Certain autoantibodies in autoimmune liver disease have prognostic implications that are under-utilized and under-developed. The goals of this review are to indicate progress in characterizing the autoantibodies with prognostic connotations and to indicate the feasibility and importance of discovering other markers. Prime source and review articles in English were selected by a Medline search through 2010. Antibodies to soluble liver antigen, actin, liver cytosol type 1, asialoglycoprotein receptor, chromatin, cyclic citrullinated peptide, and uridine glucuronosyltransferases have been associa…

Serial serum samples from 16 Italian patients presenting with acute hepatitis C virus (HCV) infections (which progressed to chronic hepatitis in six) were screened for the non-organ-specific autoantibodies most frequently associated with autoimmune hepatitis (AIH), as well as for antibodies against the hepatic asialoglycoprotein receptor (ASGP-R) and against the GOR peptide. One patient had low titres (1:10-1:80) of liver-kidney microsomal (LKM-1) antibodies during the recovery phase and three others had transient low titres of anti-smooth muscle (IgM class, 1:10) or anti-ASGP-R (1:150-1:300).…

Previous studies have revealed that desialylated forms of hCG have a high potency to inhibit both the binding of bovine TSH (bTSH) to human … f TSH and Graves' immunoglobulin G and which also display a relatively low affinity for the hepatic asialoglycoprotein receptor and, as a consequence, have a reasonable survival in the circulation. Two hCG forms isolated by sequential chromatography of crude hCG on DEAE-52 and Sephadex G-100 were of interest, since they displayed some degree of selectivity in binding to thyr …

C-type lectin domain family 10 member A (Clec10a), also called macrophage galactose-type lectin 1 (MGL1) or CD301a, is expressed on myeloid cells, including macrophages in the skin. Several pieces of evidence suggest that Clec10a physically binds to O-linked mucin-like molecules, but the functional ligand that mediates signaling in macrophages remains undetermined. We previously reported that house dust mite (HDM) treatment on the skin produced a greater amount of Toll-like receptor 4 (TLR4)-mediated inflammatory cytokines from macrophage in Clec10a-deficient mice than in wild-type (WT) mice,

The preparation of one-step kit for the 99mTc labelled galactosyl-neo-glycoalbumin (NGA) using SnCl2 reduced method was reported in this paper. We developed a method of quality control and established the optimum scheme of technetium labelling on the basis of exploring the effects of varied labelled conditions on radiochemical purity. Our data indicates : (a) the kit is sterile, pyrogen-free and no toxic effects; (b) the operation is simple and fast; (c) its period of validity lasts no less than 3 months; (d) the radiochemical purity > 95%; (e) the stability of 99mTc-NGA in vitro > 4 hours; (f

The complete amino acid sequences of alpha and beta subunits of alboaggregin-beta are presented. The alpha and beta subunits were separated by reversed-phase HPLC after reduction and S-pyridylethylation, and their sequences were determined by analysis of peptides generated by enzymatic or chemical digestion. The alpha and beta subunits consist of 133 and 123 amino acid residues, respectively. The sequences are highly homologous to each other (41.4% identity) and also to those of the alpha and beta subunits of botrocetin (a von Willebrand factor modulator) and the A and B chains of factor IX/X

BACKGROUND: Atherosclerosis is the most common cause of cardiovascular diseases. Clinical studies indicate that loss-of-function ASGR1 (asialoglycoprotein receptor 1) is significantly associated with lower plasma cholesterol levels and reduces cardiovascular disease risk. However, the effect of ASGR1 on atherosclerosis remains incompletely understood; whether inhibition of ASGR1 causes liver injury remains controversial. Here, we comprehensively investigated the effects and the underlying molecular mechanisms of ASGR1 deficiency and overexpression on atherosclerosis and liver injury in mice.

BACKGROUND AND AIMS: Metabolic dysfunction-associated steatotic liver disease (MASLD) is a leading cause of chronic liver disease with limited therapeutic options. Hepatocyte senescence is a critical driver of MASLD progression, but its upstream regulators remain poorly understood. This study aims to investigate the role and mechanism of high expression of asialoglycoprotein receptor 1 (ASGR1) in MASLD-associated hepatocyte senescence.
APPROACH AND RESULTS: ASGR1 expression was assessed in liver tissue from patients with chronic liver disease and mice with MASLD induced by a high-fat diet (HF…

In 1950, Waldenström was the first to describe a chronic form of hepatitis in young women. Subsequently, the disease was found to be associated with other autoimmune syndromes and was later termed "lupoid hepatitis" because of the presence of antinuclear antibodies. In 1965, it became designated by Mackay et al. as "autoimmune hepatitis" at an international meeting, at which the general concept of autoimmunity was endorsed by the scientific community. In the early 1960s and 1970s, the value of immunosuppressive therapy with glucocorticoids and/or azathioprine was well documented in several stu

Three preparations of purified von Willebrand factor (vWF), obtained from unrelated patients affected by type IIB von Willebrand disease, were found to have normal sialic acid content (between 129 and 170 nmol/mg of vWF, as compared with 158 +/- 17 nmol/mg in four normal preparations) and to induce platelet aggregation in the presence of physiologic levels of divalent cations and without addition of ristocetin. A monoclonal antibody that blocks the vWF binding domain of the platelet glycoprotein (GP)Ib caused complete inhibition of IIB vWF-induced aggregation. In contrast, a monoclonal antibod

PURPOSE: The hepatic asialoglycoprotein receptor is responsible for degradation of desialylated glycoproteins through receptor-mediated endocytosis. It has been shown that imaging of the receptor density using [(99m)Tc]diethylenetriamine pentaacetic acid (DTPA) galactosyl human serum albumin ([(99m)Tc]GSA) allows non-invasive determination of functional hepatocellular mass. Here we present the synthesis and evaluation of [(68)Ga]GSA for the potential use with positron emission tomography (PET).
METHODS: Labelling of GSA with (68)Ga was carried out using a fractionated elution protocol. For qu…

AIMS: ASGR1 is a hepatocyte surface glycoprotein receptor that plays a crucial role in maintaining liver homeostasis. This study investigates its role in acute liver injury (ALI) and whether it is associated with the regulation of PANoptosis-like pathway.
MATERIALS AND METHODS: An LPS/D-Gal-induced ALI model was established using ASGR1 knockout mice. Liver function, inflammation, oxidative stress, mitochondrial function, and PANoptosis-like markers were assessed. In vitro studies on primary hepatocytes examined the effects of ASGR1 knockout or overexpression on ALI, with the ROS inhibitor NAC

The hepatic Ashwell-Morell receptor (AMR) can bind and remove desialylated platelets. Here we demonstrate that platelets become desialylated as they circulate and age in blood. Binding of desialylated platelets to the AMR induces hepatic expression of thrombopoietin (TPO) mRNA and protein, thereby regulating platelet production. Endocytic AMR controls TPO expression through Janus kinase 2 (JAK2) and the acute phase response signal transducer and activator of transcription 3 (STAT3) in vivo and in vitro. Recognition of this newly identified physiological feedback mechanism illuminates the patho
