Iridium-Catalyzed Stereoselective α-Alkylation of α-Hydroxy Ketones with Minimally Polarized Alkenes.

Cationic Ir(I)-complexes modified with homochiral diphosphines promote the α-C-H addition of α-hydroxy ketones to styrenes or alkyl olefins. These processes are predicated on the hydroxyl-directed formation of an Ir-enolate. Inter- and intramolecular processes are feasible, with the latter offering stereocontrolled access to carbocycles bearing two new stereocenters. The intramolecular processes constitute rare examples of alkene-based Conia-ene reactions that are enantio- and diastereoselective.

Regio-Reversed α-Addition of Oxygen-Centered Nucleophiles to Carbazole-Based Trifluoromethyl Alkenes.

The conjugate addition of nucleophiles to electron-deficient alkenes represents one of the most powerful methods for forming chemical bonds. While this process typically demonstrates β-selectivity, the regio-reversed α-additions have largely been underexplored. In this study, we present an unprecedented α-specific nucleophilic addition of weak oxygen-centered nucleophiles to carbazole-based trifluoromethyl alkenes, achieved through a photoinduced polarity transduction pathway under mild, photocatalyst-free conditions. This reaction exhibits a broad scope, good functional group tolerance, excep…

Electrochemically Driven Direct Participation of Chalcogenophenols in 1,2-Thio/Seleno-Heteroarylation of Alkenes.

Herein, we report an electrochemically driven three-component strategy that successfully achieves direct coupling of nonpreactivated chalcogenophenols with styrenes and N-heteroarenes, enabling efficient construction of the biologically significant β-azaaryl chalcogenoether. The key to this strategy lies in an anodic oxidation-mediated deprotonation-oxidation pathway, which cleverly circumvents the thermodynamic barrier posed by the bond dissociation energy (∼360 kJ/mol) of the S/Se-H bond in chalcogenophenols. This process generates key thiyl radical intermediates from the thiolate anion in s

Diastereoconvergence in NHC/photo dual-catalysis: stereoselective boroacylation of internal electron-poor alkenes.

Achieving diastereoconvergence in synthetic chemistry, particularly in photocatalytic radical systems, remains a significant yet challenging goal. Herein, we report a diastereoconvergent radical boroacylation enabled by cooperative N-heterocyclic carbene (NHC) and photocatalysis. Notably, the reaction consistently delivers syn-diastereoselective products, regardless of the starting alkene geometry-E, Z, or an E/Z mixture-demonstrating its diastereoconvergent nature. In contrast, when the conformational dynamics are restricted, as in the case of a rigid cyclic Z-alkene, the reaction yields the

Photochemical Formal [4 + 2] Cycloaddition of α-Bromo Phenylsulfones and Acetophenones with Alkenes.

Benzocycles are the predominant skeletons in medicinal chemistry. Herein, we report a modular and practical [4 + 2] cycloaddition reaction for the photochemical synthesis of benzocyclic sulfones/ketones via sequential alkylation and arylation across an olefinic double bond. This scalable approach is compatible with the direct cyclization of α-bromo phenylsulfones or acetophenones with various alkenes, which can be expected to rapidly convert abundant alkene feedstocks into medically relevant molecules and the late-stage functionalization of pharmacologically related substrates.

CuBr2-mediated dehydrogenative [4+2] annulation of 1-naphthyl-1,3-indandiones and alkenes.

Intermolecular annulation reactions of 1-naphthyl-1,3-indandiones with alkenes proceed efficiently in the presence of a copper catalyst to generate spirocarbocycle compounds. Various spirocyclic molecules bearing an all-carbon quaternary center could be obtained by this novel method with good yields, excellent regioselectivity, and good functional group tolerance. A radical mechanism is proposed based on the HRMS analysis results of control experiments.

1,2-Alkynyl Functionalization of Unactivated Alkenes via Diverse Radical-Triggered Functional Group Migration.

We have developed a transition-metal-free radical approach for 1,2-alkynyl functionalization of unactivated alkenes through the combination of 3-exo-dig cyclization with alkynyl migration triggered by in situ-generated diverse radical precursors. This strategy provides a robust toolkit to access a variety of synthetically important α-functionalized alkynyl ketones, simultaneously installing densely functionalized carbonyl, alkynyl, and other various functional groups into the alkenes. The broad substrate scope, which includes distinctly electron-donating or electron-withdrawing alkynyl migrati…

Iodine-promoted transfer of dihydrogen from ketones to alkenes, triphenylmethyl, and diphenylmethyl derivatives.

