Microtitre plate assay for paralytic shellfish toxins using saxiphilin: gauging the effects of shellfish extract matrices, salts and pH upon assay performance.

Saxiphilin is a hydrophilic protein with a high affinity and specificity for paralytic shellfish toxins (PSTs) found in the circulatory fluid of many invertebrates and ectothermic vertebrates. Saxiphilin has been found to be closely related to the iron binding transferrins, a group of proteins that range in molecular weight between 70 and 90 kDa. One saxiphilin isoform, that from the centipede Ethmostigmus rubripes, has been used to develop a microtitre plate assay for PSTs which relies upon detection of bound tritiated saxitoxin (STX). In this study, this assay was challenged with differing c

M-Count: an application that uses machine learning object detection and color thresholding to count settled mussel larvae.

Quantification of biofouling is a complex task that often involves counting both isolated and tightly packed groups of organisms on a test surface. Mussel larvae, which settle as both individuals and as clumps, are an important fouling organism because adult mussel colonies form via the settlement of larvae, making the settlement or repellence of mussel larvae a good indicator of a surface's antifouling performance. Manual quantification methods are time-consuming, and existing automatic machine learning-based methods are poorly suited for use by coding non-experts and often lack the ability t

Synergistic regulation of aquatic microbiome assembly and function by carbon resource availability and a bivalve bioengineer.

While resource subsidies and biotic manipulation are prevalent in managed ecosystems, their interactive effects on microbial community assembly and function remain elusive. Here, we investigated how external carbon (molasses) and a filter-feeding ecosystem engineer (Anodonta woodiana) interactively shape bacterial communities in aquaculture systems using a factorial mesocosm experiment (n = 3 replicate tanks per treatment, sampled at three time points). Integrating 16 S rRNA sequencing, community-level physiological profiling, and null model analyses, we found that combined treatment was assoc

Mechanisms of lipid metabolic disorder and inflammation in the clam Ruditapes philippinarum under PAHs stress.

The mechanisms by which Polycyclic Aromatic Hydrocarbons (PAHs) induce lipid metabolic disorder and inflammation in marine invertebrates remain poorly understood. This study utilized the clam Ruditapes philippinarum during its reproductive stage as a model organism, integrating high-throughput omics, computational simulation, and confocal microscopy to elucidate the accumulation characteristics and toxicological pathways of PAHs. The results demonstrated that PAHs significantly accumulated in the digestive gland and gonads, primarily sequestered within lipid droplets. This tissue distribution

Mussel-inspired molecular strategy for ultrasensitive detection of multiple chiral molecules.

Chiral recognition in racemic mixtures remains a critical challenge in life and environmental sciences, while the practical development of chiral electrochemical sensors is often restricted by complicated fabrication processes and insufficient signal transduction. Herein, a chiral electrochemical recognizer was developed by integrating molecular imprinting with mussel-inspired dopamine chemistry, using biomass-derived porous carbon as a conductive scaffold to achieve both high enantioselectivity and enhanced electrochemical response. Benefiting from the strong interfacial adhesion of dopamine,

Long-term exposure to 17β-estradiol of the Manila clam (Ruditapes philippinarum): Shifts of gender ratio and changes of gene expression.

Estrogen is widely distributed in the aquatic environment, causing feminization, changes of gender ratio and reduced fecundity in aquatic organisms. 17β-estradiol is a potent estrogen with strong estrogenic activity and pseudo-persistence, which poses significant risks to global aquatic ecosystems. In this study, effects of long-term exposure to 17β-estradiol in the Manila clam (Ruditapes philippinarum) were investigated. The clams were treated with 17β-estradiol (10 μg/L) for 2 months and then subjected to dissection. Gonadal tissue sections were prepared for observation, and the sex ratio wa

Injectable Dual-Crosslinked Poly(oligo(Ethylene Glycol) Methacrylate) Hydrogels Inspired by Mussel Adhesion for Cutaneous Wound Healing and Functional Tissue Regeneration.

Traditional dermal wound closure methods, such as sutures and staples, are invasive, causing soft tissue trauma, increasing the likelihood of inflammation and infections. Alternatively, while existing tissue adhesives can seal and adhere to wounds, they may cause immunogenic responses, tissue necrosis, restricted movement, and wound disruption upon removal, leading to secondary injuries and scarring. Herein, injectable poly(oligo(ethylene glycol) methyl ether methacrylate) (POEGMA)-dopamine (DA) hydrogels co-crosslinked via hydrazone linkages and dopamine self-polymerization are fabricated tha

Transcriptomics as an Early Warning of Domoic Acid Exposure in Pacific Razor Clams (Siliqua patula).

