Integrin-binding sialoprotein (IBSP) is a member of the small integrin-binding ligand N-linked glycoprotein (SIBLING) family; and the whole SIBLING family is further included in a larger secretory calcium-binding phosphoprotein (SCPP) family. SIBLING proteins are known to construct a part of the non-collagenous extracellular matrices of calcified tissues, and considered to have arisen by duplication and subsequent divergent evolution of a single ancient gene. To understand the alterations of SIBLING molecules associated with the evolution of calcified tissues in vertebrates, we initiated a sea

Amphibian populations are declining globally at an alarming rate. Pesticides are among a number of proposed causes for these declines. Although a sizable database examining effects of pesticides on amphibians exists, the vast majority of these studies focus on toxicological effects (lethality, external malformations, etc.) at relatively high doses (parts per million). Very few studies focus on effects such as endocrine disruption at low concentrations. Further, most studies examine exposures to single chemicals only. The present study examined nine pesticides (four herbicides, two fungicides, …

Since the production of Dolly the sheep cloning methods for somatic cells have been thoroughly described and are becoming routine. However, … erience, our view is that mammalian somatic-cell cloning and the pioneer nuclear-transfer data from amphibians have much in common. We suggest that the only way to improve nuclear reprogramming is to modify the chromatin structure of somatic cells before nuclear transfer, to provide the oocyte with a chromosomal structure that is more compatible with the nat …

Liver and oviduct of frogs Rana temporaria and Xenopus laevis were shown to possess the activity of beta-galactoside alpha 1-2-fucosyltransferase. Extracts of these tissues as well as porcine and rat submaxillary gland homogenates catalyze the fucosylation of synthetic plant xyloglucan fragments, the disaccharide D-Gal(beta 1-2)D-Xyl and the tetrasaccharide D-Gal(beta 1-2)D-Xyl(alpha 1-6)D-Glc(beta 1-4)D-Glc; the active preparations can be used for enzymatic synthesis of biologically active oligosaccharides.

Activity of isolated auditory-nerve fibers in tree frogs (Eleutherodactylus coqui) exposed to continuous 3-min tones of different intensities at their characteristic frequencies (CFs) was recorded. Period histograms show a retardation in the preferred phase of discharge during and after the cessation of the exposure. Postexposure phase shift is concomitant with an elevation in CF thresholds and related to the level of tone exposure above threshold. Vector strength does not show comparable trends of change; postexposure shifts are related to preexposure CF thresholds. Recovery of phase retardat

1. Salicylate actions on afferent nerve activity in the Xenopus lateral line and on cochlear potentials in guinea pig were investigated. 2. In the lateral line, salicylate (0.3-2.5 mM) suppressed spontaneous activity, water motion evoked excitation and responses to L-glutamate (1-2 mM) and kainate (10-20 microM). 3. In the guinea pig, salicylate (0.6-10 mM) suppressed the compound action potential (CAP) and increased N1 latency at low but not high sound intensities. 4. In the lateral line salicylate action may involve an antagonism of the hair-cell transmitter on the afferent nerve. 5. In the

Aquatic toxicity tests were conducted to evaluate the effects of cadmium-enriched sediment on embryo-larval stages of the goldfish (Carassius auratus), leopard frog (Rana pipiens), and largemouth bass (Micropterus salmoides). Natural stream sediment was collected and enriched with cadmium to nominal concentrations of 1.0, 10.0, 100, and 1000 mg/kg. Enriched sediments were placed in Pyrex dishes and covered with 350 ml of reconstituted water. Fertilized eggs were placed in the dishes and maintained through 4 days posthatching, giving a total exposure time of 6 to 7 days. For all tests the cadmi

Ideal disease mitigation measures for wildlife are safe and benign for target species, non-target organisms, the environment, and humans. Identifying collateral (i.e., unintended) effects is a key consideration in implementing such actions. Deltamethrin dust and fipronil-laced baits represent a group of insecticides that target fleas (pulicides) and are used to control flea (Siphonaptera) vectors of the plague bacterium Yersinia pestis to protect prairie dogs (Cynomys spp.) and their plague-susceptible obligate predators, endangered black-footed ferrets (Mustela nigripes). A variety of animals

Based on long-term and often frustrating experiences with the poor quality of tapeworms (Cestoda) collected throughout the world for taxonomic and phylogenetic studies, and considering the increasing obstacles to obtaining new material, a simple, easy-to-use and illustrated methodological guide (manual) is provided. It focusses mainly on key steps in examining hosts, collecting cestodes from poikilothermous vertebrates except elasmobranchs, i.e., from ray-finned fish (Actinopterygii), amphibians and 'reptiles' (a paraphyletic group comprising all sauropsids except birds), and fixing them for s…

