Dual-targeting oral nanoparticle mucosal vaccine against brucella infection: co-targeting M cells and dendritic cells.

This study designed and evaluated a chitosan nanoparticle-based mucosal vaccine (CS-CKS9-MEV-DCpep-NPs) targeting M cells and dendritic cells against Brucella. From four immunogenic outer membrane proteins (OMP10, Omp25, Omp31, BtpB), 41 specific epitope peptides were screened and optimized via bioinformatics and ELISPOT for high antigenicity and safety. The dual-targeting strategy uses CKS9 to enhance M cell-mediated antigen uptake and DCpep, together with CS-NPs, to activate DCs and promote their maturation. This coordinates cDC1, cDC2, and pDC subsets, inducing CD8⁺ and CD4⁺ T cell response

Co-trimoxazole-induced reproductive toxicity and placental-barrier disruption: impact on cell-cell junctions and ERK signaling pathway.

Co-trimoxazole (CTX), a combination of sulfamethoxazole (SMZ) and trimethoprim (TMP), is used during pregnancies complicated by infections like Urinary Tract Infections (UTI) and Human Immunodeficiency Virus (HIV), despite potential fetal safety concerns. This study examines CTX's effects on the placenta using both in vivo and in vitro models. Pregnant mice received CTX for 4 days during both early and late gestation. On gestational day 19, tissues were collected for analysis. Furthermore, in vitro, human BeWo placental cells were exposed to non-cytotoxic concentrations of CTX for 24 h. Gene e

LST1: a novel biomarker for efferocytosis in the co-occurrence of type 2 diabetes mellitus and clear cell renal cell carcinoma.

BACKGROUND: The rising global incidence of clear cell renal cell carcinoma (ccRCC) often coincides with metabolic disorders like type 2 diabetes mellitus (T2DM). However, the association between these two conditions remains unclear. This study aimed to identify common molecular pathways and novel biomarkers for the comorbidity of T2DM and ccRCC. METHODS: Utilizing single-cell transcriptomic datasets from the GEO public database, this study identified heterogeneous cell characteristics and core subpopulations in T2DM and ccRCC. Immune infiltration was assessed using the GSEA algorithm, and a p

ICOS and OX40 tandem co-stimulation enhances CAR T-cell cytotoxicity and promotes T-cell persistence phenotype.

Chimeric Antigen Receptor (CAR) T-cell therapies have emerged as an effective and potentially curative immunotherapy for patients with relapsed or refractory malignancies. Treatment with CD19 CAR T-cells has shown unprecedented results in hematological malignancies, including heavily refractory leukemia, lymphoma, and myeloma cases. Despite these encouraging results, CAR T-cell therapy faces limitations, including the lack of long-term responses in nearly 50-70% of the treated patients and low efficacy in solid tumors. Among other reasons, these restrictions are related to the lack of targetab

[Heterotopic osteogenesis study of tissue engineered bone by co-culture of vascular endothelial cells and adipose-derived stem cells].

OBJECTIVE: To investigate the heterotopic osteogenesis of tissue engineered bone using the co-culture system of vascular endothelial cells (VECs) and adipose-derived stem cells (ADSCs) as seed cells. METHODS: The partially deproteinized biological bone (PDPBB) was prepared by fibronectin combined with partially deproteinized bone (PDPB). The ADSCs of 18-week-old Sprague Dawley (SD) rats and VECs of cord blood of full-term pregnant SD rats were isolated and cultured. Three kinds of tissue engineered bone were constructed in vitro: PDPBB+VECs (group A), PDPBB+ADSCs (group B), PDPBB+co-cultured

Co-transplantation of hippocampal neural stem cells and astrocytes and microvascular endothelial cells improve the memory in ischemic stroke rat.

BACKGROUND: Neural stem cells (NSCs) are promising for ischemia stroke because they can replace damaged or lost cells. However, the adult central nervous system (CNS) does not provide an optimal microenvironment for exogenous NSCs to survive, proliferation and differentiation. We established a co-transplantation system with NSCs and astrocyte and brain microvascular endothelial cells (BMECs) to explore whether it can improve the memory ability in ischemic stroke rat. METHODS: After building the ischemic stroke in 50 rats by middle cerebral artery occlusion and reperfusion (MCAO/R), transplant

Nicotine favors osteoclastogenesis in human periodontal ligament cells co-cultured with CD4(+) T cells by upregulating IL-1β.

