Impaired calcitonin secretion following stimulation in spasmophilic women.

Calcitonin (CT) secretion following stimulation was studied in 9 young women suffering from spasmophilia, this having been diagnosed by clinical and electromyographic criteria. The controls were 9 women, matched by age, who responded negatively to the diagnostic tests. After rapid intravenous infusion (60 sec) of 4 mg/kg/lean body mass of calcium (as gluconate), CT concentration increased measurably (p less than 0.0001) in the controls, while remaining unchanged in the spasmophilic. The average increase per cent vs. the baseline values was 225% in the controls and 34% in those with spasmophili…

Paget's disease. Reversal of severe paraparesis using calcitonin.

Relatively short-term treatment of paraparesis due to Paget's disease with subcutaneous salmon calcitonin alone produced dramatic relief of sensory loss, pain, and paraparesis. The successful outcomes in 2 patients, one with spinal cord and one with cauda equina compression, indicate a potential alternative to surgery in reversing mild-to-severe neural dysfunction in Pagets disease. The proposed mechanisms of action of calcitonin include reduction of a direct bony impingement on the neural tissue and/or a decrease of neural ischemia. It is suggested that even if paraparesis does not improve wi…

Early postoperative calcitonin-to-preoperative calcitonin ratio as a predictive marker for structural recurrence in sporadic medullary thyroid cancer: A retrospective study.

BACKGROUND: Calcitonin (Ctn) is widely used as a marker in the diagnosis, prognosis, and postoperative follow-up of patients with medullary thyroid carcinoma (MTC). The prognostic value of postoperative calcitonin-to-preoperative calcitonin ratio (CR), reflecting the change in Ctn level of response to initial treatment, remains uncertain in long-term disease outcomes. This study aims to determine the cut-off value of CR for predicting structural recurrence and assess the prognostic role of CR in patients with MTC. METHODS: We retrospectively reviewed patients with MTC in Sun Yat-sen Universit…

Cloning of two members of the calcitonin-family receptors from stingray, Dasyatis akajei: possible physiological roles of the calcitonin family in osmoregulation.

In cartilaginous fish, two cDNAs encoding calcitonin-family receptors were isolated for the first time from the stingray brain. The open reading frame of one receptor cDNA coded a 525-amino acid protein. The amino acid identity of this receptor to human calcitonin-receptor-like receptor (CRLR) is 64.5%, frog CRLR is 64.7%, and flounder CRLR is 61.2% and this was higher than to human calcitonin receptor (CTR) (46.1%), frog CTR (54.7%), and flounder CTR (48.9%). We strongly suggested that this receptor is a ray CRLR based on phylogenetic analysis. In case of the second receptor, amino acid ident…

Intermedin, a novel calcitonin family peptide that exists in teleosts as well as in mammals: a comparison with other calcitonin/intermedin family peptides in vertebrates.

Endocrine regulation in vertebrates is critical for the adaptation and regulation of homeostasis. The G protein-coupled receptor (GPCR) signaling transduction system represents one of the most ancient forms of cell surface signaling. Recently, comparative sequence analysis has aided in the identification and pairing of a variety of ligand/GPCR signaling systems. Among the ligands of type II GPCRs, the calcitonin family peptides including calcitonin, alpha-calcitonin gene-related peptide (alphaCGRP), betaCGRP, adrenomedullin, and amylin are among the best studied hormones, and the founding memb…

Computational insights into the aggregation mechanism of human calcitonin.

Human calcitonin (hCT) is a peptide hormone that regulates calcium homeostasis, but its abnormal aggregation can disrupt physiological functions and increase the risk of medullary thyroid carcinoma. To elucidate the mechanisms underlying hCT aggregation, we investigated the self-assembly dynamics of hCT segments (hCT1-14, hCT15-25, and hCT26-32) and the folding and dimerization of full-length hCT1-32 through microsecond atomistic discrete molecular dynamics (DMD) simulations. Our results revealed that hCT1-14 and hCT26-32 predominantly existed as isolated monomers with transient small-sized ol…

Expression of human calcitonin by microencapsulated recombinant myoblasts.

