Mite species and blood parasites were identified morphologically, while representative pooled mite samples were cultured on general and selective media to detect microbial pathogens. Molecular identification of mites was conducted using PCR targeting the c

Development of an Enzyme-Linked Immunosorbent Spot Assay for the Assessment of Adeno-Associated Virus Peptides to Examine Immune Safety.

Adeno-associated virus (AAV)-based gene therapies have shown promise as novel treatments for rare genetic disorders such as hemophilia A and spinal muscular atrophy. However, cellular immune responses mediated by cytotoxic (CD8+) and helper (CD4+) T cells may target vector-transduced cells as well as healthy immune cells, impacting safety and efficacy. In this study, we describe the optimization and reproducibility of interferon-γ (IFNγ)-based and interleukin-2 (IL-2)-based enzyme-linked immunosorbent spot (ELISpot) assays for measuring T cell responses against AAV peptide antigens. For method

Identification of New Microfoci and Genetic Characterization of Tick-Borne Encephalitis Virus Isolates from Eastern Germany and Western Poland.

(1) Background: Tick-borne encephalitis (TBE) is the most important tick-borne viral disease in Eurasia, although effective vaccines are available. Caused by the tick-borne encephalitis virus (TBEV, syn. Orthoflavivirus encephalitidis), in Europe, it is transmitted by ticks like Ixodes ricinus and Dermacentor reticulatus. TBEV circulates in natural foci, making it endemic to specific regions, such as southern Germany and northeastern Poland. Our study aimed to identify new TBEV natural foci and genetically characterize strains in ticks in previously nonendemic areas in Eastern Germany and West

Distinct impacts of fat and fructose on the liver, muscle, and adipose tissue metabolome: An integrated view.

OBJECTIVE: In the last years, changes in dietary habits have contributed to the increasing prevalence of metabolic disorders, such as non-alcoholic fatty liver disease (NAFLD) and type 2 diabetes mellitus (T2DM). The differential burden of lipids and fructose on distinct organs needs to be unveiled. Herein, we hypothesized that high-fat and high-fructose diets differentially affect the metabolome of insulin-sensitive organs such as the liver, muscle, and different adipose tissue depots. METHODS: We have studied the impact of 12 weeks of a control (11.50% calories from fat, 26.93% from protein

Brick tea consumption and its relationship with fluorosis in Tibetan areas.

Brick tea-type fluorosis (BTF) due to a high intake of brick tea is possible in Tibetan populations, and dental fluorosis (DF) and skeletal fluorosis (SF) are its primary manifestations. To determine the prevalence of DF and SF and their relationships with brick tea intake in Tibetan populations, a literature review was conducted for studies published between 1994 and 2021. The available evidence revealed that brick tea may be produced from older stems and leaves of the tea plant and that the fluoride content of brick tea exceeds the national standard. The harsh environment of the plateau has
