Comparison of free serum oxylipin concentrations in hyper- vs. normolipidemic men.

Oxylipins, the oxidation products of unsaturated fatty acids (FA), are potent endogenous mediators being involved in the regulation of various biological processes such as inflammation, pain and blood coagulation. Compared to oxylipins derived from arachidonic acid (AA) by cyclooxygenase action, i.e. prostanoides, only limited information is available about the endogenous levels of hydroxy-, epoxy- and dihydroxy-FA of linoleic acid (LA), AA, α-linolenic acid (ALA), eicosapentaenoic acid (EPA) and docosahexaenoic acid (DHA) in humans. Particularly, it is unknown how metabolic disorders affect e

Gestational Diabetes Mellitus Remodels the Fetal Brain Fatty Acid Profile Through Placenta-Brain Lipid Axis in C57BL/6J Mice.

BACKGROUND: Polyunsaturated fatty acids (PUFAs), especially docosahexaenoic acid (DHA), are critical for proper fetal brain growth and development. Gestational diabetes mellitus (GDM) could affect maternal-fetal fatty acid metabolism. OBJECTIVE: This study aimed to explore the effect of GDM and high-fat (HF) diet on the DHA transport signaling pathway in the placenta-brain axis and fatty acid concentrations in the fetal brain. METHODS: Insulin receptor antagonist (S961) and HF diet were used to establish an animal model of GDM. Eighty female C57BL/6J mice were randomly divided into control (

Volume-sensitive chloride current activated by hyposmotic swelling in antral gastric myocytes of the guinea-pig.

The characteristics of volume-sensitive chloride current (ICl) induced by osmotic cell swelling were studied using the whole-cell patch-clamp technique and cell diameters of antral circular guinea-pig myocytes were simultaneously measured under isosmotic and hyposmotic conditions by using a video image analysis system. At -60 mV, osmotic cell swelling (200 mosmol/l) activated a sustained inward current. Instantaneous current/voltage (I-V) relations obtained by step voltage pulses showed an outward rectification. At potentials above +40 mV, the current exhibited time-dependent decay. The outwar

Artesunate improves glucose and lipid metabolism in db/db mice by regulating the metabolic profile and the MAPK/PI3K/Akt signalling pathway.

BACKGROUND: Diabetes is a metabolic disorder characterized by chronic hyperglycaemia. Chronic metabolic abnormalities and long-term hyperglycaemia may result in a wide range of acute and chronic consequences. Previous studies have demonstrated that artesunate(ART) has antidiabetic, anti-inflammatory, antiatherosclerotic, and other beneficial effects, but the specific regulatory mechanism is not completely clear. AIM: This study investigated the effects of ART on metabolic disorders in type 2 diabetes mellitus (T2DM) model db/db mice and explored the underlying mechanisms involved. METHODS: C

Impact of perinatal α-linolenic acid deficiency combined with high-fat diet consumption on tissue-specific fatty acid profiles in rats: Maternal and offspring analysis.

BACKGROUND: An imbalance in the dietary ratio of omega-6 (n-6) to omega-3 (n-3) polyunsaturated fatty acids (PUFAs) and/or a high-fat diet (HFD) may disrupt the metabolic conversion of essential PUFAs, such as linoleic acid (LA) and α-linolenic acid (ALA), into their long-chain derivatives (LC-PUFAs), thereby altering their accumulation in various tissues. To determine the effects of low maternal intake of ALA and/or an HFD on FA composition in maternal tissues (liver, adipose tissue, and erythrocytes) and offspring tissues (liver, erythrocytes, adipose tissue, heart, kidney, intestine, muscle

Evaluating dietary starch concentrations in reduced-forage rations fed to lactating dairy cattle.

Increasing frequency of severe weather events such as droughts and floods may reduce the inventory of high-quality forage available for feeding dairy cattle. Thus, optimizing rumen health and milk production when feeding reduced-forage rations is critical for dairy farmers and nutritionists. Our experiment investigated the effects of replacing NDF with starch in diets with 12.6% forage NDF on rumen metabolism, rumen inflammation, feed intake, and milk production. We hypothesized that increasing dietary starch would increase inflammatory cytokines and alter T lymphocytes in rumen tissue. We als

Lacticaseibacillus rhamnosus B6 alleviates metabolic dysfunction-associated fatty liver disease by suppressing intestinal LPS synthesis and regulating lipid metabolism.

INTRODUCTION: Metabolic dysfunction-associated fatty liver disease (MAFLD) has become a global epidemic with an unclear etiology and no effective therapeutic options. Disruption of the gut-liver axis driven by intestinal dysbiosis is closely implicated in MAFLD pathogenesis, making gut microbiota-targeted probiotic interventions promising preventive strategies. METHODS: Lacticaseibacillus rhamnosus B6, a probiotic strain isolated from homemade Bulgarian fermented milk, synthesizes immunomodulatory macromolecules and regulates the intestinal flora. In the present study, we comprehensively inve

Lipidomic signatures of heat tolerance in mungbean [Vigna radiata(L.) Wilczek] reveal reduced fatty acid unsaturation.

Heat stress is a major environmental constraint limiting plant growth and productivity, including in mungbean (Vigna radiata). Although physiological, biochemical, and molecular responses of mungbean to elevated temperature have been widely investigated, the role of heat stress-induced leaf lipidome reconfiguration in thermotolerance remains insufficiently understood. In this study, we examined physiological, biochemical, and lipid metabolic responses to heat stress in two contrasting mungbean genotypes: PI425243 (heat-tolerant) and PI223002 (heat-sensitive), grown under non-stress (34/25 °C d

Mediterranean Diet Pattern: Potential Impact on the Different Altered Pathways Related to Cardiovascular Risk in Advanced Chronic Kidney Disease.

BACKGROUND: Cardiovascular disease (CVD) remains the most common cause of mortality in chronic kidney disease (CKD) patients. Several studies suggest that the Mediterranean diet reduces the risk of CVD due to its influence on endothelial function, inflammation, lipid profile, and blood pressure. Integrating metabolomic and proteomic analyses of CKD could provide insights into the pathways involved in uremia-induced CVD and those pathways modifiable by the Mediterranean diet. METHODS: We performed metabolomic and proteomic analyses on serum samples from 19 patients with advanced CKD (aCKD) and

[Desaturases of fatty acids (FADS) and their physiological and clinical implication].

States associated with insulin resistance, as overweight/obesity, type 2 diabetes mellitus (DM2), cardiovascular diseases (CVD), some cancers and neuropsychiatric diseases are characterized with a decrease of long-chain polyunsaturated fatty acids (LC-PUFA) levels. Amounts of LC-PUFA depend on the exogenous intake of their precursors [linoleic (LA) and α-linolenic acid (ALA)] and by rate of their metabolism, which is influenced by activities of enzymes, such as Δ6-desaturase (D6D, FADS2), D5D, FADS1, elongases (Elovl2, -5, 6).Altered activities of D5D/D6D were described in plenty of diseases,