Periodontitis, which is the main cause of tooth loss, is one of the most common chronic oral diseases in adults. Tooth loss is mainly a result of alveolar bone resorption, which reflects an increased osteoclast formation and activation. Osteoclast formation in periodontal tissue is a multistep process driven by osteoclastogenesis supporting cells such as human periodontal ligament (PDL) cells and CD4(+) T cells. Inflammatory cytokines, such as interleukin-1β (IL-1β), can induce osteoclastogenesis by affecting the expression of receptor activator of NF-κB ligand (RANKL) and osteoprotegerin (OPG

Proliferation and differentiation of neural stem cells co-cultured with cerebral microvascular endothelial cells after oxygen-glucose deprivation.

Various stem cells, including neural stem cells (NSCs), have been extensively studied in stroke models, but how to increase neuronal differentiation rate of NSCs remains unresolved, particularly in a damaged environment. The purpose of this study was to investigate the effects of cerebral microvascular endothelial cells (CMECs) on the neurogenesis of NSCs with or without oxygen-glucose deprivation (OGD). The NSCs acquired from primary culture were immunostained to prove cell purity. Survival and proliferation of NSCs were determined after the co-culture with CMECs for 7 days. After removing th

Characterization of lymphocytic infiltrates in progressive multifocal leukoencephalopathy: co-localization of CD8(+) T cells with JCV-infected glial cells.

We characterized inflammatory infiltrates in archival brain biopsy and autopsy samples from 26 HIV(+) and 20 HIV(-) patients with progressive multifocal leukoencephalopathy (PML). The predominant inflammatory cells were CD8(+) T lymphocytes. We defined CD8(+) T cell distribution with regard to JCV-infected glial cells, PML lesions and the extent of demyelination. In most samples from either HIV(+) and HIV(-) patients, we found positive correlations between the parenchymal CD8(+) T cells and JCV-infected glial cells and conversely, negative correlations between the perivascular CD8(+) T cells a

Co-expression of the squamous cell carcinoma antigens 1 and 2 in normal adult human tissues and squamous cell carcinomas.

Squamous cell carcinoma antigen (SCCA) serves as a serological marker for advanced squamous cell carcinomas (SCCs) and as an indicator of therapeutic response. Recent molecular studies show that the SCCA is transcribed by two almost identical tandemly arrayed genes, SCCA1 and SCCA2. These genes are members of the high molecular weight serine proteinase inhibitor (serpin) superfamily. Although SCCA1 and SCCA2 are 92% identical at the amino acid level, they have distinct biochemical properties. Paradoxically, SCCA1 is an inhibitor of papain-like cysteine proteinases, such as cathepsins L, S, and

Restrictin: a chick neural extracellular matrix protein involved in cell attachment co-purifies with the cell recognition molecule F11.

We report here the characterization of restrictin, a novel chick neural extracellular matrix glycoprotein associated with the cell recognition molecule F11. Immunoaffinity chromatography using monoclonal antibody 23-13 directed to restrictin yield a major relative molecular mass band at 170 x 10(3) and minor bands at 160, 180, 250 and 320 x 10(3) which are immunologically related to each other. Neural cells attach on immobilized restrictin in a short-term adhesion assay. This adhesion can be blocked specifically by monoclonal or polyclonal antibodies to restrictin but not by antibodies to F11

Mechanism of mast cell deficiency in mutant mice of mi/mi genotype: an analysis by co-culture of mast cells and fibroblasts.

Mutant mice of mi/mi genotype are osteopetrotic and are deficient in mast cells. The osteopetrosis of mi/mi mice can be cured by bone marrow transplantation from congenic normal (+/+) mice, and therefore, the cause of the osteopetrosis is attributed to a defect of osteoclasts. Since both osteoclasts and mast cells are the progeny of multipotential hematopoietic stem cells, we examined whether mast cells were defective in mi/mi mice. In spite of the deficiency of mast cells in tissues of mi/mi mice, mast cells did develop when spleen cells of mi/mi mice were cultured with pokeweed mitogen-stimu

Immunity to coccidiosis: T-cell control of infection with Eimeria vermiformis in mice does not require co-operation with inflammatory cells.

The necessity for co-operation between lymphocytes and myeloid-derived inflammatory cells in the mediation of anti-coccidial immunity was investigated using mice infected with Eimeria vermiformis. Reciprocal exchange of immune lymphocytes between H-2 compatible strains of contrasting susceptibility to infection (resistant BALB/B and susceptible C57BL/10) resulted in successful transfer of immunity in both homologous and heterologous exchanges. Recipients of immune cells, whatever their original response phenotype, expressed a high degree of immunity to infection, indicating that the differenti

Tailored Monolayer Co-Assembly for Enhanced Efficiency and Stability in Inverted Perovskite Solar Cells.

