Synthesis and selected properties of oligonucleotidyl-(Pm----O)-amino acids.

N-Cbz-Ser(OMe)-(Pm----O)-d(TpT) was synthesized as a diastereomeric mixture of approx. (1:1) by reacting d(TpT) with 2,4,6-triisopropylbenzenesulfonyl chloride and N-Cbz-Ser-OMe. The phosphoesteric bond of N-Cbz-Ser (OMe)-(Pm----O)-d(TpT) was found to be stable in acid (1 N HCl, 1 h, 37 degrees C), but labile in alkaline solution (0.1-1 N NaOH, 1 h, 37 degrees C). The products of alkaline hydrolysis were determined to be d(TpT) and the amino acid derivatives. Furthermore, the phosphotriester N-Cbz-Ser(OMe)-(Pm----O)-d(TpT) was more labile than the diesteric analogue N-Cbz-Ser (OMe)-pdT. The in

Plasma amino acids in dogs with osteochondrosis or hip dysplasia.

Radiography of the hip joints of mature dogs has shown that hip dysplasia is quite common in many middle and large size breeds in Norway. The heritability of hip dysplasia has been estimated in several studies to be between 0.2 and 0.6 (Hedhammar et al 1979). This indicates that environmental factors are important for development and severity of hip dysplasia.

Mechanisms controlling renal hemodynamics and electrolyte excretion during amino acids.

Our purpose was to investigate the mechanisms by which increased plasma amino acids elevate renal blood flow (RBF) and glomerular filtration rate (GFR). Since transport of amino acids and Na+ is linked in the proximal tubule, we hypothesized that increased amino acids might stimulate proximal tubular Na+ reabsorption (PRNa) and thus increase RBF and GFR by a macula densa feedback mechanism. A solution of four amino acids (Ala, Ser, Gly, Pro) was infused intravenously (0.075 mmol X kg-1 X min-1 total) into anesthetized dogs with normal kidneys (NK) and with kidneys in which the tubuloglomerular…

The nephron in transport of sodium, amino acids, and glucose.

The ability of the kidney to maintain overall control of sodium and fluid balance appears greater than the sum of its parts, yet microperfusion and dissection studies that have assigned particular transport processes to individual segments of the nephron are helping to clarify a number of clinical states. Among those that can be so localized are certain congenital disorders accompanied by glycosuria and aminoaciduria.

Neonatal citrllinemia: treatment with keto-analogues of essential amino acids.

A patient with neonatal onset citrullinemia survived to 8 months of age when treated with a mixture of essential amino acids and their keto-analogues. The initial plasma citrulline concentration was 2.7 mM; the blood ammonia concentration was greater than 500 muM. During the first week of therapy, the blood ammonia concentration became normal and that of plasma citrulline was reduced by almost 50%. It was possible to institute progressive increases in dietary calories and protein; growth and developmetn with resolution of almost all clinical signs of disease ensued. The patient died at 8 month…

Poly(amino acid) Dot-Based Fluorescent Receptor Array for Multiplexed Sensing of Saccharides and Amino Acids.

Inspired by protein-based odorant recognition in the mammalian olfactory system, we constructed a biomimetic fluorescent sensor array using poly(amino acid) dots synthesized from tyrosine, alanine, and cysteine. Saccharides and amino acids are two categories of structurally analogous small molecules that coexist widely in biological and food matrices (e.g., energy drinks, nutritional supplements, fermented products). Their simultaneous monitoring is challenging because of their completely different functional groups. While most studies target individual detection of such organic compounds, rap…

Fecal Amino Acid Profiles Exceed Accuracy of Serum Amino Acids in Diagnosing Pediatric Inflammatory Bowel Disease.

In this prospective intention-to-diagnose pilot study, we aimed to assess accuracy of serum and fecal amino-acids to discriminate de novo pediatric inflammatory bowel disease (IBD) and non-IBD children. Patients with suspected IBD were allocated the IBD (n = 11) or non-IBD group (n = 8) following laboratory testing or endoscopy according to the revised Porto-criteria. Fecal calprotectin levels were obtained, an additional blood and fecal sample were collected. Fecal and serum amino-acid profiles were analyzed using high performance-liquid chromatography. Nine fecal amino-acids (alanine [area u

Effect of essential amino acid supplementation on muscle and plasma free amino acids in chronic uremia.

In chronic uremia patients on an 18-g protein diet for 2 to 3 weeks, the intracellular free amino acid pattern in muscle was grossly abnormal, with low concentrations of threonine, valine, carnosine, and high concentrations of aspartate, glutamine, glycine, alanine, and taurine. When the protein-poor diet was supplemented with essential amino acids (2 to 3 times that for minimum requirements) and histidine, these abnormalities were partly corrected. Other abnormalities, however, were observed, such as low intracellular concentrations of phenylalanine and tyrosine, and high concentrations of ly…

Elevation of amino acids in the interstitial space of the rat brain following infusion of large neutral amino and keto acids by microdialysis: leucine infusion.

A microenvironment similar to that found in maple syrup urine disease was created in the brain of free-moving, awake rats by the infusion of leucine into the brain using microdialysis. To determine the effects on amino acid homeostasis the eluate of the probe was analyzed. Perfusion with leucine elevated the interstitial levels of large neutral amino acids suggesting hetero exchange of large neutral amino acids from neuronal cells into the interstitial space. The data also demonstrated the inter relationship of leucine and glutamine, both of which may be nitrogen sinks in the brain. Elevation …

Decreased branched-chain amino acids and elevated fatty acids during antecedent hypoglycemia in type 1 diabetes.

INTRODUCTION: Hypoglycemia is a major limiting factor in achieving recommended glycemic targets for people with type 1 diabetes. Exposure to … ry. RESULTS: In total, 68 metabolites were studied. On day 1, concentrations of the branched-chain amino acids, leucine (p=3.8×10-3) and isoleucine (p=2.2×10-3), decreased during hypoglycemia. On day 2, during hypoglycemia, five amino acids (including leucine and isoleucine) significantly decreased, and two fatty acids (tetradecanoic and oleic acids) signifi …

Branched-Chain Amino Acids and Branched-Chain Keto Acids in Hyperammonemic States: Metabolism and as Supplements.

In hyperammonemic states, such as liver cirrhosis, urea cycle disorders, and strenuous exercise, the catabolism of branched-chain amino acids (BCAAs; leucine, isoleucine, and valine) is activated and BCAA concentrations decrease. In these conditions, BCAAs are recommended to improve mental functions, protein balance, and muscle performance. However, clinical trials have not demonstrated significant benefits of BCAA-containing supplements. It is hypothesized that, under hyperammonemic conditions, enhanced glutamine availability and decreased BCAA levels facilitate the amination of branched-chai…

Design, Synthesis, and Applications of Fluorescent Amino Acids in Bioimaging.

Fluorescent unnatural amino acids (FlAAs) provide a powerful molecular bridge between small-molecule fluorophores and biomacromolecules, enabling minimally perturbative optical interrogation of peptides and proteins. Over the past decade, substantial progress has been made in the rational design, synthesis, and application of FlAAs, driven by advances in organic synthesis, C-H functionalization, and translation-based incorporation technologies. This review summarizes contemporary strategies for constructing FlAAs, with a particular emphasis on structure-property relationships and modular desig…
