Alterations in ADAMTS12 gene expression in salivary glands of radioiodine-131-administered rats.

PURPOSE: The aim of this study was to evaluate the alterations in ADAMTS12 expression after radioiodine-131 (RAI)-induced salivary gland damage. MATERIALS AND METHODS: A total of 30 Wistar male albino rats (260±45 g, 6 months old) were studied for ADAMTS12 gene expression levels and histological changes in the parotid and submandibular salivary glands of rats after the administration of RAI. A series of healthy rats were used as controls. A 3 mCi (111 MBq) dose of RAI was administered to rats in group 1 (n=6), group 2 (n=6), group 3 (n=6), and group 4 (n=6) to induce salivary gland damage. Ev

Activation of transforming growth factor beta 1 through integrin alpha V beta 6 and its molecular insights into cancer progression, and future directions.

Cancer metastasis is the last stage of cancer and can often lead to lethality. Cancer cells invade distal organs and establish an entirely new host microenvironment. Among several cancer-influencing mediators, TGF-β has been observed as a potential pro-oncogene to provoke metastasis. Among the 3 isoforms of TGF-β, TGF-β1 is identified as a strong promoter of several cancers, including colorectal, lung, gastric, and prostate cancer. TGF-β1 is always made by cells as an inactive, latent complex, which must be activated to function. A crucial activator of TGF-β is integrin αVβ6, which has a stron

Restoring disc matrix homeostasis: Dual-miRNA and human platelet lysate as a novel therapeutic strategy.

Intervertebral disc (IVD) degeneration is the predominant cause of low back pain, resulting from progressive extracellular matrix (ECM) degradation and establishment of a pro-catabolic microenvironment. microRNA (miRNA) delivery has potential to promote ECM restoration and curb the catabolic milieu. We previously demonstrated a robust anti-catabolic effect through the delivery of a dual-miRNA injectable therapy in ex vivo organ culture models of degeneration. However, to further enhance regenerative outcomes, additional stimulation, such as supplementation with human platelet lysate (HPL) may

Cooperative Mechanism of ADAMTS/ ADAMTSL and Fibrillin-1 in the Marfan Syndrome and Acromelic Dysplasias.

The term "fibrillinopathies" gathers various diseases with a wide spectrum of clinical features and severity but all share mutations in the fibrillin genes. The first described fibrillinopathy, Marfan syndrome (MFS), is a multisystem disease with a unique combination of skeletal, thoracic aortic aneurysm (TAA) and ocular features. The numerous FBN1 mutations identified in MFS are located all along the gene, leading to the same pathogenic mechanism. The geleophysic/acromicric dysplasias (GD/AD), characterized by short stature, short extremities, and joint limitation are described as "the mirror

ADAMTS18 deficiency leads to abnormal brain methylation metabolism, dysregulated neuroinflammatory response, and unsound blood-brain barrier structure in mice.

ADAMTS (A Disintegrin and Metalloproteinase with Thrombospondin motifs) family is a group of secretory proteases involved in the maintenance of central nervous system (CNS) homeostasis and neuronal disease. ADAMTS18 is a member of this family and has been linked to the integrity of the human brain's white matter. However, the cellular and molecular basis of ADAMTS18 in brain metabolism and homeostasis remains unclear. In this study, a total of 47,719 genes were identified in 8 independent wild type (WT) and Adamts18 knockout (KO) mouse brain samples using brain transcriptomic analysis. The abu

Analysis of genetic and clinical factors associated with buprenorphine response.

BACKGROUND: Buprenorphine, approved for treating opioid use disorder (OUD), is not equally efficacious for all patients. Candidate gene studies have shown limited success in identifying genetic moderators of buprenorphine treatment response. METHODS: We studied 1616 European-ancestry individuals enrolled in the Million Veteran Program, of whom 1609 had an ICD-9/10 code consistent with OUD, a 180-day buprenorphine treatment exposure, and genome-wide genotype data. We conducted a genome-wide association study (GWAS) of buprenorphine treatment response [defined as having no opioid-positive urine

Method for Generation of adamtsl4 Knock-Out Zebrafish Lines Using CRRISPR/Cas9 System.

Congenital ectopia lentis (CEL) is a rare pediatric ocular disorder characterized by zonular fiber defects leading to lens dislocation and is genetically heterogeneous. Among known causes, biallelic mutations in ADAMTSL-4 represent the second most common genetic contributor, frequently associated with ectopia pupillae (EP)-a distinct and clinically significant feature. However, the mechanisms by which ADAMTSL-4 mutations lead to these ocular abnormalities remain poorly understood, partly due to the lack of effective animal models. In this study, we generated adamtsl-4 knock-out zebrafish lines

The extracellular protease AdamTS-B is a dosage-sensitive regulator of unicellular tracheal branch morphogenesis.

Epithelial tube morphogenesis requires coordinated interactions between epithelial cells and their extracellular environment, yet the extracellular mechanisms that regulate this process remain poorly understood. The Drosophila trachea provides an excellent model for investigating how extracellular remodeling contributes to epithelial tube morphogenesis. AdamTS-B, an extracellular ADAMTS protease, is expressed in the embryonic trachea from early to late stages of tracheal development. Loss of AdamTS-B caused mild defects in unicellular branch organization, whereas overexpression produced ectopi

Weill-Marchesani Syndrome, a Rare Presentation of Severe Short Stature with Review of the Literature.

