Inhibition of casein kinase II reduces TGFβ induced fibroblast activation and ameliorates experimental fibrosis.

OBJECTIVES: Casein kinase II (CK2) is a constitutively active serine/threonine protein kinase that plays a key role in cellular transformation and tumorigenesis. The purpose of the study was to characterise whether CK2 contributes to the pathologic activation of fibroblasts in patients with SSc and to evaluate the antifibrotic potential of CK2 inhibition. METHODS: Activation of CK2, JAK2 and STAT3 in human skin and in experimental fibrosis was analysed by immunohistochemistry. CK2 signalling was inhibited by the selective CK2 inhibitor 4, 5, 6, 7-Tetrabromobenzotriazole (TBB). The mouse model

Factors contributing to the resistivity of a higher casein diet against choline deficiency-induced hyperhomocysteinemia in rats.

The mechanism by which feeding a higher casein diet results in resistance to choline deprivation-induced hyperhomocysteinemia was investigated in rats. Plasma homocysteine concentration was significantly lower in rats fed a 30% casein diet (30C) than in rats fed a 10% casein diet (10C). Choline deprivation did not enhance plasma homocysteine concentration in rats fed 30C, while it significantly enhanced plasma homocysteine concentration in rats fed 10C. The choline deprivation-induced enhancement of plasma homocysteine concentration in rats fed 10C was significantly suppressed by methionine su

Relative efficacy of casein or soya protein combined with palm or safflower-seed oil on hyperuricaemia in rats.

Diets that ameliorate the adverse effects of uric acid (UA) on renal damage deserve attention. The effects of casein or soya protein combined with palm or safflower-seed oil on various serum parameters and renal histology were investigated on hyperuricaemic rats. Male Wistar rats administered with oxonic acid and UA to induce hyperuricaemia were fed with casein or soya protein plus palm- or safflower-seed oil-supplemented diets. Normal rats and hyperuricaemic rats with or without allopurinol treatment (150 mg/l in drinking water) were fed with casein plus maize oil-supplemented diets. After 8

Optimization of production of Brucella abortus S19 culture in bioreactor using soyabean casein digest medium.

A method of cultivating Brucella abortus S19 culture in bioreactor was attempted using three different media. Culture conditions in bioreactor were optimized by varying agitation and aeration parameters. Varying the aeration ranging from 0.5 vvm to 0.8 vvm and agitation rate ranging from 250 rpm to 400 rpm during bacterial growth was found to yield highest viable count within 48 hours of culture period. A count of > 1 x 10(11) CFU per ml within 48 to 60 hours post seeding was obtained consistently in all five consecutive batches (P > 0.05) with 6 x 10(11) CFU per ml being the maximum yield whe

Biochemical and pharmacological aspects of two bradykinin-potentiating peptides obtained from tryptic hydrolysis of casein.

Peptides that display bradykinin-potentiating activity have been obtained from a number of distinct sources, such as snake venoms, fibrinogen, and casein. This paper describes the characterization of two new peptides generated by tryptic hydrolysis of casein. No homology was found with other known vasoactive or vasopotentiating peptides, especially by the lack of Ile-Pro-Pro motif. The peptides EMPFPK and YPVEPFTE, corresponding to the gamma casein sequence (108-113 and 114-121, respectively), displayed a selective potentiating activity on isolated guinea pig ileum for bradykinin. Besides, the

Immunochemical demonstration of alpha(s1)- and beta-casein in mouse mammary glands at early stages of pregnancy.

We generated monoclonal antibodies (MAbs) against mouse alpha(s1)- and beta-casein and used them to survey casein immunochemically in mammary glands of mice at peri-coitous and pregnant stages. Two MAb-producing hybridoma cells, designated MCalpha1 cell and MCbeta1 cell, were established. Each antibody, when used in Western blotting, recognized specifically mouse alpha(s1)- and beta-casein among a wide spectrum of proteins of both a lactating mammary homogenate and mouse skim milk. Immunohistochemistry revealed alpha(s1)- and beta-casein in sections of lactating mammary glands. Staining was fo

Tal-1 induces T cell acute lymphoblastic leukemia accelerated by casein kinase IIalpha.

Ectopic activation of the TAL-1 gene in T lymphocytes occurs in the majority of cases of human T cell acute lymphoblastic leukemia (T-ALL), yet experiments to date have failed to demonstrate a direct transforming capability for tal-1. The tal-1 gene product is a serine phosphoprotein and basic helix-loop-helix (bHLH) transcription factor known to regulate embryonic hematopoiesis. We have established a transgenic mouse model in which tal-1 mis-expression in the thymus results in the development of clonal T cell lymphoblastic leukemia/lymphoma. Thus, overexpression of tal-1 alone can be transfor

[Effect of additional administration of essential amino acids on assimilability of casein hydrolysate in rats with toxic hepatitis].

