Evaluation of anti-inflammatory and antinociceptive effects of D-002 (beeswax alcohols).

D-002, a mixture of six higher aliphatic alcohols purified from beeswax, displayed anti-inflammatory effects in carrageenan-induced pleurisy and cotton pellet granuloma in rats. The aim of the present study was to confirm the anti-inflammatory properties of D-002 and to explore its potential analgesic effects. Xylene-induced mouse ear oedema was used to assess the anti-inflammatory effect, acetic acid-induced writhing and hot plate responses for the analgesic activity, and the open field and horizontal rotarod tests for motor performance. For anti-inflammatory tests, mice were randomised into …

Potentially important nighttime heterogeneous chemistry: NO3 with aldehydes and N2O5 with alcohols.

We report the first measurements of the reactive uptake of NO(3) with condensed-phase aldehydes. Specifically, we studied NO(3) uptake on so … imated to range from 1.9-7.5 h. We also carried out detailed studies of N(2)O(5) uptake kinetics on alcohols. We show that uptake coefficients of N(2)O(5) for five different organics at 293 K varied by more than 2 orders of magnitude, ranging from 3 × 10(-4) to 1.8 × 10(-2). We show that the uptake coefficients correlate with √(D(alcohol)(OH concentration) …

Formation of H3O+ from alcohols and ethers induced by intense laser fields.

The processes of H(3)O(+) production from alcohols (ethanol, 2-propanol, 1-propanol, 2-butanol) and ethers (diethyl ether and ethyl methyl ether), and their deuterium-substituted species, by intense laser fields (800 nm, 100 fs, approximately 1 x 10(14) W/cm) were investigated through time-of-flight (TOF) mass spectrometry. H(3)O(+) formation was observed for all these compounds except for ethyl methyl ether. From the analysis of TOF signals of H((3-n))D(n)O(+) (n = 0, 1, 2, and 3) that have expanding tails with increasing flight time, it has been confirmed that the reaction proceeds through m…

Initial studies using aliphatic beta-nitro alcohols for therapeutic corneal cross-linking.

PURPOSE: Corneal collagen cross-linking through UVA-riboflavin photochemistry (UVAR) has been shown to be an effective treatment for keratoconus and related keratectasias. In recent studies using sclera, the authors observed that short-chain aliphatic beta-nitro alcohols can cross-link collagenous tissue under physiologic conditions. Thus, this study was undertaken to evaluate these agents as potential pharmacologic alternatives to UVAR. METHODS: Porcine corneal strips (8 x 4 mm) and corneoscleral complexes were cross-linked using 1 to 100 mM 2-nitroethanol (2ne), 2-nitro-1-propanol (2nprop),…

Lead-induced catalase activity differentially modulates behaviors induced by short-chain alcohols.

Acute lead administration produces a transient increase in brain catalase activity. This effect of lead has been used to assess the involvement of brain ethanol metabolism, and therefore centrally formed acetaldehyde, in the behavioral actions of ethanol. In mice, catalase is involved in ethanol and methanol metabolism, but not in the metabolism of other alcohols such as 1-propanol or tert-butanol. In the present study, we assessed the specificity of the effects of lead acetate on catalase-mediated metabolism of alcohols, and the ability of lead to modulate the locomotion and loss of the right…

Induction of suppressed ADH activity in Drosophila pachea exposed to different alcohols.

Laboratory-reared males of the cactophilic Drosophila pachea exhibit a spontaneous and sex-specific suppression of alcohol dehydrogenase (ADH) activity within 4 days after eclosion. A lack of ADH activity also is usually seen in wild-caught males, although relatively high activity is always seen in female flies. In the present study we examined the effectiveness of different alcohols and related compounds, including several found naturally in necroses of the host cactus, to induce suppressed ADH activity in wild males of D. pachea and to serve as enzyme substrates. The primary alcohols (methan…

Inhibition of glycine receptor function of native neurons by aliphatic n-alcohols.

The inhibitory effects of n-alcohols (methanol to dodecanol) on glycine-activated currents were studied in neurons freshly dissociated from the ventral tegmental area of neonatal rats using whole-cell patch-clamp recording technique. Ethanol enhanced and depressed glycine-activated currents in 35% and 45%, respectively, of neurons of ventral tegmental area of neonatal rats. In this report, we extended our focus of ethanol-induced inhibition of glycine currents to other straight-chain alcohols. Aliphatic n-alcohols, which have carbon numbers less than nine, suppressed glycine currents in 45% (7…

Distinct actions of alcohols, barbiturates and benzodiazepines on GABA-activated chloride channels.

Similarities and differences in the actions of alcohols, barbiturates and benzodiazepines on GABA-activated chloride channels are reviewed. This question has been approached using behavioral measures, the uptake of 36Cl- by isolated brain membranes and the function of GABA-activated chloride channels expressed in Xenopus oocytes using brain mRNA. Although drugs from all three classes augment GABA-dependent chloride conductances, there are a number of differences between these drugs. Studies of genetic differences in sensitivity to sedative drugs and development of tolerance and cross-tolerance…

Biosynthesis of long-chain alcohols by developing and regenerating rat sciatic nerve.

Cell-free preparations of rat sciatic nerve were found to catalyze the reduction of fatty acid to alcohol in the presence of NADPH as reducing cofactor. The reductase was membrane-bound and associated primarily with the microsomal fraction. When fatty acid was the substrate, ATP, coenzyme A (CoA), and Mg2+ were required, indicating the formation of acyl CoA prior to reduction. When acyl CoA was used as substrate, the presence of albumin was required to inhibit acyl CoA hydrolase activity. Fatty acid reductase activity was highest with palmitic and stearic acids, and somewhat lower with lauric

Engineering g-C3N4/CsPbCl3 Heterojunctions for Selective Photocatalytic Oxidation of Benzylic Alcohols.