BACKGROUND Short stature is the second most common reason for referral to a pediatric endocrinology clinic. Numerous genetic causes have been identified. Weill-Marchesani syndrome (WMS) is one of the rare genetic disorders that cause short stature. It is caused by homozygous mutations in the FBN1 gene, ADAMTS10 gene, ADAMTS17 gene, or LTBP2 gene. Despite genetic heterogeneity, WMS is clinically homogeneous. It is characterized by short stature, brachydactyly, joint stiffness, ocular abnormalities, mainly microspherophakia and glaucoma, and occasionally cardiac defects. CASE REPORT A 9-year-old

Thrombosis, Microangiopathies, and Inflammation.

Thrombotic microangiopathy (TMA) is characterized by the presence of microangiopathic hemolytic anemia and thrombocytopenia. There are several disorders with varied etiopathogenesis, both genetic and acquired, that result in TMA. The neutrophils play an important role in inflammation and thrombosis through the formation of neutrophil extracellular traps (NETs). NETs are formed in response to a variety of stimuli including infections, chemical factors, and platelet activation. The classic TMA, thrombotic thrombocytopenic purpura (TTP) is caused by a severe deficiency of ADAMTS-13 (a disintegrin

The Protective Effect of Ligustilide in Osteoarthritis: An in Vitro and in Vivo Study.

BACKGROUND/AIMS: Osteoarthritis is a degenerative joint disease characterized by cartilage degeneration and a chondrocyte inflammatory response that induces an inflammatory environment closely linked to extracellular matrix (ECM) degradation. Ligustilide (LIG) is a major component of the herb Radix Angelicae Sinensis, with demonstrated anti-inflammatory effects. To confirm whether LIG has an equally inhibitory effect on inflammation in human osteoarthritis chondrocytes, we performed in vivo and in vitro experiments to validate the above conjectures and determine the relevant mechanisms. Mutations in ADAMTSL4 cause a unique form of autosomal-recessive congenital ectopia lentis.

Several unique mutations of ADAMTSL4 leading to congenital ectopia lentis (CEL) have been previously reported by our team. The purpose of this study is to find out the possible mechanism of a recurrent novel intronic variant in ADAMTSL4 led to CEL. Twelve novel ADAMTSL4 mutations with a unique form congenital ectopic lentis were detected previously by panel-based NGS. Genetic analysis verified a novel heterozygous ADAMTSL4 variation c.2177+4A > G on Intron 11 in two unrelated patients with iris and lens abnormalities. MINI-Gene assay showed two splicing modes of mRNA that process two different

Aucubin and Tanshinol from Bu-Shen-Huo-Xue-Fang (BSHXF) improve intervertebral disc degeneration through affecting nucleus pulposus cell.

Intervertebral disc degeneration (IDD) is characterized by the abnormal synthesis of extracellular matrix (ECM) proteins, increased inflammatory cytokines, and upregulated degradative enzymes. According to the Traditional Chinese Medicine (TCM) concept, the degenerative disease of the intervertebral disc falls under the heading of "stagnation syndrome." Bu-Shen-Huo-Xue-Fang (BSHXF) is commonly applied to the treatment of IDD. In the present study, IDD models were established in rats, and degeneration, including morphological and histopathological alterations, the loss of collagen and proteogly

The biology and pathophysiology of the proatherogenic metalloprotease ADAMTS7.

The ADAMTS (a disintegrin and metalloproteinase with thrombospondin type-1 motifs) metalloprotease family comprises 19 members, and they play critical roles in development, angiogenesis and coagulation. These enzymes are also pivotal in the turnover of extracellular matrix proteins in various tissues, and their dysregulation has been linked to diseases such as arthritis, atherosclerosis, cancer and inflammation. These secreted zinc metalloproteases are composed of multiple domains and are localized to the extracellular matrix and present in circulation. The metalloprotease member 7 of this fam

MMPs, ADAMs and ADAMTSs are associated with mammalian sperm fate.

Metalloproteinases include matrix metalloproteinases and disintegrin metalloproteinases. They are important members of the ECM degradation and reconstruction process and are associated with tissue development and disease. The ECM is a three-dimensional network of large molecules consisting of a variety of proteins. It is a physical scaffold for organs, and all types of cells can be found within the ECM. The testicle, where sperm are produced, is an organ that is constantly in dynamic flux. Metalloproteinases can regulate testicular tissue development and the maturation of sperm by affecting th

Extracellular metalloproteinases in neural crest development and craniofacial morphogenesis.

The neural crest (NC) is a population of migratory stem/progenitor cells that is found in early vertebrate embryos. NC cells are induced during gastrulation, and later migrate to multiple destinations and contribute to many types of cells and tissues, such as craniofacial structures, cardiac tissues, pigment cells and the peripheral nervous system. Recently, accumulating evidence suggests that many extracellular metalloproteinases, including matrix metalloproteinases (MMPs), a disintegrin and metalloproteinases (ADAMs), and ADAMs with thrombospondin motifs (ADAMTSs), play important roles in va