The effect of certain amino acids on assimilability of casein hydrolysate was studied in albino rats with toxic hepatitis. It is established that in case of this pathology casein hydrolysate is uncapable of correcting completely the disturbed protein and nucleic metabolism. An additional administration under these conditions of amino acids, methionine in particular, changes the negative nitrogeneous balance to the positive one, that evidences for an increase in assimilability of the introduced casein hydrolysate, lowers the excretion of amino nitorgen with urea and rises the DNA content in the

Discovery and Synthesis of Selective Casein Kinase 1δ Degraders for Suppressing Oncogenic NRAS-Mutant Melanoma.

Activating NRAS mutations occur in approximately 20% of melanomas but remain largely undruggable due to the lack of effective targeted therapies. We previously identified casein kinase 1δ (CK1δ) as a critical upstream regulator that stabilizes oncogenic NRAS mutants. However, no selective CK1δ inhibitors have advanced to clinical evaluation. Here, we report the discovery of 8u, a selective CK1δ degrader derived from PF-670462. Compound 8u potently degrades CK1δ in NRASQ61R melanoma cells (DC50 = 1.25 μM, Dmax = 74%) via the ubiquitin-proteasome system, sparing against the closely related isofo

Rennet-Induced Mixed Yak-Cow Casein Micelle Gels: Gelation Properties and Formation Mechanisms.

Rennet-induced yak casein gels have longer gelation time but higher elastic modulus than bovine casein gels, which limits cheese processing efficiency. In this study, yak and bovine casein micelles were mixed at ratios of 100:0, 75:25, 50:50, 25:75, and 0:100 (denoted as Y100, Y75, Y50, Y25, and Y0, respectively). HPLC was used to monitor the relative peak area of released glycomacropeptide (GMP) during κ-casein hydrolysis (normalized to the 60-min maximum) to characterize rennet catalysis kinetics, and the effects on rheology, texture, and microstructure were analyzed. The results showed that

Digested casein-stabilized co-dissolution of calcium and iron in oxalate system for enhanced metal bioavailability.

Oxalate severely impairs calcium and iron bioavailability by forming insoluble precipitates in the gastrointestinal tract, posing challenges for mineral nutrition in plant-based diets. This study investigated the thermodynamic and kinetic interactions between calcium, iron, and oxalate, and explored whether digested casein could enhance mineral bioavailability under oxalate challenge. Thermodynamic analysis revealed that iron(II) oxalate exhibited higher solubility product (Ksp) and weaker association (Ka = 4655) compared to calcium oxalate (Ka = 25126) at 25 ℃. Conductometric titration demons

Structurally optimized photo-crosslinkable casein-PEG hydrogels for enhanced encapsulation and functional stability of resveratrol.

Casein-based gel delivery systems often exhibit limited loading efficiency and functional performance under conventional formulation approaches. We present a novel UV-induced photo-crosslinking strategy for generating porous casein-polyethylene glycol gels, enhancing resveratrol stabilization and enabling controlled delivery during processing and storage. Structural characterization by FTIR and 13C NMR supported the formation of an enhanced covalent network via ester and amide linkages. Formulation optimization showed that 10% casein achieved a crosslinking degree of 93.32 ± 0.43% (p < 0.05),

Cocaine-Induced Changes in the BRD4 Interactome Identifies Casein Kinase 1 Epsilon as a Therapeutic Target.

Recent research has revealed that BRD4, a bromodomain and extra terminal domain (BET) epigenetic "reader" protein, plays an essential role in regulating behavioral and molecular responses to cocaine. To date, the roles of BRD4 and other BET proteins in substance use disorder (SUD) models have been mainly studied using small molecule inhibitors that block the bromodomain interactions with acetylated histones. In other disease models, non-bromodomain BRD4-protein interactions have also been shown to play an important role in BRD4's molecular functions, but these interactions have yet to be studi

Opioid peptides in autism spectrum disorder and gluten-free casein-free diet as a therapeutic approach.

According to the Diagnostic and Statistical Manual of Mental Disorders, Fifth Edition, Autism Spectrum Disorder (ASD) is characterized by persistent difficulties in social communication and interaction, together with restricted and repetitive behaviors, interests, or activities. The diagnosis of ASD currently relies on comprehensive assessments of developmental history and behavioral patterns, as there are no validated laboratory tests for a definitive clinical diagnosis. While evidence-based interventions are largely restricted to educational and behavioral programs, many individuals with ASD

Tuning the Gel Network Structure and Rheology of Acid-Induced Casein Gels via Thiol Blocking.

This study systematically investigates how thiol-disulfide interactions influence the structure and mechanical properties of casein gels. Acid gels were prepared from suspensions of micellar casein (MC) powder that were heat-treated at 70 °C. Thiol groups were variably blocked with N-ethylmaleimide (NEM). The gels were characterized using stress-strain measurements, rheological analyses, and confocal microscopy. The stress-strain curves exhibited a biphasic behavior, with an initial linear elastic phase followed by a linear plastic region and a nonlinear failure zone. Compared to control sampl