Self-assembled monolayers (SAMs) are extensively employed as hole-selective interlayers in inverted perovskite solar cells (PSCs), yet their incomplete coverage and poor molecular ordering often induce interfacial defects and exacerbate non-radiative recombination. To overcome these limitations, we developed a bulky methylthio-functionalized carbazole-based SAM, (2-(3,6-bis(bis(4-(methylthio)phenyl)amino)-9H-carbazol-9-yl)ethyl)phosphonic acid (S-2PACz), and implemented a sequential co-assembly strategy utilizing cysteine (Cys). In this tailored architecture, Cys is pre-anchored via  carboxyl

A human endothelial and adipose stem cell-based co-culture model for venous malformations.

Venous malformations (VMs) are developmental defects in vascular morphogenesis characterized by enlarged vein-like channels and defective perivascular cell coverage. The majority of VMs are caused by somatic mutations in endothelial cell (EC) receptor tyrosine kinase TIE2, but current medicinal options are limited. The common bottleneck when testing new therapeutic strategies and identifying efficient drug candidates is the absence of a reliable and robust model for replicating the characteristics of VMs. We report here on an advanced in vitro model for recapitulating cellular and molecular pa

A Modified Co-Culture System for Understanding Granulosa-Theca Cell Interactions in the Bovine Ovary.

A reliable co-culture of granulosa (GCs) and theca cells (TCs) has the potential to facilitate a more comprehensive understanding of the interactions, signaling pathways, and substrate exchange between both cell compartments in the bovine ovarian follicle. Utilizing commercially available cell culture inserts, a reproducible co-culture model of bovine GCs and TCs was established. It was observed that primary theca and granulosa cells require considerably more time to attach to the insert membrane compared to cell lines. Initially, the TCs were seeded onto the inverted insert membrane, with a t

Optimizing co-expression strategies for the simultaneous display and secretion of monoclonal antibodies in CHO cells.

Mammalian cell-based dual display and secretion (dualDS) systems enable both surface display of antibodies for binding analysis and simultaneous secretion for functional testing within the same cells. While powerful, their efficiency depends heavily on how light (LC) and heavy chains are co-expressed. In this study, we compared three vector designs in Chinese Hamster Ovary cells: (1) multiple promoters (MP), (2) internal ribosomal entry sites (IRES), and (3) a minimal furin cleavage sequence linked to the 2A peptide (Fm-2A). Vectors were stably integrated into a genomic landing pad using recom

Differential PARP inhibitor responses in BRCA1-deficient and resistant cells in competitive co-culture.

Synthetic lethality describes a genetic relationship where the loss of two genes results in cell death, but the loss of one of those genes does not. Drugs used for precision oncology can exploit synthetic lethal relationships; the best described are PARP inhibitors which preferentially kill BRCA1-deficient tumours preferentially over BRCA1-proficient cells. New synthetic lethal targets are often discovered using genetic screens, such as CRISPR knockout screens. Here, we present a competitive co-culture assay that can be used to analyse drugs or gene knockouts with synthetic lethal effects. We

Helminth extracellular vesicles co-opt host monocytes to drive T cell anergy.

Parasitic helminths secrete extracellular vesicles (EVs) into their host tissues to modulate immune responses, but the underlying mechanisms are poorly understood. We demonstrate that Ascaris EVs are efficiently internalised by monocytes in human peripheral blood mononuclear cells and increase the percentage of classical monocytes. Furthermore, EV treatment of monocytes induced a novel anti-inflammatory phenotype characterised by CD14+, CD16-, CC chemokine receptor 2 (CCR2-) and programmed death-ligand 1 (PD-L1)+ cells. In addition, Ascaris EVs induced T cell anergy in a monocyte-dependent mec

Single is not combined: The role of Co and Ni bioavailability on toxicity mechanisms in liver and brain cells.

The two trace elements cobalt (Co) and nickel (Ni) are widely distributed in the environment due to the increasing industrial application, for example in lithium-ion batteries. Both metals are known to cause detrimental health impacts to humans when overexposed and both are supposed to be a risk factor for various diseases. The individual toxicity of Co and Ni has been partially investigated, however the underlying mechanisms, as well as the interactions of both remain unknown. In this study, we focused on the treatment of liver carcinoma (HepG2) and astrocytoma (CCF-STTG1) cells as a model fo