A novel approach to treat post-menopausal osteoporosis is proposed by engineering cell lines for the secretion of human calcitonin (hCT) and which would then be suitable for implantation in different allogeneic hosts. Mouse myoblast were transfected with the cDNA for hCT using the liposome-based gene delivery technique and clones secreting of human calcitonin were isolated. Human calcitonin expression was detected by ELISA. Western blot and immunohistochemical analyses also confirmed the plasmid, pcDNA3-hCT, had been transfected into the cells. Upon enclosure in microcapsules, which are biocom…

Effects of hyperprolactinemia on calcitonin secretion in male rats.

The role of prolactin (PRL) in calcitonin (CT) release by the thyroid C cell in male rats was studied. Anterior pituitary (AP)-grafted male rats were characterized by hyperprolactinemia. Brain cortex (CX)-grafted male rats were used as control animals. AP- and CX-grafted rats were infused intravenously with CaCl2 and bled from the jugular catheter at 0, 30, 60, and 120 minutes following the CaCl2 challenge. Rat thyroid gland was incubated with or without 3-isobutyl-1-methylxanthine (IBMX) at 37 degrees C for 30 minutes. Thyroid C cells were incubated in culture medium at 37 degrees C for 60 mi…

Amylin is an agonist of the renal porcine calcitonin receptor.

The cloned renal porcine calcitonin (pCT) receptor cDNA expressed by transient transfection in COS-1 cells or stable transfection in HEK-293 cells was assayed for interaction with CT, amylin, and CT gene-related peptide. Both [125I]salmon CT ([125I]sCT) and [125I]rat amylin displayed specific binding to transfected cells, and in both cases, pCT and rat amylin were equipotent in competing for binding. sCT was most potent in binding competition assays, whereas human CT and rat or human CT gene-related peptide did not compete. Despite the greater apparent affinity of sCT for receptor binding, sCT…

Rectal salmon calcitonin for the treatment of postmenopausal osteoporosis.

In a 2-year study, we examined bone mass and calcium metabolism in 36 elderly women with moderate osteoporosis. The study period comprised 1 year of observation, during which the women received no treatment affecting calcium metabolism, and 1 year of treatment, during which all participants received daily salmon calcitonin (sCT) 100 IU rectally and calcium 500 mg. During the observational period a significant bone loss of 1.5% was seen in the forearm (P less than 0.01), whereas the spinal bone mass was virtually unchanged. After institution of treatment, the bone loss was arrested in the forea…

Calcitonin: chronotherapeutic effect on osteopenia in the ovariectomized rat.

This chronotherapeutic study has revealed no clear evidence to support the concept that calcitonin promotes bone formation in intact or OVX'd rats. Rather, the efficacy of CT-impaired bone resorption appeared to be optimal in OVX'd rats treated during the daytime, and least effective during the first half of the environmental dark span. These findings indicate a benefit for calcitonin chronotherapy in this animal model where the cellular kinetics resemble those documented in early development of postmenopausal osteoporosis.

[Treatment of primary osteoporosis with calcium and salmon calcitonin].

Fifty-nine consecutive patients (19 men, 40 women, mean age 60.8 [27-80] years) with primary osteoporosis were studied to see if there was any significant gain in bone mass after treatment with salmon calcitonin. All the patients were given 1 g calcium by mouth every morning. Group 1 (n = 20) received no other specific medication while group 2 (n = 19) were given 100 I.U. calcitonin subcutaneously every second evening and group 3 (n = 20) received the same dose every evening. The pain reported by the patients was subdivided into four severity grades, and analgesic consumption was recorded. In …

[Postoperative treatment of phantom pain and causalgias with calcitonin].

Results of IV calcitonin treatment in patients suffering from postoperative phantom limb pain (n = 12) or causalgia following peripheral nerve lesions (n = 4) are reported. All patients were complained of severe pain after a traumatic event or amputation, with disturbed sleep in many cases. After only 1-2 infusions 10 patients with phantom limb pain (83%) were discharged from hospital pain-free. Pain was effectively reduced by up to 5 infusions in 2 patients (17%). A follow-up for maximally 24 months showed a recurrence of pain in only 4 patients with obvious stump problems or reamputations. T…