As oceans warm, harmful algal blooms (HABs) are expected to increase, including blooms of Pseudo-nitzschia, a diatom that produces domoic acid (DA), which is a potent neurotoxin. Regulatory limits for human consumption (0.075-0.1 mg/kg/day; acute exposure) exist for the Pacific razor clam; however, fisheries currently do not have regulatory limits for chronic low-level exposure to DA even though razor clams can retain DA for over a year after an algal bloom. For bivalves, exposure to marine toxins may disrupt important cellular processes, leading to concerns about effects on their overall heal

Environmental factors driving microcystin contamination of estuarine bivalve populations downstream of freshwater cyanobacterial blooms.

Estuaries are dynamic environments that provide vital habitat to ecologically and commercially important bivalves. In some cases, freshwater tributaries can introduce cyanobacteria and associated cyanotoxins into estuaries that may subsequently accumulate in estuarine bivalves. Temporarily open/closed estuaries (TOCEs), which only experience tidal input for limited periods of time, may be particularly vulnerable to the accumulation of cyanotoxins in bivalves as they can be subject to freshwater input without tidal flushing and may experience lower salinities and cyanobacterial blooms. This stu

The potential ameliorative role of Dimercaptosuccinic acid against the toxicity of Titanium Dioxide Nanoparticles on Caelatura nilotica clams.

The prevalent use of nanoparticles has adverse negative effects on biosystems. Subsequently, this study aimed to use Caelatura nilotica to assess the ecotoxicity of TiO2 NPs and how Dimercaptosuccinic acid (DMSA) improves these effects. Two concentrations of TiO2 NPs (25 and 150 µg/L) were used for 28 days. TiO2 NPs bioaccumulation, gonadal weight, gonado-somatic index, and histopathological alterations of gonads were determined. The tissues' accumulation of TiO2 NPs was concentration-time-dependent: it was 78.5 ± 28.93 μg/g dry weight in the exposed clams to 150 µg/L TiO2 NPs after 4 weeks of

Evaluation of the accumulation of the iodinated contrast agents diatrizoic acid and iohexol in Dreissena polymorpha mollusks.

Mollusks are very sensitive to aquatic environmental alterations and then, are important bio-indicators for monitoring the contamination of water bodies. Iodinated X-ray contrast media (ICMs) are ubiquitously present in the aquatic environment, primarily due to their high consumption for diagnosis purposes, high injection levels, low biodegradability, and low removal rates by wastewater treatment plants. Although these compounds are assumed to be of low toxicity, aquatic organisms are continuously exposed to these agents, which may result in adverse effects as ICMs can act as iodine source and

Metal bioaccumulation and transfer in benthic species based on isotopic signatures of individual amino acids in South China Sea cold seep environments.

Cold seeps are deep-sea 'oases' with dense and dominant coexisting populations of large mussels and tubeworms under extreme environments. Under such natural source of high metal concentrations, the present study investigated the metal bioaccumulation and transfer with trophic positions in six benthic species by the isotopic δ15N and δ13C signatures in the active Haima cold seep, South China Sea. Comparing the isotopic signatures of bulk-tissue and amino acids by compound-specific isotopic analysis (CSIA-AA), we found that the bulk trophic (TPB) values in the benthos except mussels were signifi

Sulfadiazine, sulfamethazine and sulfachloropyridazine removal using three different porous materials: Pine bark, "oak ash" and mussel shell.

This work focuses on studying the efficacy of three different by-products to adsorb three antibiotics (sulfadiazine, SDZ; sulfamethazine, SMT; sulfachloropyridazine, SCP). These antibiotics can be considered pollutants of the environment when they reach water, as well as in cases where they are spread on soils through irrigation or contained in sewage sludge or livestock manure. In this study, batch-type adsorption/desorption experiments were performed for each of the three sulfonamides, adding 7 different concentrations of the antibiotics, going from 1 to 50 μmol L-1, and with contact time of

Neandertals on the beach: Use of marine resources at Grotta dei Moscerini (Latium, Italy).