Herein, a novel class of transfer hydrogenation agent, cycloheptanone, was successfully employed in metal-free hydrogenation facilitated by iodine. A series of alkenes, triphenylmethyl derivatives, and diphenylmethyl derivatives were reduced to the desired compounds in moderate to excellent yields. The transfer hydrodeuteration of alkenes using α-deuterated cyclododecanone exhibited high regioselectivity. Preliminary mechanism studies confirmed the origins of the two hydrogen atoms involved in the reduction of alkenes. The current study paves the way for the use of ketones as unique transfer h…

Photocatalyzed Defluorinative Dichloromethylation of α-CF3 Alkenes Using CHCl3 as the Radical Source.

A visible-light-induced defluorinative dichloromethylation of α-CF3 alkenes was developed with cheap and readily accessible chloroform simultaneously as a dichloromethylation reagent and reaction medium, leading to the facile preparation of new polyhalogenated scaffolds. Notably, the change from CHCl3 to CDCl3 offers a straightforward pathway for accessing the deuterated analogues with excellent degrees of D incorporation. Mechanistic studies suggested the reaction underwent a radical addition of the dichloromethyl radical with alkenes, followed by sequential single-electron transfer and deflu…

Molecular Diversity via Tetrasubstituted Alkenes Containing a Barbiturate Motif: Synthesis and Biological Activity.

The synthesis of a molecularly diverse library of tetrasubstituted alkenes containing a barbiturate motif is described. Base-induced condensation of N1-substituted pyrimidine-2,4,6(1H,3H,5H)-triones with 5-(bis(methylthio)methylene)-2,2-dimethyl-1,3-dioxane-4,6-dione gave 3-substituted 5-(methylthio)-2H-pyrano[2,3-d]pyrimidine-2,4,7(1H,3H)-triones ('pyranopyrimidinones'), regioselectively. A sequence of reactions involving ring-opening of the pyran moiety, displacement of the methylthio group with an amine, re-formation of the pyran ring, and after its final cleavage with an amine, gave tetras…

Metal-free oxysulfenylation of alkenes with 1-(arylthio)pyrrolidine-2,5-diones and alcohols.

β-Alkoxy sulfides are widely used as versatile building blocks in organic synthesis. Therefore, it is highly desirable to develop a convenient and efficient method for oxysulfenylation of alkenes. In this communication, an easy and efficient metal-free approach to β-alkoxy sulfides has been developed. The protocol uses readily available 1-(arylthio)pyrrolidine-2,5-diones and alcohols as the oxysulfenylating agents, chloroform as the solvent, and no ligand, additive and exclusion of air were required. Therefore, the present method provides a useful strategy for synthesis of β-alkoxy sulfides.

Electrophilic bromination of alkenes: environmental, health and safety aspects of new alternative methods.

More than twenty new alternative methods for bromination of alkenes have been evaluated taking into consideration their resource demands, waste production as well as environmental, health and safety aspects. The cost of bromine and the substances designated to circumvent the application of molecular bromine have also been taken into account. As bromine is only one of several problematic substances being used, its avoidance-by applying bromine supported on solid material or by performing the in situ generation of bromine-does not significantly reduce the technological requirements. On the contr…

Energy Transfer-Mediated Carboamination of Alkenes to Access γ-Amino-1,3-dicarbonyl Scaffolds.

A visible light-induced, energy transfer-mediated strategy for the carboamination of alkenes was developed using a rationally designed enyl oxime ester-based bifunctional reagent, enabling the efficient construction of structurally diverse γ-amino-1,3-dicarbonyl compounds. The novel bifunctional reagent can be facilely prepared from readily available feedstock chemicals, and the rationale of this structural design relies on the resonance-driven spin center shift (SCS) to generate the key carbon-centered radical. A wide range of alkenes participated in this transformation smoothly, affording th…

Visible-Light-Induced Aziridination of Unactivated Alkenes via Energy-Transfer Cleavage of Sulfilimines.

Herein, we report that the aziridination of alkenes proceeds via visible-light-mediated energy-transfer-induced cleavage of N-sulfonyl sulfilimines, which serve as bench-stable nitrene precursors with a recoverable dibenzothiophene moiety. This strategy proves to be efficient, delivering good to excellent yields for mono- to tetrasubstituted unactivated alkenes and styrenes, with its applicability further extended to pharmaceuticals. Mechanistic studies support an energy-transfer pathway involving a free triplet nitrene intermediate.

Photoredox-Catalyzed Single-Carbon Insertion for De Novo Indene Synthesis from Tetrasubstituted Alkenes.

A visible-light photoredox-catalyzed single-carbon insertion of tetrasubstituted alkenes is developed using hypervalent iodine diazo reagents. The reaction proceeds via a radical-polar crossover manifold under mild conditions, enabling intramolecular arene capture of photogenerated carbocations from acyclic precursors to afford polysubstituted indenes through a cascade cyclization. This strategy exploits weakly nucleophilic arenes as internal carbocation traps, enabling efficient de novo construction of fused carbocyclic frameworks. The reaction exhibits broad functional group tolerance and pr…