Pathophysiological effects of clinical chytridiomycosis in amphibians include disorders of cutaneous osmoregulation and disruption of the ability to rehydrate, which can lead to decreased host fitness or mortality. Less attention has been given to physiological responses of hosts where enzootic infections of Batrachochytrium dendrobatidis (Bd) do not cause apparent population declines in the wild. Here, we experimentally tested whether an enzootic strain of Bd causes significant mortality and alters host water balance (evaporative water loss, EWL; skin resistance, R(s); and water uptake, WU) i…

The success of ex situ survival assurance populations as tools for amphibian conservation depends on the health and reproductive success of founder populations. Necropsy examination and histopathology of animals that die in assurance populations are useful for the identification of population-limiting disease problems and can help to direct applied research efforts in areas such as amphibian husbandry and nutrition. This study reviewed postmortem findings in 167 frogs from 13 species that died in a large Panamanian rescue and survival assurance population between 2006 and 2011. Common problems

The use of hormonally induced spermatozoa expressed in urine (HISu) is a valuable component of reproduction technologies for amphibians. Five protocols for sampling HISu from the European common frog (Rana temporaria) were compared: (1) pituitary extracts, (2) 0.12 µg g⁻¹ luteinising hormone-releasing hormone analogue (LHRHa), (3) 1.20 µg g⁻¹ LHRHa, (4) 11.7 IU g⁻¹ human chorionic gonadotrophin (hCG) and (5) 23.4 IU g⁻¹ hCG (g⁻¹ = per gram bodyweight). From 1 to 24h after administration we assessed the number and concentration of spermatozoa in spermic urine and in holding water, and in urine …

Amphibian skin has been the source of a wide variety of biologically active substances, but less than one-third of the known genera of amphibians have been probed for such active substances. Skins of 21 genera of anurans from Thailand have now been investigated for noxious secretions, toxic substances, and alkaloids. Four genera of bufonid toads (Bufo, Ansonia, Leptophryne, Pedostipes) were toxic due to the presence of bufadienolides or bufadienolide-like compounds. Two species of ranid frogs (Rana raniceps, Rana signata) were toxic, perhaps due to the presence of toxic peptide(s). Two species…

Dorsal (D), lateral (L and R), and ventral (V) portions of the endoderm of blastulae ofAmbystoma mexicanum of different age (stages 8+ to 10-) were combined with ectodermal caps of stage 8+ blastulae. All V and most L and R portions induced only ventrocaudal mesodermal structures - "ventral" type of mesoderm induction. Almost all D portions induced much more voluminous structures of predominantly axial character - "dorsal" type of mesoderm induction. The difference in mesoderm-inducing capacity of the dorsal as against the lateral and ventral endoderm is probably purely quantitative in charact

Managing species in an uncertain future is a reality for natural resource decision makers. Climate change is expected to exacerbate threats such as habitat loss and disease, and cause phenological mismatches, but there is uncertainty in the magnitude of these effects. Amphibians are among the most threatened taxa on earth, and most species in North America are uniquely tied to water availability for breeding, larval development, thermal refugia, and food availability. Changes in water availability and temperature may result in phenological mismatches with one or more of these processes. Thus, …

Critical periods of development are time points that are especially sensitive to disruptions. Critical periods are important for microbiome assembly and the development of an effective immune system and, therefore, can have large and lasting impacts on host health, even through later life stages. Here, we investigated how a disruption to the microbiome of amphibians (treatment with a cocktail of six antibiotics and one antifungal compound) during early life and subsequent introduction to microbes at different developmental stages influences microbiome assembly and the development of the immune…

BACKGROUND: The pancreas exhibits diverse structures and roles across vertebrates. The pancreas has evolved to include both endocrine and exocrine cells, a change that occurred during the transition from fish to amphibian. This event emphasizes the evolutionary significance of amphibians. However, research has focused predominantly on anuran amphibians, with urodeles, such as newts, remaining underexplored. In this study, we investigated the development of the pancreas using Pleurodeles waltl as a model species of urodele.
RESULTS: The newt pancreas consists of a single organ with exocrine ti…

The skin of amphibians is a rich source of peptides with a wide range of biological activities. They are stored in secretory granules in an inactive form. Upon stimulation, they are secreted together with proteases into the skin. Once activated, they rapidly exert their biological effects, including fighting microorganisms and predators, while their excess is immediately destroyed by the released proteases. To keep bioactive peptides in their initial form, it is necessary to inhibit these enzymes. Several inhibitors for this purpose have previously been mentioned; however, there has not been a…