Excavated in 1949, Grotta dei Moscerini, dated MIS 5 to early MIS 4, is one of two Italian Neandertal sites with a large assemblage of retouched shells (n = 171) from 21 layers. The other occurrence is from the broadly contemporaneous layer L of Grotta del Cavallo in southern Italy (n = 126). Eight other Mousterian sites in Italy and one in Greece also have shell tools but in a very small number. The shell tools are made on valves of the smooth clam Callista chione. The general idea that the valves of Callista chione were collected by Neandertals on the beach after the death of the mollusk is

Recent advances in toxicological research of nanoplastics in the environment: A review.

Nanoplastics have attracted increasing attention in recent years due to their widespread existence in the environment and the potential adverse effects on living organisms. In this paper, the toxic effects of nanoplastics on organisms were systematically reviewed. The translocation and absorption of nanoplastics, as well as the release of additives and contaminants adsorbed on nanoplastics in the organism body were discussed, and the potential adverse effects of nanoplastics on human health were evaluated. Nanoplastics can be ingested by organisms, be accumulated in their body and be transferr

Explorations on the ecological role of toxin secretion and delivery in jawless predatory Polychaeta.

Motivated by biotechnological prospects, there is increasing evidence that we may just be scraping the tip of the iceberg of poisonous marine invertebrates, among which the Polychaeta are promising candidates for bioprospecting. Here we show that an inconspicuous phyllodocid uses toxins in its uncanny feeding strategy. The worm, a jawless active predator characterised by its bright green colour, preys on larger invertebrates (including conspecifics) by extracting tissue portions with its powerful proboscis through suction. The animal is even able to penetrate through the valves and plates of l

Prewetting dichloromethane induced aqueous solution adhered on Cassie superhydrophobic substrates to fabricate efficient fog-harvesting materials inspired by Namib Desert beetles and mussels.

Namib Desert beetles harvest water from harsh environments by using their hydrophilic-hydrophobic dorsal surfaces. Generally, Cassie-state superhydrophobic materials are chosen as substrates to prepare bioinspired (super)hydrophilic/(super)hydrophobic patterned surfaces. However, due to the low adhesion and strong repellency, aqueous solution cannot be directly set on Cassie superhydrophobic materials until the dropping volume is larger than 6.5 μL. Therefore, arranging a (super)hydrophilic substance on Cassie superhydrophobic substrates to construct (super)hydrophilic/superhydrophobic pattern

Access and Binding of H2S to Hemeproteins: The Case of HbI of Lucina pectinata.

Hydrogen sulfide (H2S) was recently discovered as a gasotransmitter, capable of coordinating to the heme iron of hemeproteins. H2S is unique for its ability to render varying concentrations of the nucleophilic conjugate bases (HS(-) or S(2-)), either as free or bound species with expected outcomes on its further reactivity. There is no direct evidence about which species (H2S, HS(-), or S(2-)) coordinates to the iron. We performed computer simulations to address the migration and binding processes of H2S species to the hemoglobin I of Lucina pectinata, which exhibits the highest affinity for t

Assessing the effects of trematode infection on invasive green crabs in eastern north america.

A common signature of marine invasions worldwide is a significant loss of parasites (= parasite escape) in non-native host populations, which may confer a release from some of the harmful effects of parasitism (e.g., castration, energy extraction, immune activation, behavioral manipulation) and possibly enhance the success of non-indigenous species. In eastern North America, the notorious invader Carcinus maenas (European green crab) has escaped more than two-thirds its native parasite load. However, one of its parasites, a trematode (Microphallus similis), can be highly prevalent in the non-n

Metabolomic, behavioral, and reproductive effects of the synthetic estrogen 17 α-ethinylestradiol on the unionid mussel Lampsilis fasciola.

The endocrine disrupting effects of estrogenic compounds in surface waters on fish, such as feminization of males and altered sex ratios, may also occur in aquatic invertebrates. However, the underlying mechanisms of action and toxicity, especially in native freshwater mussels (Order Unionoida), remain undefined. This study evaluated the effects of a 12-day exposure of 17 α-ethinylestradiol (EE2), a synthetic estrogen in oral contraceptives commonly found in surface waters, on the behavior, condition, metabolism, and reproductive status of Lampsilis fasciola. Adult mussels of both sexes were e