The selective oxidation of benzylic alcohols to carbonyl compounds is a fundamental transformation in the synthesis of fine chemicals and pharmaceuticals, yet conventional methods often rely on stoichiometric oxidants and generate significant waste. Herein, we report a sustainable heterogeneous photocatalytic system based on engineered g-C3N4/CsPbCl3 heterojunctions for the aerobic oxidation of benzylic alcohols under visible-light irradiation. A series of heterojunction photocatalysts with different compositions was prepared and evaluated, identifying the material containing 10%W/W CsPbCl3 as…

Orthogonal nucleophile-ordered assembly for modular access to enantiopure 1,2-amino alcohols.

The programmed assembly of multiple common chemicals with minimal reactivity differences into complex chiral architectures with high selecti … erse native nucleophile pairs, providing a platform and modular access to valuable chiral 1,2-amino alcohols and 1,2-diols with exceptional regio-, chemo-, Z- and enantiocontrol. This work demonstrates how subtle reactivity differences can be systematically harnessed for programmable complex synthesis from fundamental feedstocks.

Photocatalytic Selective Oxidation of Alcohols to Acids by Carbon Dots in O2.

Photocatalytic oxidation of alcohols to acids represents the eco-friendly and sustainable route for producing high-value chemicals. Developing an efficient and mild photocatalytic system for this target is still a great challenge, especially in selective oxidation control. Herein, vanillic acid-based carbon dots (VA-CDs) are conveniently prepared as an efficient photocatalyst for the selective oxidation of alcohols to acids. This photocatalytic method exhibits mild reaction conditions and high catalytic activity. Various functional-group-substituted alcohols could be smoothly oxidized into cor…

Dynamic "Deactivation" mechanism enables epoxides ring-opening to prepare α-substituted alcohols.

The selective synthesis of α-substituted alcohols from epoxides remains challenging. A promising strategy involves Lewis acid (LA)-catalyzed Meinwald rearrangement of epoxides, followed by 1,2-addition to in situ-generated aldehydes. However, generalizing this approach is complicated by a mechanistic contradiction: LAs promote the rearrangement but also facilitate side reactions of inherently enolizable aldehydes. Here, we show that NaI stabilizes these aldehydes by forming reversible α-oxy iodides, temporarily suppressing side reactions while maintaining their availability for Ni-catalyzed 1,…

Unspecific Peroxygenase-Based Chemo-Enzymatic Synthesis of Amino Alcohols for Pharmaceutical Preparation.

This study explores the use of unspecific peroxygenases for the enantioselective preparation of amino alcohol-based pharmaceuticals and their respective precursors. The aim of this work was to develop a practical concept by simplifying the otherwise complex synthesis approach, thereby expanding the application of unspecific peroxygenases toward chemo-enzymatic synthesis. To introduce hydroxy groups at specific positions within protected amines, the unspecific peroxygenase (UPO) was used, which facilitated the enantioselective synthesis toward shorter overall reaction pathways and thus may serv

A Synergistic C2+ Alcohols/Olefins-Intermediated Pathway Boosts CO2 Hydrogenation to Aromatics.

The rational design of highly efficient catalysts and the development of novel reaction pathways are eternal themes and central challenges in the field of chemical synthesis. Here, we design a dual-engine catalytic system for CO2 hydrogenation to aromatics via a synergistic C2+ alcohols/olefins pathway. The dual-engine catalytic system initiated by FeCo active sites and CuZnAl promoters guarantees the continuous C2+ alcohols/olefins supply, delivering a record-breaking aromatics yield (31.1%) with the aid of aromatization component H-ZSM-5. Multiple characterization and theoretical simulations…

Construction of Chiral Quaternary Carbons via Enzymatic Epoxidation of Tertiary Allylic Alcohols.

A chemoenzymatic strategy to construct aryl-substituted chiral quaternary carbon centers has been established. Directed evolution of P450BM3 enables highly enantioselective epoxidation of tertiary allylic alcohols (up to 99% ee), which is a formidable challenge in chemical synthesis. The resulting chiral epoxides undergo acid-catalyzed semipinacol rearrangement with retention of chirality, affording enantioenriched cyclic ketones. This approach is applied to the total synthesis of (-)-sinoracutine, demonstrating the possibility of divergent synthesis of complex molecules.

A Unified Platform for Direct Alkynylation from Alcohols via Deoxygenative Metallaphotoredox Catalysis.

Alkyne motifs are privileged functional groups in medicinal chemistry and serve as key handles in bioorthogonal click chemistry. Herein, we report a general metallaphotoredox deoxygenative C(sp3)-C(sp) coupling strategy that enables the direct conversion of alcohols into alkynes using alkynyl bromides as coupling partners. This protocol operates under redox-neutral, mild conditions via NHC-mediated alcohol activation and nickel catalysis, enabling the generation of alkyl radicals from alcohols and their subsequent cross-coupling with alkynyl electrophiles. The method exhibits broad substrate s…

Green thermocatalytic ammonia synthesis assisted by catalytic hydrogen transfer reactions from alcohols.

This preliminary work opens a possible innovative route to investigating a paradigm shift in ammonia synthesis. It involves a hydrogen donor molecule (HDM) to favor ammonia synthesis at low pressure and temperature (1 bar, T < 200 °C), subsequently reducing the environmental footprint of NH3 synthesis.
