Glucose prevents the acquisition of the capacitated state in pig spermatozoa.

BACKGROUND: Mammalian spermatozoa need to undergo a process named capacitation to be able to fertilize an oocyte. During their journey in the female tract, spermatozoa obtain energy while exposed to a changing environment containing a variety of metabolic substrates. The energy requirements for sperm capacitation are species-specific. In addition, the available energy source can hinder the process of sperm capacitation and eventually the acrosome reaction. OBJECTIVES: To evaluate whether the metabolic substrates available in the in vitro sperm capacitation medium allow or interfere with the p…

Oral D-Aspartate Treatment Improves Sperm Fertility in Both Young and Adult B6N Mice.

D-Aspartate (D-Asp) treatment improved the fertility of young male C57BL/6N mice in vivo revealing a direct role on capacitation, acrosome reaction, and fertility in vitro in young males only. We investigated whether the positive effect of D-Asp on fertility could be extended to adult males and evaluated the efficacy of a 2- or 4-week-treatment in vivo. Therefore, 20 mM sodium D-Asp was supplied in drinking water to males of different ages so that they were 9 or 16 weeks old at the end of the experiments. After sperm freezing, the in vitro fertilization (IVF) rate, the birth rate, hormone leve…

Cytochemical study of spermiogenesis and mature spermatozoa in the lizard Tropidurus itambere (Reptilia, Squamata).

The present study was undertaken to detect essential components of spermatozoa by ultrastructural and cytochemical analyses of testis and epididymis of the lizard Tropidurus itambere at different points of its annual reproductive cycle. Cytochemical investigations of spermiogenesis have not been performed so far in Squamata and are scarce for lower vertebrates. Essential components are: 1) polysaccharides, identified by PAS staining, abundantly present in Sertoli cell elongations, acrosomal vesicles and the acrosome of sperm cells; 2) glycoconjugate variations, labeled by different lectins and

Identification of an anti-sperm auto-monoclonal antibody (Ts4)-recognized molecule in the mouse sperm acrosomal region and its inhibitory effect on fertilization in vitro.

We previously established an anti-mouse sperm auto-monoclonal antibody, Ts4, which shows immunoreactivity against several kinds of glycoprot … d anti-Naglu pAb negatively affected mouse fertilization in vitro. In addition, Ts4 inhibited sperm acrosome reaction induced by heparan sulfate. The Ts4-recognized fucosylated agalactobiantennary complex-type glycan with bisecting N-acetylglucosamine and Naglu on cauda epididymal spermatozoa may play a role in the process of fertilization.

Post-testicular development of a novel membrane substructure within the equatorial segment of ram, bull, boar, and goat spermatozoa as viewed by atomic force microscopy.

Atomic force microscopy has been used to investigate changes in the plasma membrane overlying the head region of mammalian spermatozoa (bull … oa. The equatorial subsegment develops during epididymal maturation, and following induction of the acrosome reaction with Ca(2+) ionophore A23187, its topography changes from a finely ridged appearance to that resembling truncated papillae. A monoclonal antibody to the equatorial subsegment binds only to permeabilized spermatozoa, suggesting that the subsegment is …

Spermatogenic cell-specific SPACA4 is essential for efficient sperm-zona pellucida binding in vitro.

Fertilization is a complex and highly regulated process that involves a series of molecular interactions between sperm and oocytes. However, the mechanisms of proteins involved in human fertilization, such as that of testis-specific SPACA4, remain poorly understood. Here we demonstrated that SPACA4 is a spermatogenic cell-specific protein. SPACA4 is expressed during spermatogenesis, upregulated in early-stage spermatids, and downregulated in elongating spermatids. SPACA4 is an intracellular protein that locates in the acrosome and is lost during the acrosome reaction. Incubation with antibodie…

The role of Mitogen activated protein kinase (MAPK) in sperm functions.

The generation of mature spermatozoa in the epididymis includes the activation of the MAPK cascade in a complex manner. MAPKs are thought to be involved in the regulation of transcription and ectoplasmic specialization (ES) in the testis. MAPKs also regulate mature spermatozoa flagellar motility, hyperactivation and the acrosome reaction. Here we review the current data regarding the functions of MAPKs in spermatogenesis and in mature spermatozoa.

Sperm-egg binding requires a multiplicity of receptor-ligand interactions: new insights into the nature of gamete receptors derived from reproductive tract secretions.

Although the broad concepts of fertilisation are well defined, our understanding of the biochemical mechanisms underlying sperm-egg binding is limited. Early studies of fertilisation in the mouse implicated beta-1,4-galactosyltransferase-1 (GalT) as a sperm receptor for oligosaccharide ligands on the zona pellucida glycoprotein, ZP3. Binding of multiple ZP3 oligosaccharides induces GalT aggregation, leading to G-protein activation and initiation of the acrosome reaction. Consistent with this, GalT-null sperm show greatly reduced binding to ZP3 and are unable to undergo a zona-induced acrosome …
