Targeting cattle for malaria elimination: marked reduction of Anopheles arabiensis survival for over six months using a slow-release ivermectin implant formulation.

BACKGROUND: Mosquitoes that feed on animals can survive and mediate residual transmission of malaria even after most humans have been protected with insecticidal bednets or indoor residual sprays. Ivermectin is a widely-used drug for treating parasites of humans and animals that is also insecticidal, killing mosquitoes that feed on treated subjects. Mass administration of ivermectin to livestock could be particularly useful for tackling residual malaria transmission by zoophagic vectors that evade human-centred approaches. Ivermectin comes from a different chemical class to active ingredients

Effects of climate and different management strategies on Aedes aegypti breeding sites: a longitudinal survey in Brasília (DF, Brazil).

OBJECTIVE: To determine the influence of climate and of environmental vector control with or without insecticide on Aedes aegypti larval ind … ypti adults for the area is described. CONCLUSION: In a moderately infested area such as Brasilia, insecticides do not improve environmental vector control. Rather, infestations could be further reduced by focusing on residences and containers particularly at risk. The nature of the link between climate and larval population should be investigated in larger-s …

Functional Profiling of the A-Family of Venom Peptides from the Wolf Spider Lycosa shansia.

The venoms of spiders from the RTA (retro-lateral tibia apophysis) clade contain diverse short linear peptides (SLPs) that offer a rich source of therapeutic candidates. Many of these peptides have insecticidal, antimicrobial and/or cytolytic activities, but their biological functions are unclear. Here, we explore the bioactivity of all known members of the A-family of SLPs previously identified in the venom of the Chinese wolf spider (Lycosa shansia). Our broad approach included an in silico analysis of physicochemical properties and bioactivity profiling for cytotoxic, antiviral, insecticida

Rational spatio-temporal strategies for controlling a Chagas disease vector in urban environments.

The rational design of interventions is critical to controlling communicable diseases, especially in urban environments. In the case of the Chagas disease vector Triatoma infestans, successful control is stymied by the return of the insect after the effectiveness of the insecticide wanes. Here, we adapt a genetic algorithm, originally developed for the travelling salesman problem, to improve the spatio-temporal design of insecticide campaigns against T. infestans, in a complex urban environment. We find a strategy that reduces the expected instances of vector return 34-fold compared with the c

[Toxic effect of beta-cipermethrin, deltamethrin and fenitrothion in colonies of Triatoma dimidiata (Latreille, 1811) and Triatoma maculata (Erichson, 1848) (Hemiptera, Reduviidae)].

INTRODUCTION: The susceptibility to insecticides of triatomine species must be evaluated because of their involvement in the transmission of the Chagas disease. In each region with Chagas endemicity, evaluation of insecticide response is necessary to predict the success of the control campaigns. OBJECTIVE: The baseline susceptibility was determined for the active principles deltamethrin, beta-cypermethrin and fenitrothion in nymphs of first and fifth instar of Triatoma dimidiata and nymphs of first instar of Triatoma maculata. MATERIALS AND METHODS: The insecticide activity in triatomines wa…

Recent advances in the study of knockdown resistance mutations in Aedes mosquitoes with a focus on several remarkable mutations.

The Aedes mosquito, which transmits the dengue fever virus and other viruses, has acquired resistance to pyrethroid insecticides in a naturally selective manner. Massive use of insecticides has led to the worldwide expansion of resistant populations. The major factor in pyrethroid resistance is knockdown resistance (kdr) caused by amino acid mutation(s) in the voltage-gated sodium channel, which is the target site of this insecticide group. Some kdr mutations can lead to a dramatic increase in resistance, and multiple mutations can increase the level of pyrethroid resistance by 10 to several-h…

Long-term efficacy of fluralaner (Exzolt®) in Gallus gallus domesticus against epidemiologically relevant triatomines (Hemiptera: Reduviidae: Triatominae): a potential complementary strategy for Chagas disease control.

BACKGROUND: Chagas disease control relies primarily on vector control using pyrethroid insecticide sprays with residual action in domestic and peridomestic environments. However, the necessity for repeated applications and the development of pyrethroid resistance in some countries have undermined this strategy. Alternative control tools are needed, and host-targeted systemic insecticides offer an alternative approach by creating toxic blood meals for hematophagous vectors. In peridomestic settings, chickens are major blood meal sources for triatomines. This study evaluated the insecticidal act…

Novel Approach in the Synthesis of Some Pyrimidines with Eminent Toxicological Effects towards Aphis gossypii (Glover, 1887) and Outstanding Safety against Earthworms (Aporrectodea caliginosa).

This study reports the synthesis and bioevaluation of ten novel pyranothiazolopyrimidine derivatives. Their insecticidal efficacy against Aphis gossypii (Glover, 1887) revealed compounds 3 and 10 as lead candidates, exhibiting toxicity comparable to the commercial standard acetamiprid insecticide. Notably, these compounds demonstrated a markedly safer ecotoxicological profile toward the earthworms (Aporrectodea caliginosa), with LC50 values orders of magnitude higher than acetamiprid over 5- and 10-day exposures. Application also induced measurable changes in soil chemistry. These findings ide

Impact of plant monoterpenes on insect pest management and insect-associated microbes.

The fight against insect pests primarily relies on the utilization of synthetic insecticides. However, improper application of these chemicals can lead to detrimental effects on both the environment and human health, as well as foster the development of insect resistance. Consequently, novel strategies must be implemented to address the challenges stemming from the prolonged use of synthetic insecticides in agricultural and public health environments. Certain strategies involve the combination of crop protectants, which not only enhance insecticidal effectiveness but also reduce application ra…

Anopheles coluzzii from Sakassou, Central Côte d'Ivoire show aggravated resistance to pyrethroids and organophosphate but are fully susceptible to clothianidin.

BACKGROUND: Insecticide-based interventions, including insecticide-treated nets (ITNs) and indoor residual spraying (IRS), are central to malaria vector control in sub-Saharan Africa. In Côte d'Ivoire, increasing insecticide resistance in the key malaria vector, Anopheles gambiae sensu lato (An. gambiae s.l.), has been reported across the country, potentially compromising the current ITN-based control strategy. To assess the feasibility of supplementing control efforts with IRS using clothianidin as an alternative insecticide in areas with high malaria prevalence, we have examined the intensit

Exposure to low-concentration fipronil impairs survival, behavior, midgut morphology and physiology of Aedes aegypti larvae.

The Aedes aegypti mosquito is a vector for various arboviruses, including dengue and yellow fever. Insecticides, such as pyrethroids and organophosphates, are widely used to manage and control these insects. However, mosquitoes have developed resistance to these chemicals. Therefore, this study aimed to investigate the effects of the commercial formulation of fipronil (Tuit® Florestal; 80% purity) on the survival, behavior, morphology, and proteins related to signaling pathways of the midgut in A. aegypti larvae under controlled laboratory conditions. Significant reductions in immature surviva…
